Mutations in GBA which are causative of Gaucher disease in their biallelic form, are the most common genetic risk factor for Parkinson's disease (PD). The diagnosis of PD relies upon clinically defined motor features which appear after irreversible neurodegeneration. Prodromal symptoms of PD may provide a means to predict latent pathology, years before the onset of motor features. Previous work has reported prodromal features of PD in GBA mutation carriers, however this has been insufficiently sensitive to identify those that will develop PD. The Remote Assessment of Parkinsonism Supporting Ongoing Development of Interventions in Gaucher Disease (RAPSODI GD) study assesses …

Galleria Mellonella Larvae Exhibit A Weight-Dependent Lethal Median Dose When Infected With Methicillin-Resistant Staphylococcus Aureus, Poppy J. Hesketh-Best, Michelle V. Mouritzen, Kayleigh Shandley-Edwards, Richard A. Billington, Mathew Upton

ABSTRACT Galleria mellonella is a recognised model to study antimicrobial efficacy; however, standardisation across the scientific field and investigations of methodological components are needed. Here, we investigate the impact of weight on mortality following infection with Methicillin-resistant Staphylococcus aureus (MRSA). Larvae were separated into six weight groups (180–300 mg at 20 mg intervals) and infected with a range of doses of MRSA to determine the 50% lethal dose (LD50), and the ‘lipid weight’ of larvae post-infection was quantified. A model of LD50 values correlated with weight was developed. The LD50 values, as estimated by our model, were further tested in …

Measuring Individual And Group Flow In Collaborative Improvisational Dance, Klara Łucznik, Jon May
Measuring Individual And Group Flow In Collaborative Improvisational Dance, Klara Łucznik, Jon May
School of Psychology
This study investigated flow experience during group collaborations of creative dance improvisation, where group flow was defined as periods when most members of a group reported a flow experience. Sixteen dancers took part in the experimental sessions, performing improvisational tasks in groups of four. We chose two different types of dance task, hypothesising that dancers’ external focus upon the group's shared surroundings and awareness of others might facilitate group flow experience; while an internal focus upon one's own mental imagery might inhibit it. A novel, video-stimulated recall method to assess flow experience was used to track time patterns and shared …
Good Enough For Governance? Audit And Marine Biodiversity Offsetting In Australia, Holly J. Niner, Samuel Randalls
Good Enough For Governance? Audit And Marine Biodiversity Offsetting In Australia, Holly J. Niner, Samuel Randalls
School of Biological and Marine Sciences
Biodiversity offsetting is often presented as a way to manage competing demands of environmental protection and economic development. It is premised on the transparent demonstration of how aims of no net loss of biodiversity (NNL) or similar are met in practice. This is complicated in marine systems where ecological predictions are commonly highly uncertain, knowledge of ecological restoration is low and administrative governance is complex. Drawing on a case study of marine biodiversity offsetting in Australia, this paper shows how these uncertainties pose practical challenges for both producers and consumers of marine biodiversity offsets, needing to progress with decision-making while …

A Comparison Of The Quality Of Image Acquisition Between Two Different Sidestream Dark Field Video-Microscopes, Jonny Coppel, Vassiliki Bountziouka, Daniel Martin, Edward Gilbert-Kawai
Peninsula Medical School
AbstractSidestream dark field (SDF) imaging enables direct visualisation of the microvasculature from which quantification of key variables is possible. The new MicroScan USB3 (MS-U) video-microscope is a hand-held SDF device that has undergone significant technical upgrades from its predecessor, the MicroScan Analogue (MS-A). The MS-U claims superior quality of sublingual microcirculatory image acquisition over the MS-A, however, this has yet to be robustly confirmed. In this manuscript, we therefore compare the quality of image acquisition between these two devices. The microcirculation of healthy volunteers was visualised to generate thirty video images for each device. Two independent raters, blinded to the …

BackgroundCurrent NHS policy encourages an integrated approach to provision of mental and physical care for individuals with long term mental health problems. The ‘PARTNERS2’ complex intervention is designed to support individuals with psychosis in a primary care setting.AimThe trial will evaluate the clinical and cost-effectiveness of the PARTNERS2 intervention.Design & settingThis is a cluster randomised controlled superiority trial comparing collaborative care (PARTNERS2) with usual care, with an internal pilot to assess feasibility. The setting will be primary care within four trial recruitment areas: Birmingham & Solihull, Cornwall, Plymouth, and Somerset. GP practices are randomised 1:1 to either (a) the PARTNERS2 …

Background Despite oxygen being the commonest drug administered to critically ill patients we do not know which oxygen saturation (SpO2) target results in optimal survival outcomes in those receiving mechanical ventilation. We therefore conducted a feasibility randomised controlled trial in the United Kingdom (UK) to assess whether it would be possible to host a larger national multi-centre trial to evaluate oxygenation targets in mechanically ventilated patients. Methods We set out to recruit 60 participants across two sites into a trial in which they were randomised to receive conservative oxygenation (SpO2 88–92%) or usual care (control – SpO2 ≥96%). The primary …

Judgemental rationality is infrequently referenced within discussions of Realist Evaluations. Judgemental rationality refers to researchers’ capacity to assess which, potential, meanings provide the most credible explanations. In evaluation work, rationale for analysis is provided, though rarely do we see how an evaluator made judgements between competing theories, and which theories were discarded and why. We provide a worked example of the application of judgemental rationality. The Engager intervention offered support to prison leavers with common mental health problems. The data for 24, purposively sampled, participants from the intervention arm of the trial were integrated. Bhaskar’s DREIC, a five-step analytical procedure, …

Abstract Background Research into the effects of asthma treatments on the extra-pulmonary symptoms of severe asthma is limited by the absence of a suitable questionnaire. The aim was to create a questionnaire suitable for intervention studies by selecting symptoms that are statistically associated with asthma pathology and therefore may improve when pathology is reduced. Methods Patients attending a specialist asthma clinic completed the 65-item General Symptom Questionnaire (GSQ-65), a questionnaire validated for assessing symptoms of people with multiple medically unexplained symptoms. Lung function (FEV1%) and cumulative oral corticosteroids (OCS) calculated from maintenance dose plus exacerbations were obtained from clinic records. …

Homocysteine (Hcy) is a breakdown product of methionine metabolism. The risk of cardiovascular disease (CVD) correlates with an increase in plasma Hcy levels. The aim of this study was to investigate whether 1% methionine supplementation of adult rats altered intracellular reactive oxygen species (ROS) generation, intracellular Ca2+ content, and contractile activity in freshly isolated cardiomyocytes. This was measured under normal conditions and during oxidative stress in freshly isolated cardiomyocytes. Single rat cardiomyocytes from both sexes were isolated by enzymatic and mechanical dispersion techniques. Fluorescence microscopy was used to measure ROS production and intracellular Ca2+ concentration. Cell contraction was measured using …
