Isolation, Structure Elucidation, And Synthesis Of Natural Products From Marine Cyanobacteria, Keren Solomon

This thesis describes the isolation, structure elucidation, and synthesis of natural products from marine cyanobacteria. A crude extract from a cyanobacterium collected in Curacao showed selective affinity for the dopamine D5 receptor in a screen against a panel of CNS receptors. Due to the high similarity of the D5 and D1 receptor, to date there are no known ligands that differentiate them. Attempts to purify the compound responsible for this affinity led to the isolation of the known compound caylobolide A. A second extract from a cyanobacterium collected in Panama underwent bioassay-guided fractionation and yielded the novel …

Abstract
Loneliness is a subjective experience characterized by a profound decrease in the amount and the quality of desired social connectedness. Community-dwelling older adults are particularly vulnerable to loneliness because of age-related changes such as the loss of driving privileges, loss of a spouse, living alone, socioeconomic changes caused by retirement, and a lack of access to meaningful social activities. These factors diminish an older adult’s sense of belonging, the quality of their social relationships, and the degree to which they are socially engaged. Major depressive disorder (MDD) is the second most common mental health condition and a leading cause of disability in the world. It is theorized that MDD develops from a combination of biological, psychological, and social stressors. The condition is typically treated using pharmaceuticals and psychotherapy. However, not all individuals with MDD have access to or choose to use these treatments, or may prefer to incorporate therapeutic lifestyle changes such as exercise, sleep, and healthy eating. Hypoplastic left heart syndrome (HLHS) is a severe and rare form of congenital heart disease with no known cause that decreases life expectancy and occurs during fetal cardiac development. Without treatment HLHS is fatal. HLHS consists of the underdevelopment of the left-sided heart structures leading to the failure of the left ventricle to support the systemic circulation resulting in single-ventricle physiology (Mussa & Barron, 2016). Prior to the 1980s HLHS was a universally lethal condition until 1982, when surgical management became available (Mussa & Barron, 2016). Accurate population estimates are critical to inform conservation management of species. Incomplete sampling can lead to population underestimates and lacking conservation efforts. Population surveys are important for assessing human and animal welfare to allow for targeted conservation action. Recent ring-tailed lemur (Lemur catta) surveys have led researchers to conclude that L. catta populations are crashing. This has generated much attention and alarm, but may also be based on incomplete information. To better understand population dynamics, more thorough sampling is needed. Here, we survey five such sites to reassess the presence and abundance of L. catta in these locations. …

On January 1, 2022, mandatory genetically engineered (GE) food disclosure labeling will be required nationwide in the United States. To date, the only mandatory GE labeling law implemented in the U.S. was Act 120 in Vermont. This thesis examines the consumer purchasing response to the implementation of Vermont Act 120 using store-level scanner data of food purchases. I measure the effects of Vermont Act 120 on the grocery store sales of non-GMO, organic, and GE- labeled products in Vermont. Co-occurrence of risk for impoverished families is common, but less is known about how compounded risk influences parenting behavior. Mothers (n = 167) and their two-year-old children were visited at home and engaged in a game aimed to elicit everyday parenting behavior. Mothers endorsed experience of sociodemographic and psychosocial risks. Two unique cumulative risk indices were created from these variables. Regression analyses assessed the relation between the risk indices and positive and negative parenting behavior. Latent class analysis examines classes of risk experience on the same indicators. Type 2 diabetes (T2D) results when pancreatic beta cells are unable to secrete adequate amounts of insulin in response to elevated blood glucose levels. Genome-wide association studies (GWAS) have linked single nucleotide polymorphisms (SNPs) in >250 regions of the human genome to a higher risk for developing T2D. These T2D-associated GWAS SNPs are predominantly non-coding (~90%) and enriched at islet regulatory elements, implicating altered transcriptional regulation in islets as a molecular mechanism underlying T2D risk. A vicious cycle has arisen in San Francisco’s Tenderloin (TL) neighborhood, in which poverty contributes to significant health disparities, thwarting optimal health outcomes, thus further worsening the underlying poverty which takes on a generational and urban character. TL residents face core socioeconomic and public health challenges with regard to the following themes: access to and coordination of care and services; food security (including healthy eating and active living); housing security; safety from violence and trauma; social, emotional, and behavioral health; poverty; and infrastructure. Anthropogenic global warming is causing unprecedented changes to occur within Arctic ecosystems. As sea ice continues to decline in both thickness and extent, ice-adapted species will be particularly affected. Specifically, Arctic seals are reliant on sea ice during critical life-history stages and many of their preferred prey depend on reliable patterns in annual sea ice cover. Thus, ongoing changes will likely affect the ability of Arctic seals to carry out key life-history tasks and maintain positive energy balance. This research investigates maternal nutritional status through the analysis of young infants (N=27) that are likely pre-weaned from two Roman occupational periods at Put Dragulina. Because young infants are solely dependent on the nutritional status of their mother while in utero and postpartum through the consumption of breast milk, the presence of skeletal pathologies positively associated to nutritional deficiencies on the remains of young infants can be analyzed to reconstruct the nutritional status of the infant’s mother. This study finds that 89% (N=24) of young infants, regardless of occupational period, presented with skeletal pathologies consistent with nutritional deficiencies. These results …

