Testing Flowering Perennial Plants In A Bermudagrass (Cynodon Spp.) Lawn, Paige Elizabeth Boyle, Michelle Wisdom
Testing Flowering Perennial Plants In A Bermudagrass (Cynodon Spp.) Lawn, Paige Elizabeth Boyle, Michelle Wisdom
Horticulture Faculty Publications and Presentations
Most pollinating insects require a season-long succession of floral resources to fulfill life-cycle requirements. Incorporating forbs into turfgrass sites may create a season-long sequence of flowers to support foraging pollinators. However, persistence of forbs in warm-season turfgrasses such as bermudagrass (Cynodon spp.) may be affected by the competitive nature of the turfgrass and routine management practices such as mowing. A 2-year study was conducted to evaluate seven forbs (Bellis perennis L., Lotus corniculatus L., Prunella vulgaris L., Trifolium fragiferum L. ‘Fresa’, Trifolium repens L. ‘Durana’ and ‘Resolute’, Trifolium subterraneum L.) for persistence and ability to produce floral resources …

Calcium (Ca) signaling modulates sodium (Na) transport in plants; however, the role of the Ca sensor calmodulin (CaM) in salt tolerance is elusive. We previously identified a salt-responsive calmodulin (HvCaM1) in a proteome study of barley (Hordeum vulgare) roots. Here, we employed bioinformatic, physiological, molecular, and biochemical approaches to determine the role of HvCaM1 in barley salt tolerance. CaM1s are highly conserved in green plants and probably originated from ancestors of green algae of the Chlamydomonadales order. HvCaM1 was mainly expressed in roots and was significantly up-regulated in response to long-term salt stress. Localization analyses revealed that HvCaM1 is an …

Method Development For Identifying Counterfeit Cigarettes Using Environmental Pollen Analysis, Tiffany Chan
Student Theses
Cigarette counterfeiting negatively impacts government revenue and undermines tobacco control policies. Tracking geographical origins and potential trade routes of counterfeit cigarettes is important for investigation illegal trade in tobacco products. Traditional pollen extraction techniques provide clear residues of pollen trapped on tobacco leaves and used for geographical identification; however, such methods are time-consuming and utilize concentrated chemicals. This study developed a simple, fast and environmentally friendly method for extraction of pollen grains used in differentiating genuine and counterfeit cigarettes. Experimental factors that may affect pollen recovery including number of cigarettes, extraction solution, and extraction temperature, were optimized. The optimum parameters …
Developing A Maceration Method For Isolation Of Dna From Cancellous Bones, Bryan Bernal
Developing A Maceration Method For Isolation Of Dna From Cancellous Bones, Bryan Bernal
Student Theses
Prior to isolating DNA, it is necessary to remove any biological materials that may be present on commingled remains. Manual maceration of the outer surface is time-consuming. This study will address this issue by developing a simple method for processing bone samples prior to DNA isolation. A liquid-based technique was applied to macerate bone fragments by incubating bone fragments in an enzyme solution, removing potential contaminants, followed by a bone tissue disruption. Swine (Sus scrofa) bones were used to simulate human bones. Microscopic analysis suggested this method was effective for surface material removal. The methods effect on the …

Hostile Attributions To Ambiguous Situations: A Vignette Study Of The Mediating Role Of Hostile Attributions In The Relationship Between Community Violence Exposure And Youths’ Aggressive Behaviors, Hyun Kim
Student Theses
The effect of exposure to community violence has received increasing attention in recent years, given its prevalence and negative psychological and behavioral consequences on adolescents, including aggression (Lambert et al., 2018). The existing literature links violence exposure to hostile interpretation of situations (Dodge et al., 1990; Huesmann, 1988), and hostile interpretations to aggressive responses (Bradshaw et al., 2009; Crick & Dodge, 1994; Dodge et al., 1990). However, most studies have focused on parenting practices and within home violence. Few studies have examined the possible mediating role of hostile attribution bias in the relationship between community violence exposure and youth aggression, …

Mock Jurors' Perceptions Of "I Don't Know" Answers In Child Testimony, Leanza Greenlee
Mock Jurors' Perceptions Of "I Don't Know" Answers In Child Testimony, Leanza Greenlee
Masters Theses and Doctoral Dissertations
Attorneys questioning child witnesses often ask complex questions that negatively impact children’s accuracy and consistency. Research has shown that instructing children to answer confusing questions with “I don’t know” can improve their accuracy, but little research has examined the impact of using this strategy on jurors’ perceptions of child witness credibility. The present study assessed 702 mock jurors’ perceptions of a 4- or 10-year-old child witness in a fabricated sexual assault trial transcript. Number of “don’t know” responses were manipulated, and half the jurors were told about the “I don’t know” instruction. Results demonstrated that greater numbers of “I don’t …

