Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has killed more than 4 million humans globally, but there is no bona fide Food and Drug Administration–approved drug-like molecule to impede the COVID-19 pandemic. The sluggish pace of traditional therapeutic discovery is poorly suited to producing targeted treatments against rapidly evolving viruses. Here, we used an affinity-based screen of 4 billion DNA-encoded molecules en masse to identify a potent class of virus-specific inhibitors of the SARS-CoV-2 main protease (Mpro) without extensive and time-consuming medicinal chemistry. CDD-1714, the initial three-building-block screening hit (molecular weight [MW] = 542.5 g/mol), was a potent inhibitor (inhibition …

Posttraumatic fibrotic scarring is a significant medical problem that alters the proper functioning of injured tissues. Current methods to reduce posttraumatic fibrosis rely on antiinflammatory and anti-proliferative agents with broad intracellular targets. As a result, their use is not fully effective and may cause unwanted side effects. Our group previously demonstrated that extracellular collagen fibrillogenesis is a valid and specific target to reduce collagen- rich scar buildup. Our previous studies showed that a rationally designed antibody that binds the C-terminal telopeptide of the α2(I) chain involved in the aggregation of collagen molecules limits fibril assembly in vitro and reduces scar …

Background Black men and women are at higher risk for, and suffer greater morbidity and mortality from, atherosclerotic cardiovascular disease (ASCVD) compared with adults of European Ancestry (EA). Black patients with familial hypercholesterolemia are at particularly high risk for ASCVD complications because of lifelong exposure to elevated levels of low-density-lipoprotein cholesterol. Methods and Results This retrospective study analyzed ASCVD prevalence and risk factors in 808 adults with heterozygous familial hypercholesterolemia from 5 US-based lipid clinics, and compared findings in Black versus EA patients. Multivariate logistic regression models were used to determine the strongest predictors of ASCVD as a function of …

Bark beetles vector symbiotic fungi and the success of these mutualisms may be limited by competition from other microbes. The outcome of fungal competition is strongly influenced by the physical and chemical conditions of the wood they inhabit. These conditions are in turn subject to climatic variation. In particular, wood moisture content (MC) influences fungal competition and, therefore, could help determine environmental suitability for thousand cankers disease (TCD) caused by Geosmithia morbida and its vector Pityophthorus juglandis. We conducted competition experiments in Juglans nigra wood that was naturally or artificially colonized by G. morbida and other fungi over a range …

Transactions of the Nebraska Academy of Sciences and Affiliated Societies
Soapweed Yucca (Yucca glauca) is a conspicuous and common shrub in the Great Plains of North America, characterized by tall woody flower stalks, large flowers and seed pods, and dense masses of ground-level evergreen leaves. These plant structures can provide a variety of resources or functions to animals. In general, studies focus on single species associated with Y. glauca. We examined three groups of vertebrates that interacted with Y. glauca and the functions this plant provided for organisms in western Nebraska. We experimentally examined small mammals in areas with and without Y. glauca, and we descriptively …
Tracing The Roots Of Disjunction : Dutertismo And The Discourse Of Liberal Democracy In The Philippines, Kristine Marie Tayo Reynaldo
Tracing The Roots Of Disjunction : Dutertismo And The Discourse Of Liberal Democracy In The Philippines, Kristine Marie Tayo Reynaldo
Lingnan Theses (MPhil & PhD)
This dissertation examines Dutertismo as an event in the context of what it put forwards as the problem-spaces of Philippine postcolonial modernity: nationalism, democracy, and development. Specifically, it investigates the substance of the contemporary disjunction between the discourse of liberal democracy, which often frames critiques of Dutertismo, and Philippine social experience. Its objective is to examine the underlying causes of the democratic "crisis" that Dutertismo represents, both material/structural and discursive/ideological.
“Dutertismo,” a populist political movement and discourse, is characterized by its subversion of the inherited norms and mores of Philippine society, and consolidation of popular support for a charismatic leader …

