Our laboratory initiated a comprehensive research program to investigate the phytochemical composition and biological effects of several botanical based natural products including pomegranate (Punica granatum), red maple (Acer rubrum), polyphenol-enriched maple (Acer saccharum) syrup extract, and various phytocannabinoids (Cannabis sp.). We hypothesize that these bioactive natural products can exert beneficial biological effects via their antioxidant and anti-inflammatory effects.

A large body of evidence shows the harmful effects of cigarette smoke to oral and systemic health. More recently, a link between smoking and susceptibility to coronavirus disease 2019 (COVID-19) was proposed. COVID-19 is due to infection with severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), which uses the receptor ACE2 and the protease TMPRSS2 for entry into host cells, thereby infecting cells of the respiratory tract and the oral cavity. Here, we examined the effects of cigarette smoke on the expression of SARS-CoV-2 receptors and infection in human gingival epithelial cells (GECs). In 2014, the American Council on Education (ACE) launched an initiative to achieve gender parity among U.S. school presidencies by 2030. In terms of unmet needs for people with disabilities (PWD), dental care is number three behind residential care and employment (Fisher, 2012). Regardless of disability, oral healthcare traverses all disability diagnoses and can be a significant factor in quality of life (Deckler, 2011; Griffin et al., 2007; Norwood & Slayton, 2013). Glycosaminoglycans (GAGs) are linear, polydisperse polysaccharides consisting of alternating amino sugars and either a uronic acid or galactose residue. They are ubiquitous components of the extracellular matrix and are present on cell surfaces and in basement membranes. GAGs are highly negatively charged as a result of many sulfate and carboxylic acid groups, and are extremely heterogeneous in both their structure and in their physicochemical properties. This complexity in structure is attributed to the alterations in residue types, glycosidic bond linkage, sulfation levels, sulfation positions, and chain lengths during biosynthesis. Cancer-associated deaths account for the second-highest mortality rates in the United States. Primary modalities of treatment often include surgery, radiation, and chemotherapy, and may also incorporate targeted therapy and immunotherapy. However, resistance to these treatments remains high, resulting in disease reoccurrence and poor survival rates. While apoptosis or cell death of tumor cells is the ideal outcome for anti-cancer therapy, this is often not the case, and in fact cancer cells may upregulate several pathways, such as autophagy and senescence, as a means to undergo alternative cell fate and evade apoptotic cell death. Bisphosphoglycerate mutase (BPGM) is a part of the erythrocyte glycolysis system. Specifically, it is a central enzyme in the Rapoport-Leubering pathway, a side glycolytic pathway involved in the regulation of the concentration of the natural allosteric effector of hemoglobin (Hb), 2,3-bisphosphoglycerate (2,3-BPG). BPGM catalyses the synthesis and hydrolysis of 2,3-BPG through its synthase and phosphatase activities. The synthase activity is the main role of BPGM, while the phosphatase activity is low and is activated by the physiological effector, 2-phosphoglycolate (2-PG) with the latter mechanism poorly understood.

