Free-Floating Immunostaining Of Mouse Brains, Longlong Tu, Nan Zhang, Kristine M Conde, Jonathan C Bean, Chunmei Wang, Yong Xu
Children’s Nutrition Research Center Staff Publications
Immunohistochemical staining of mouse brains is a routine technique commonly used in neuroscience to investigate central mechanisms underlying the regulation of energy metabolism and other neurobiological processes. However, the quality, reliability, and reproducibility of brain histology results may vary among laboratories. For each staining experiment, it is necessary to optimize the key procedures based on differences in species, tissues, targeted proteins, and the working conditions of the reagents. This paper demonstrates a reliable workflow in detail, including intra-aortic perfusion, brain sectioning, free-floating immunostaining, tissue mounting, and imaging, which can be followed easily by researchers in this field.

Journal of Applied Research on Children: Informing Policy for Children at Risk
Food insecurity is a growing concern both locally and globally. Food insecurity has a number of causative factors. The food delivery system is laden with many inequities and fragilities. We will outline a framework of the food delivery system and the areas where inequities exist.
Climate change is an equally expansive topic. Changes to our climate have a myriad of impacts on our food delivery systems and in turn can exacerbate our already tenuous delivery system. Continued impacts on our environment creates even more imbalance and can subsequently exacerbate food inequities further.

Internet Journal of Allied Health Sciences and Practice
Purpose: Knowledge translation (KT) in the health system is critical for the delivery of evidence-based practice. Supporting allied health professionals to plan and implement KT, using strategies that broadly reach across multiple geographical locations of the workforce, are needed. We piloted KT group telementoring via videoconference as an innovative solution to support and empower a vastly dispersed workforce.
Methods: The 6-month Knowledge Translation Support Service (KTSS) involved monthly, one-hour, virtual group-based support of clinician-led KT projects within state-run hospital and health services. Animals utilise a wide range of regulatory systems to control the conditions within their bodies or homeostasis. These regulatory systems control for example, body temperature, nutritional state, water balance, social interactions and fear and these systems react to environmental and endogenous stimuli so as to correct or prevent displacements from the optimal range. The behavioural and physiological responses utilised by the animal are usually proportional to the challenge to homeostasis. The stress response commences with the central nervous system perceiving a potential challenge to homeostasis. Ground-based animal models have been used extensively to understand the effects of microgravity on various physiological systems. Among them, hindlimb suspension (HLS), developed in 1979 in rats, remains the gold-standard and allows researchers to study the consequences of total unloading of the hind limbs while inducing a cephalic fluid shift. While this model has already brought valuable insights to space biology, few studies have directly compared functional decrements in the muscles of males and females during HLS. Objective: The lack of a validated and cross-culturally equivalent scale for measuring individual-level water insecurity has prevented identification of those most vulnerable to it. Therefore, we developed the 12-item Individual Water InSecurity Experiences (IWISE) Scale to comparably measure individual experiences with access, use, and stability (reliability) of water. Here, we examine the reliability, cross-country equivalence, and cross-country and within-country validity of the scale in a cross-sectional sample.
Methods: IWISE items were implemented by the Gallup World Poll among nationally representative samples of 43 970 adults (15 y) in 31 low-income and middle-income countries (LMICs). Internal consistency was assessed …

We present the Aquatic Symbiosis Genomics Project, a global collaboration to generate high quality genome sequences for a wide range of eukaryotes and their microbial symbionts. Launched under the Symbiosis in Aquatic Systems Initiative of the Gordon and Betty Moore Foundation, the ASG Project brings together researchers from across the globe who hope to use these reference genomes to augment and extend their analyses of the dynamics, mechanisms and environmental importance of symbiosis. Applying large-scale, high-throughput sequencing and assembly technologies, the ASG collaboration will assemble and annotate the genomes of 500 symbiotic organisms – both the “hosts” and the microbial …

Background: In Pakistan, the prevalence of stunting among children under-five years has remained above WHO critical thresholds (≥30%) over the last two decades.
Objective: We hypothesized that an unconditional cash transfer (UCT) combined with lipid-based nutrient supplement (LNS) and/or social and behavior change communication (SBCC) will prevent stunting among children 6-23 months of age.

A study was undertaken to determine the effects of differing levels of nitrogen (N) application (0 (N1), 25 (N2), 50 (N3) and 75 kgN/ha (N4)) during late autumn (T1), early (T2) and late winter (T3) on the nutritive characteristics and nitrate content of perennial ryegrass. Changes in crude protein (CP) for all treatments at each application time were similar irrespective of level of N application. At the commencement of all treatments, the existing CP content was highest in N3, followed by N2, N1 and N0. Nitrate content decreased throughout T1 primarily due to dry conditions, whilst during T2, levels for …

The objectives of this experiment were to evaluate the influence of N fertilizer, age of regrowth, and season on concentration of N in cell-wall fractions of three warm-season perennial grasses (limpograss, [Hemarthria altissima (Poir.) Stapf et C.E. Hubb.], bermudagrass [Cynodon spp.], and bahiagrass [Paspalum notatum Flügge]. The herbage neutral detergent insoluble nitrogen (NDIN) fraction composed almost half of total N in these grasses. Though acid detergent insoluble nitrogen (ADIN) concentrations generally were 80 g kg-1 of total N or less, this fraction is indigestible and unavailable and composes a significant portion of a nutrient that may …

Background
Parenting interventions and conditional cash transfer (CCT) programmes are promising strategies to reduce the risk of violence against children, but evidence of the effectiveness of combining such programmes is lacking for families in low- and middle-income countries with children over two years of age. This study examined the effectiveness of a locally adapted parenting programme delivered as part of a government CCT system to low-income families with children aged two to six years in Metro Manila, Philippines.

It is common knowledge nowadays that consuming sugar-sweetened beverages (“SSBs”) is associated with obesity, diabetes, and other poor health outcomes. 40% of the American public consumes 300-800 calories a day from soda. Unsurprisingly, over 40% of the American public is obese, with roughly 30% falling into the overweight category. Governments have sought to combat this issue by levying a tax on sugar-sweetened beverages to raise their price and deter their consumption. A trial was conducted on five psammophyte shrubs -Caragana intermedia, C. korshinskii, Hedysarum scoparium, H. laeve, Salix psammophylla in the post-fruit period in Ningxia, China. Chemical composition, palatability and in vitro digestibility were studied systematically. The feeding value was evaluated through Fuzzy multifactorial model. According to chemical composition, palatability and in vitro digestibility, C. intermedia, had the highest feeding value and C .korshinskii the lowest.

The objective of this study was to evaluate dry matter ruminal degradation kinetics, of prairie grass (Bromus catharticus Vahl.) and annual ryegrass (Lolium multiflorum), by using in sacco technique, on seven different harvesting dates. The forage was incubated in nylon bags, suspended in the rumen of four Hereford steers with a rumen fistula. The dry matter ruminal degradation was calculated through the exponential model P=a+b(1-exp(-ct)). The article presents the results of studying the sum of concentrations of cholecalciferol isoforms (vit. D3) + ergocalciferol (vit. D2) in the blood serum of Bunyodkor FC players with different morphofunctional phenotypes by seasons of the year. Epidemiological studies of the status of vitamin 25 (OH) D in the blood serum of 53 football players were carried out taking into account the maximum total solar radiation (Qmonth, kilowatt hours /m2) in the climatic conditions of Tashkent city. A significant prevalence of Vitamin D deficiency in summer, autumn and winter and deficiency in spring was established among young football players.
