The purpose of this course is to introduce students to the etiology and classification of mental disorders as defined by the Diagnostic and Statistical Manual of Mental Disorders, Fifth Edition (DSM-5). Students will learn to utilize diagnostic information to conduct diagnostic interviews, conceptualize the presenting problem, and write diagnostic assessments in order to facilitate case management, treatment plan development, and therapeutic interventions with people seeking/receiving mental health counseling services. Instructional methodology will include lectures, case studies presented in class, diagnostic activities using dyadic and small group activities, and other instructional modalities designed to facilitate the learning process.

“We are not alone!” (“Ui aa natto aroon!”) was the shared response of Natsume Fusanosuke and his colleague Takekuma Kentarō when, as if visited by extraterrestrials, they encountered a kindred spirit in Scott McCloud’s Understanding Comics (first published in Japanese as Mangagaku in 1998; retranslated in 2020).1 Natsume and his fellow critics and scholars had published their own definitive study of how to understand comics in 1995: the now out-of-print co-authored How to Read Manga (Manga no yomikata, Takarajima), for which Natsume made a considerable contribution. The Gram-negative bacterium, Salmonella enterica is a prominent etiologic agent of both diarrheal disease and enteric fever that encompasses over 2500 serovars, including S. Typhimurium (STm) and S. Typhi (STy). S. enterica is transmitted through contaminated food and water and, following ingestion, invades the gastrointestinal (GI) mucosa. The bacterium uses flagellar-based motility to target microfold (M) cells overlying gut-associated lymphoid tissues known as Peyer’s patches in the small intestine. Entry into Peyer’s patch tissues is a pivotal step in the infection process, as the bacterium can then disseminate systemically in the host. Given the rise in antibiotic resistance amongst S. …

Group II introns are self-splicing mobile elements that are thought to be the evolutionary ancestors of eukaryotic retrotransposons, the spliceosome, and spliceosomal introns. Yet, little is known about how group II introns have coevolved within their native hosts or about the corresponding host biology.Using the Ll.LtrB intron in its native host, Lactococcus lactis, I first sought to identify host factors that influence retrotransposition. Since retrotransposition can be costly to the host organism, group II introns must achieve a delicate balance between self-proliferation and host preservation. In this dissertation I provide a pre-steady-state kinetic characterization of an antibiotic-resistant mutant of a Staphylococcus aureus DNA polymerase. Staphylococcus aureus is one of the most common causes of infections in humans, and is widely known for its ability to acquire resistance to most antibiotics. Staphylococci infections pose a significant burden to the healthcare system and increase mortality, as more than 95% of Methicillin-Resistant S. aureus (MRSA) infections do not respond to first-line antibiotics. This thesis describes a chemical method for alternative oligonucleotide purification that is non-chromatographic and gel-free and allows to routinely synthesize and purify long functional RNA strands. The purification of long RNAs is based on the bio-orthogonal inverse electron demand Diels-Alder (IEDDA) chemistry between trans-cyclooctene (TCO) and tetrazine (Tz). Target oligonucleotide strands are selectively tagged with Tz and can be captured and purified from the failure sequences with immobilized TCO. RNA strands are synthesized on solid support through a photolabile linker to avoid the loss of Tz tag. Lead (Pb) is ubiquitous in the environment and exposure can have adverse health effects even at low concentrations, particularly in young children. Blood lead levels (BLLs) are the best indicator of recent Pb exposure. Based upon data from the NHANES, the Centers for Disease Control and Prevention’s (CDC) blood Pb (BPb) reference value, defined as the 97.5th percentile of children 1-5 years old, was lowered from 10 μg/dL to 5 μg/dL in 2012. According to more recent NHANES data, the 97.5th percentile has decreased to 3.5 μg/dL. If the CDC recommends a further lowering of the BPb reference value, many …

