Cytochrome P450 2D6 (CYP2D6) is a highly polymorphic gene whose protein product metabolizes more than 20% of clinically used drugs. Genetic variations in CYP2D6 are responsible for interindividual heterogeneity in drug response that can lead to drug toxicity and ineffective treatment, making CYP2D6 one of the most important pharmacogenes. Prediction of CYP2D6 phenotype relies on curation of literature-derived functional studies to assign a functional status to CYP2D6 haplotypes. As the number of large-scale sequencing efforts grows, new haplotypes continue to be discovered, and assignment of function is challenging to maintain. To address this challenge, we have trained a convolutional neural …

Hysteretic Behavior In Voltage-Gated Channels, Carlos A. Villalba-Galea, Alvin T. Chiem

School of Pharmacy Faculty Articles

An ever-growing body of evidence has shown that voltage-gated ion channels are likely molecular systems that display hysteresis in their activity. This phenomenon manifests in the form of dynamic changes in both their voltage dependence of activity and their deactivation kinetics. The goal of this review is to provide a clear definition of hysteresis in terms of the behavior of voltage-gated channels. This review will discuss the basic behavior of voltage-gated channel activity and how they make these proteins into systems displaying hysteresis. It will also provide a perspective on putative mechanisms underlying hysteresis and explain its potential physiological relevance. …

The Experience Of Learning: Early Adolescents In Organized Youth Programs, Jennifer A. Skuza Nov 2020

The Experience Of Learning: Early Adolescents In Organized Youth Programs, Jennifer A. Skuza

Journal of Human Sciences and Extension

Although researchers and policymakers have focused on school as the critical place in which learning occurs, organized youth programs offer different environments in which early adolescents can learn. However, early adolescent learning in organized youth programs is an under-researched area of learning, which may limit the ability of youth development practitioners to respond appropriately to early adolescent learning needs. The purpose of this article is to describe the experience of learning in an organized youth program by finding meaning in early adolescents’ lived experience through a phenomenological methodology. For youth in this study, learning in an organized program is an …

Journal of Human Sciences and Extension

The need for physical activity promotion is ubiquitous, and offering physical activity interventions in community settings may reach people where they live, work, and play. Reaching people and providing evidence-based programming is exactly what the Cooperative Extension System has accomplished over the last century. In 2014, federal policy brought physical activity promotion into the mission of Extension. Although this policy marks the beginning of concerted efforts for physical activity promotion, several states have had strong success in this area. Borrowing from these efforts, this paper reports strengths, challenges, and opportunities for physical activity promotion in Extension.

Effect Of Catholyte Concentration On Current Production During Chocolate Industry Wastewater Treatment By A Microbial Fuel Cell, Diana Rahayuning Wulan, Suprihanto Notodarmojo

Makara Journal of Technology

Microbial fuel cells (MFCs) use bacteria as a catalyst to oxidize organic and/or inorganic substrates and produce electric currents. Wastewater could function as an electron donor in the anode chamber and, thus, represents a very promising energy source. Catholytes, as electron acceptors, influence power production in MFCs by increasing the availability of electrons. The present research aims to determine the influence of catholyte concentration on current production in an aerobic two-chamber MFC. Aerobic treatment was carried out in the two-chamber MFC operated in an incubator at 37  1 °C with and without aeration in the cathode chamber. Wastewater from …

Makara Journal of Technology

Hepatitis B virus (HBV) infection is one of the main causes of liver damage, which can also lead to chronic hepatitis B (CHB) infection. More than 240 million individuals worldwide are chronic carriers of HBV. Among individuals with CHB who are untreated, approximately 15% – 40% will progress to liver cirrhosis or cancer. The interactions between HBV and host immune response play significant roles in the progression of CHB. CHB can be generally divided into four different clinical phases: immune tolerance (IT), immune clearance, inactive carrier, and Hepatitis B surface antigen (HBsAg)-negative reactivation phase (ENEG). Many studies showed that interleukins …

Fabrication And Characterisation Of Highly Aligned Fibrous Assemblies, Tan Chin Joo

Student Works (2020-2029)

Vascular grafting has been an effective remedy to vascular diseases for decades. Autografting by using the veins or arteries in the patient’s body is found to have patency rate for years. However, autografting may not be possible when compatible vessels are not available. In this case, synthetic vascular grafts are found to be effective replacements. Nevertheless, synthetic grafts with diameter less than 4 mm becomes challenging due to thrombogenicity and mismatch of mechanical properties with the native vessels. Along this line, the ability to develop new biostable synthetic materials suitable for smaller grafts and the ability to evaluate their resulting …

