Characterization Of Ovarian Tissue Culture And Investigation Of Embryonic Response To Temperature Stress Using A Shp2 Phosphatase Inhibitor In Ciona Intestinalis, Rose Jacobson

This dissertation is comprised of two parts, the first is cell culture. In this work I describe the successful primary culture of Ciona intestinalis ovary tissue. Improved cell proliferation in 3D Petri Dish® system over culture treated plastic is shown, as well as the efficacy of using C. intestinalis hemolymph as a media additive. The seeded ovarian cells assemble into multicellular aggregations, which mimic the morphology of in vivo ovaries. It is also important to understand the molecular pathways in which marine organisms handle stress and how these pathways change in high stress situations.

The preoccupation with reproduction in Victorian literature has, by and large, received little critical attention. This dissertation seeks to contribute to Victorian scholarship and examines the representations of reproduction in Victorian literature in relation to the contemporaneous desire for progress. Adopting a historiographical approach, this dissertation analyzes five Victorian novels: Charlotte Brontë’s Jane Eyre, George Eliot’s Middlemarch, George Du Maurier’s Trilby, Thomas Hardy’s Jude the Obscure, and Sarah Grand’s The Heavenly Twins. This dissertation is an extended prescription for the inclusion of social justice in high school biology (HSB) curricula. Chapter 1 is context: a review of social injustice (mainly racism) in the US, and the specious scientific justifications for it. Chapter 2 explores how three veteran teachers introduce social justice in HSB classes. A case-study, semi-constructed interview-based approach is used. Chapter 3, the culmination of these ideas, adduces a practical curriculum module that provides the theory, lessons and activities for HSB student exploration the inferior science used to sustain racial inequities. Winter flounder (Pseudopleuronectes americanus) has historically supported productive commercial and recreational fisheries throughout its range in the northwest Atlantic. As a small flatfish that inhabits estuaries, bays, and the coastal zone, this species is vulnerable to shifting environmental conditions in many of the habitats critical to its life cycle. At the southern edge of the species range, the Southern New England/Mid-Atlantic Bight winter flounder stock entered a steep decline during the 1980s following a period of overfishing. New active pharmaceutical ingredients (APIs) continue to be discovered from the natural world. These compounds can be further developed into a range of useful products such as drugs, supplements, or cosmeceuticals. To determine the biological target of the API, the new compounds are evaluated for their biological activity through a combination of in silico, in vitro, and in vivo analyses. Convalescent plasma has been used worldwide to treat patients hospitalized with coronavirus disease 2019 (COVID-19) and prevent disease progression. Despite global usage, uncertainty remains regarding plasma efficacy, as randomized controlled trials (RCTs) have provided divergent evidence regarding the survival benefit of convalescent plasma. Here, we argue that during a global health emergency, the mosaic of evidence originating from multiple levels of the epistemic hierarchy should inform contemporary policy and healthcare decisions. Indeed, worldwide matched-control studies have generally found convalescent plasma to improve COVID-19 patient survival, and RCTs have demonstrated a survival benefit when transfused early in the disease course but …

Advocates of decarcation often focus their critiques on imprisonment for non-violent offences. In this vein, current advocacy efforts to end mandatory sentences in Canada tend to carve out “serious violent offences” as not part of a reform agenda. In this chapter, Debra Parkes sketches out the contours of an argument for why feminists might not want to cede that ground, why anti-carceral feminism might involve centering our analysis on the most, rather than the least, serious crimes – starting with those who are serving life sentences for murder. The institution of property arises in the tension between autonomy and community. It serves not simply to demarcate spaces of individual control and authority, but also to balance individual with collective interests. Private property and common property emphasize individual and collective interests, respectively, but the bifurcation may not be as stark as it appears. Condominium constructs separate titles to individual units, and these private interests are carefully mapped in a constituting plan that marks their boundaries. Objective:

Anterior segment ischemia (ASI) is a serious complication of strabismus surgery and other ocular procedures. The purpose of this study is to evaluate variation of the long posterior artery (LPCA). These variations may predispose patients to anterior segment ischemia following strabismus surgery, and proper mapping may provide surgical “danger zones” for other invasive ocular procedures.

Subjects:

A total of 100 preserved cadaveric eyeballs from University of Nebraska Medical Center (UNMC), Creighton University, and Kansas City University (KCU) were utilized for this study.

Four million square kilometers of seabed within the sovereignty of Pacific Island nations are currently under contract for mineral exploration or exploitation. Over a million additional square kilometers of the non-sovereign seafloor are licensed for such use. Historically, these licenses have served to establish “squatters’ rights” in anticipation of a distant future when the industry would develop the machinery to exploit oceanic mineral wealth. That moment has arrived, with the first seafloor mining machines rolling off production lines in 2015-2016. Judicial independence is a fixture of American government, but its structure has never been fully understood. As long as the federal judiciary has survived episodic attacks with its independence intact, there has been no pressing need to know how or why. But a confluence of cyclical, sustained, and sudden developments now threatens the federal judiciary’s autonomy in arguably unprecedented ways and demands a more comprehensive analysis of judicial independence and its vulnerabilities. Employment discrimination laws make the “simple but momentous” declaration that it is illegal to deny employment on the basis of race, sex, religion, or other key aspects of identity. But when employees who have been treated unfairly turn to the courts for relief, courts rarely assess whether their claims meet the statutory standard. Instead, they funnel the evidence through a convoluted body of judge-made law known as McDonnell Douglas burden shifting.

