MicroRNAs are important regulators in plant developmental processes and stress responses. In this study, we generated a series of maize STTM166 transgenic plants. Knock-down of miR166 resulted in various morphological changes, including rolled leaves, enhanced abiotic stress resistance, inferior yield-related traits, vascular pattern and epidermis structures, tassel architecture, as well as abscisic acid (ABA) level elevation and indole acetic acid (IAA) level reduction in maize. To profile miR166 regulated genes, we performed RNA-seq and qRT-PCR analysis. A total of 178 differentially expressed genes (DEGs) were identified, including 118 up-regulated and 60 down-regulated genes. These DEGs were strongly enriched in cell …

Department of Radiology Faculty Papers
BACKGROUND: Combined hepatocellular-cholangiocarcinoma (CHC) is a rare type of primary liver cancer. Due to its complex histopathological characteristics, the imaging features of CHC can overlap with those of hepatocellular carcinoma (HCC) and intrahepatic cholangiocarcinoma (ICC).
AIM: To investigate the possibility and efficacy of differentiating CHC from HCC and ICC by using contrast-enhanced ultrasound (CEUS) Liver Imaging Reporting and Data System (LI-RADS) and tumor biomarkers.
METHODS: Between January 2016 and December 2019, patients with histologically confirmed CHC, ICC and HCC with chronic liver disease were enrolled. The diagnostic formula for CHC was as follows: (1) LR-5 or LR-M with elevated alpha-fetoprotein …

Sybil Defense Using Efficient Resource Burning, Diksha Gupta
Computer Science ETDs
In 1993, Dwork and Naor proposed using computational puzzles, a resource burning mechanism, to combat spam email. In the ensuing three decades, resource burning has broadened to include communication capacity, computer memory, and human effort. It has become a well-established tool in distributed security. Due to the cost attached to utilizing resource burning mechanism, these have not been popularized in domains apart from cryptocurrency.
In this dissertation, we design efficient resource burning based Sybil defense techniques for permissionless systems. As a first step, we identify existing resource burning mechanisms in literature in Chapter 2. Additionally, we enumerate numerous open problems …

Print Culture, Digital Culture, Poetics And Hermeneutics: Discussion With J. Hillis Miller, Liyuan Zhu
Print Culture, Digital Culture, Poetics And Hermeneutics: Discussion With J. Hillis Miller, Liyuan Zhu
CLCWeb: Comparative Literature and Culture
This paper is a response to Hillis Miller’s query on the author’s essay “Hillis Miller on the End of Literature.” The author basically agrees with Miller’s view on the shift from print culture to digital culture, explaining the special cultural context under which Chinese scholars emphasize the visual turn. Based on the rapid development of Chinese online literature, the author points out that print culture does not rival but coexists with digital culture. On the other hand, drawing on Aristotle’s Poetics and insights of several leading figures of contemporary hermeneutics, the author contends that Miller’s dichotomy of poetics (form) and …

One of the greatest difficulties with evolutionary approaches in the study of stone tools (lithics) has been finding a mechanism for tying culture and biology in a way that preserves human agency and operates at scales that are visible in the archaeological record. The concept of niche construction, whereby organisms actively construct their environments and change the conditions for selection, could provide a solution to this problem. In this review, we evaluate the utility of niche construction theory (NCT) for stone tool archaeology. We apply NCT to lithics both as part of the “extended phenotype” and as residuals or precipitates …

Dysregulated Interferon Response Underlying Severe Covid-19, Leann Lopez, Peter C. Sang, Yun Tian, Yongming Sang
Agricultural and Environmental Sciences Faculty Research
Innate immune interferons (IFNs), including type I and III IFNs, constitute critical antiviral mechanisms. Recent studies reveal that IFN dysregulation is key to determine COVID-19 pathogenesis. Effective IFN stimulation or prophylactic administration of IFNs at the early stage prior to severe COVID-19 may elicit an autonomous antiviral state, restrict the virus infection, and prevent COVID-19 progression. Inborn genetic flaws and autoreactive antibodies that block IFN response have been significantly associated with about 14% of patients with life-threatening COVID-19 pneumonia. In most severe COVID-19 patients without genetic errors in IFN-relevant gene loci, IFN dysregulation is progressively worsened and associated with the …

Sjögren syndrome (SS) is an immunologically complex, chronic autoimmune disease targeting lacrimal and salivary glands. Nonobese diabetic (NOD) mice spontaneously develop inflammation of lacrimal and salivary glands with histopathological features similar to SS in humans including focal lymphocytic infiltrates in the affected glands. The innate immune signals driving lymphocytic infiltration of these glands are not well-defined. Here we evaluate the role of Toll-like receptor (TLR) 7 in the development of SS-like manifestations in NOD mice. We created a Tlr7 knockout NOD mouse strain and performed histological and gene expression studies to characterize the effects of TLR7 on autoimmunity development. TLR7 …

Angiotensin-converting enzyme 2 (ACE2) is the cellular receptor for the Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) that is engendering the severe coronavirus disease 2019 (COVID-19) pandemic. The spike (S) protein receptor-binding domain (RBD) of SARS-CoV-2 binds to the three sub-domains viz. amino acids (aa) 22-42, aa 79-84, and aa 330-393 of ACE2 on human cells to initiate entry. It was reported earlier that the receptor utilization capacity of ACE2 proteins from different species, such as cats, chimpanzees, dogs, and cattle, are different. A comprehensive analysis of ACE2 receptors of nineteen species was carried out in this study, and the …