Highly sensitive and specific plasma biomarkers for Alzheimer's disease (AD) have the potential to improve diagnostic accuracy in the clinic and facilitate research studies including enrollment in prevention and treatment trials. We recently reported CSF tau hyperphosphorylation, especially on T217, is an accurate predictor of β-amyloidosis at asymptomatic and symptomatic stages. In the current study, we determine by mass spectrometry the potential utility of plasma p-tau isoforms to detect AD pathology and investigate CSF and plasma tau isoforms' profile relationships. Plasma tau was truncated as previously described in CSF. CSF and plasma measures of p-tau-217 and p-tau-181 were correlated. No …

A comprehensive understanding of spaceflight factors involved in immune dysfunction and the evaluation of biomarkers to assess in-flight astronaut health are essential goals for NASA. An elevated neutrophil-to-lymphocyte ratio (NLR) is a potential biomarker candidate, as leukocyte differentials are altered during spaceflight. In the reduced gravity environment of space, rodents and astronauts displayed elevated NLR and granulocyte-to-lymphocyte ratios (GLR), respectively. To simulate microgravity using two well-established ground-based models, we cultured human whole blood-leukocytes in high-aspect rotating wall vessels (HARV-RWV) and used hindlimb unloaded (HU) mice. Both HARV-RWV simulation of leukocytes and HU-exposed mice showed elevated NLR profiles comparable to spaceflight …

The Use Of Spanish Moss As A Biological Indicator To Examine Relationships Between Metal Air Pollution, Vegetation Cover, And Environmental Equity In Tampa, Florida, Yousif Abdullah

USF Tampa Graduate Theses and Dissertations

Studies of inequality in exposure to less common air pollutants, like metals, are often limited by the costs of high spatial resolution measurements. Spanish moss (Tillandsia usneoides) is a promising bioindicator for measuring air pollution due to its lower cost, enabling capture of time-average environmental concentrations at high spatial resolution. This study had three major aims. First, I aimed to use Spanish moss as a bioindicator to characterize ambient concentrations of selected metals (Ti, Cr, Mn, Co, Ni, Cd, Hg, Pb, As, and Sb) in Tampa, Florida. My second goal was to determine the impact of vegetation cover on metals …

Salt Dependence Of Thermodynamic Stability Of A Cold-Active Dna Polymerase I Fragment, Xinji Zhu

LSU Doctoral Dissertations

P. ingrahamii is a halo-psychrophilic bacterium isolated from Arctic sea ice. We have cloned and purified the large fragment of the cold-active DNA polymerase I from P. ingrahamii, named Klenpin. The objective of this project is to directly compare the thermodynamic stability of Klenpin, and the salt dependence of that stability, with Klenow and Klentaq; two homologous polymerases from a mesophile (E. coli) and a thermophile (Thermus aquaticus).

We first examined the effects of salts on the thermal stability (Tm) of Klenpin and Klenow across the Hofmeister series. Significantly different trends were observed on …

Congenital Muscular Dystrophy-Associated Inflammatory Chemokines Provide Axes For Effective Recruitment Of Therapeutic Adult Stem Cell Into Muscles, Vitali Alexeev, Jacquelyn Olavarria, Paolo Bonaldo, Luciano Merlini, Olga Igoucheva

Department of Dermatology and Cutaneous Biology Faculty Papers

Background: Congenital muscular dystrophies (CMD) are a clinically and genetically heterogeneous group of neuromuscular disorders characterized by muscle weakness. The two most prevalent forms of CMD, collagen VI-related myopathies (COL6RM) and laminin α2 deficient CMD type 1A (MDC1A), are both caused by deficiency or dysfunction of extracellular matrix proteins. Previously, we showed that an intramuscular transplantation of human adipose-derived stem cells (ADSC) into the muscle of the Col6a1-/- mice results in efficient stem cell engraftment, migration, long-term survival, and continuous production of the collagen VI protein, suggesting the feasibility of the systemic cellular therapy for COL6RM. In order for this …

Diarrheal disease remains a major cause of childhood mortality and morbidity causing poor health and economic outcomes. In low-resource settings, young children are exposed to numerous risk factors for enteric pathogen transmission within their dwellings, though the relative importance of different transmission pathways varies by pathogen species. The objective of this analysis was to model associations between five household-level risk factors-water, sanitation, flooring, caregiver education, and crowding-and infection status for endemic enteric pathogens in children in five surveillance studies. Data were combined from 22 sites in which a total of 58,000 stool samples were tested for 16 specific enteropathogens using …
