Spring/Summer 2020: A Norbertine At Yale, Mike Dauplaise Jul 2020

Spring/Summer 2020: A Norbertine At Yale, Mike Dauplaise

St. Norbert College Magazine 2019-2023

The Rev. Matthew Dougherty ’09 arrived at St. Norbert College on a path he confesses was headed more toward atheism than the priesthood. That all began to change after late-night discussions with some fellow Madelaine/Lorraine Hall residents that covered questions as deep as whether there were a God, and what would that mean for the living? Dougherty was ordained a priest in 2015 and is now a Ph.D. candidate at Yale University in the school’s department of ecology and evolutionary biology. He spoke about his journey with Mike Dauplaise ’86.

Developing A Virus-Like Particle (Vlp)-Based Vaccine For Neisseria Gonorrhoeae, Ashvini Vaidya Jul 2020

Developing A Virus-Like Particle (Vlp)-Based Vaccine For Neisseria Gonorrhoeae, Ashvini Vaidya

Biomedical Sciences ETDs

Neisseria gonorrhoeae (Ng) is a sexually transmitted bacterium that infects human reproductive mucosal surfaces. Due to the risk of complications in untreated cases and evolving antibiotic resistance, a prophylactic vaccine is critical for preventing infection. There are four Ng outer-membrane proteins that contribute to Ng survival and resistance: PorB, BamA, TbpA, and MtrE. Each protein contains surface-exposed loops that are targets of bactericidal antibodies. Vaccines were developed by displaying Ng epitopes on the surface of bacteriophage virus-like particles (VLP). VLPs are immunogenic, multivalent, self-assembling complexes composed of viral structural proteins. Antigenic peptide sequences can be displayed on the surface …

Ornate Box Turtles (Terrapene Ornata) Copulating In Water: An Incidental Observation Or Ancestral Behavior, Ashley Forrester, Michael Rohde, Mary Harner, Carter Kruse, Keith Geluso

Transactions of the Nebraska Academy of Sciences and Affiliated Societies

Herein, we report the first observation of the terrestrial Ornate Box Turtle (Terrapene ornata) copulating in water. To understand whether such an event reflected an isolated incident, we compiled observations reported in the literature of copulatory behaviors in water for other North American turtles. Our literature review revealed that other species of box turtles in North America, the Coahuilan Box Turtle (Terrapene coahuila) and Gulf Coast Box Turtle (Terrapene carolina major), occasionally also copulate in water. The vast majority of turtles in North America copulate in water, as most are semi-aquatic, including all species …

The Personification Of The Perfect Citizen: The English Political Cartoon, Colonial Anxiety, And Identity During The American Revolution, Sarah Johns Jul 2020

The Personification Of The Perfect Citizen: The English Political Cartoon, Colonial Anxiety, And Identity During The American Revolution, Sarah Johns

History Summer Fellows

When studying the American Revolution, there is a variety of written source materials from the actors involved that have been used to decipher the many social and political changes that occurred throughout the conflict; however, imagery, especially political cartoons, can be key to uncovering avenues of cultural debate that highlight these changes in new and more detailed ways. With Great Britain experiencing its golden age of political caricature during the late 18th century, what might these images have to say about gender and race during this tumultuous period? In this project, I argue that British political cartoons were essential …

Exploration Of Novel Catalytic Methods: Merging Nitroxides And Visible-Light Energy To Perform Oxidative Functionalization Reactions, Jyoti Nandi Jul 2020

Exploration Of Novel Catalytic Methods: Merging Nitroxides And Visible-Light Energy To Perform Oxidative Functionalization Reactions, Jyoti Nandi

