Systemic lupus erythematosus (SLE) is a chronic, multisystem, autoimmune inflammatory disease with genomic and non-genomic contributions to risk. We hypothesize that epigenetic factors are a significant contributor to SLE risk and may be informative for identifying pathogenic mechanisms and therapeutic targets. To test this hypothesis while controlling for genetic background, we performed an epigenome-wide analysis of DNA methylation in genomic DNA from whole blood in three pairs of female monozygotic (MZ) twins of European ancestry, discordant for SLE. Results were replicated on the same array in four cell types from a set of four Danish female MZ twin pairs discordant …

Rapid And Direct Action Of Lipopolysaccharide (Lps) On Skeletal Muscle Of Larval Drosophila, Rachel Potter, Alexis Meade, Samuel Potter, Robin L. Cooper
Biology Faculty Publications
The endotoxin lipopolysaccharide (LPS) from Gram-negative bacteria exerts a direct and rapid effect on tissues. While most attention is given to the downstream actions of the immune system in response to LPS, this study focuses on the direct actions of LPS on skeletal muscle in Drosophila melanogaster. It was noted in earlier studies that the membrane potential rapidly hyperpolarizes in a dose-dependent manner with exposure to LPS from Pseudomonas aeruginosa and Serratia marcescens. The response is transitory while exposed to LPS, and the effect does not appear to be due to calcium-activated potassium channels, activated nitric oxide synthase …

The SARS-CoV-2 pandemic, and the likelihood of future coronavirus pandemics, emphasized the urgent need for development of novel antivirals. Small-molecule chemical probes offer both to reveal aspects of virus replication and to serve as leads for antiviral therapeutic development. Here, we report on the identification of amiloride-based small molecules that potently inhibit OC43 and SARS-CoV-2 replication through targeting of conserved structured elements within the viral 5′-end. Nuclear magnetic resonance–based structural studies revealed specific amiloride interactions with stem loops containing bulge like structures and were predicted to be strongly bound by the lead amilorides in retrospective docking studies. Amilorides represent the …

Autosomal dominant polycystic kidney disease (PKD) is a hereditary disorder affecting multiple organs, including the heart. PKD has been associated with many cardiac abnormalities including the arrhythmogenic remodeling in clinical evaluations. In our current study, we hypothesized that Pkd2 gene mutation results in structural and functional defects in the myocardium. The structural and functional changes of Pkd2 mutant hearts were analyzed in the myocardial-specific Pkd2 knockout (KO) mouse. We further assessed a potential role of TGF-b1 signaling in the pathology of Pkd2-KO hearts. Hearts from age-matched 6-month-old MyH6•Pkd2wt/wt (control or wild-type) and MyH6 …

Neuromusculoskeletal (NMS) models can aid in studying the impacts of the nervous and musculoskeletal systems on one another. These computational models facilitate studies investigating mechanisms and treatment of musculoskeletal and neurodegenerative conditions. In this study, we present a predictive NMS model that uses an embedded neural architecture within a finite element (FE) framework to simulate muscle activation. A previously developed neuromuscular model of a motor neuron was embedded into a simple FE musculoskeletal model. Input stimulation profiles from literature were simulated in the FE NMS model to verify effective integration of the software platforms. Motor unit recruitment and rate coding …

Examining The Role Of Buzzing Time And Acoustics On Pollen Extraction Of Solanum Elaeagnifolium, Mandeep Tayal, Rupesh R. Kariyat
School of Integrative Biological & Chemical Sciences (Formerly Dept. of Biology)
Buzz pollination is a specialized pollination syndrome that requires vibrational energy to extract concealed pollen grains from poricidal anthers. Although a large body of work has examined the ecology of buzz pollination, whether acoustic properties of buzz pollinators affect pollen extraction is less understood, especially in weeds and invasive species. We examined the pollination biology of Silverleaf nightshade (Solanum elaeagnifolium), a worldwide invasive weed, in its native range in the Lower Rio Grande Valley (LRGV) in south Texas. Over two years, we documented the floral visitors on S. elaeagnifolium, their acoustic parameters (buzzing amplitude, frequency, and duration of buzzing) and …

Nature-based solutions are widely advocated for freshwater ecosystem conservation and restoration. As increasing amounts of river restoration are undertaken, the need to understand the ecological response to different measures and where measures are best applied becomes more pressing. It is essential that appraisal methods follow a sound scientific approach. Here, experienced restoration appraisal experts review current best practice and academic knowledge to make recommendations and provide guidance that will enable practitioners to gather and analyse meaningful data, using scientific rigor to appraise restoration success. What should be monitored depends on the river type and the type and scale of intervention. …

IGC Proceedings (1977-2023)
In northeastern Patagonia where grain crops often led to harvest failure, low yields and a high drought risk, a lamb production system was established. On a 10 years old agriculture wheat cropping was interrupted in 1996, allowing re-vegetation through exclusion of grazing. Natural grassland recovered, prevailing the species Stipa tenuis, Piptochaetium napostaense, Stipa ambigua. Part of the plot was sown to wheat grass (Thinopyrum ponticum). In 1999, to improve sheep feeding, oat (Avena sativa) and barley (Hordeum vulgare) were sown for grazing during winter-spring season. In this system Corriedale sheep, Texel x australian …

