Mechanistic Insight Into Β-Lactamase Catalysis, Inhibitor Design And Resistance, Michael Trent Kemp Nov 2021

Mechanistic Insight Into Β-Lactamase Catalysis, Inhibitor Design And Resistance, Michael Trent Kemp

USF Tampa Graduate Theses and Dissertations

The emergence of antibiotic resistance and spread of Gram negative bacteria poses a very real health threat to the public. The main mode of resistance within Gram negative bacteria is the production of β-lactamase enzymes that catalyze the breakdown of β-lactam antibiotics through a hydrolysis mechanism. Once the β-lactam ring is hydrolyzed and opened, the drug loses its efficacy, which allows for the bacteria to grow and proliferate uninhibited. These β-lactamase enzymes are organized into four categories based on the Ambler classification, with classes A, C and D being denoted as serine-based β-lactamase enzymes. Class B is composed of metalloenzymes …

Mitogen-activated protein kinase 6 (MAPK6) is an atypical MAPK. Its function in regulating cancer growth remains elusive. Here, we reported that MAPK6 directly activated AKT and induced oncogenic outcomes. MAPK6 interacted with AKT through its C34 region and the C-terminal tail and phosphorylated AKT at S473 independent of mTORC2, the major S473 kinase. mTOR kinase inhibitors have not made notable progress in the clinic. Our identified MAPK6-AKT axis may provide a major resistance pathway. Besides repressing growth, inhibiting MAPK6 sensitized cancer cells to mTOR kinase inhibitors. MAPK6 overexpression is associated with decreased overall survival and the survival of patients with …

Regional Expansion And Evaluation Of Potential Chemical Control For Invasive Apple Snails (Pomacea Maculata) In Southwest Louisiana, Julian M. Lucero

LSU Master's Theses

The integration of monitoring and chemical control is an efficient strategy for managing invasive apple snails, Pomacea maculata, in the rice (Oryza sativa L.) and crawfish systems of southwest Louisiana. However, their current distribution, expansion rates, and susceptibility to chemical control methods in this area are not well known. This study evaluated the expansion of P. maculata in southwest Louisiana and assessed potential chemical control for P. maculata among toxicity assays using various application rates. The effects of potential chemical control were also assessed on a non-target species, the red swamp crawfish (Procambarus clarkii). P. maculata …

Angiotensin Receptor-Neprilysin Inhibition In Acute Myocardial Infarction, Marc A Pfeffer, Douglas L Mann, Et Al

Open Access Publications

BACKGROUND: In patients with symptomatic heart failure, sacubitril-valsartan has been found to reduce the risk of hospitalization and death from cardiovascular causes more effectively than an angiotensin-converting-enzyme inhibitor. Trials comparing the effects of these drugs in patients with acute myocardial infarction have been lacking.

METHODS: We randomly assigned patients with myocardial infarction complicated by a reduced left ventricular ejection fraction, pulmonary congestion, or both to receive either sacubitril-valsartan (97 mg of sacubitril and 103 mg of valsartan twice daily) or ramipril (5 mg twice daily) in addition to recommended therapy. The primary outcome was death from cardiovascular causes or incident …

Sedation And Acute Encephalopathy In A Pediatric Patient Following Ingestion Of Delta-8-Tetrahydrocannabinol Gummies, Peter Akpunonu, Regan A. Baum, Andrew Reckers, Blake Davidson, Ross Ellison, Matthew Riley, Jordan Trecki, Roy Gerona

Emergency Medicine Faculty Publications

BACKGROUND Delta-8 tetrahydrocannabinol (delta-8 THC) is an isomer of delta-9-tetrahydrocannabinol (delta-9 THC), the primary psychoactive cannabinoid in the marijuana plant. Typically found at lower concentrations in marijuana, delta-8 THC exhibits psychoactive properties similar to delta-9 THC. Products containing delta-8 THC are readily available across the US and currently there is a lack of available confirmatory testing specific to delta-8 THC as there is cross-reactivity to other naturally occurring cannabinoids in standard immunoassays. Pediatric exposures to this substance are on the rise.

