Chulalongkorn University Theses and Dissertations (Chula ETD)
The purpose of this study was to develop Eudragit® -nanoparticles (NPs) eye drop formulations containing fenofibrate (FE) for diabetic retinopathy and age-related macular degeneration. Due to the limited aqueous solubility of FE, it may hamper the ocular bioavailability and results in low efficiency to treat such diseases. Hence, the increased solubility of FE was determined by complexation with cyclodextrins (CDs) and CD derivatives The solubility determination was evaluated by phase-solubility technique with heating method. Among CD tested, randomly methylated-b-CD (RMbCD) exhibited the best solubilizer with the highest complexation efficiency for FE. Moreover, the water-soluble polymer i.e., Soluplus® (SOL), poloxomer 407 …

Smart food packaging based on biosensors has been attracting more and more interest to the industrial community because of the concerns of food quality and safety. A food packaging with biosensor has a scope to enable real-time monitoring of microbial breakdown products of packaged foods. Furthermore, one of the biggest challenges in implementing biosensor for smart packaging materials is the development of bio-sensing active materials that can leverage their electrical, thermal, biodegradable and other functional properties. In this regard, nanocellulose-based activated carbon (NAC) nanocomposite was developed using the activated carbon and nanocellulose gel using the casting method with their different …
Profiling Of Fda-Approved And Clinical Trial Drugs Revealed Shared Cytotoxicity And Collateral Sensitivity In Resistant (H69ar) And Non-Resistant (H69) Small Cell Lung Cancer Cells. (Drug Repurposing In Cancer Chemotherapy), Pius Reyderg Agyemang
Electronic Theses and Dissertations
Some cancers are capable of “spitting out” drugs being fed to them, metaphorically speaking, becoming resistant to what were previously effective chemotherapeutics. In small-cell lung cancer (SCLC), an overexpression of a membrane protein (MRP1) and its transport activity can lead to chemotherapy failure. However, this study showed that certain drugs are selectively cytotoxic (exhibit collateral sensitivity) to MRP1-overexpressed SCLC (H69AR) cells. In this study, three drugs (Erlotinib, Pyrimethamine, Fludarabine) were identified to exhibit a dose-dependent collateral sensitivity on H69AR with IC50 values of ~3.5 μM, ~2 μM, and ~20 μM respectively. Halting the transport activity of the MRP1 with 25 …
Free-Living Diazotrophs And The Nitrogen Cycle In Natural Grassland Revealed By Culture Dependent And Independent Approaches, Amrit Koirala
Free-Living Diazotrophs And The Nitrogen Cycle In Natural Grassland Revealed By Culture Dependent And Independent Approaches, Amrit Koirala
Electronic Theses and Dissertations
Biological nitrogen fixation contributes to half of the global supply of nitrogen to the biosphere. It is carried out by a diverse group of prokaryotes called diazotrophs via the nitrogenase enzyme. Nitrogen fixation research is focused on the narrow group of symbiotic diazotrophs, and the vast majority of free-living diazotrophs which contribute significantly to fixed nitrogen are yet to be explored. The goal of this research was to access phylogeny of diazotrophs considering the most up-to-date genomic information and apply that knowledge to understand the diversity of free-living diazotrophs in a natural grassland ecosystem, both by culture dependent and independent …

The importance of next generation sequencing (NGS) rises in cancer research as accessing this key technology becomes easier for researchers. The sequence data created by NGS technologies must be processed by various bioinformatics algorithms within a pipeline in order to convert raw data to meaningful information. Mapping and variant calling are the two main steps of these analysis pipelines, and many algorithms are available for these steps. Therefore, detailed benchmarking of these algorithms in different scenarios is crucial for the efficient utilization of sequencing technologies. In this study, we compared the performance of twelve pipelines (three mapping and four variant …