The molecular mechanisms that drive hematopoietic stem cell functional decline under conditions of telomere shortening are not completely understood. In light of recent advances in single-cell technologies, we sought to redefine the transcriptional and epigenetic landscape of mouse and human hematopoietic stem cells under telomere attrition, as induced by pathogenic germline variants in telomerase complex genes. Here, we show that telomere attrition maintains hematopoietic stem cells under persistent metabolic activation and differentiation towards the megakaryocytic lineage through the cell-intrinsic upregulation of the innate immune signaling response, which directly compromises hematopoietic stem cells’ self-renewal capabilities and eventually leads to their exhaustion. …

Lu Xun’S Imagery And Its Visual Representations, Naiwu Zhang
Theoretical Studies in Literature and Art
Lu Xun’s imagery refers to the visual construction of Lu Xun. Different images of Lu Xun are constructed via visual presentations such as biography, animation and spectacle. The process of shaping Lu Xun’s images is marked by heterogeneity, and particularly by the escape of images as the most prominent feature in their dissemination. Although Lu Xun’s image and Lu Xun himself are closely related, the transition from the literary world to the verbal-pictorial world forms a unique context of image as well as image modeling. “Serenity” and “spectacle” are the two faces of Janus of the image modeling of Lu …

Brown and beige adipocytes are specialized to dissipate energy as heat. Sgk2, encoding a serine/threonine kinase, has been identified as a brown and beige adipocyte-specific gene in rodents and humans; however, its function in brown/beige adipocytes remains unraveled. Here, we examined the regulation and role of Sgk2 in brown/beige adipose tissue thermogenesis. We found that transcriptional coactivators PGC-1α and NT-PGC-1α activated by the β3 adrenergic receptor-cAMP-PKA pathway are recruited to the Sgk2 promoter, triggering Sgk2 transcription in response to cold. SGK2 elevation was closely associated with increased serine/threonine phosphorylation of proteins carrying the consensus RxRxxS/T phosphorylation site. However, despite cold-dependent …

(1) Background: Intracortical microelectrodes (IMEs) are essential to basic brain research and clinical brain–machine interfacing applications. However, the foreign body response to IMEs results in chronic inflammation and an increase in levels of reactive oxygen and nitrogen species (ROS/RNS). The current study builds on our previous work, by testing a new delivery method of a promising antioxidant as a means of extending intracortical microelectrodes performance. While resveratrol has shown efficacy in improving tissue response, chronic delivery has proven difficult because of its low solubility in water and low bioavailability due to extensive first pass metabolism. (2) Methods: Investigation of an …

During ovarian follicular development, granulosa cells proliferate and progressively differentiate to support oocyte maturation and ovulation. To determine the underlying links between proliferation and differentiation in granulosa cells, we determined changes in 1) the expression of genes regulating DNA methylation and 2) DNA methylation patterns, histone acetylation levels and genomic DNA structure. In response to equine chorionic gonadotropin (eCG), granulosa cell proliferation increased, DNA methyltransferase (DNMT1) significantly decreased and Tet methylcytosine dioxygenase 2 (TET2) significantly increased in S-phase granulosa cells. Comprehensive MeDIP-seq analyses documented that eCG treatment decreased methylation of promoter regions in approximately 40% of the genes in granulosa …

Nutritive Quality Of Tropical Grasses Sown Under A Pine Plantation, D. Gutmanis, Antonio João Lourenço, V. B. G. Alcantara, Maria T. Colozza
IGC Proceedings (1977-2023)
A trial was conducted in Brazil to evaluate the nutritive quality of 6 tropical grasses: tanzania (Panicum maximum cv. Tanzania), green-panic (P. maximum var. Trichoglume), aruana (P. maximum cv. Aruana), brizanta (Brachiaria brizantha), humidicola (B. humidicola) and tifton-85 (Cynodon dactylon cv. Tifton 85), planted under two different densities of pines (Pinus elliottis): 200 and 400 stems/ha, as well as in full sunlight. The results showed a significant increase in protein content and a significant decrease in NDF and cellulose contents as tree density increased. Comparing the values obtained under shading conditions …

A trial was conducted in Brazil to evaluate the performance of 6 tropical grasses: tanzania (Panicum maximum cv. Tanzania), green-panic (P. maximum var. Trichoglume), aruana (P. maximum cv. Aruana), brizanta (Brachiaria brizantha), humidicola (B. humidicola) and tifton-85 (Cynodon dactylon cv. Tifton 85), planted under two different densities of pines (Pinus elliottis): 200 and 400 stems/ha, as well as in the full sunlight. The results showed that the dry matter yield decreased as shading increased. The grasses tanzania and brizanta were the most tolerant to shading. Although N concentration increased with …

Two Centuries On: The Iconic ‘Mermaid’ Boab Tree At Careening Bay In The Kimberley, Kevin Kenneally
Talking Heads Seminar Series
In September 1820, Lieutenant Phillip Parker King’s hydrographic survey vessel His Majesty’s Cutter Mermaid was leaking badly and in danger of sinking. King careened his cutter in a sheltered bay on the north-west Kimberley coast, now known as Careening Bay. During their stay, the ship’s carpenter inscribed ‘HMC MERMAID 1820’ on a large boab tree behind the beach. Two hundred years later, it is difficult to visualise the courage, skill and ingenuity of the Mermaid’s officers and crew in their unassisted maritime survey work on remote parts of the Australian coast. Likewise, the hardships they endured. But the importance of …