Doctoral Dissertations

With the growing demand for green and sustainable chemistry, it is imperative to develop novel catalytic methods that involve mild conditions and less toxic reagents. Described herein, is a new paradigm for oxidation and oxidative functionalization methodologies by harvesting visible-light energy. It consists of a dual catalytic system of an environmentally friendly oxidant, 4-acetoamido-2,2,6,6-tetramethylpiperidine N-oxyl (also known as ACT) with photoredox catalysis. The integration of these two forms of catalysis has been realized for the oxidative amidation of aldehydes, furnishing N-acylated heterocycles. Extension of this process to the oxidative amidation of alcohols via the intermediacy of an aldehyde …

Microrna-29 Family: A Positive Regulator Of Osteoclastogenesis, Henry Hrdlicka Jul 2020

Microrna-29 Family: A Positive Regulator Of Osteoclastogenesis, Henry Hrdlicka

Doctoral Dissertations

Osteoclasts are large multinucleated bone cells of the myeloid lineage. Osteoclasts secrete proteases and create an acidic environment, causing bone resorption. Osteoclast differentiation, driven by MCSF and RANKL, is tightly regulated by numerous factors, including microRNAs (miRNAs, miRs). miRNAs are short non-coding RNAs that post-transcriptionally regulate gene expression by binding to complementary mRNA sequences, leading to mRNA degradation or destabilization. The aim of these studies was to investigate the function of the miRNA-29-3p family in osteoclasts both in vivo and in vitro.

We generated mice which globally express a miR-29-3p tough decoy which competitively inhibits miR-29-3p activity. The examination …

Respiratory syncytial virus (RSV) infection can cause severe bronchiolitis in infants requiring hospitalization, whereas the elderly and immunocompromised are prone to RSV-induced pneumonia. RSV primarily infects lung epithelial cells. Given that no vaccine against RSV is currently available, we tested the ability of the epithelial-barrier protective cytokine interleukin-22 (IL-22) to control RSV production. When used in a therapeutic modality, IL-22 efficiently blunted RSV production from infected human airway and alveolar epithelial cells and IL-22 administration drastically reduced virus titer in the lungs of infected newborn mice. RSV infection resulted in increased expression of LC3B, a key component of the cellular …

Militarizing Marriage : West African Soldiers’ Conjugal Traditions In Modern French Empire, Sarah J. Zimmerman Jul 2020

Militarizing Marriage : West African Soldiers’ Conjugal Traditions In Modern French Empire, Sarah J. Zimmerman

Ohio University Press Open Access Books

Following tirailleurs sénégalais’ deployments in West Africa, Congo, Madagascar, North Africa, Syria-Lebanon, Vietnam, and Algeria from the 1880s to 1962, Militarizing Marriage historicizes how African servicemen advanced conjugal strategies with women at home and abroad. Sarah J. Zimmerman examines the evolution of women’s conjugal relationships with West African colonial soldiers to show how the sexuality, gender, and exploitation of women were fundamental to the violent colonial expansion and the everyday operation of colonial rule in modern French Empire.

These conjugal behaviors became military marital traditions that normalized the intimate manifestation of colonial power in social reproduction across the empire. Soldiers’ …

School of Medicine Faculty Research

Schizophrenia is a complex disorder with many comorbid conditions. In this study, we used polygenic risk scores (PRSs) from schizophrenia and comorbid traits to explore consistent cluster structure in schizophrenia patients. With 10 comorbid traits, we found a stable 4-cluster structure in two datasets (MGS and SSCCS). When the same traits and parameters were applied for the patients in a clinical trial of antipsychotics, the CATIE study, a 5-cluster structure was observed. One of the 4 clusters found in the MGS and SSCCS was further split into two clusters in CATIE, while the other 3 clusters remained unchanged. For the …

Type 1 cGMP-dependent protein kinases (PKGs) play important roles in human cardiovascular physiology, regulating vascular tone and smooth-muscle cell phenotype. A mutation in the human PRKG1 gene encoding cGMP-dependent protein kinase 1 (PKG1) leads to thoracic aortic aneurysms and dissections. The mutation causes an arginine-to-glutamine (RQ) substitution within the first cGMP-binding pocket in PKG1. This substitution disrupts cGMP binding to the pocket, but it also unexpectedly causes PKG1 to have high activity in the absence of cGMP via an unknown mechanism. Here, we identified the molecular mechanism whereby the RQ mutation increases basal kinase activity in the human PKG1α and …

