Exosomes Secreted Under Hypoxia Enhance Stemness In Ewing’S Sarcoma And Transform Microenvironment Cells, Matthew J. Kling
Exosomes Secreted Under Hypoxia Enhance Stemness In Ewing’S Sarcoma And Transform Microenvironment Cells, Matthew J. Kling
Theses & Dissertations
Intercellular communication between tumor cells and stroma within the hypoxic microenvironment promote aggressiveness and poor patient prognoses in ways that remain unclear. Here we show that hypoxic Ewing’s sarcoma (EWS) cells release exosomes that promote sphere formation, a stem-like phenotype, in EWS cells by enhancing survival and a cancer-associated phenotype in fibroblasts (CAFs). Given that hypoxia enriches for stem-like cells, we exposed EWS cells to hypoxia and hypoxic EWS-derived exosomes and assessed the stem cell phenotype. Hypoxia and hypoxic EWS-derived exosomewere found to enhance TIC formation in vitro and in vivo by reprogramming EWS cells. Analysis of the hypoxic exosomal …

Currently, a global pandemic era of public health concerns is going on with the Coronavirus Disease 2019 (COVID-19), which is caused by severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2). The first case of COVID-19 was reported from Wuhan’s Huanan seafood market in China late December 2019. Bats, pangolins, and snakes have been nominated as salient carriers of the virus. Thanks to its high pathogenicity, it can cause severe respiratory infections. Fever, dry cough, sore throat, pneumonia, septic shock, and ground-glass opacities are the foremost clinical manifestations of COVID-19. Immunocompromised patients are at high risk for COVID-19 infection and may lead to …

Representing And Performing Pilgrimage In A Comic Book: On The Camino, Lucrezia Lopez
International Journal of Religious Tourism and Pilgrimage
Over the last few decades, comic books and graphic novels have become valid literary sources within the humanities and social sciences. This paper addresses a new creative and performative artistic expression of the European pilgrimage route the Camino de Santiago in Spain through examining the autobiographical graphic novel On the Camino, written by the Norwegian cartoonist Jason (2017), which introduces a new way of sharing pilgrimage experiences through combining pictorial and literary devices. Here, the focus is on the dynamic paradigm that arises beyond the ‘fixed sequential images’ in the graphic novel and the role of readers in the spatial …

“This Trip Is Very Meaningful To Me, So I Want To Remember It Forever”: Pilgrim Tattoos In Santiago De Compostela, Christian Kurrat, Patrick Heiser
International Journal of Religious Tourism and Pilgrimage
Pilgrim tattoos have come into fashion: in Santiago de Compostela, the destination of all Ways of St. James, tattoo studios are springing up and in social networks, corresponding photographs can be found more and more often. In this paper we present the results of a survey of pilgrims who have been tattooed after their pilgrimage (N=256). It turns out that certain symbols and body parts are particularly popular among pilgrim tattoos. The tattooing practice of pilgrims also depends strongly on age, nationality and previous tattoos. The central features of the pilgrimage itself, on the other hand, have only a weak …

The Imprint Of The Pilgrimage An Ethnography Of A Tattoo Studio In Jerusalem, Mustafa Diktaş
International Journal of Religious Tourism and Pilgrimage
Several major religions exhibit complex attitudes towards self-mutilation and adornment. Judaism, Islam, and Christianity forbid marking the body and associate it with sin. Still, many people apparently have continued to feel a need for confirmation of their religion and their religious journeys by marking their bodies. This ethnographic study focuses on the recent situation in pilgrimage tattooing, utilising the local and daily processes of a tattoo shop called Razzouk Tattoo located in the Christian quarter of the old city of Jerusalem. The aim of this paper is not only to give a panoptic view of the tattoo studio but also …
Symbolism Of Temple Gates In Ancient Israel, Talitha Hart
Symbolism Of Temple Gates In Ancient Israel, Talitha Hart
Studia Antiqua
The gates of the city and the temple establish boundaries between inner and outer space, while also allowing access to an area that is clearly separated from its surroundings. Throughout ancient Israel, the city gate was seen as representing economic activity, belonging, justice, and strength. I would argue that the gate of the temple represented many of the same things and was seen in a similar way. I have decided to include the tabernacle, as well as both Solomon’s and Herod’s temples, in this analysis, as they seem to have been seen in a similar light even if they were …
Covenant Peoples, Covenant Journeys: Archetypal Similarities Between The Noah, Abraham, And Moses Narratives, Jeremy Madsen
Covenant Peoples, Covenant Journeys: Archetypal Similarities Between The Noah, Abraham, And Moses Narratives, Jeremy Madsen
Studia Antiqua
The stories of Noah, Abraham, and Moses display remarkable similarities. All three follow a narrative pattern where God appears in theophany to a prophet-patriarch figure, God forms a covenant with this prophet-patriarch and his people to bring them to a new land, and God guides them on a divinely-assisted journey until they reach that land. Rather than being the result of typological shaping or historical resemblance, the narrative similarities between these three stories are most likely indicative of a common narrative archetype, which this paper titles the covenant journey archetype. The thrice-fold repetition of this archetype within the Pentateuch attests …

