Trypanosome Lytic Factor (TLF) is a primate-specific high-density lipoprotein (HDL) complex that, through the cation channel-forming protein apolipoprotein L-1 (APOL1), provides innate immunity to select kinetoplastid parasites. The immunoprotective effects of TLF have been extensively investigated in the context of its interaction with the extracellular protozoan Trypanosoma brucei brucei, to which it confers sterile immunity. We previously showed that TLF could act against an intracellular pathogen Leishmania, and here we dissected the role of TLF and its synergy with host-immune cells. Leishmania major is transmitted by Phlebotomine sand flies, which deposit the parasite intradermally into mammalian hosts, where neutrophils are …

Hypoxia-induced pulmonary hypertension (PH) is one of the most common and deadliest forms of PH. Fibroblast growth factor receptors 1 and 2 (FGFR1/2) are elevated in patients with PH and in mice exposed to chronic hypoxia. Endothelial FGFR1/2 signaling is important for the adaptive response to several injury types and we hypothesized that endothelial FGFR1/2 signaling would protect against hypoxia-induced PH. Mice lacking endothelial FGFR1/2, mice with activated endothelial FGFR signaling, and human pulmonary artery endothelial cells (HPAECs) were challenged with hypoxia. We assessed the effect of FGFR activation and inhibition on right ventricular pressure, vascular remodeling, and endothelial-mesenchymal transition …

PURPOSE: Treatment of advanced melanoma is a clinical challenge. Natural killer (NK) cells are a promising cellular therapy for T cell-refractory cancers, but are frequently deficient or dysfunctional in patients with melanoma. Thus, new strategies are needed to enhance NK-cell antitumor responses. Cytokine-induced memory-like (ML) differentiation overcomes many barriers in the NK-cell therapeutics field, resulting in potent cytotoxicity and enhanced cytokine production against blood cancer targets. However, the preclinical activity of ML NK against solid tumors remains largely undefined.

EXPERIMENTAL DESIGN: Phenotypic and functional alterations of blood and advanced melanoma infiltrating NK cells were evaluated using mass cytometry. ML NK …

Rethinking Immigration Enforcement, Peter L. Markowitz Sep 2021

Rethinking Immigration Enforcement, Peter L. Markowitz

Articles

As the nation turns the page away from the dark chapter of President Trump's relentless assault on immigrants, it is time to take stock of the nation's unprecedented immigration enforcement regime. During its relatively short existence, the Immigration and Customs Enforcement agency (ICE) has deported more than twice as many people as were deported in the entire previous history of the United States. The human and fiscal costs of ICE's mass deportation agenda are astronomical, and there is almost universal agreement across the political spectrum that the United States' immigration enforcement apparatus is badly broken. That, however, is where the …

Poly(methyl methacrylate) (PMMA) is used to manage bone loss in revision total knee arthroplasty (rTKA). However, the application of PMMA has been associated with complications such as volumetric shrinkage, necrosis, wear debris, and loosening. Glass polyalkenoate cements (GPCs) have potential bone cementation applications. Unlike PMMA, GPC does not undergo volumetric shrinkage, adheres chemically to bone, and does not undergo an exothermic setting reaction. In this study, two different compositions of GPCs (GPCA and GPCB), based on the patented glass system SiO2-CaO-SrO-P2O5-Ta2O5, were investigated. Working and setting times, pH, ion release, …

The Theory Of Multiple Intelligences In A Special Education Music Classroom, Daniel Passadino Sep 2021

The Theory Of Multiple Intelligences In A Special Education Music Classroom, Daniel Passadino

Masters Theses

Special education students involved in music classes currently face a lack of inclusivity. Various methods have been used to promote inclusivity in the music classroom. This paper focuses on how Howard Gardner’s multiple intelligence theory promotes inclusivity in the special education music classroom. The multiple intelligence theory streamlines a special education music curriculum by highlighting each child’s strongest intelligence. To assess a child’s intelligences, they must participate in music-related tests. The tests include mini lessons which cater to the multiple intelligences. For example, when children are taught using visuals, some will not be able to learn as effectively if they …

Eviction: A Story Of Displacement, Kaila O. Eisenkraft

Massachusetts Office of Public Collaboration Publications

About one-third of the US household population lives in rented housing. Among the housing disputes that occur between tenants and landlords, the removal of tenants from their property – that is, their displacement – by landlords through evictions poses one of the greatest threats to housing stability. The prevalence and high stakes of eviction elevate the importance of examining this phenomenon and the efficacy of various interventions, such as financial assistance, legal representation, and mediation.