Positive Regulation Of Tnfr1 Signaling Via Sh3 Recognition Motif, Fatma Ece Çopuroğlu, Fatma Zehra Hapi̇l Zevkli̇ler, Şükran Burçak Yoldaş, Osman Ni̇dai̇ Özeş
Turkish Journal of Biology
TNF is a pleiotropic cytokine and shows its biological function by binding to its receptors called TNFR1 and TNFR2. While TNFR1 induces apoptosis by activation of caspase-8 via the "death domain", it also activates IKKα/β, MKK3/6, MKK4/7 by activation of TAK1. Although the TNFR1 signaling pathway is known by in large, it is not known how AKT and MAPKs p38, ERK1/2, and JNK1/2 are activated. The presence of a proline-rich PPAP region, (P448PAP451, a binding site for the SH3 domain-containing proteins) very close to the C-terminus promoted us to determine whether this region has any role in the TNFR1 signal …

Negative Regulation Of Diminutive Cancer Regulator Through Differentiation And Microrna Pathway Components In Drosophila Cells, Sumira Malik
Turkish Journal of Biology
Drosophila model is intensively studied for the development of cancer. The diminutive (dMyc), a homolog of the human MYC gene, is responsible for cell- apoptosis and its upregulation is responsible for determining the fate of cancerous growth in humans and Drosophila model. This work implores the requirement of dMyc and its expression as one of the major regulator of cancer with other proteins and repression of dMyc mRNA in Drosophila S2 cells. Here we report protein complex of Argonaute 1 (AGO1), Bag of marbles (Bam), and Brain tumor (Brat) proteins and not the individual factor of this complex repression of …

Compensatory Expression Regulation Of Highly Homologous Proteins Hnrnpa1 Andhnrnpa2, Yan Chang, Xiao Feng Lu, Jia Ying Qiu
Turkish Journal of Biology
Heterogeneous nuclear ribonucleoprotein (HNRNP) A1 and A2 are the most abundant HNRNPs with nearly identical functions, and play important roles in regulating gene expression at multiple levels (i.e. transcription, posttranscription, and translation). However, the expression and regulation mechanism of HNRNPA1 and A2 themselves remain unclear. In this study, the amino acid sequences of HNRNPA1 and HNRNPA2 were compared and found to have 78% and 86% homology in key functional domains. Transfection of HEK293 cells with small interfering RNA and overexpression vectors of HNRNPA1 and HNRNPA2 demonstrated that HNRNPA1 and HNRNPA2 paralogs regulate each other?s expression in a compensatory manner at …

Turkish Journal of Biology
Inflammation has a dual effect: it can protect the body and destroy tissue and cell as well. The purpose of this experiment was to determine the role of IL-1R1 in liver regeneration (LR) after partial hepatectomy (PH) in aged mice. The wild-type (WT, n = 36) and the IL-1R1 knockout (KO, n = 36) 24-month-old C57BL/6J mice underwent two-thirds PH; 33 WT mice underwent sham operation. Liver coefficient was calculated by liver/body weight. The mRNA and protein expressions of genes were evaluated by quantitative realtime polymerase chain reaction (qRT-PCR) and Western blotting methods, respectively. Compared with WT mice, liver coefficient …

Reconstitution Of Pol Ii (G) Responsive Form Of The Human Mediator Complex, Murat Alper Cevher
Turkish Journal of Biology
RNA polymerase II (Pol II) is a 12 subunit protein complex from yeast to human that is required for gene expression. Gdown1 containing Pol II [Pol II (G)] is a special form of Pol II that is catalytically inactive and heavily depends on the 30-subunit Mediator complex for its activator and basal dependent function in vitro. Here we report for the first time, the identification and the generation of a 15-subunit human Mediator complex via the novel multibac baculovirus expression system that is fully responsive to Pol II (G). Our results show complete recovery of Pol II (G) dependent transcription …

Polyhydroxyalkanoate Accumulation In Streptomyces Coelicolor Affected By Sco7613 Gene Region, Zeynep Demi̇r Öksüz, Tuğrul Doruk, Nevi̇n Yağci, Sedef Tunca Gedi̇k
Turkish Journal of Biology
Polyhydroxyalkanoate (PHA) is stored as an important carbon and energy source in bacterial cells. For biomedical applications, gram-positive bacteria can be better sources of PHAs, since they lack outer membrane lipopolysaccharide. Although gram-positive Streptomyces coelicolor A3(2) has been indicated as a high potential PHA producer, phaC gene that encodes the key enzyme PHA synthase in the metabolic pathway is not determined in its genome. BLAST search results of the GenBank database argued that SCO7613 could specify a putative polyhydroxyalkanoate synthase (PhaC) responsible for PHA biosynthesis. Deduced amino acid sequence of SCO7613 showed the presence of conserved lipase box like sequence, …