CASE REPORT We present a case with laboratory confirmation of a previously healthy 2-year-old girl ingesting approximately 15 mg/kg …

Department of Agronomy and Horticulture: Faculty Publications

Genome-wide DNA polymorphism analysis and molecular marker development are important for forward genetics research and DNA marker-assisted breeding. As an ideal model system for Panicoideae grasses and an important minor crop in East Asia, foxtail millet (Setaria italica) has a high-quality reference genome as well as large mutant libraries based on the “Yugu1” variety. However, there is still a lack of genetic and mutation mapping tools available for forward genetics research on S. italica. Here, we screened another S. italica genotype, “SSR41”, which is morphologically similar to, and readily cross-pollinates with, “Yugu1”. High-throughput resequencing of “SSR41” …

A Novel Method For Ecg Signal Classification Via One-Dimensional Convolutional Neural Network, Xuan Hua, Jungang Han, Chen Zhao, Haipeng Tang, Zhuo He, Qinghui Chen, Shaojie Tang, Jinshan Tang, Weihua Zhou

Faculty Publications

This paper develops an end-to-end ECG signal classification algorithm based on a novel segmentation strategy and 1D Convolutional Neural Networks (CNN) to aid the classification of ECG signals and alleviate the workload of physicians. The ECG segmentation strategy named R-R-R strategy (i.e., retaining ECG data between the R peaks just before and after the current R peak) is used for segmenting the original ECG data into segments to train and test the 1D CNN models. The novel strategy mimics physicians in scanning ECG to a greater extent, and maximizes the inherent information of ECG segments for diagnosis. The performance of …

Let It Rip: The Mechanics Of Self-Bisection In Asexual Planarians Determines Their Population Reproductive Strategies, T. Goel, Danielle Ireland, Vir Shetty , '22, Christina Rabeler, P. H. Diamond, Eva-Maria S. Collins

Biology Faculty Works

Asexual freshwater planarians reproduce by transverse bisection (binary fission) into two pieces. This process produces a head and a tail, which fully regenerate within 1–2 weeks. How planarians split into two offspring—using only their musculature and substrate traction—is a challenging biomechanics problem. We found that three different species, Dugesia japonica, Girardia tigrina and Schmidtea mediterranea, have evolved three different mechanical solutions to self-bisect. Using time lapse imaging of the fission process, we quantitatively characterize the main steps of division in the three species and extract the distinct and shared key features. Across the three species, planarians actively alter …

BACKGROUND: Problems continue to exist with the reporting quality and risk of bias in search methods and strategies in systematic reviews and related review types. Peer reviewers who are not familiar with what is required to transparently and fully report a search may not be prepared to review the search components of systematic reviews, nor may they know what is likely to introduce bias into a search. Librarians and information specialists, who have expertise in searching, may offer specialized knowledge that would help improve systematic review search reporting and lessen risk of bias, but they are underutilized as methodological peer …

Efficient Management Of Nitrogen And Phosphorus At Centralized Water Reclamation Facilities, Helene Kassouf Nov 2021

Efficient Management Of Nitrogen And Phosphorus At Centralized Water Reclamation Facilities, Helene Kassouf

USF Tampa Graduate Theses and Dissertations

A variety of process configurations are employed at municipal water reclamation facilities (WRFs), such as 5-stage Bardenpho, oxidation ditch, modified Ludzack-Ettinger (MLE) process, and sidestream recycle. However, all of these configurations face certain challenges in achieving optimum treatment of nitrogen and phosphorus. In this dissertation, the driving objectives were to: (1) quantitatively assess the fate of nitrogen and phosphorus (mass fluxes) at a treatment facility that employs biological nutrient removal, aerobic digestion, and sidestream recycle, (2) evaluate the performance of a microbial fuel cell (MFC) as a technology to remove nitrogen from sidestreams at a treatment plant that employs that …

The Role Of Cpeb2 Alternative Splicing In Tnbc Metastasis, Shaun C. Stevens Nov 2021

The Role Of Cpeb2 Alternative Splicing In Tnbc Metastasis, Shaun C. Stevens

USF Tampa Graduate Theses and Dissertations

Breast cancer is the second leading cause of cancer-related deaths for women in the U.S. Although the overall 5-year survival rate for breast cancer is 90%, this rate drops substantially for triple-negative breast cancer (TNBC) due to its high metastatic potential. Furthermore, there is a lack of targeted therapeutics for TNBC, and clinical trials have been largely unsuccessful. These characteristics validate the need for identifying novel therapeutic targets for the treatment of TNBC. The study of alternative splicing (AS) has emerged as a powerful tool to elucidate the molecular underpinnings driving cancer.

Our lab has identified cytoplasmic polyadenylation element-binding protein …

Revisiting Darwin’S Little Pond As A Method To Liberate Phosphorus From Apatite Under Prebiotic Hadean Earth Conditions, Jennifer Lago Nov 2021

Revisiting Darwin’S Little Pond As A Method To Liberate Phosphorus From Apatite Under Prebiotic Hadean Earth Conditions, Jennifer Lago

USF Tampa Graduate Theses and Dissertations

Phosphorus is an essential element for life as we know it. Phosphorus, mainly in the form of phosphate, is key to biologic functions such as genetic material, energy production, and cellular framework. As phosphorus is key to so many important biological functions it is of no wonder the question of how phosphorus was incorporated into life initially is a fundamental question in how life began.