Pharmacology, Physiology and Toxicology
Obesity has been a worldwide epidemic for decades. Despite the abundant increase in knowledge regarding the etiology and pathogenesis of obesity, the prevalence continues to rise with estimates predicting considerably higher numbers by the year 2030. Obesity is characterized by an abnormal lipid accumulation, however, the physiological consequences of obesity are far more concerning. The development of the obesity phenotype constitutes dramatic alterations in adipocytes, along with several other cellular mechanisms which causes substantial increase in systemic oxidative stress mediated by reactive oxygen species (ROS). These alterations promote a chronic state of inflammation in the body caused by the redox …

Testing The Combined Module Swapping And Repair By Modification Strategies: A Step Toward Universal Toolbox, Catherine A. Martini
Testing The Combined Module Swapping And Repair By Modification Strategies: A Step Toward Universal Toolbox, Catherine A. Martini
Biology Theses
The crux of synthetic biology is the engineering of biological components to modulate the activity of specific DNA-based promoter(s) to drive gene expression; thus, providing a means to modulate pathways between signal detection and cellular response. However, the longstanding frustration of synthetic biologists has been the inability to transfer those engineered components between cellular systems — this lack of “modular universality” or “universal toolbox” impedes research by forcing a metaphorical reinvention of the wheel in new systems. The modular swapping strategy expanded the available “toolbox” with customizable hybrid repressors formed from “swapping” the DNA-recognition modules (DRMs) and environmental-sensing modules (ESMs) …

Off mainland Portugal, the common dolphin (Delphinus delphis) is the most sighted cetacean, although information on this species is limited. The Atlantic coast of Southern Portugal is characterized by an intense wind-driven upwelling, creating ideal conditions for common dolphins. Using data collected aboard whale-watching boats (1929 sightings and 4548 h effort during 2010–2014), this study aims to understand the relationships between abundance rates (AR) of dolphins of different age classes (adults, juveniles, calves and newborns) and oceanographic [chlorophyll a (Chl-a) and sea surface temperature (SST)] variables. Over 70% of the groups contained immature animals. The AR of adults was negatively …

I once met a nun who ran an orphanage in Saigon during the Vietnam War. She told me that near the end of the war, they routinely and without hesitation lied about the children waiting for adoption. They’d say anything to get the kids adopted and away from the bombs and the guns. You know who’s better at padding resumes to promote adoption? Those people who write for animal shelters.

Searching For New Analgesics Without Addiction Risks, Mohammad Atiqur Rahman
Theses and Dissertations
Opioids are widely used to treat acute and chronic pain. But opioid addiction to these compounds can cause social and life-threatening health problems, including the risk of overdose. In this thesis, I evaluated IBNtxA (3-iodobenzoyl naltrexamine), a novel mu opioid receptor (MOR) agonist structurally related to the classical MOR antagonist naltrexone, in drug discrimination studies in order to better understand its subjective effects and more thoroughly its abuse liability. IBNtxA represents an intriguing lead compound for preclinical drug development specifically targeting MOR splice variants, potentially creating effective analgesics with reduced side effects. These results indicate that IBNtxA produces potent antinociception …

Complement Activation By C-Reactive Protein Is Critical For Protection Of Mice Against Pneumococcal Infection, Sanjay K. Singh, Donald N. Ngwa, Alok Agrawal
ETSU Faculty Works
C-reactive protein (CRP), a component of the innate immune system, is an antipneumococcal plasma protein. Human CRP has been shown to protect mice against infection with lethal doses of Streptococcus pneumoniae by decreasing bacteremia. in vitro, CRP binds to phosphocholine-containing substances, such as pneumococcal C-polysaccharide, in a Ca2+-dependent manner. Phosphocholine-complexed human CRP activates the complement system in both human and murine sera. The mechanism of antipneumococcal action of CRP in vivo, however, has not been defined yet. In this study, we tested a decades-old hypothesis that the complement-activating property of phosphocholine-complexed CRP contributes to protection of mice against pneumococcal infection. …