Lyme disease is a bacterial pathogen that spreads from animals to humans through bites from blacklegged ticks (Ixodes scapularis). The disease is a public health problem causing both physical suffering and increased health care spending since the 1980s. A snapshot of Lyme disease coverage in the popular media shows that most people prioritize conversations about Lyme disease diagnosis and treatment over prevention concerns. Anthropology has recently examined peoples’ anxieties and debates regarding the diagnosis and treatment of Lyme disease in the United States. Differentiated Instruction, an approach to teaching that encourages educators to adjust their content, pedagogy, and assessment based on the needs of individual students, lacks suggestions for how to target specific student groups, such as “high-risk” students. Researchers in the field of evolutionary developmental psychology have begun to focus on stress-adapted students (who are described in a similar way as “high risk” students) and has made suggestions about how to play to the strengths of the cognitive enhancements, or “hidden talents” of these students. The life sequence and human capital accumulation of women have changed in the US. This dissertation intends to propose three interrelated studies that investigate the possible trajectories of human capital accumulation after women’s transition to motherhood, the determinants of these trajectories, and the implications for women and their children’s wellbeing. Driven by the life course perspective, the cumulative advantage and disadvantage theory, and Bourdieu’s cultural framework, this dissertation uses mothers’ vocational training experiences to identify distinct trajectories over time in the first study. Introduction. Academic work on vegans is biased towards studying educated White women, despite evidence that vegans having varying identities. The lack of diversity in research has implications for our understanding of psychological issues that affect vegans. Data regarding mental health of vegans is mixed, due to a failure to account for differing identities and experiences. This study sought to replicate and extend existing research (i.e., DelTosta et al., 2019) on predictors of vicarious traumatization among mental health counseling trainees and tested empirically the effects on trainees’ quality of life. The current investigation examined relations among personal distress empathy (PDE), the supervisory working alliance (SWA), mindfulness, vicarious traumatization (VT), and quality of life (QoL) in a sample of 294 mental health counseling trainees in the United States and Canada. Since its inception, residential treatment has been expected to continuously evolve and adapt. Most recently, change in the field has revolved around accountability and the incorporation of evidence-based practices. Among those practices include trauma-informed approaches as an effort to meet the ever-changing and increasingly significant needs of youth in care. Although randomized control-trials remain the gold standard for demonstrating effectiveness in residential treatment remains, program evaluation has been acknowledged as a more viable assessment tool.

Ricin is a highly lethal protein toxin derived from the seeds of the castor plant, Ricinus communis. It is a Type II ribosome inactivating protein (RIP), meaning it is a heterodimer with one subunit, ricin toxin B (RTB), that mediates cell surface attachment and intracellular trafficking and a second subunit, ricin toxin A (RTA), that irreversibly shuts down protein synthesis in the cytosol. During trafficking, RTA and RTB necessarily separate in the endoplasmic reticulum, wherein RTA unfolds and translocates into the cytosol where it refolds into an enzymatically active conformation. Emotion regulation and executive functioning are intricate processes that allow individuals to initiate, inhibit, or modulate emotions and behaviors in service of personally-relevant goals in familiar, novel, and ambiguous contexts. Research has demonstrated that exercise improves emotion regulation and executive functioning. Recently, researchers have started investigating whether mindful exercise – physical exercise that incorporates an inwardly directed, non-judgmental, contemplative focus – explicitly strengthens these capabilities. Following its discovery in 1947, Zika virus garnered little attention in the global stage until 2015, when a rapidly spreading epidemic brought the virus, and its devastating clinical manifestations to light. Zika virus pathologies include Congenital Zika Syndrome, a known causative of microcephaly, as well as spontaneous miscarriages and the development of Guillain Barré syndrome. Focusing polycapillary optics yield high gains in intensity and increased spatial resolution for a variety of clinical, lab-based, synchrotron, or in situ analysis applications. In this dissertation we investigate the extension of two applications of focusing polycapillary optics. The first is the application of polycapillary optics in radiation therapy. This discussion includes measurements and calculation of dose for focused beam orthovoltage therapy. Body fluids, such as blood and semen, can provide invaluable information to a criminal investigation. As such, it is imperative that body fluid evidence is properly detected and identified. Our laboratory has developed a methodology to universally detect and identify body fluids using Raman spectroscopy and chemometrics. This methodology was developed under ideal conditions, which are rare in the field. In 1969, the enrichment of mobile element repeat sequences led Britton and Davidson to propose the hypothesis that gene expression in higher eukaryotes is regulated through the exaptation of mobile elements. In this work, I have explored the hypothesis that mobile elements can also be harnessed by the host to regulate asymmetric cell division, thereby determining cell fate. Phosphodiesterases (PDEs) are integral components of 3’,5’-cyclic adenosine monophosphate (cAMP) signaling pathways by degrading cAMP to modulate the concentration, duration, and localization of the cAMP signal which maintains the specificity of cAMP pathways. The human pathogen, Mycobacterium tuberculosis (Mtb) has a unique cAMP network architecture with at least 15 adenylyl cyclases (ACs) that generate cAMP, but only one characterized PDE, Rv0805, which is found exclusively in pathogenic mycobacteria. Rv0805 can influence Mtb cAMP levels but the absence of Rv0805 orthologs in non-pathogenic mycobacteria and apparent separation of Rv0805 from cAMP directed roles led to numerous questions surrounding PDE function in …