Becoming A Place Of Unrest : Environmental Crisis And Ecophenomenological Praxis, Robert Booth
Ohio University Press Open Access Books
The key to mitigating the environmental crisis isn’t just based on science; it depends upon a profound philosophical revision of how we think about and behave in relation to the world.
Our ongoing failure to interrupt the environmental crisis in a meaningful way stems, in part, from how we perceive the environment—what Robert Booth calls the "more-than-human world.” Anthropocentric presumptions of this world, inherited from natural science, have led us to better scientific knowledge about environmental problems and more science-based—yet inadequate—practical “solutions.” That’s not enough, Booth argues. Rather, he asserts that we must critically and self-reflexively revise how we perceive …

Aberrant activation of the nucleotide-binding domain and leucine-rich repeat related (NLR) family, pyrin domain containing 3 (NLRP3) inflammasome drives the development of many complex inflammatory diseases, such as obesity, Alzheimer’s disease, and atherosclerosis. However, no medications specifically targeting the NLRP3 inflammasome have become clinically available. Therefore, we aim to identify new inhibitors of the NLRP3 inflammasome in this study. Methods: Vesicle-like nanoparticles (VLNs) were extracted from garlic chives and other Allium vegetables and their effects on the NLRP3 inflammasome were evaluated in primary macrophages. After garlic chive-derived VLNs (GC-VLNs) were found to exhibit potent anti-NLRP3 inflammasome activity in cell culture, …

The potential for Blue Carbon ecosystems to combat climate change and provide co-benefits was discussed in the recent and influential Intergovernmental Panel on Climate Change Special Report on the Ocean and Cryosphere in a Changing Climate. In terms of Blue Carbon, the report mainly focused on coastal wetlands and did not address the socio-economic considerations of using natural ocean systems to reduce the risks of climate disruption. In this paper, we discuss Blue Carbon resources in coastal, open-ocean and deep-sea ecosystems and highlight the benefits of measures such as restoration and creation as well as conservation and protection in helping …

Towards A Biblical Foundation For A Philosophy Of Library Sociality, Terry Dwain Robertson
Journal of Adventist Libraries and Archives
In this essay, I focus on the sociality of librarianship in the academic setting. How should we as library workers think about the communities we serve? What mindset should guide our interactions with the people who seek our services? How should library workers think about how we collaborate to serve our public? As a profession, librarianship has a solid public service orientation, evidenced by its best community outreach, advocacy, and hospitality practices. Can Biblical teachings emphasized within the Seventh-day Adventist faith community be foundational for this ethos? Does faith integration matter? Correlating insights from Tuomela on group agency, Lankes on …

The Onset Of Exercise-Associated Hyponatremia And Individual Differences In Inappropriate Arginine Vasopressin Excretion: A Review Of Proposed Mechanisms, Michelle Stehman, Stephen A. Maris
Topics in Exercise Science and Kinesiology
Topics in Exercise Science and Kinesiology Volume 2: Issue 1, Article 10, 2021. Exercise-associated hyponatremia (EAH) has been reported to develop during endurance events such as triathlons and marathons. As these events become more popular, the incidence of developing EAH also increases. The development of EAH is commonly associated with the overconsumption of hypotonic fluids such as water and tends to be more prevalent in females. There is also evidence to suggest the inappropriate secretion of arginine vasopressin (AVP) leading to water retention may predispose an individual for developing EAH, especially when coupled with the overconsumption of fluids. Recent research …

Resistance to anthracnose, dry matter yield (DMY) and seed yield (SY) was assessed for germplasm and breeding lines of Stylosanthes in Brazil, India and China. Overall, Stylosanthes guianensis produced higher DMY than S. scabra, S. capitata and S. macrocephala at most sites in Brazil. Data from China suggest that there are high yielding anthracnose resistant S. guianensis lines that can reduce the reliance on CIAT 184. S. seabrana might also prove successful. S. seabrana in India produced the highest DMY and SY and it can form nodules with native Bradyrhizobium strains. Regional differences in resistance within accessions stress the …

As part of the recent electrification of the transportation industry, internal combustion engines are being coupled with or replaced by electric motors. This movement towards an electrified drivetrain poses new noise, vibration, and harshness (NVH) challenges related to electric motors. In this paper, the acoustic signature of an electric motor was analyzed to obtain a better understanding of the sound generated by these motors. This work provides an insight into an acoustic measurement technique that can be used to identify certain frequency bands that significantly contribute to the perceived sound. In the first part, the structural response of the motor …

