Body fluids, such as blood and semen, can provide invaluable information to a criminal investigation. As such, it is imperative that body fluid evidence is properly detected and identified. Our laboratory has developed a methodology to universally detect and identify body fluids using Raman spectroscopy and chemometrics. This methodology was developed under ideal conditions, which are rare in the field. Body fluid evidence can be found on substrates that produce a large Raman signal that can hinder detection of body fluid traces, which produce a weak signal. In 1969, the enrichment of mobile element repeat sequences led Britton and Davidson to propose the hypothesis that gene expression in higher eukaryotes is regulated through the exaptation of mobile elements. In this work, I have explored the hypothesis that mobile elements can also be harnessed by the host to regulate asymmetric cell division, thereby determining cell fate. Retrotransposons are ubiquitous eukaryotic mobile elements that transpose through an RNA intermediate. Phosphodiesterases (PDEs) are integral components of 3’,5’-cyclic adenosine monophosphate (cAMP) signaling pathways by degrading cAMP to modulate the concentration, duration, and localization of the cAMP signal which maintains the specificity of cAMP pathways. The human pathogen, Mycobacterium tuberculosis (Mtb) has a unique cAMP network architecture with at least 15 adenylyl cyclases (ACs) that generate cAMP, but only one characterized PDE, Rv0805, which is found exclusively in pathogenic mycobacteria. Rv0805 can influence Mtb cAMP levels but the absence of Rv0805 orthologs in non-pathogenic mycobacteria and apparent separation of Rv0805 from cAMP directed roles led to numerous questions surrounding PDE function in …

Translation initiation is a critical step in the process of protein synthesis. The canonical way of translation initiation involves ribosomes being recruited to the 7-methyl guanosine cap present at the 5’end of the untranslated region (5’ UTR) of the RNAs. However, viral RNAs and some cellular mRNAs lack this 5’ cap structure and thus deploy an alternate non-canonical translation initiation mechanism. In non-canonical translation initiation, ribosome recruitment is facilitated by the RNA secondary structures called Internal Ribosome Entry Site (IRES) present most often in the 5’ UTR. Methamphetamine (MA) is a psychostimulant drug of addiction that induces a wide range ofeffects. Among its many side effects is the potentiation of cellular, physiological, and behavioral changes in the stress response system. Upon exposure, the hypothalamic-pituitary-adrenal (HPA) axis is initiated. This process entails many factors. Receiving input from other brain regions, the paraventricular nucleus (PVN) of the hypothalamus secretes corticotropin-releasing factor (CRF) and arginine vasopressin into the anterior pituitary gland. These together stimulate the synthesis and release of adrenocorticotropic hormone, which circulates via the blood stream to cause the release of glucocortoicoids, the stress hormone (CORT). When trying to understand risk factors for a disease, even before we know the causal agents, it is necessary to create a surveillance data set that answers the questions of who, when, and where, and includes any potential covariates which may either promote or prevent the disease. There are a wide variety of surveillance data. In this dissertation, I defend the view that, despite empathy’s susceptibility to problematic biases, we can and should cultivate empathy to aid our understanding of our own values and the values of others. I argue that empathy allows us to critically examine and potentially revise our values by considering concrete moral problems and our own moral views from the perspective of another person. Appropriately calibrated empathy helps us achieve a critical distance from our own moral perspective and is thus tied to impartiality in moral inquiry. Objective: This study examined the moderating role of racial conflict among the relation between experiences of racial discrimination and mental health (psychological distress and psychological well-being) for the multiracial sub-group. Furthermore, this study was among the first to also explore how multifaceted identity choice/racial self-identification was associated with racism, racial tension, and mental health. Methods: Two hierarchical multiple regressions were conducted to assess the potential moderation effect of racial conflict on the relationships between racism and mental health for 242 multiracial adults using survey design and recruitment methods through Amazon’s Mechanical Turk and social media. Adverse childhood experiences, such as interparental conflict exposure, have long-term negative effects across various domains, including psychological and physiological health. Research reveals a link between interparental conflict and coordination between stress response systems, specifically sympathetic and parasympathetic nervous systems (SNS and PNS, respectively). Despite this knowledge, much less is known about potential protective factors or intervention strategies within this area. Viral epitranscriptomics is a novel field of RNA biology. Chemical groups are appended to RNA nucleotides under different conditions in cells, and I examine these changes using a mass spectrometry-based approach. Various changes are found to occur in post-transcriptional modifications (PTMs) during viral infection. The field primarily studies a small number of PTMS, such as m6A, while I explore a variety of novel modifications and their impact on viral infection. Hepatitis C virus (HCV) infection induces a variety of modification changes, including the novel presence of dimethylcytosine (m5Cm/m44C). “Meeting Places: The Entanglements of Poetry and Science in the Modern American Imagination” explores how Ralph Waldo Emerson, W.E.B. Du Bois, William Carlos Williams, and Muriel Rukeyser responded to the rise of modern science and industrial technology by reflecting on the similarities and differences