Iron deficiency chlorosis (IDC) is a frequent problem in soybean [Glycine max (L.) Merr.] production on calcareous soils. Greenhouse and soil incubation experiments were established to evaluate pyritic lignite efficacy to supply Fe as compared to Fe-EDDHA and Fe-EDTA sources across contrasting IDC tolerance varieties grown on a slightly acid and a calcareous soil. Soybean IDC incidence was influenced by the soil properties and variety tolerance. All iron sources increased plant dry matter accumulation on the Leeper soil, whereas on the Marietta soil only lignite at 0.672 kg ha-1 produced an increase. Human activity affects white-tailed deer (Odocoileus virginianus) movement and habitat selection during all times of the year, but knowledge is limited regarding how human risk affects white-tailed deer during the summer and winter. During spring and summer, variation in patch selection varied. Natural vegetation was selected for early in the year, with anthropogenic forages being important for deer use during the summer. During the winter, deer responded to different levels of risk. As hunter risk increased on the landscape, deer altered selection of the landscape. Deer avoided areas that were heavily used by hunters, using areas containing less hunter risk. …

Post-Harvest Reduction Of Salmonella In Pork Trimming, Dishnu Sajeev

The objective of the current study was to determine the efficacy of 3% acetic acid in reducing Salmonella in pork trimming and the effects of such treatment on meat quality. For 15-s dipping and 5-log CFU/pork cube inoculation, only 0.2- to 0.3-log reduction was observed (P ≤ 0.026). Acetic acid worked best at 75 s and 50°C, providing 1.4-log reduction (P < 0.001), damaging Salmonella cell membranes. When an inoculated pork cube was placed at the geometrical center of 2.3-kg pork trimming, dipping at 50°C for 75 s only reduced Salmonella by 0.2 log (P = 0.040). Although dipping slightly increased lightness (P < 0.001) and decreased redness (P ≤ 0.008) on the meat surface, no inside color change was detected (P = 0.120). Neither lipid oxidation (TBARS, P = 0.644), protein solubility (P = 0.187), nor water-holding capacity (P = 0.076) were affected by treatments.

Controlling pathogenic presence in broilers has become a priority in the poultry industry to prevent economic losses due to disease and infection, as well as the possible contamination of chicken products. The use of antibiotics reduces the incidence of infections; however, their removal from production initiated the search for suitable alternatives. Probiotic ineed supplements have been widely evaluated as alternatives. Probiotic use has improved broiler performance, reduced pathogenic loads, and stimulated the immune system at later life stages. Diet is a key factor in the development and progression of Inflammatory Bowel Disease (IBD). A variety of diets have been studied with IBD patients. This cross-sectional survey identified current healthcare practitioner views on different diets and their efficacy with IBD patients. Diets were rated on awareness, compliance, and contributors to success by participants (n = 181). Frequencies were conducted, and ANOVA with Duncan pairwise comparison or chi-square analysis were used to determine significant differences. Most participants (96%) and 98% of registered dietitians (RD) considered using diet to help treat IBD patients. RDs perceived the low fiber or low residue …