BACKGROUND: Cerebral glucose hypometabolism is consistently observed in individuals with Alzheimer's disease (AD), as well as in young cognitively normal carriers of the Ε4 allele of Apolipoprotein E (APOE), the strongest genetic predictor of late-onset AD. While this clinical feature has been described for over two decades, the mechanism underlying these changes in cerebral glucose metabolism remains a critical knowledge gap in the field.
METHODS: Here, we undertook a multi-omic approach by combining single-cell RNA sequencing (scRNAseq) and stable isotope resolved metabolomics (SIRM) to define a metabolic rewiring across astrocytes, brain tissue, mice, and human subjects expressing APOE4.
RESULTS: Single-cell …

The vascular endothelium is present within metabolic organs and actively regulates energy metabolism. Here we show osteocalcin, recognized as a bone-secreted metabolic hormone, is expressed in mouse primary endothelial cells isolated from heart, lung and liver. In human osteocalcin promoter-driven green fluorescent protein transgenic mice, green fluorescent protein signals are enriched in endothelial cells lining aorta, small vessels and capillaries and abundant in aorta, skeletal muscle and eye of adult mice. The depletion of lipoprotein receptor-related protein 1 induces osteocalcin through a Forkhead box O -dependent pathway in endothelial cells. Whereas depletion of osteocalcin abolishes the glucose-lowering effect of low-density …

The mammalian DNA methylome is formed by two antagonizing processes, methylation by DNA methyltransferases (DNMT) and demethylation by ten-eleven translocation (TET) dioxygenases. Although the dynamics of either methylation or demethylation have been intensively studied in the past decade, the direct effects of their interaction on gene expression remain elusive. Here, we quantify the concurrence of DNA methylation and demethylation by the percentage of unmethylated CpGs within a partially methylated read from bisulfite sequencing. After verifying 'methylation concurrence' by its strong association with the co-localization of DNMT and TET enzymes, we observe that methylation concurrence is strongly correlated with gene expression. …

Military installations are valuable in global biodiversity conservation as they secure representative ecosystems from land conversion and protect many threatened or endangered species. Selecting suitable areas for biodiversity conservation within military installations is a challenging problem as this must not impede military training activities. The issue gets more complicated when considering multiple cohabiting species in a metacommunity with species dependency. In this paper, we present an example for the conservation of two cohabiting species, Gopher Tortoise (GT) and Gopher Frog (GF), located within the boundaries of a military installation, Fort Stewart, Georgia, United States. The GF depends on both locations …

Disrupted systemic copper (Cu) homeostasis underlies neurodegenerative diseases with early symptoms including olfactory dysfunction. This study investigated the impact of Cu dyshomeostasis on olfactory function, adult neurogenesis, and neurochemical balance. Models of Cu deficiency (CuD) and Cu overload (CuO) were established by feeding adult rats with Cu-restricted diets plus ip. injection of a Cu chelator (ammonium tetrathiomolybdate) and excess Cu, respectively. CuD reduced Cu levels in the olfactory bulb (OB), subventricular zone (SVZ), rostral migratory stream (RMS), and striatum, while CuO increased Cu levels in these areas. The buried pellet test revealed both CuD and CuO prolonged the latency to …

Background
Environmental fluctuation during embryonic and fetal development can permanently alter an organism’s morphology, physiology, and behaviour. This phenomenon, known as developmental plasticity, is particularly relevant to reptiles that develop in subterranean nests with variable oxygen tensions. Previous work has shown hypoxia permanently alters the cardiovascular system of snapping turtles and may improve cardiac anoxia tolerance later in life. The mechanisms driving this process are unknown but may involve epigenetic regulation of gene expression via DNA methylation. This work aimed to determine the genetic diversity of Colletotrichum gloeosporioides infecting Stylosanthes spp. in Brazil, China and India. A total of 132 isolate originating from S. seabrana, S. macrocephala, S. capitata, S. scabra, and S. guianensis were used. Four major genetic groups were identified from an analysis of genetic diversity using selection-neutral DNA markers. Group 1 contained 20 isolates and this may represent a genotype that migrated from the center of diversity in Brazil and Colombia to Australia, Thailand and India. Group 2 consisted of 66 Brazilian isolates and group 3 had 19 isolates from …
