INTRODUCTION: Cachexia contributes to increased mortality and reduced quality of life in Chronic Obstructive Pulmonary Disease (COPD) and may be associated with underlying gene expression changes. Our goal was to identify differential gene expression signatures associated with COPD cachexia in current and former smokers.
METHODS: We analyzed whole-blood gene expression data from participants with COPD in a discovery cohort (COPDGene, N = 400) and assessed replication (ECLIPSE, N = 114). Gene Patents, Drug Prices, And Scientific Research: Unexpected Effects Of Recently Proposed Patent Eligibility Legislation, Charles Duan

Recently, Congress has considered legislation to amend § 101, a section of the Patent Act that the Supreme Court has held to prohibit patenting of laws of nature, natural phenomena, and abstract ideas. This draft legislation would expand the realm of patent-eligible subject matter, overturning the Court’s precedents along the way. The draft legislation, and movement to change this doctrine of patent law, made substantial headway with a subcommittee of the Senate holding numerous roundtables and hearings on the subject.
This article considers some less-discussed consequences of that draft leg- islative proposal. The legislation likely opens the door to patenting …

The discovery and development of new and potentially nonaddictive pain therapeutics requires rapid, yet clinically relevant assays of nociception in preclinical models. A reliable and scalable automated scoring system for nocifensive behavior of mice in the formalin assay would dramatically lower the time and labor costs associated with experiments and reduce experimental variability. Here, we present a method that exploits machine learning techniques for video recordings that consists of three components: key point detection, per frame feature extraction using these key points, and classification of behavior using the GentleBoost algorithm. Pulmonary fibrosis is characterized by fibroblasts persisting in an activated form, producing excessive fibrous material that destroys alveolar structure. The second messenger molecule cyclic 3',5'-adenosine monophosphate (cAMP) has antifibrotic properties, and prostaglandin E2 (PGE2) can stimulate cAMP production through prostaglandin E (EP)2 and EP4 receptors. Although EP receptors are attractive therapeutic targets, the effects of long-term exposure to PGE2 have not been characterized. To determine the effects of long-term exposure of lung fibroblasts to PGE2, human fetal lung (HFL)-1 cells were treated for 24 h with 100 nM PGE2 or other cAMP-elevating agents. cAMP levels stimulated by acute exposure to …

Congenital heart disease (CHD) is the most common major birth defect in children, affecting nearly 3% of children and is the leading cause of infant mortality. Heterotaxy (Htx) is a disorder of left-right (LR) patterning, in which organs, including the heart, are mispatterned relative to the LR axis. Htx is associated with severe forms of CHD, but its genetic causes remain largely undefined. Exome sequencing studies of large patient cohorts have recently revealed hundreds of CHD/Htx candidate disease genes, which now need to be validated in experimental systems for causality in order to better understand the causes and biological mechanisms …

When health policy experts noticed that health outcomes for African Americans were consistently worse than those of their White counterparts, many in the health care community assumed that the poor outcomes could be blamed on poverty and lifestyle choices. Subsequent research told a different story. Studies repeatedly showed that neither money, nor marriage, nor educational achievement protect African American men, women, or children from poor health. Instead, the disparities were more likely explained by racism. Specifically, multiple studies have shown that experiencing racism has been linked to increased infant and maternal mortality rates, elevated stress levels, and an increased risk …

