Dielectrophoresis (DEP) is an established technique used as a label-free method to manipulate microorganisms by exploiting the interaction of targeted microorganisms with a non-uniform electric field. The strength and direction of the DEP force on a targeted cell, specifically Komagataeibacter xylinus (K. xylinus), is dependent on the frequency of the applied electric field as well as the dielectric properties of the bacterial cell and suspending media.

Heterocyclic molecules that contain nitrogens and consist of four, five and six-membered rings such as azetidinones (eg: β-lactams), pyrroles, indoles, azoles, triazoles, oxadiazines, and oxadiazinones, etc. are ubiquitous in the use of pharmaceutical fields. This also reflected in the recently FDA approved unique small-molecule pharmaceuticals, where about 90% of those molecules contain a nitrogen heterocycle. Particularly, the vicinal diamino motif has been appeared in leading drug directing candidates such as penicillin, cephalosporins, and oseltamivir (Tamiflu) over the past years.

The main portion of this thesis describes two distinct applications of guanidinium sulfation crystallization: use of a flat guanidinium chloride for enantiomeric identification and a chiral guanidinium bromide for diastereomeric resolution. Additionally, the following two chapters includes the modification of naturally occurring materials for pollution remediation and the synthesis of biodegradable polyesters for biomedical imaging.The importance of chirality can be found in a variety of fields including pharmaceuticals, fragrances, food additives, agrochemicals, and forensics. ABSTRACTThis dissertation begins with the introduction of genetics, genomics, and the Healthcare Genetics (HCG) model. The five concepts of the HCG model, genetics/genomics, education, environment, clinical testing and therapeutics, and ethical/legal/social implications (ELSI) are defined. These concepts are threaded throughout the manuscripts and dissertation chapters. Chapter II defines molecular testing and differences between single-gene and multi-gene testing. The Kaplan Meier Curve and its use in research, to illustrate survival over time, is discussed. Spider silks are remarkable biomaterials, possessing a variety of desirable mechanical and material properties. Humans have long-sought to utilize these materials. Spiders produce a range of different types of silks for different purposes, such as web-building, egg casing, and prey swathing. Because spiders are difficult to farm, a synthetic biological approach must be used to produce silk-like proteins. This research is intended to improve the production of piriform, a specific type of spider silk. This was done with targeted-improvements to microbial metabolism and recovery methodologies. Long known as a toxic gas, carbon monoxide (CO) has been recently redefined as a health beneficial molecule produced endogenously in humans. The biological activity of CO has been extensively examined in inflammatory conditions, vasoprotection, anti-apoptotic and anticancer studies, although the mechanisms of action are still not well defined. Because CO is a gaseous molecule, a straightforward administration would be inhalation. However, this delivery method is limited to use in hospital settings under professional supervision. Anaerobic processes are being explored for the production of bioenergy in the form of biomethane and biohydrogen in a "waste-to-value" approach that considers wastes as a resource. Laboratory scale Biomethane potential (BMP) tests and Up-Flow Anaerobic Sludge Blanket (UASB) reactors can be used to indicate the potential recovery of bioenergy from industry wastes. Shepherdia ×utahensis ‘Torrey’ (Elaeagnaceae) is a hybrid of two native actinorhizal plants in the Intermountain West, S. argentea (silver buffaloberry) and S. rotundifolia (roundleaf buffaloberry). Due to actinorhizal symbiosis, atmospheric nitrogen (N2) can be converted to ammonium, a bioavailable form. Actinorhizal plants have great value in sustainable nursery production and urban landscape use. However, nitrogen fertilizer negatively affects the nodulation of actinorhizal plants. Coast redwood (Sequoia sempervirens) is the tallest species in the world, frequently attaining heights greater than 300 ft. The unique characteristics of the redwoods has led to the establishment of several preservation areas including national and state parks. Understanding how natural variation in flow regimes influences stream ecosystem structure and function is critical to the development of effective stream management policies and actions. Spatial variation in flow regimes is well understood for stream reaches in mesic regions, but a more robust characterization of flow regimes in arid regions is needed, especially to support biological monitoring and assessment programs. An Evaluation of the Trauma-Informed Approach Implemented at a Title I Elementary School assesses the merits of the implementation of an intervention to support the social emotional ability and stability of students within elementary schools; called the Trauma Informed Approach. Many children attending elementary schools bring diverse and complex experiences to the classroom setting. Students with multiple adverse childhood experiences attend the educational system. Many students have a more difficult time concentrating and succeeding due to anxiety and lack of physiological needs being met. This project focuses on how one school district uses quarterly assessments to identify standards students have not mastered. This project identifies how some schools utilize quarterly assessment data with varying results. One problem I have identified is a lack of a consistent expectation from district leadership, regarding the expectation for how schools should use the assessment program. Despite the creation and implementation of laws intended to support and protect Deaf individuals, stories of limited opportunities and oppression within the workplace still exist and are pervasive. Current research in regard to Deaf individuals’ upward mobility includes a discussion of cultural capital, Imposter Syndrome, and navigational capital. To further understand the experiences of Deaf individuals, the research team conducted a mixed-methods study utilizing surveys and interviews. The results provided insight regarding challenges experienced by the participants in either-or-both their education and employment. The data suggests that the use of navigational capital was the most significant predictor for upward mobility.

