Analysis Of The Functional Relationship Of Protein Kinase Families Using Phospho-Proteomics Data, David A. Parra Peña

Theses and Dissertations

As cancer research advances, Mass-spectrometry based proteomics is becoming a widely used technique for proteome characterization. Phosphoproteomics is a specific type of proteomics that characterizes proteins with the reversible post-translational modification of phosphorylation PTM), which has allowed the identifications of thousands of phosphorylation sites. These phosphorylation sites, also known as substrates, are known to interact with a protein type named kinases. Studies have shown that abnormal phosphorylation activity is related to cancer diseases. Moreover, these kinases are divided into families, based on the similarity of their catalytic domain, as this part of their amino acid sequence determines a large part …

Caspase-3 and -7 are executioner caspases whose enzymatic activity is necessary to complete apoptotic cell death. Here, we questioned whether endothelial cell infection leads to caspase-3/7-mediated cell death. Pulmonary microvascular endothelial cells (PMVECs) were infected with Pseudomonas aeruginosa (PA103). PA103 caused cell swelling with a granular appearance, paralleled by intracellular caspase-3/7 activation and cell death. In contrast, PMVEC infection with ExoY+ (PA103 ΔexoUexoT::Tc pUCPexoY) caused cell rounding, but it did not activate intracellular caspase-3/7 and it did not cause cell death. However, ExoY+ led to a time-dependent accumulation of active caspase-7, but not caspase-3, …

Here, we report the generation of a Cre-recombinase (iCre) transgenic rat, where iCre is driven using a vascular endothelial-cadherin (CDH5) promoter. The CDH5 promoter was cloned from rat pulmonary microvascular endothelial cells and demonstrated ~60% similarity to the murine counterpart. The cloned rat promoter was 2,508 bp, it extended 79 bp beyond the transcription start site, and it was 22,923 bp upstream of the translation start site. The novel promoter was cloned upstream of codon-optimized iCre and subcloned into a Sleeping Beauty transposon vector for transpositional transgenesis in Sprague-Dawley rats. Transgenic founders were generated and selected for iCre expression. Crossing …

Tumor-Derived Exosomes Drive Immunosuppressive Macrophages In A Pre-Metastatic Niche Through Nf-Kβ Dependent Glycolytic Metabolic Reprogramming., Samantha M. Morrissey

Electronic Theses and Dissertations

The formation of a pre-metastatic niche is a fundamental requirement for primary tumor metastasis. One of the defining characteristics of a pre-metastatic niche is infiltration of immunosuppressive macrophages. However, how these macrophages acquire their immunosuppressive phenotype remains largely unexplored. Here, we demonstrate that tumor-derived exosomes (TDE) polarize macrophages towards an immunosuppressive phenotype characterized by increased PD-L1 expression through NF-kB-dependent metabolic reprogramming in mice and humans. While NF-κB has previously been shown to act as a direct transcription factor for PD-L1, we report a novel mechanism where TDE-induced NF-κB activation drives PD-L1 expression by augmenting the glycolytic capacity of macrophages through …

Roles Of Endothelial Cell Heat Shock Protein A12b And Β-Glucan, A Reagent For Trained Immunity In The Regulation Of Inflammation In Sepsis, Fei Tu

Electronic Theses and Dissertations

Sepsis is dysregulated host immune response to infection causing life-threatening organ dysfunction. Endothelial cell dysfunction and uncontrolled inflammatory responses are two contributors for sepsis-induced mortality. The crosstalk between endothelial and immune cells plays a critical role in the pathophysiology of sepsis. Therefore, understanding the mechanism of interaction between endothelial and immune cells will provide novel information to develop therapeutic strategies for sepsis.

Pathogen associated moleculear patterns (PAMPs) and/or damage associated molecular patterns (DAMPs) produced during sepsis, activate endothelial cells to increase the expression of adhesion molecules, attracting immune cell infiltration into the tissues. Uncontrolled inflammatory responses during the early phase …

More recently, many wetlands are being restored in an attempt to regain their ecosystem service. This study seeks to determine the effects of restored wetlands on local bat habitat use. Bat activity was found to be significantly higher around the wetlands when compared to distant grassy fields; however, no significant difference was found among the restored wetlands and a remote cattle farm containing multiple water features. Geospatial models of bat distribution and bat foraging were produced using machine learning that showed higher …

Axonal Regrowth Of Olfactory Sensory Neurons After Chemical Ablation And Removal Of Axonal Debris By Microglia, Rudy Chapman Aug 2020

Axonal Regrowth Of Olfactory Sensory Neurons After Chemical Ablation And Removal Of Axonal Debris By Microglia, Rudy Chapman

Electronic Theses and Dissertations

Olfactory sensory neurons (OSNs) are contained within the olfactory epithelium (OE) and are responsible for detecting odorant molecules in the air. The exposure of OSNs to the external environment is necessary for their function, but it also leaves them exposed to potentially harmful elements and thus results in a high turnover rate. Despite the high turnover, the olfactory sense is maintained throughout life through the division of a population of stem cells that produce new OSNs both during normal turnover and after an injury occurs in the OE. When new OSNs are born, they must extend axons from the OE …

Effect Of Electrolyte Ph On The Electrosynthesis Of Hydrogen Peroxide On Carbon Black-Based Gas Diffusion Electrodes, Spencer Lindsay

All Theses

The electrochemical synthesis of H2O2 by the cathodic reduction of O2 is an on-site alternative to the current industrial scale method of production. Carbon-based gas diffusion electrodes (GDEs) are selective to H2O2 synthesis and inexpensive compared to their precious metal counterparts. The research presented here examines the role electrolyte pH in the cathode has on the efficiency of H2O2 electrosynthesis, particularly as it pertains to the rate of H2O2 production on the electrode and subsequent degradation in the cathode chamber. From these results, the optimization of the cathode surface and environmental conditions is then considered.

The overall performance was dependent …

Investigating Rad51 And Dmc1 In Entamoeba Histolytica: Homologous Recombinases With Drug Target Potential, Rachel Evelyn Ham

All Theses

Entamoeba histolytica is the causative agent of amebic dysentery and is prevalent in developing countries. It has a biphasic lifecycle: active, virulent trophozoites and dormant, environmentally-stable cysts. Only cysts are capable of establishing new infections, and are spread by fecal deposition. Many unknown factors influence stage conversion, and synchronous encystation of E. histolytica is not currently possible in vitro. E. histolytica infections are treated with nitroimidazole drugs, such as metronidazole (Flagyl™). However, several clinical isolates have shown metronidazole resistance. Enhancing the amebicidal mechanisms of metronidazole through drug combination therapy may allow for more effective treatment. Metronidazole may cause amebic death …

Taxonomy Of Echinostoma Revolutum And 37-Collar-Spined Echinostoma Spp.: A Historical Review, Jong-Yil Chai, Jaeeun Cho, Taehee Chang, Bong-Kwang Jung, Woon-Mok Sohn

Harold W. Manter Laboratory of Parasitology: Library Materials

Echinostoma flukes armed with 37 collar spines on their head collar are called as 37-collar-spined Echinostoma spp. (group) or ‘Echinostoma revolutum group’. At least 56 nominal species have been described in this group. However, many of them were morphologically close to and difficult to distinguish from the other, thus synonymized with the others. However, some of the synonymies were disagreed by other researchers, and taxonomic debates have been continued. Fortunately, recent development of molecular techniques, in particular, sequencing of the mitochondrial (nad1 and cox1) and nuclear genes (ITS region; ITS1-5.8S-ITS2), has enabled us to obtain highly …