This qualitative study used a phenomenological approach to capture the lived experience of non-relational foster families navigating the Colorado public education system. While most research on children in foster care looks at all sub-populations of foster care combined, this is the first known study to focus solely on non-relational foster families navigating the education system. I explored non-relational foster parents experience navigating the education system for the school-age children in their care through a semi-structured two-interview approach. MftC is a radical S-adenosyl L-methionine (SAM) enzyme that catalyzes the first step in the production of mycofactocin. It is a ribosomally synthesized post-translationally modified (RiPP) redox cofactor shown to be essential for the survival of bacteria in the Mycobacterium genus in the presence of cholesterol as a carbon source and for the sequestration of ethanol. MftC catalyzes the C – terminal decarboxylation of tyrosine and the subsequent cross-linking of tyrosine to the penultimate valine on the precursor peptide MftA. Epithelial tubules form critical structures in various body tissues; how- ever, since they are difficult to access experimentally, their architecture and dynamics are not well understood. Here we examine the dynamic remodeling of epithelial tubes in vivo using a novel and uniquely accessible model system: the extracorporeal vasculature of Botryllus schlosseri (sea squirt). In Botryllus, massive retraction of blood vessels can be triggered without loss of barrier function, through (i) disrupting collagen crosslinking in the basement membrane using β-aminopropionitrile (BAPN); or (ii) disrupting the integrin pathway through inhibition of focal adhesion kinase (FAK). Public and Population Health Faculty Publications and Presentations
A portable system based on a non-destructive sterilization method was developed that consists of a chamber producing ozone concentration levels reported in the scientific literature to deactivate 99.9% of all microorganisms. The system includes a small cordless 30mg/h ozone generator placed inside a 20L enclosed chamber. Personal protective equipment is loaded into the chamber and automatically exposed to an ozone concentration up to 18 ppm for 30 minutes. PPE is then removed and ventilated in open air to allow residual ozone to degrade to oxygen. The paper describes the design and performance characteristics of this technology.

Undergraduate Research Posters 2020
Terminal differentiation in erythroid cells culminates in enucleation, a type of cytokinesis, where the nucleus is expelled from the cell. Improper cytokinesis and enucleation is due to a mutation in Erythroid Krüppel-like Factor (EKLF), an erythroid transcription factor, leading to Congenital Dyserythropoeitic Anemia IV (CDA’s) whose pathway remains to be understood. If EKLF is knocked out of mouse erythroid cells, the cells exhibit a failure in cytokinesis and enucleation with more megakaryocyte production. EKLF does have a role in allowing megakaryocyte erythroid cells to enter into erythropoiesis if the megakaryopoiesis genes are suppressed. Although, it is unknown if EKLF suppress …

The PITX2 adjacent noncoding RNA (PANCR) is an intergenic long noncoding RNA adjacent to the PITX2 gene. The PITX2 gene functions as a regulator for bilateral symmetry during development. PANCR is only expressed in the left atrium of the heart and in the eye in humans. PANCR has been studied with its effects in atrial fibrillation in the heart, but very little is known about its expression in the human eye. This project hypothesizes that the dysregulation of PANCR, which causes mutations in the PITX2 gene, will cause deformities to the human eye, similar to those of Axenfeld-Rieger Syndrome, a …

Introduction: During the last decade, the results of several clinical studies suggested that oral probiotics may potentially improve oral health. The first study examining the effects of probiotics on oral health demonstrated that almost every patient with gingivitis, periodontitis or pregnancy gingivitis, had significant improvements in measurable periodontal indices when they were treated with a locally administered culture supernatant of a Lactobacillus acidophilus strain. This finding sparked several other studies to further examine the potential for treating oral diseases, such as, combating halitosis, oral candidiasis, and dental caries with probiotics. Decreased sleep can negatively affect mental and physical health and can significantly disrupt participation in daily occupations and leisure interests. Even though sleep is an area of occupation, there is limited evidence on the effectiveness of occupational therapy interventions for sleep. The purpose of this embedded mixed methods research study was to identify how disruptions in sleep affected daily occupational performance and to understand the effectiveness of occupational therapy- directed non-pharmacological sleep interventions for three participants in a community re-entry program for brain injury. Background: Autism Spectrum Disorder (ASD) is a prevalent neurodevelopmental disorder in the U.S (about 1 in 54 children). Treating children with ASD in the dental office can be challenging due to their impairments in social interactions, communication and aversive behavior. Visual Support Schedules are visual aids or picture cues that have been beneficial for the behavior management of patients with ASD.