Giftedness In Young Children: What Do Parents And Teachers Know?, Katie Goodman
Departmental Honors & Graduate Capstone Projects
Abstract
Past research has explored how young children express giftedness as well as how parents and teachers can best meet the needs of these children once they have been identified. However, there is a distinct lack of literature on what parents and teachers actually know about how giftedness manifests in the youngest learners. This research investigates the background knowledge that teachers and parents of preschoolers hold on the notion of giftedness. Parents and teachers of young children were surveyed using online survey software by contacting a diverse set of private preschools within the Northern Virginia area. Parents and teachers at …

Lymphatic vasculature is an integral part of digestive, immune and circulatory systems. The homeobox transcription factor PROX1 is necessary for the development of lymphatic vessels, lymphatic valves (LVs) and lymphovenous valves (LVVs). We and others previously reported a feedback loop between PROX1 and vascular endothelial growth factor-C (VEGF-C) signaling. PROX1 promotes the expression of the VEGF-C receptor VEGFR3 in lymphatic endothelial cells (LECs). In turn, VEGF-C signaling maintains PROX1 expression in LECs. However, the mechanisms of PROX1/VEGF-C feedback loop remain poorly understood. Whether VEGF-C signaling is necessary for LV and LVV development is also unknown. Here, we report for the …

The Clean Air Act: How It Can Be Localized To Promote Both Environmental And Social Justice, Tate Kirk
Seattle Journal of Technology, Environmental, & Innovation Law
Legislators attempt to achieve intended goals by enacting laws that provide for regulatory enforcement. However, many times laws are unable to achieve their stated goals and in some ways may create new or exacerbate existing issues. Luckily, upon review, many of these issues can be fixed with quick modifications to either their implementation or enforcement mechanisms. In its current form, the Clean Air Act does not effectively account for differences in regional climate patterns, and, moreover, it perpetuates environmental injustice. If local governments were given more autonomy to enforce the Clean Air Act, they could shape its enforcement to more …

Wildlife Trade And Covid-19: Towards A Criminology Of Anthropogenic Pathogen Spillover, Piers Beirne Phd
Department of Criminology
The general remit of this paper is the role of wildlife trade in pathogen spillover. Its underlying assumption is that, so far from being the exclusive domain of the life sciences, the study of pathogen spillover will be greatly enhanced by multi-perspectival approaches, including One Health and those employed here, namely, non-speciesist green criminology and critical animal studies. The paper moves from discussions of zoonosis, anthroponosis and wildlife trade to the emergence of COVID-19 in Wuhan, China. The paper recommends the abolition of all wildlife trade and the reclamation of wildlife habitat and broaches discussion of the extension of legal …

Assessing Moringa Oleifera Plants As Exposure Pathways For Heavy Metals In Highly Contaminated Areas, Marissa Louise Mayfield*
Biology Theses
This study examined the uptake of heavy metals by Moringa oleifera trees in both field and laboratory settings. The main goal of this study was to determine whether using Moringa, grown in heavily contaminated areas, as food or herbal supplements increases the risk of heavy metal exposure. The field component of this study was carried out in the Southern part of Kabwe, Zambia where a lead-zinc mine has left a legacy of severe environmental pollution in the neighboring areas. Moringa trees have several medicinal properties which is one of the reasons they were first introduced to the area. The trees …

An Invasive Species As A Threat To Native Biodiversity: Larval Competition Between Native Anurans And An Invasive Treefrog, Mckenzie Wasley
Electronic Theses and Dissertations
Cuban Treefrogs (Osteopilus septentrionalis) are established in Florida and Louisiana and have invasive potential further westward, possibly impacting native anurans at multiple life stages. In anurans, competition at the larval stage can decrease adult fitness through slower development and smaller size at metamorphosis, ultimately decreasing recruitment rates. To examine the potential impacts of O. septentrionalis at the larval life-history stage, I quantified activity levels and growth of their tadpoles and those of two anurans native to eastern Texas: Green Treefrogs (Hyla cinerea) and Gulf Coast Toads (Incilius nebulifer). Tadpoles of the three species were …

Prenatal Alcohol Exposure Alters Transcriptional Expression Of Stress And Tlr4-Related Immune Signaling Factors In The Brain Following Early Life Stress, Chaselyn D. Ruffaner-Hanson
Biomedical Sciences ETDs
Background: Depression and anxiety disorders, believed to be caused by stress-related neuropeptides and pro-inflammatory molecules in the brain, are prevalent comorbidities of fetal alcohol spectrum disorders (FASD).
Methods: Brain tissue was collected following a 4-hour maternal separation stress during the early postnatal period, postnatal days 10 and 12, from mice subjected to 1st and 2nd trimester prenatal alcohol exposure (PAE).
Hypothesis: We hypothesized that PAE leads to glucocorticoid receptor insensitivity, stress molecule overexpression, and potentiated toll-like receptor 4 (TLR4) pathway inflammatory responses in the brain following early life stress.
Results: PAE and stress increased mRNA expression of TLR4 …