Effects Of Freshwater Crayfish On Influenza A Virus Persistence In Water, J. Jeffrey Root, Jeremy W. Ellis, Susan A. Shriner

United States Department of Agriculture Wildlife Services: Staff Publications

Several investigations have recently assessed the ability of some aquatic invertebrates to act as tools for avian influenza A virus (IAV) surveillance as well as their potential role(s) in IAV ecology. Because of this, as well as the high IAV seroprevalence rates noted in select mesocarnivores that commonly inhabit aquatic and semi-aquatic habitats, we evaluated the effects that freshwater crayfish have on IAV in water at three dose levels and monitored for the presence of IAV in crayfish tissues (gill and green gland) and haemolymph at multiple time points. At relatively high, medium 432 and low (approximately 10 , 10 …

Geological Factors Impacted Cadmium Availability And Use As An Alternative Cofactor For Zinc In The Carbon Fixation Pathways Of Marine Diatoms, Naman Srivastava, Stephanie J. Spielman, Shaunna M. Morrison, Eli K. Moore

School of Earth & Environment Departmental Research

Transition metal cofactors are crucial for many biological processes. Despite being primarily considered to be toxic, the transition metal cadmium (Cd) was discovered to be a substitute for zinc (Zn) in photosynthetic carbon fixation pathways in marine diatoms. However, it is not known how conditions in the geosphere impacted Cd availability and its incorporation as an alternative metal cofactor for phytoplankton. We employed mineral chemistry network analysis to investigate which geochemical factors may have influenced the availability of Cd and Zn during the putative time period that alternative Cd-based pathway evolved. Our results show that Zn minerals are more chemically …

© 2020, The Author(s), under exclusive licence to Springer Nature Limited. The recent discovery of N6-methyladenine (N6-mA) in mammalian genomes suggests that it may serve as an epigenetic regulatory mechanism1. However, the biological role of N6-mA and the molecular pathways that exert its function remain unclear. Here we show that N6-mA has a key role in changing the epigenetic landscape during cell fate transitions in early development. We found that N6-mA is upregulated during the development of mouse trophoblast stem cells, specifically at regions of stress-induced DNA double helix destabilization (SIDD)2–4. Regions of SIDD are conducive to topological stress-induced unpairing …

"A Comparison Of Variable Selection Methods Using Bootstrap Samples From Environmental Metal Mixture Data", Paul-Yvann Djamen

Mathematics & Statistics ETDs

In this thesis, I studied a newly developed variable selection method SODA, and three customarily used variable selection methods: LASSO, Elastic net, and Random forest for environmental mixture data. The motivating datasets have neuro-developmental status as responses and metal measurements and demographic variables as covariates. The challenges for variable selections include (1) many measured metal concentrations are highly correlated, (2) there are many possible ways of modeling interactions among the metals, (3) the relationships between the outcomes and explanatory variables are possibly nonlinear, (4) the signal to noise ratio in the real data may be low. To compare these methods …

Background: Although native to North America, the invasion of the aphid-like grape phylloxera Daktulosphaira vitifoliae across the globe altered the course of grape cultivation. For the past 150 years, viticulture relied on grafting-resistant North American Vitis species as rootstocks, thereby limiting genetic stocks tolerant to other stressors such as pathogens and climate change. Limited understanding of the insect genetics resulted in successive outbreaks across the globe when rootstocks failed. Here we report the 294-Mb genome of D. vitifoliae as a basic tool to understand host plant manipulation, nutritional endosymbiosis, and enhance global viticulture. Results: Using a combination of genome, RNA, …