Where The Zigzags Go: A Microhabitat Analysis For The Plethodon Dorsalis Complex Within The Tennessee River Gorge, Tegan Childers
Masters Theses and Doctoral Dissertations
Understudied high-biomass animals such as the Northern Zigzag salamander (Plethodon dorsalis) are known contributors to essential nutrient cycles within their respective habitats. Knowing that these sensitive creatures serve as bioindicators for the overall health and quality of many ecosystems, we should aim to comprehend their habitat requirements. Doing so could ultimately allow conservationists to set more accurate baselines for various mitigation and land management practices. Due to our location within the southeast amphibian biodiversity hot spot, sampling of P. dorsalis is conducted in the Tennessee River Gorge (TRG). Sampling of survey polygons began on October 28, 2018 and concluded on …
An Examination Of Parental Locus Of Control, Stress, Social Support, And Depression Among Parents Of Children And Adolescents With Intellectual Disabilities, Brandi Ellis
Dissertations
Caregivers (i.e., mothers and fathers) of youths with ID seem to be at a much higher risk of having symptoms of depression than caregivers of typically developing youths (Cantwell, Muldoon, & Gallagher, 2015; Giallo et al., 2015; Hu et al., 2010; Lee, 2013). A concurrent link has been found between parental locus of control (PLOC) and depression; however, there is a lack of support for a longitudinal association between these constructs among caregivers of youths with ID (Campis, Lyman, & Prentice-Dunn, 1986; Coyne & Thompson, 2011; Freed & Tompson, 2011; Lloyd & Hastings, 2009). Researchers have hypothesized that the lack …

Development, Expansion And Role Of Myeloid-Derived Suppressor Cells In Post-Sepsis Immune Suppression, Tuqa Alkhateeb
Development, Expansion And Role Of Myeloid-Derived Suppressor Cells In Post-Sepsis Immune Suppression, Tuqa Alkhateeb
Electronic Theses and Dissertations
Myeloid-derived suppressor cells (MDSCs) numbers increase significantly in sepsis and are associated with high mortality rates. These myeloid cell precursors promote immunosuppression, especially in the late (post sepsis) stage. However, the mechanisms that underlie MDSC expansion and programming are not completely understood. To investigate these mechanisms, we used a cecal-ligation and puncture (CLP) mouse model of polymicrobial sepsis that progresses from an early/acute proinflammatory phase to a late/chronic immunosuppressive phase. Previous studies in our laboratory showed that microRNA (miR)-21 and miR-181b elevate levels of the transcription factor nuclear factor 1 (NFI-A) that promotes MDSC expansion. We report here that miR-21 …

Talk This Way: A Look At The Historical Conversation Between Hip-Hop And Christianity, Joshua Swanson
Talk This Way: A Look At The Historical Conversation Between Hip-Hop And Christianity, Joshua Swanson
Electronic Theses and Dissertations
Christianity and Hip-Hop culture are often said to be at odds with one another. One is said to promote a lifestyle of righteousness and love, while the other is said to promote drugs, violence, and pride. As a result, the public has portrayed these two institutions as conflicting with no willingness to resolve their perceived differences. This paper will argue that there has always been a healthy conversation between Hip-Hop and Christianity since Hip-Hop’s inception. Using sources like Hip-Hop lyrics, theologians, historians, autobiographies, sermons, and articles that range from Ma$e to Tipper Gore, this paper will look at the conversation …

Unbinding Of Testosterone From The Androgen Receptor And Two Bf-3 Site Mutants, Muniruzzaman Chowdhury
Unbinding Of Testosterone From The Androgen Receptor And Two Bf-3 Site Mutants, Muniruzzaman Chowdhury
Theses and Dissertations
The androgen receptor (AR) can be activated by molecules binding in the steroid binding pocket. It can also be regulated allosterically by cofactors binding to the AF-2 surface site as well as by exogenous small molecules binding to the BF-3 surface site. Recent data indicated that mutations in the BF-3 site changed the amount of the endogenous steroid needed to activate the receptor, indicating that mutations of BF-3 can cause an allosteric effect. Molecular dynamics and steered molecular dynamics simulations were used to study the allosteric effect of the mutations on AR structure and the unbinding pathways of testosterone (TES) …

