Body tissue and venom glands from an eastern population of the scorpion Centruroides vittatus (Say, 1821) were homogenized and molecular constituents removed to characterize putative sodium β toxin gene diversity, RT-qPCR, transcriptomic, and proteomic variation. We cloned sodium β toxins from genomic DNA, conducted RT-qPCR experiments with seven sodium β toxin variants, performed venom gland tissue RNA-seq, and isolated venom proteins for mass spectrophotometry. We identified greater than 70 putative novel sodium β toxin genes, 111 toxin gene transcripts, 24 different toxin proteins, and quantified sodium β toxin gene expression variation among individuals and between sexes. Our analyses contribute to …

Cytokine Exposure Mediates Transcriptional Activation Of The Orphan Nuclear Receptor Nur77 In Hematopoietic Cells, Orsola Di Martino, Haixia Niu, Gayla Hadwiger, Margaret A Ferris, John S Welch
Open Access Publications
The orphan nuclear receptor Nur77 is an immediate-early response gene that based on tissue and cell context is implicated in a plethora of cellular processes, including proliferation, differentiation, apoptosis, metabolism, and inflammation. Nur77 has a ligand-binding pocket that is obstructed by hydrophobic side groups. Naturally occurring, cell-endogenous ligands have not been identified, and Nur77 transcriptional activity is thought to be regulated through posttranslational modification and modulation of protein levels. To determine whether Nur77 is transcriptionally active in hematopoietic cells in vivo, we used an upstream activating sequence (UAS)-GFP transgenic reporter. We found that Nur77 is transcriptionally inactive in vivo in …

Recent Advances In Non-Enzymatic Electrochemical Detection Of Hydrophobic Metabolites In Biofluids, Zahra Panahi, Luciana Custer, Jeffrey M. Halpern
New Hampshire EPSCoR
This review focuses on recent advances in non-enzymatic electrochemical biosensors for detection of hydrophobic metabolites. Electrochemical approaches have been widely applied in many established and emerging technologies and a large range of electrochemical biosensors have been used for detection of various hydrophobic metabolites. Despite the progress made in this field, some problems still exist, specifically, electrochemical detection of hydrophobic biomarkers can be challenging in complex biological fluids. In this review, we have highlighted some of the most representative surface modification technologies that have been employed in electrochemical biosensors to counter the problems of poor sensitivity and selectivity towards hydrophobic metabolites. …

There are numerous publications demonstrating that plant polyphenols can reduce oxi-dative stress and inflammatory processes in the brain. In the present study we have investigated the neuroprotective effect of plant extract isolated from the roots of L. gmelinii since it contains a rich source of polyphenols and other biologically active compounds. We have applied an oxidative and inflammatory model induced by NMDA, H2O2, and TNF‐α in human primary neurons and astrocytes, and mouse cerebral endothelial cell (CECs) line in vitro. The levels of ROS generation, NADPH oxidase activation, P‐selectin expression, and activity of ERK1/2 were evaluated by quan-titative immunofluorescence analysis, …

It is challenging to associate features such as human health outcomes, diet, environmental conditions, or other metadata to microbial community measurements, due in part to their quantitative properties. Microbiome multi-omics are typically noisy, sparse (zero-inflated), high-dimensional, extremely non-normal, and often in the form of count or compositional measurements. Here we introduce an optimized combination of novel and established methodology to assess multivariable association of microbial community features with complex metadata in population-scale observational studies. Our approach, MaAsLin 2 (Microbiome Multivariable Associations with Linear Models), uses generalized linear and mixed models to accommodate a wide variety of modern epidemiological studies, including …

Biodegradable stents have tremendous theoretical potential as an alternative to bare metal stents and drugeluting stents for the treatment of obstructive coronary artery disease. Any bioresorbable or biodegradable scaffold material needs to possess optimal mechanical properties and uniform degradation behavior that avoids local and systemic toxicity. Recently, molybdenum (Mo) has been investigated as a potential novel biodegradable material for this purpose. With its proven moderate degradation rate and excellent mechanical properties, Mo may represent an ideal source material for clinical cardiac and vascular applications. The present study was performed to evaluate the mechanical performance of metallic Mo in vitro and …

Assessing The Impact Of Vaccination On Covid-19 In South Africa Using Mathematical Modeling, Musyoka Kinyili, Justin B. Munyakazi, Abdulaziz Y. A. Mukhtar
Applied Mathematics & Information Sciences
The unexpected continuing mushrooming tendency of the COVID-19 epidemic calls for alarm in the entire globe especially with the cropping up of more divergent contagious variants being witnessed. On top of the many non-pharmaceutical measures put in place for containment of the pandemic, pharmacological measures have been incorporated in the battle against the SARS-CoV-2 especially with the commencement of vaccination in the early December 2020. This study develops a deterministic compartmental model that incorporates vaccination as a measure to combat the spread of COVID-19 epidemic. We use the model particularly to assess the potential impact of vaccination in shattering the …

On Dynamical Analysis Of A Prey-Diseased Predator Model With Refuge In Prey, Srinivasarao Thota, Abayomi Ayoade
Applied Mathematics & Information Sciences
In this paper, we propose a mathematical model of a prey-diseased predator model with refuge in prey system. In other words, a prey-predator model with an infectious disease in the prey population is formulated. This model is constituted by a system of three nonlinear ordinary differential equations of first order, which describe the interaction between the infected prey, non-infected prey and predator. The equilibria of the system are derived and the stability analyses of the disease-free and the endemic equilibria are conducted.