Although most causes of death and morbidity in premature infants are related to immune maladaptation, the premature immune system remains poorly understood. We provide a comprehensive single-cell depiction of the neonatal immune system at birth across the spectrum of viable gestational age (GA), ranging from 25 weeks to term. A mass cytometry immunoassay interrogated all major immune cell subsets, including signaling activity and responsiveness to stimulation. An elastic net model described the relationship between GA and immunome (R=0.85, p=8.75e-14), and unsupervised clustering highlighted previously unrecognized GA-dependent immune dynamics, including decreasing basal MAP-kinase/NFkB signaling in antigen presenting cells; increasing responsiveness of …

Psychosis, a break in reality which is manifested as hallucinations, delusions or the disruption in thought process, is the hallmark of schizophrenia. Despite novel pharmacotherapy advancements of antipsychotic medications that have resulted in some patients having the ability to return to social settings and thereby decreasing psychotic symptoms and reducing hospital admissions, there is still a sub-population of patients who remain symptomatic. Treatment-resistant schizophrenia is defined as failure of treatment with at least two different antipsychotics with the proper length of treatment and titration. Clozapine has been heralded as a drug to resolve the puzzle of treatment-resistant schizophrenia. Clozapine has …

Exploiting Group Structures To Infer Social Interactions From Videos, Maksim Bolonkin Sep 2021

Exploiting Group Structures To Infer Social Interactions From Videos, Maksim Bolonkin

Dartmouth College Ph.D Dissertations

In this thesis, we consider the task of inferring the social interactions between humans by analyzing multi-modal data. Specifically, we attempt to solve some of the problems in interaction analysis, such as long-term deception detection, political deception detection, and impression prediction. In this work, we emphasize the importance of using knowledge about the group structure of the analyzed interactions. Previous works on the matter mostly neglected this aspect and analyzed a single subject at a time. Using the new Resistance dataset, collected by our collaborators, we approach the problem of long-term deception detection by designing a class of histogram-based features …

A Religious Double Standard: Post-9/11 Challenges To Muslims’ Religious Land Usage, Asma T. Uddin Sep 2021

A Religious Double Standard: Post-9/11 Challenges To Muslims’ Religious Land Usage, Asma T. Uddin

Michigan Journal of Race and Law

Muslims in the United States face real limits on their religious freedom. Numerous influential individuals and organizations even posit that Islam is not a religion and that, therefore, Muslims do not have rights to religious freedom. The claim is that Islam is a political ideology that is intent on taking over the country and subverting Americans’ constitutional rights. This narrative has gained momentum since the attacks of September 11, 2001 and continues to be amplified and disseminated by a well-funded cadre of anti-Muslim agitators. One area where its effects can be seen clearly is in religious land use, where a …

Lipid-Nanoparticle-Encapsulated Mrna Vaccines Induce Protective Memory Cd8 T Cells Against A Lethal Viral Infection., Cory J Knudson, Pedro Alves-Peixoto, Hiromi Muramatsu, Colby Stotesbury, Lingjuan Tang, Paulo J C Lin, Ying K Tam, Drew Weissman, Norbert Pardi, Luis J. Sigal

Department of Microbiology and Immunology Faculty Papers

It is well established that memory CD8 T cells protect susceptible strains of mice from mousepox, a lethal viral disease caused by ectromelia virus (ECTV), the murine counterpart to human variola virus. While mRNA vaccines induce protective antibody (Ab) responses, it is unknown whether they also induce protective memory CD8 T cells. We now show that immunization with different doses of unmodified or N(1)-methylpseudouridine-modified mRNA (modified mRNA) in lipid nanoparticles (LNP) encoding the ECTV gene EVM158 induced similarly strong CD8 T cell responses to the epitope TSYKFESV, albeit unmodified mRNA-LNP had adverse effects at the inoculation site. A single immunization …

Conventional cross-sectional imaging done shortly after radioembolization of hepatocellular carcinoma (HCC) does not reliably predict long-term response to treatment. This study evaluated whether quantitative contrast-enhanced ultrasound (CEUS) can predict the long-term response of HCC to yttrium-90 (Y-90) treatment. Fifteen patients underwent CEUS at three time points: immediately following treatment and 1 and 2 wk post-treatment. Response 3-6 mo after treatment was categorized on contrast-enhanced magnetic resonance imaging by two experienced radiologists using the Modified Response Evaluation Criteria in Solid Tumors. CEUS data were analyzed by quantifying tumor perfusion and residual fractional vascularity using time-intensity curves. Patients with stable disease on …