between the poetic and scientific imaginations. Across four chapters, I show how the growth of the natural and social sciences from the mid-nineteenth to mid-twentieth century spurred these writers to rethink the social and cultural functions of literature in a democratic society. Females with autism spectrum disorder are an underrepresented group in the autism literature that is in need of greater understanding. Growing evidence suggests that autistic females have a unique clinical presentation of symptoms and they are more frequently camouflaging their autism characteristics to fit in with peers. While support for social camouflaging theory builds, little attention has been paid to the effects of camouflaging on outcomes for females with autism. Per- and polyfluoroalkyl substances (PFAS) constitute a broad class of anthropogenic chemicals that have been used in numerous industrial and consumer-product applications since the late 1940s. The environmental persistence of these compounds in combination with their widespread commercial use have led PFAS to be recognized as contaminants of concern on the global scale. Despite regulatory efforts calling for the decreased use of certain perfluorocarboxylic and -sulfonic acids referred to as “legacy” PFAS, these compounds continue to be detected in both environmental and biological matrices. Youth elopement from out-of-home placement settings, an act colloquially referred to as AWOL, is a frequent and extremely risky outcome for youth and a key concern for child welfare entities. AWOL is hypothesized here as a maladaptive coping response to the experience of out-of-home placement as a crisis-inducing event. Conceiving of AWOL as a response to a crisis state redefines apparent risk factors as crisis origins and enables an understanding of which youth may experience a removal and out-of-home placement event as crisis inducing. Primary cilia are hair-like protrusions that stem from the basal bodies in the cytoplasm and extend into the extracellular space to sense signals. Intraflagellar transport (IFT) functions to transport cargo molecules into and out of the ciliary compartment to assemble, maintain, and disassemble the cilia. Major depressive disorder is one of the most pervasive psychiatric illnesses in the United States. Women are at greater risk for developing depression, particularly during their childbearing years. Approximately 17% of new mothers develop postpartum depression within 4 weeks after parturition. The risk for postpartum depression is even greater in women who do not breastfeed or stop breastfeeding early. Major depressive disorder and postpartum depression share the same symptomology and common etiological bases. There is robust evidence that early childhood adversity has wide ranging impacts on mental health, physical health and well-being in adulthood. Executive functioning (EF) deficits may partially mediate these effects, predisposing children to risky behavior and poor decision making. A growing body of research indicates that there is a link between early adversity and EF. However, more research is needed with younger children and with children at higher risk of multiple adversities. Long noncoding RNAs (lncRNAs) are emerging as key molecules regulating several cellular pathways and contributing to the development and progression of different diseases including cancer. The epithelial-to- mesenchymal transition (EMT), a molecular reprogramming process that plays a critical role in cancer metastasis, involves a complex interaction between protein coding and non-coding transcripts including lncRNAs. Understanding the biological significance of those lncRNAs of relatively unknown function, may help to uncover potential therapeutic targets for the aggressive subtypes of breast and other cancers. This dissertation considers various examples of change and continuity in biology. In particular, it focuses on the conceptual possibilities during the fission of microorganisms and genes when they multiply or replicate. Among the kinds of conceptual revision that are outlined in the first two chapters, the three chapter argues that certain concepts such as death, reproduction and organism have limited applicability in certain domains of biology. The fourth chapter investigates whether synthetic genomes should be considered parts of genetic lineages. It concludes that synthetic genomes are parts of the lineages upon which they were modeled. In this dissertation, I review the fundamental processes and mechanisms for bacterial DNA replication, especially the mechanisms employed by high-fidelity DNA polymerases to replicate the genome. Unlike the prototypical bacterial system from Escherichia coli which uses a single C-family polymerase, DNA polymerase IIIα (Pol IIIα), to replicate the genome, low-GC content Gram-positive bacteria utilize two essential C-family polymerases, PolC and DnaE. PolC and DnaE work cooperatively to replicate the genome, with DnaE initiating synthesis from RNA-primers and PolC performing the bulk synthesis. It is well documented that children with autism spectrum disorder (ASD) tend to exhibit more problem behaviors compared to typical peers; moreover, parents of children with ASD are more likely to report experiencing greater parental and family stress. The current study set out to shed light on the ways and degree to which problem behaviors in children with ASD may impact multiple aspects of family life. Organophosphates are a group of insecticides that are commonly used as chemical warfare agents (CWAs), since they can act as nerve agents by inhibiting acetylcholinesterase. Acetylcholinesterase is a vital enzyme of the central nervous system (CNS). This enzyme facilitates the breakdown of acetylthiocholine into thiocholine and acetate. The major goal of the research will be to provide a preliminary rapid, sensitive, and reliable method for sensing organophosphates. Current methods are enzyme inhibition-based – that is, a decrease in absorbance or signal yields a positive indicator for the presence of analyte.