Staphylococcus aureus has maintained its clinical relevance as a major cause of hospital and community acquired infections globally with a high burden of antimicrobial resistance (AMR). Though reported, the burden of infection, antimicrobial resistance and molecular epidemiology of S. aureus are not well defined in Kenya. This descriptive review evaluated reported data on the detection and characterization of S. aureus infections in Kenya. Published data between 2000 and 2020 were evaluated. S. aureus isolation frequencies varied from 1% in blood specimens to 52.6% among skin and soft tissues infections while MRSA rates ranged from 1% to 84.1%. While penicillin resistance …

This special issue is dedicated to the memory of Prof. Dwight David Amparo Diestro, who died on June 2, 2020 at the age of 55. Prof. Diestro, a historian of Southern Luzon, was an Associate Professor of history in the Department of Social Sciences (DSS) of UPLB. He was one of the great pillars of DSS, serving as Department Chair for three terms in a total of 10 years. He was the first chair of the Southern Luzon Studies Center and the motivating force behinds its progress.

The objectives of this research were to evaluate the antibacterial activity and to determine the chemical composition of a list of medicinal plants used by Native Americans in California. Artemisia californica, Mimulus aurantiacus, Equisetum telmateia, Equisetum hyemale, and Marah fabacea were selected from a list of plants reported as having been used for ailments related to infections by tribes located in California. The extracts obtained through steam distillation from E. telmateia, E. hyemale and M. fabacea were assayed for in vitro antibacterial activity against 16 Gram-negative and 6 Gram-positive bacteria using disk diffusion assays and measuring the diameters of inhibition …

Objective: The influence of childhood trauma has been found to be related to difficulties in emotion regulation and distress tolerance in young adulthood (Berenz et al., 2018a, 2018b). Research has shown that childhood abuse and adversities such as neglect or emotional abuse results in impaired processes related to the development of emotion regulation and efficient interpersonal skills, while also resulting in symptoms reflecting disordered affective self-regulation (Cloitre et al., 2009; Shipman, Edwards, Brown, Swisher, & Jennings, 2005; Shipman, Zeman, Penza, & Champion, 2000). There is evidence in rodents to suggest that theacrine-based supplements modulate tissue sirtuin activity as well as other biological processes associated with aging. Herein, we examined if a theacrine-based supplement (termed NAD3) altered sirtuin activity in vitro while also affecting markers of mitochondrial biogenesis. The murine C2C12 myoblast cell line was used for experimentation. Following 7 days of differentiation, myotubes were treated with 0.45 mg/mL of NAD3 (containing ~2 mM theacrine) for 3 and 24 h (n = 6 treatment wells per time point). Relative to control (CTL)-treated cells, NAD3 treatments increased (p < 0.05) Sirt1 mRNA levels at 3 h, as well as global sirtuin activity at 3 and 24 h. Follow-up experiments comparing 24 h NAD3 or CTL treatments indicated that NAD3 increased nicotinamide phosphoribosyltransferase (NAMPT) and SIRT1 protein levels (p < 0.05). Cellular nicotinamide adenine dinucleotide (NAD+) levels were also elevated nearly two-fold after 24 h of NAD3 versus CTL treatments (p < 0.001). Markers of mitochondrial biogenesis were minimally affected. Although these data are limited to select biomarkers in vitro, these preliminary findings suggest that a theacrine-based supplement can modulate select biomarkers related to NAD+ biogenesis and sirtuin activity. However, these changes did not drive increases in mitochondrial biogenesis. While promising, these data are limited to a rodent cell line and human muscle biopsy studies are needed to validate and elucidate the significance of these findings.

This idea has been well-tested in plants and some animal groups, but not for fishes. Microorganisms can potentially colonise volcanic rocks using the chemical energy in reduced gases such as methane, hydrogen (H2) and carbon monoxide (CO). In this study, we analysed soil metagenomes from Chilean volcanic soils, representing three different successional stages with ages of 380, 269 and 63 years, respectively. A total of 19 metagenome-assembled genomes (MAGs) were retrieved from all stages with a higher number observed in the youngest soil (1640: 2 MAGs, 1751: 1 MAG, 1957: 16 MAGs). Satellite constellation design is a complex, highly constrained, and multidisciplinary problem. Unless optimization tools are used, tradeoffs must be conducted at the subsystem level resulting in feasible, but not necessarily optimal, system designs. As satellite technology advances, new methods to optimize the system objectives are developed. This study is based on the development of a representative regional remote sensing constellation design. This thesis analyses the design process of an electrooptic satellite constellation with regional coverage considerations using system-level optimization tools. Hand-crafting effective and efficient structures for recurrent neural networks (RNNs) is a difficult, expensive, and time-consuming process. To address this challenge, this work presents three nature-inspired (NI) algorithms for neural architecture search (NAS), introducing the subfield of nature-inspired neural architecture search (NI-NAS). These algorithms, based on ant colony optimization (ACO), progress from memory cell structure optimization, to bounded discrete-space architecture optimization, and finally to unbounded continuous-space architecture optimization. These methods were applied to real-world data sets representing challenging engineering problems, such as data from a coal-fired power plant, wind-turbine power generators, and aircraft flight data recorder (FDR) data.

This thesis argues RuPaul’s Drag Race (RPDR, 2009–) positions itself as a homonormative pathway to LGBTQ+ social inclusion through privileging neoliberal selfbranding and commodity activist practices that reify privileged raced, classed, and sexuality identity markers. Utilizing interdisciplinary and intersectional cultural studies methods to conduct a textual analysis, I examine how RPDR produces homonormative LGBTQ+ identities through the commodification and standardization of drag cultures. In conversation with existing RPDR scholars, I critically survey RPDR’s gender biases and prosocial messaging as an example of brand culture’s reification of hegemony and homonormativity within LGBTQ+ communities. It is estimated that 16% of men and 26% of women have experienced child sexual abuse (CSA), and studies have shown a strong association with long-term psychosocial consequences. The use of psychoeducational groups with survivors of CSA has been found to produce favorable outcomes; however, no meta-analyses have been conducted on studies assessing the use of psychoeducational groups to treat victims of CSA.