Some Properties And Application Of Modified Jacknified Liu-Type Negative Binomial Ridge Regression, Huda M. Alshanbari, Irsa Sajjad, Javid Gani Dar, Abd Al-Aziz Hosni El-Bagoury, Sundus Naji Al-Aziz
Applied Mathematics & Information Sciences
The aim of this paper is to introduce a new estimator for the modified Jackknified Liu Type Negative Binomial. As in the presence of multicollinearity, the Maximum Likelihood Estimator (MLE) is unable to produce valid statistical inference. So, this paper is designed to solve the problem of multicollinearity. Several ridge regression estimators are used for this purpose. Moreover, Monte Carlo simulation and real life data set are applied on the proposed and existing estimators to evaluate the performance of proposed estimator in the case of MSE. The results reveal that our proposed estimator has best performance among all other estimators …

A Framework Of Paracellular Transport Viananoparticles-Induced Endothelial Leakiness, Myeongsang Lee, Nengyi Ni, Huayuan Tang, Yuhuan Li
Open Access Publishing Fund
Nanomaterial-induced endothelial leakiness (NanoEL) is an interfacial phenomenon denoting the paracellular transport of nanoparticles that is pertinent to nanotoxicology, nanomedicine and biomedical engineering. While the NanoEL phenomenon is complementary to the enhanced permeability and retention effect in terms of their common applicability to delineating the permeability and behavior of nanoparticles in tumoral environments, these two effects significantly differ in scope, origin, and manifestation. In the current study, the descriptors are fully examined of the NanoEL phenomenon elicited by generic citrate-coated gold nanoparticles (AuNPs) of changing size and concentration, from microscopic gap formation and actin reorganization down to molecular signaling pathways …

Exploring The Insights Of Science Within The Context Of Asian Christianity Through The Work Of William R. Stoeger, S.J., Lawrence Ng Yew Kim
Jesuit School of Theology Dissertations
The work to relate theology and the findings of modern science, a long-standing one among western scholars, has been, for historical and cultural reasons, largely absent within the context of Asia. Asian theologians engaged in this field appear to do so by employing the perspective of their North American and European colleagues but without contextualizing it within an Asian context, which has been shaped by deeply rooted Asian philosophical foundations and religious traditions.
This dissertation offers a strategy to contextualize the insights of science within the Sinic context of Southeast Asia by appealing to the methodology of a Jesuit priest, …

Context-specific Bayesian networks (i.e. directed acyclic graphs, DAGs) identify context-dependent relationships between variables, but the non-convexity induced by the acyclicity requirement makes it difficult to share information between context-specific estimators (e.g. with graph generator functions). For this reason, existing methods for inferring context-specific Bayesian networks have favored breaking datasets into subsamples, limiting statistical power and resolution, and preventing the use of multidimensional and latent contexts. To overcome this challenge, we propose NOTEARS-optimized Mixtures of Archetypal DAGs (NOTMAD). NOTMAD models context-specific Bayesian networks as the output of a function which learns to mix archetypal networks according to sample context. The archetypal …

Roman L. Hruska U.S. Meat Animal Research Center: Reports
Bovine viral diarrhea virus’s (BVDV) entry into bovine cells involves attachment of virions to cellular receptors, internalization, and pH-dependent fusion with endosomal membranes. The primary host receptor for BVDV is CD46; however, the complete set of host factors required for virus entry is unknown. The Madin-Darby bovine kidney (MDBK) cell line is susceptible to BVDV infection, while a derivative cell line (CRIB) is resistant at the level of virus entry. We performed complete genome sequencing of each to identify genomic variation underlying the resistant phenotype with the aim of identifying host factors essential for BVDV entry. Three large compound deletions …

South American freshwater ichthyofauna is taxonomically the most diverse on the planet, yet its diversity is still vastly underestimated. The Amazon basin alone holds more than half of this diversity. The evidence of this underestimation comes from the backlog of morphologically distinct, yet undescribed forms deposited in museum collections, and from DNA-based inventories which consistently identify large numbers of divergent lineages within even well-studied species groups. In the present study, we investigated lineage diversity within the Geophagus sensu stricto species group. To achieve these objectives, we analyzed 337 individuals sampled from 77 locations within and outside the Amazon basin representing …

Why Model Order Reduction, Salvador Robles, Martine Ceberio, Vladik Kreinovich
Departmental Technical Reports (CS)
Reasonably recently, a new efficient method appeared for solving complex non-linear differential equations (and systems of differential equations). In this method -- known as Model Order Reduction (MOR) -- we select several solutions, and approximate a general solution by a linear combination of the selected solutions. In this paper, we use the known explanation for efficiency of neural networks to explain the efficiency of MOR techniques.

Kinematic Metric Spaces Under Interval Uncertainty: Towards An Adequate Definition, Vladik Kreinovich, Olga Kosheleva, Victor Selivanov
Departmental Technical Reports (CS)
In the physical space, we define distance between the two points as the length of the shortest path connecting these points. Similarly, in space-time, for every pair of events for which the event a can causally effect the event b, we can define the longest proper time t(a,b) over all causal trajectories leading from a to b. The resulting function is known as kinematic metric. In practice, our information about all physical quantities -- including time -- comes from measurement, and measurements are never absolutely precise: the measurement result V is, in general, different from the actual (unknown) value v …