During this time a prebiotic phosphorus source would need to have originated in rock, as phosphorus has not volatile source on Earth. The most prevalent mineral source on a prebiotic Earth would likely have been …

Unraveling The Role Of Novel G5 Peptidase Family Proteins In Virulence And Cell Envelope Biogenesis Of Staphylococcus Aureus, Stephanie M. Marroquin

USF Tampa Graduate Theses and Dissertations

Virulence factors and the bacterial cell envelope are two important components in S. aureus pathogenesis and survival. More importantly, understanding the regulation of these cellular processes is crucial to further understanding and combating this successful pathogen. To date, numerous factors have been identified as playing a role in the regulation of Agr activity in S. aureus, including transcription factors, antisense RNAs, and host elements. Herein we investigate the product of SAUSA300_1984 (termed MroQ), a transmembrane G5 peptidase family protein, as a novel effector of this system. Using a USA300 mroQ mutant we observed a drastic reduction in proteolysis, hemolysis, and …

Investigating The Role And Localization Of B Cells In The Red-Eared Slider, Trachemys Scripta, Whitney Hurst

Theses and Dissertations

The immune system of humans has been studied extensively, but very little is known about the immune response in reptiles. Our lab has been working to better understand the immune system of a pond turtle, Trachemys scripta (red-eared slider). Turtles are known for their longevity and their immune system should play an important role in their ability to survive. Our main interest is understanding B cell function and location in slider turtles. B cells are a type of white blood cell that play a vital role in the immune response of humans, with the majority secreting antibodies that mark invading …

Better Together: Analysis Of Integrated Acoustic And Visual Methods When Surveying A Cetacean Community, L. Dalpaz, A. D. Paro, F. G. Daura-Jorge, M. Rossi-Santos, T. F. Norris, S. N. Ingram, L. L. Wedekin

School of Biological and Marine Sciences

Understanding the relative performance of data collection methods is critical for the production of robust results in any biological field study. This is particularly relevant for monitoring studies of rare and inconspicuous species, such as some cetaceans. Here, we compared how passive acoustic and visual survey methods performed when surveying a multispecies cetacean community in a large-scale survey area and whether the simultaneous application of these methods leads to complementary results. For both methods we compared (1) frequency of unscheduled survey stoppages, (2) detection ranges, (3) success in identifying species, (4) precedence of either method in synchronous detections, (5) detection …

Photosynthetic Pigments Of Co-Occurring Northeast Atlantic Laminaria Spp. Are Unaffected By Decomposition, Ls Wright, A Foggo

School of Biological and Marine Sciences

Along Northeast Atlantic coasts, the genus Laminaria dominates kelp forests. Two species, L. digitata and L. hyperborea, are cold temperate whilst the third, L. ochroleuca, is warm temperate. In northern France and the southern British Isles, all 3 species now co-occur, with a gradual rise in predominance of L. ochroleuca evident over recent decades due to climate-driven range shifts. Despite recent focus on the export of photosynthetically viable macroalgal detritus, the effect of decomposition on detrital pigment concentration remains unknown for northern hemisphere kelps. Furthermore, notwithstanding their importance in determining habitat suitability, the photosynthetic pigments of all 3 species have …

Sugarcane (Saccharum spp.) is a prominent source of sugar and serves as bioenergy/biomass feedstock globally. Multiple biotic and abiotic stresses, including drought, salinity, and cold, adversely affect sugarcane yield. G-protein-coupled receptors (GPCRs) are components of G-protein-mediated signaling affecting plant growth, development, and stress responses. Here, we identified a GPCR-like protein (ShGPCR1) from sugarcane and energy cane (Saccharum spp. hybrids) and characterized its function in conferring tolerance to multiple abiotic stresses. ShGPCR1 protein sequence contained nine predicted transmembrane (TM) domains connected by four extracellular and four intracellular loops, which could interact with various ligands and heterotrimeric G …

Poorness is the main factor of children forced to work, this job is to meet the needs of live to his or her family. Indonesia has provided a rule of law for children who are forced to work, but on the condition that the work does not interfere with education, does not harm its health and safety. But the fact is that there are still employers who dare to hire children for reasons of lower paid wages, but not necessarily the jobs given to the child are lighter or sometimes even as heavy as adults. "Dharmasisya” Jurnal Program Magister Hukum FHUI

The number of smokers and cigarette sales in Indonesia has been increasing every year. In parallel with that, the latent risk faced by society is increased. In the socio-cultural context, the cigarette industry has been transformed into a hegemony whose expansion is unstoppable. The circulation of cigarette products is still become an unresolvable problem. Such problem expands from merely legal issues to the ethical dimension. The corporate social responsibility carried out by the corporation tends to be in favor of promotional motive, or creating false public reputation, which makes its implementation often deviate from its philosophy. Inevitably, the smoking phenomenon …
