Bacterial endophytes, while residing in host-plant tissues, offer a wide array of plant growth promotion benefits such as nitrogen fixation to impart nitrogen (N) nutrition, phosphate (P) solubilization to make soil-bound phosphate more mobile in soil for plant uptake, can suppress pathogenic microbes such as fungus and produce plant growth hormones to offset stress incurred by the plant. Hesperia dacotae (Skinner, 1911) (Insecta: Lepidoptera), also known as Dakota skipper, is a northern Great Plains species of butterfly associated with tall or mixed grass prairies. Its range once extended from southern Manitoba, southeastern Saskatchewan, North Dakota, South Dakota, Minnesota, to Iowa and Illinois. Now H. dacotae is only found in small isolated pockets in southern Manitoba, southeastern Saskatchewan, North Dakota, South Dakota, and Minnesota. The species is often observed on alkaline prairies with poor soil not desirable for cultivation. Thus, many of the historical H. dacotae are used for pasturing cattle or as hay prairie. Influenza remains a global health risk and challenge. Currently, NA inhibitors are extensively used to treat influenza, but their efficacy is compromised by the emergence of drug resistant variants. Antibodies targeting influenza A virus surface glycoproteins are critical components of influenza therapeutic agents and may provide alternative strategies to the existing countermeasures. This study characterized both polyclonal and monoclonal antibodies produced from the transchromosomic (Tc) cattle platform. A polyclonal antibody, designated SAB-100, was generated from Tc cattle after immunized with H1N1,H3N2, and influenza B virus. Auxin and cytokinin are major plant hormones that orchestrate plant development. These hormones are involved in cell division and differentiation and their spatio‐temporal balance controls organ initiation and maturation. Even though independent transcriptional responses to these hormones have been characterized, their concomitant outputs have not been described. We developed methodology to determine auxin-cytokinin relative output (ACRO) ratios at cellular resolution. Hormone-specific fluorescent markers that localize to the nucleus were used to report transcriptional outputs of auxin and cytokinin. Multiphoton microscopy and image data processing were used to quantify their relative ratio at cellular level in three-dimensional tissue volumes. This strategy …

Infectious respiratory disease has long been identified as the cause of bighorn sheep (Ovis canadensis) historical declines and extirpations, and Mycoplasma ovipneumoniae (Movi) is the primary pathogen inducing disease and mortality. Population-level effects of pneumonia events range from mild to extirpation. Variable individual response to pathogen exposure emerges from dynamic interactions between competing evolutionary processes within the host and pathogen. Understanding impacts of this evolutionary warfare is essential to assessing long-term impacts of pathogen invasion and developing appropriate countermeasures to protect population health. Cellulose Acetate (CA) based biomaterials are being used as substrates for bone ingrowth applications due to their nontoxic and nonirritant nature coupled with optimum morphology and stiffness. Electrospinning with additives and/or post-treatment has emerged as a viable protocol to further improve mechanical properties of CA and expand its utility. Herein, we highlight the role of potassium chloride (KCl) in association with airdrying to enhance the elastic modulus and tensile strengths of CA fibers. Sulfur mustard is a highly toxic and dangerous vesicant that has been utilized as a chemical warfare agent (CWA) since World War I. Despite its extensive history, an effective antidote to sulfur mustard exposure still does not exist. With detectable levels of unreacted sulfur mustard lasting days after initial contact, a window of opportunity exists to administer a “scavenger” to affected individual immediately and in the days following exposure in hopes of reducing harm by neutralizing unreacted sulfur mustard. In eukaryotes, small RNAs (sRNAs) are key regulators of RNA silencing which is also known as RNA interference (RNAi). RNAi is an essential mechanism known in plants, animals and fungi that regulates various biological activities, including defense against foreign nucleic acids and viruses. Fungal pathogens, such as Sclerotinia sclerotiorum, severely limit crop production in all parts of the world. Sclerotinia sclerotiorum cause white mold infection, affecting all dicotyledonous plants including many economically important field crops, vegetables, and floriculture. Macrophages maintain tissue homeostasis by identifying and eliminating threats within their tissue microenvironment. Pattern recognition receptors allow macrophages to recognize and internalize specific ligands while macropinocytosis allows the internalization of all extracellular solutes from their environment. Without pattern recognition receptors, pathogens may grow unchecked if they cannot be detected, and without macropinocytosis, macrophages struggle to detect and move around their environment. This thesis presents the results of CRISPR/Cas9 whole-genome screens used to identify the regulators of both endocytosis (Chapter 2) and macropinocytosis (Chapter 3). Alcohol is a widely available drug that is often abused. Studies have shown alcohol-induced memory loss in humans and animals; however, the large amount of alcohol often necessary to observe such loss and use of involuntary drinking paradigms in animals makes translation difficult. The current study was designed to look at voluntary binge-like drinking behavior and memory in rodents. We anticipated decreases in working memory function following consumption of binge-like alcohol in mice and expected a greater deficit in mice experiencing acute withdrawal during the memory task. The Role Of Gender In The Perception Of Lipreading, Emma Richards

The experiment examines the gender differences in lipreading ability. We tested 25 fluent English-speaking adults with normal hearing and vision. Each participant was asked to watch four videos testing their lipreading ability with matched and nonmatched gendered speakers. The participants were asked to transcribe 40 sentences, which were scored on the number of target words correct. It was found that participants who watch the videos with matched genders perceived more target words than in the mismatched gender videos. The results could explain previous inconsistencies in earlier research of whether ability to lipread is improved by the gender of the speaker.

Respiratory syncytial virus (RSV) is a negative-sense, single-strand RNA virus that affects the upper and lower respiratory system in humans. Currently, RSV is the leading cause of infant mortality worldwide, while also infecting target populations of the elderly and immunocompromised. Significant efforts towards vaccine development have been made due to limited therapeutic options, however, physical instabilities of RSV may hinder movement in vaccine development. We hypothesize that strain-specific differences in stability likely attribute to differences in RSV F protein, as proposed by previous studies about the intrinsically instability of RSV. Adequate language exposure is crucial for speech-language development in children. Previous research suggests that decreased language exposure leads to an increased risk for speech-language developmental delays. This study aims to explore the language exposure of two arguably language-deprived environments: Neonatal Intensive Care Units (NICU) and severe-to-profound hearing loss. This is a two-part study that examines caregiver-infant interactions through the analysis of infant-directed speech (IDS). In the first part of the study, researchers transcribed and analyzed pre-recorded Language Environment Analysis (LENA) audio recordings of five (5) infants with severe-to-profound hearing loss (6-months post cochlear implantation). A known metabolic pathway to neutralize metals uses the Phytochelatin Synthase (PCS) enzyme to catalyze the production of phytochelatins. The scorpionate ligand tris(pyrazolyl)borate (Tp) has contributed to the understanding of coordination and organometallic chemistry and has proven to be useful in the synthesis of metalloprotein models. Studies focusing on the similar ligand, tris(pyrazolyl)methane (Tpm), have recently been made possible with improved synthetic techniques. Characterization of the Tpm ligand and its metal derivatives, [TpmW(CO)3L]+ and [TpmMo(CO)3L]+, unveiled unexpected activity in the 1H NMR chemical shift of the methane hydrogen after the coordination of a seventh ligand.
