Multiple myeloma (MM) is a debilitating hematologic malignancy of terminally differentiated plasma cells in the bone marrow (BM). Daratumumab (DARA) is an FDA-approved high-affinity monoclonal antibody targeting CD38 that has shown promising therapeutic efficacy in relapsed and refractory MM. Despite the well-established clinical efficacy of DARA, not all heavily pretreated patients respond to DARA monotherapy, and the majority of patients who initially respond eventually progress. This may be due to upregulation of pathways that inhibit DARA-mediated tumor killing. Antibody drug conjugates (ADCs) combine the highly targeted tumor antigen recognition of antibodies with cell killing properties of chemotherapy for effective internalization …

Calcium signaling is essential for regulating many biological processes. Endoplasmic reticulum inositol trisphosphate receptors (IP3Rs) and the mitochondrial Ca2+ uniporter (MCU) are key proteins that regulate intracellular Ca2+ concentration. Mitochondrial Ca2+ accumulation activates Ca2+-sensitive dehydrogenases of the tricarboxylic acid (TCA) cycle that maintain the biosynthetic and bioenergetic needs of both normal and cancer cells. However, the interplay between calcium signaling and metabolism is not well understood. Mosquitoes are known to be one of the deadliest creatures in the world. There have been several studies that aim to identify mosquito presence and species using various techniques. The most common ones involve automatic identification of mosquito species from the sounds produced by flapping its wings. The development of these important concepts and technologies can help reduce the spread of mosquito-borne diseases. This paper presents a simple model based on mean and interquartile values that aim to solve the mosquito classification. Lipopolysaccharides (LPSs) cause lethal endotoxemia if not rapidly cleared from blood circulation. Liver sinusoidal endothelial cells (LSEC) systemically clear LPS by unknown mechanisms. We discovered that LPS clearance through LSEC involves endocytosis and lysosomal inactivation via Stabilin-1 and 2 (Stab1 and Stab2) but does not involve TLR4. Cytokine production was inversely related to clearance/endocytosis of LPS by LSEC. When exposed to LPS, Stabilin double knockout mice (Stab DK) and Stab1 KO, but not Stab2 KO, showed significantly enhanced systemic inflammatory cytokine production and early death compared with WT mice. Despite extensive research and aggressive therapies, glioblastoma (GBM) remains a central nervous system malignancy with poor prognosis. The varied histopathology of GBM suggests a landscape of differing microenvironments and clonal expansions, which may influence metabolism, driving tumor progression. Indeed, GBM metabolic plasticity in response to differing nutrient supply within these microenvironments has emerged as a key driver of aggressiveness. Additionally, emergent biophysical and biochemical interactions in the tumor microenvironment (TME) are offering new perspectives on GBM metabolism. Perivascular and hypoxic niches exert crucial roles in tumor maintenance and progression, facilitating metabolic relationships between stromal and tumor cells. Alterations in extracellular …

In 2019, a large-scale fire occurred in the Amazon rainforest, causing serious damage to the ecosystem and to humans as well. Abstract. The release of legacy contaminants such as fallout radionuclides (FRNs) in response to glacier retreat is a process that has received relatively little attention to date, yet may have consequences as a source of secondary contamination as glaciers melt and down-waste in response to a warming climate. The presence of FRNs in glacier-fed catchments is poorly understood in comparison to other contaminants, yet there is now emerging evidence from multiple regions of the global cryosphere for substantially augmented FRN activities in cryoconite. Enhancing Student Learning Of Global Warming Through Reflective Writing, Liang Zeng, Guang Zeng

The National Academy of Science has published studies showing strong scientific evidence that global warming is caused by human consumption of fossil fuels, yet recent surveys have shown young adults in the U.S. are disengaged or disagree with this fact. Accordingly, the United Nations Educational, Scientific, and Cultural Organization (UNESCO) published learning objectives to educate the world population on global warming and renewable energy by 2030. In this paper, we introduce a reflective writing activity physics educators can employ to foster a deeper understanding of global warming in introductory college physics and physical science courses, without overloading their teaching time.

Background:
Worldwide, coffee is one of the most popular beverages consumed. Several studies have suggested a protective role of coffee, including reduced risk of Alzheimer’s disease (AD). However, there is limited longitudinal data from cohorts of older adults reporting associations of coffee intake with cognitive decline, in distinct domains, and investigating the neuropathological mechanisms underpinning any such associations.