Glycosaminoglycans (GAGs) are linear polysaccharides whose disaccharide building blocks consist of an amino sugar (D-glucosamine) and either uronic acid (D-glucuronic acid or L-iduronic acid) or galactose. Nearly all GAGs (with the exception of hyaluronic acid) are covalently attached to proteins, forming proteoglycans, and are ubiquitous in nature. The monomeric units, chain length, degree and pattern of sulfation, and epimerization are important properties that determine the influence of these molecules. Existing management efforts take a piecemeal approach, addressing individual stressors, managing the systems for individual benefits, and protecting individual species. These disjointed efforts are doomed to fail. Loss of motor function from spinal cord injuries (SCI) results in loss of independence. Rehabilitation efforts are targeted to enhance the ability to perform activities of daily living (ADLs), but outcomes from physical therapy alone are often insufficient. Neuromodulation techniques that induce neuroplasticity may push the limits on recovery. Neuromodulation by intermittent theta burst transcranial magnetic stimulation (iTBS) induces neuroplasticity by increasing corticomotor excitability, though this has most frequently been studied with motor targets and on individuals not in need of rehabilitation. It is estimated that there are 2.8 million antibiotic-resistant infections and 35,000 deaths in the United States every year. Such microorganisms include Acinetobacter, Enterobacterioceae, Pseudomonas, Staphylococcus and Mycobacterium. Alternative treatment methods are, thus, necessary to treat such infections. Bacteriophages are viruses of bacteria. In a lytic infection, the newly formed phage particles lyse the bacterium and continue to infect other bacteria. Whereas immune modulation has proven beneficial in multiple sclerosis (MS), we hypothesized that targeting down-stream modulators of neurogenesis and subsequent remyelination may offer an additional, if not superior, point of intervention in an attempt to repair damage and recover lost function. Exosomes (EXs) released from these stem cells could be one of the major contributors, as they are known to convey the benefit of one cell to another cell via microRNAs (miRNA). With the intent to create an optimized monoamine oxidase-B (MAO-B) inhibitor, researchers in this investigation synthesized derivatives of the benzylidene oxindole scaffold in order to determine a lead molecule for further drug development. Previous work in this laboratory group evaluated a similar scaffold, the chalcone structural unit. As this class of compounds was determined to possess the ability to permeate the blood brain barrier (BBB) and to act as potent MAO-B inhibitors, it was posited that scaffold hopping from this previously studied molecular skeleton to benzylidene oxindoles would provide analogous results. Cisplatin is a DNA damage-based chemotherapeutic drug widely used to treat various types of cancers; however, the treatment's toxicity restricts its efficiency. Studies have shown that the circadian rhythm controls the DNA damage response and affects the repair pathways of cisplatin-induced DNA damage. Circadian clock modulation, therefore, has been proposed to be a potential mechanism for enhancing cisplatin tolerability. Here we used clock-enhancing molecules to evaluate the effect of pharmacological clock modulation on cisplatin cytotoxicity. Discrete subaortic stenosis (DSS) is an obstructive cardiac disease characterized by the formation of a thin fibromuscular ring of tissue in the left ventricular (LV) outflow tract (LVOT). Although its etiology is unknown, the association between DSS, pediatric age, and morphological LVOT abnormalities points to a potential hemodynamic etiology. Here, altered subaortic flow dynamics, stemming from deviated LVOT anatomy, may promote lesion formation through local fluid shear stress abnormalities triggering membrane formation. This study characterizes the functional dependence of drag on Reynolds number for a deformed vacuum lighter-than-air vehicle. The commercial computational fluid dynamics (CFD) code, FLUENT, is used to preform large eddy simulations (LES) over a range of Reynolds numbers; only Reynolds numbers less than 310,000 are considered. While the overarching goal is drag characterization, general flow-field physics are also discussed, including basic turbulence spectra. All large eddy simulations are preceded by a Reynolds-averaged Navier-Stokes (RANS) simulation using Menter’s shear stress transport (SST) model. This thesis examines one angle of state-society relations in authoritarian states through the lens of Chinese nongovernmental organizations (NGOs) and their legal environment. While grassroots organizations have not prompted political liberalization in China, they have not been entirely co-opted by the party-state either. Through an examination of policy changes, a study of 120 organizations, and a case study of a non-profit incubator, this thesis explores whether the political environment for China’s domestic NGOs varies by geographic region and issue area. Attached algae are ubiquitous components of lake benthic habitats wherever sufficient light reaches submerged surfaces. Attached algae interact with heterotrophic bacteria and fungi to form complex biofilms (“periphyton”) that provide a nutritious food source for consumers and influence biogeochemical cycling by regulating redox potential at the sediment-water interface. Despite their ecological importance, there are limited data on the role of periphyton in the Laurentian Great Lakes. Cyanobacteria are important primary producers, but large cyanobacterial harmful algal blooms (HABs) have many negative ecological and health impacts and are becoming increasingly common. Honeoye Lake (New York, USA) is a shallow, eutrophic lake characterized by increasingly frequent HABs. Nitrogen (N) and phosphorus (P) loads often drive HABs in lakes, and sediment processes can contribute to N removal (e.g., denitrification) or loading (e.g., N fixation, remineralization).