Linkage Of Nanosecond Protein Motion With Enzymatic Methyl Transfer By Nicotinamide N-Methyltransferase, Yahui Jing, Yiting Cheng, Fangya Li, Yuping Li, Fan Liu, Jianyu Zhang
Turkish Journal of Biology
Nicotinamide N-methyltransferase (NNMT), a key cytoplasmic protein in the human body, is accountable to catalyze the nicotinamide (NCA) N1-methylation through S-adenosyl-L-methionine (SAM) as a methyl donor, which has been linked to many diseases. Although extensive studies have concerned about the biological aspect, the detailed mechanism study of the enzyme function, especially in the part of protein dynamics is lacking. Here, wild-type nicotinamide N-methyltransferase together with the mutation at position 20 with Y20F, Y20G, and free tryptophan were carried out to explore the connection between protein dynamics and catalysis using time-resolved fluorescence lifetimes. The results show that wild-type nicotinamide N-methyltransferase prefers …

A Cytoplasmic Escapee: Desmin Is Going Nuclear, Ecem Kural Mangit, Ni̇lüfer Düz, Pervi̇n Ruki̇ye Di̇nçer
Turkish Journal of Biology
It has been a long time since researchers have focused on the cytoskeletal proteins' unconventional functions in the nucleus. Subcellular localization of a protein not only affects its functions but also determines the accessibility for cellular processes. Desmin is a muscle-specific, cytoplasmic intermediate filament protein, the cytoplasmic roles of which are defined. Yet, there is some evidence pointing out nuclear functions for desmin. In silico and wet lab analysis shows that desmin can enter and function in the nucleus. Furthermore, the candidate nuclear partners of desmin support the notion that desmin can serve as a transcriptional regulator inside the nucleus. …

The history of racism and prejudice in the United States is not limited to segregation, slavery, and discrimination. Medical and scientific racism continue to disproportionately affect Black and Indigenous People of Color (BIPOC). BIPOC have been used for scientific advancement without full disclosure of potential harm or adequate compensation. Scientists and medical providers have dehumanized minority individuals by disrespecting their autonomy and targeting vulnerable populations. This thesis consists of a literature review that surrounds the history of medical and scientific exploitation of people of color, ethical guidelines surrounding research, eugenics, and how assumptions affect the quality of care that minority …

DNA/RNA synthesis precursors are especially vulnerable to damage induced by reactive oxygen species occurring following oxidative stress. Guanosine triphosphates are the prevalent oxidized nucleotides, which can be misincorporated during replication, leading to mutations and cell death. Here, we present a novel method based on micro-Raman spectroscopy, combined with ab initio calculations, for the identification, detection, and quantification of oxidized nucleotides at low concentration. We also show that the Raman signature in the terahertz spectral range (<100 >cm(-1)) contains information on the intermolecular assembly of guanine in tetrads, which allows us to further boost the oxidative damage detection limit. Eventually, we …100>

Problem
Private sector Emergency Medical Technicians (EMTs) are first responders who respond to traumatic incident scenes along with firefighters and police officers. However, news agencies and the public do not acknowledge private sector EMTs with the same vigor or accolade as public sector EMTs, nor are the risks to life, health, and mental well-being recognized among this population. Thus, the research literature has been sparse regarding how private sector EMTs experience PTSD, and what unique risk factors and needs may be present.
Purpose Statement
The purpose of this study was to examine the relationship between PTSD symptoms and coping skills, …

The Problem
Correlational studies suggest that gender, attitudes to mathematics, mathematics performance, the number of college mathematics courses taken, and mathematics teacher efficacy beliefs may be related to mathematics anxiety among preservice teachers (Boyd et al., 2014; Gresham, 2018; Jaggernauth & Jameson-Charles, 2015). Exploratory studies of mathematics anxiety and teacher self-efficacy among elementary teachers exist in Trinidad and Tobago (Jaggernauth 2012; Jaggernauth & Jameson-Charles, 2015). However, the differences in samples, and weakness in methodology, may make it challenging to generalize the results to the preservice teacher population. In addition, no studies have found which variable or combination of variables is …