This paper will focus on a historical perspective on cool season forage production, plant breeding methods for cool season forages, major cool season forage selection criteria, some examples of significant achievements, and a future perspective. Topics similar to ours have been discussed at recent previous meeting of this Congress (Humphreys, 1997; Van Wijk et al., 1993); however, we will strive to avoid “plowing the same ground twice”. Pasture and forage plant breeding is complicated by the perennial nature of the plants, the diversity of environments in which improved cultivars will be used, and the complex criteria of merit involved, criteria that necessarily include some measure of impact on the efficiency of animal production. While pasture plant breeding in the temperate zone is a demonstrably productive activity, the record of success for the tropical species -- "success" measured by release and adoption of bred cultivars -- is less convincing, in spite of four decades of activity in numerous public sector breeding programs and a large published literature. After very active years of pasture and forage research at major institutes, interest in tropical forage genetic resources has drastically declined. Apparently, the early phases of collecting and evaluation were much more valued than conserving and keeping the germplasm available for future generations. Accumulated data are not easily accessible and, therefore, the knowledge of tropical forage genetic resources is progressively being lost. This worldwide decrease in activity and loss of knowledge is due to declining resources. Yield adaptability and stability of thirty five alfalfa genotypes (Medicago sativa L.) were evaluated at Sertãozinho, SP, Brazil, considering different environments (seasons of year) from 1996 to 1999. A randomized complete block design with three replications was used. Scott-Knott’s test was performed to compare genotypes averages. Adaptability and stability were determined by Eberhart and Russell's procedures. Two diverse groups of genotypes with mean dry matter yield were identified: Araucana, F686, MH4 e BR2 were more responsive under favorable environments, but SW 8210, Alto, Monarca SP, Victoria SP, Florida 77, P5888, MH15, BR1, BR3, SW 9210A presented broad adaptability and …

Topics in Exercise Science and Kinesiology Volume 2: Issue 1, Article 12, 2021. Validity refers to the ability of a device to measure what it was intended to measure. Therefore, purpose of this study was to assess the validity and reliability of a novel vertical jump height tool designed by Brower Timing Systems (Salt Lake City, Ut). The Brower vertical jump system was compared to the Vertec jump tester. A convenience sample (n=67) of college students performed three maximum countermovement jumps, with the average score being recorded. Data was collected simultaneously for both devices. The project began in 2004 when Alex Jospe ’06, a Nordic skier who traveled Maine roads to meets, decided to use skills learned in a GIS class taught by Associate Professor of Environmental Studies Philip Nyhus. Jospe used data supplied by state transportation officials to map moose-collision hot zones. On a trip to Vermont, the map came in handy. “She came back all excited and said, ‘I saw a moose right where my map said I would,’” Nyhus recalled.

Acute ischemic stroke (AIS) is a common devastating disease that has increased yearly in absolute number of cases since 1990. While mechanical thrombectomy and tissue plasminogen activator (tPA) have proven to be effective treatments, their window-of-efficacy time is very short, leaving many patients with no viable treatment option. Over recent years there has been a growing interest in stimulating the facial nerves or ganglions to treat AIS. Pre-clinical studies have consistently demonstrated an increase in collateral blood flow (CBF) following ganglion stimulation, with positive indications in infarct size and neurological scores. (Non)Parallel Developmental Mechanisms In Vertebrate Appendage Reduction And Loss, Samantha Swank, Thomas Sanger, Yoel E. Stuart

Appendages have been reduced or lost hundreds of times during vertebrate evolution. This phenotypic convergence may be underlain by shared or different molecular mechanisms in distantly related vertebrate clades. To investigate, we reviewed the developmental and evolutionary literature of appendage reduction and loss in more than a dozen vertebrate genera from fish to mammals. We found that appendage reduction and loss was nearly always driven by modified gene expression as opposed to changes in coding sequences. This paper uses the author’s experiences of teaching the Filipino module of a multidisciplinary video game development class as a case study in teaching Filipino culture and identity as an element of video game development. Accurately measuring the brain concentration of a neurotherapeutic is critical in determining its pharmacokinetic profile in vivo. Biologics are potential therapeutics for neurologic diseases and biologics fused to an antibody targeting a transcytosis receptor at the Blood-Brain Barrier, designated as antibody-biologic fusion proteins, are Blood-Brain Barrier penetrating neurotherapeutics. The use of sandwich immunosorbent assays to measure concentrations of antibody-biologic fusion proteins in brain homogenates has become increasingly popular. The raw brain homogenate contains many proteins and other macromolecules that can cause a matrix effect, potentially interfering with the limit of detection of such assays and reduce the overall sample …