For this study, we will be looking to identify positive selection in eight genomes of the bacterial plant pathogen Xylella fastidiosa. A previous study used a branching method that identified 2 genes with positive selection along with a site-specific method identifying 34 genes showing positive selection. This current study focused specifically on the site-specific method, resulting in 28 genes (of 1,039 tested) showing positive selection. Of the 28 genes showing positive selection, 12 of them come from the pathogenicity, virulence and cellular structural categories. The remaining genes are found in the biosynthesis, metabolism, macro metabolism, and cellular process categories. …
Genetic Analysis Of Pi3k And Mtor Inhibition In U87mg Glioblastoma Cell Line, Carl G. Litif
Genetic Analysis Of Pi3k And Mtor Inhibition In U87mg Glioblastoma Cell Line, Carl G. Litif
Theses and Dissertations
NVP-BEZ235 is a Glioblastoma Multiform chemotherapeutic dual PI3K/mTOR pathway inhibitor created in 2008 and has since been proven experimentally to induce pluripotentcy in oncological cell populations. The inhibition of PI3K and mTOR has shown to coerce phenotypes associated with stem cell markers, most notably OCT4. It is necessary to understand the genetic composure of how PI3K/mTOR inhibited tumor cells are bypassing the canonical pathway for proliferation and growth and utilizing other parallel sources for tumor invasion into other neural regions. Taking a genetic approach with RNA-sequencing allowed us to gain insight into how glioblastoma interact with cytoskeleton factors MAPK4 and …
Optimizing Species Selection For Forest Restoration In The Lower Rio Grande Valley, Faeqa Mohsin
Optimizing Species Selection For Forest Restoration In The Lower Rio Grande Valley, Faeqa Mohsin
Theses and Dissertations
Over the past four decades, the Lower Rio Grande Valley has been reforested with over 3.3 million native thornscrub plants in old agricultural land through transplanting native species. However, assessments of LRGV forest restoration efforts are scarce, and to address the knowledge gap, this study examined species-specific responses to restoration interventions (RIs) aimed at minimizing seedling mortality for optimizing species selection. A critical period for forest restoration in old croplands is the seedling stage, where high mortality occurs due to the interactive effects of abiotic stress, compromised soil nutrient status from intensive agriculture, and mammalian herbivory. So, we evaluated seedling …
The Long-Term Persistence And Tissue Tropism Of Brazilian Zika Virus In Monodelphis Domestica, Marisol Morales
The Long-Term Persistence And Tissue Tropism Of Brazilian Zika Virus In Monodelphis Domestica, Marisol Morales
Theses and Dissertations
Zika virus (ZIKV) rose as a major public health concern due to the congenital abnormalities of infants born to infected mothers during the 2015 Latin American outbreak. While animal models such as transgenic mice and nonhuman primates are used to study ZIKV pathogenesis, they display various limitations. Additionally, the persistence and tissue distribution of long-term ZIKV infection remains unknown. We describe the laboratory opossum, Monodelphis domestica, as a potential new model for ZIKV pathogenesis research to address these issues. We utilized immunohistochemistry, RT-PCR, and ELISA to show that: infant animals inoculated intracerebrally were susceptible to infection, ZIKV infection persisted …

Aquatic Stage Mosquito Biocontrol: Effects Of Entomopathogenic Fungi Blastospores And Conidia On Aedes Aegypti Pupae, Ricardo Alberto Ramirez Garcia Rojas
Theses and Dissertations
Fungal entomopathogens present an opportunity to produce mosquito controls on-site, from remote locations to developing communities. Locally produced biopesticides have the potential to empower communities to develop internal disease vector control initiatives to protect local public health. We tested the effectiveness of the conidia solution and blastospore solution of Metarhizium anisopliae E9, Metarhizium anisopliae E1037, Isaria fumosorosea A97, and Isaria fumosorosea 9901 in controlling Aedes aegypti pupae. We observed that entomopathogenic fungi liquid blastospore solution induce mortality in Aedes aegypti pupae at a significantly greater rate than conidia solution and control treatment.
However, further analysis of testing methods used in …

Effects Of Seasonal Variations And Heat Exposure On Antioxidant Expression And Redox Status In The American Oyster, Crassostrea Virginica: Field And Laboratory Studies, Md Sadequr Rahman
Theses and Dissertations
Increasing atmospheric temperatures influence physiological functions in aquatic organisms. In this study, I observed the effects of seasonal (field study) and laboratory heat exposure (16, 22, 26, and 30°C for 1-week) on prooxidant-antioxidant homeostasis in gills and digestive glands of American oyster. Histological analysis showed an increase in mucus secretion with elevated temperatures in both organs. Immunohistochemical analysis showed a significant increase of heat shock protein-70, dinitrophenyl, and 3-nitrotyrosine protein expressions with higher seasonal and laboratory temperatures indicating heat-induced oxidative and nitrosative stress. This was concomitant with gradual rise in cellular apoptosis. Interestingly, catalase and superoxide dismutase expressions also increased …
Exploring Plant-Plant Interactions And Nutrient Manipulation As Strategies For Thorn Scrub And Thorn Forest Restoration, Edgar D. Vasquez
Exploring Plant-Plant Interactions And Nutrient Manipulation As Strategies For Thorn Scrub And Thorn Forest Restoration, Edgar D. Vasquez
Theses and Dissertations
Restoration of thorn scrub and thorn forest communities in South Texas is hindered by harsh environmental conditions and hypercompetitive invasive grasses. The success of thorn scrub/thorn forest restoration efforts depends on increasing seedling survival and growth by reducing stress after transplantation. Four experiments evaluated initial effects of plant-plant interactions and nutrient manipulation on thorn forest restoration. High density planting along with the use of nurse plants look promising; survival was generally higher under these conditions and growth was unaffected or in the case of high-density planting, promoted. Nutrient sequestration by sorghum had little effect on seedling performance and cover of …