In 1999, Professor Stephen Burbank wrote an article entitled The Architecture of Judicial Independence. It is a foundational piece that gave structure to what was then an understudied field. At the heart of that article is a profound insight: stable and enduring judicial systems are the product of forces in constructive tension. This Essay turns attention from actuarial risk assessment tools as a reform to the inclination for a technical sentencing reform more broadly. When situated in the context of technical guidelines created to structure and regulate judicial discretion in the 1980s and beyond, the institutionalization of an actuarial risk assessment at sentencing is both an old and new idea. This article examines four areas in which the SEC, for more than a decade, resisted reform or impeded shareholders’ access to sought-after environmental, social, and governance (ESG) information. These areas are: (1) the SEC’s refusal to act on several rulemaking petitions submitted during the years 2009 to 2018, which called for expanded ESG disclosure; (2) the SEC’s grudging promulgation of rules concerning social disclosures as required by Congress in the Dodd-Frank Act of 2010; (3) the SEC’s 2020 revisions to SEC Rule 14a-8, which make the submission of shareholder proposals more difficult, thereby thwarting investor efforts to raise ESG concerns; …

Sepsis is a disorder that targets the microcirculation with significant effects at blood-tissue barriers such as the blood-brain barrier (BBB) and gut-vascular barrier (GVB). Intestinal barrier dysfunction is thought to be one of the most important contributors to multi-organ dysfunction in sepsis. As a common site of infection, the impaired gut allows the dissemination of bacteria, which exacerbates the disease pathophysiology, and can lead to long-term morbidity and mortality. The hemlock woolly adelgid (HWA), Adelges tsugae Annand (Hemiptera: Adelgidae), is an invasive insect that is causing mortality of eastern hemlock trees, Tsuga canadensis, and Carolina hemlock trees, Tsuga caroliniana, across the eastern United States. To protect these ecologically important tree species, a neonicotinoid insecticide, imidacloprid, is commonly used. Spatial reasoning is required for many topics in undergraduate mathematics as well as other STEM fields. In calculus specifically, there are few interventions that assess students’ spatial skills and provide remediation. Haptic technology is a novel approach to spatial skills remediation due to its unfamiliarity to students and its flexibility with regard to models. Imidacloprid is the most widely used insecticide in the world and has been found to impact non-target taxa in systems in which it is applied. It is used as the primary treatment method in the protection of eastern hemlocks (Tsuga canadensis) from impacts of hemlock woolly adelgid (HWA; Adelges tsugae), an invasive insect in the U.S. that is causing widespread mortality of hemlock trees. In this study, we investigated whether imidacloprid applied in eastern hemlock forests is impacting woodland salamander and ant communities.

Fast Radio Bursts (or FRBs) are millisecond-duration transients of extragalactic origin. They exhibit dispersion caused by propagation through an ionized medium, and quantified by Dispersion Measure (DM). Around 800 FRBs (24 repeaters) have been discovered; so far, 24 FRBs have been confidently associated with a host galaxy. In this thesis, we discuss multiple new FRB search and analysis techniques and the corresponding tools that enable us to search for FRBs harder, localize them better, and classify candidates faster.

Non-native invasive insect pests can have dramatic impacts on native ecosystems, feeding on plant foliage, wood, or sap. Little is known, however, about how fruit-targeting NNIIPs may affect native ecosystems. Spotted wing Drosophila (Drosophila suzukii Matsumura, SWD) is a recently introduced invasive vinegar fly that parasitizes the fruits of many plant species in the United States. While its activity in agricultural systems is well-documented, little is known about its activity in forest ecosystems, despite growing evidence of its presence and parasitism of fruits there. Background: Bilingual speech production studies have highlighted that level of proficiency influences the acoustic-phonetic representation of phonemes in both languages (MacKay, Flege, Piske, & Schirru 2001; Zárate-Sández, 2015). The results for bilingual speech production reveal that proficient/early bilinguals produce distinct acoustic properties for the same phoneme in each language, whereas less proficient/late bilinguals produce acoustic properties for a phoneme that is closer to the native language (Flege et al., 2003; Fowler et al., 2008). This dissertation consists of three essays on energy and agricultural commodity price analysis: 1) Natural Gas Price Forecasting in a Changing World; 2) The Effect of EIA Storage Announcement on Natural Gas Returns: A Comprehensive Analysis; and 3) Forecasting the U.S. Season-Average Farm Price of Corn: Derivation of an Alternative Futures based Forecasting Model.

The first essay evaluates the performances of various individual and composite forecasting models when predicting natural gas prices in the United States.
