Theses & Dissertations

The main objective of this body of work is to develop polymeric drug delivery systems for cancer radiotherapy and the decorporation of radiological materials. The polymeric systems for radiotherapy were evaluated in vitro and in vivo (in normal, prostate, and ovarian cancer mice models). The polymeric system for radionuclide decorporation was also evaluated in vitro and in a normal mouse model. Due to its attractive properties, the N-(2-Hydroxypropyl) methacrylamide (HPMA) copolymer was utilized as the main carrier for the developed systems.

Chapter 1 provides an overview of prostate and ovarian cancer, targeted radiopharmaceuticals, nanomedicine-based drug delivery for cancer, HPMA copolymers, …

Targeted Therapies In Select Gastrointestinal Cancers And Cancer Cachexia, Scott Mulder Dec 2020

Targeted Therapies In Select Gastrointestinal Cancers And Cancer Cachexia, Scott Mulder

Theses & Dissertations

Hepatocellular carcinomas exhibit metabolic alterations to support their proliferative and biosynthetic needs. We identified that elevated expression of the mitochondrial oxidative carboxylase, malic enzyme 2 (ME2), correlates with poorer hepatocellular carcinoma patient survival. Hepatocellular carcinoma patient tumors with high ME2 expression exhibit transcriptomic alterations indicative of PI3K/AKT/mTOR and c-Myc signaling as well as elevated central carbon, fatty acid, and redox metabolism pathways. Depletion of ME2 in the hepatocellular carcinoma cell line PLC or in the livers of mice treated with diethylnitrosamine to chemically induce hepatocellular carcinomas, results in impaired proliferation and reduced tumor formation. Additionally, the loss of …

Temporal Thermal Heterogeneity As A Potential Evolutionary Driver Of Thermoregulation In The Generalist Grasshopper Melanoplus Differentialis From Five Sites In The Midwestern And Southern United States, Devin Preston Dec 2020

Temporal Thermal Heterogeneity As A Potential Evolutionary Driver Of Thermoregulation In The Generalist Grasshopper Melanoplus Differentialis From Five Sites In The Midwestern And Southern United States, Devin Preston

LSU New Orleans Theses and Dissertations

Environmental drivers of the evolution of thermoregulation, especially thermal fluctuation, have been incompletely investigated. I used a combination of field and laboratory studies to determine if thermal heterogeneity selected for greater thermal tolerance or ability to thermoregulate in a generalist grasshopper by testing upper and lower thermal limits, behavioral thermoregulation, and acclimation. I measured these from individuals originating from sites that had varying degrees of thermal fluctuation over time. I found partial support for thermal heterogeneity as an evolutionary driver of thermal tolerance, behavioral thermoregulation, and acclimation. However, whether or not differences in these metrics conformed to my hypothesis varied …

Assessing Differentiation Of Self In Multiple Cultural Contexts: Initial Validation Of The Multicultural Differentiation Of Self Inventory (Mdsi), Nasima Rahman Khan Dec 2020

Assessing Differentiation Of Self In Multiple Cultural Contexts: Initial Validation Of The Multicultural Differentiation Of Self Inventory (Mdsi), Nasima Rahman Khan

LSU New Orleans Theses and Dissertations

Researchers have shown that current instruments used to assess an individual’s level of differentiation of self or differentiation are biased towards an individualistic cultural context, therefore current instruments lack cultural validity (Alaedein, 2008; Chung & Gale, 2006, 2009; Kim et al., 2015; O’Hara & Meteyard, 2011). When counseling from Bowen’s family systems theoretical perspective, accurate assessment of an individual’s differentiation is integral for treatment planning and intervention (Bowen & Kerr, 1988). To address the existing gap in assessment of differentiation within multiple cultural contexts (i.e., individualistic, collectivistic, transcultural), the purpose of the present research was to develop the Multicultural Differentiation …

Scavenging Of Animal Carcasses By Gumaga Nigricula (Sericostomatidae, Trichoptera), An Apparent Herbivore, Stephanie M. Carlson, Pablo Rodriguez-Lozano, Hana Moidu, Robert A. Leidy Dec 2020

Scavenging Of Animal Carcasses By Gumaga Nigricula (Sericostomatidae, Trichoptera), An Apparent Herbivore, Stephanie M. Carlson, Pablo Rodriguez-Lozano, Hana Moidu, Robert A. Leidy

Western North American Naturalist

Caddisflies within the genus Gumaga are generally considered shredders (i.e., consumers of leaf litter) or grazers (i.e., consumers of algae). We report on observations of Gumaga nigricula scavenging animal carcasses within and adjacent to isolated pools along intermittent reaches of Coyote Creek, Santa Clara County, California, USA. In particular, during the summer of 2018, we observed G. nigricula scavenging fish carcasses within isolated pools as well as bits of flesh from the bones of a black-tailed deer (Odocoileus hemionus columbianus) deposited on the stream bank. To the best of our knowledge, our observations represent the first published record …

'Some Few Miles From Edinburgh': Commemorating The Scenes Of The Gentle Shepherd In Ramsay Country, Craig Lamont Dec 2020

'Some Few Miles From Edinburgh': Commemorating The Scenes Of The Gentle Shepherd In Ramsay Country, Craig Lamont

Studies in Scottish Literature

Traces the history of Ramsay commemoration, from the obelisk at Penicuik with an inscription from 1759 onwards, and successive attempts to identify actual settings for scenes and incidents in his ballad opera The Gentle Shepherd, in illustrations (notably by David Allan for the Foulis edition of 1788), other editions and memoirs, and competing contributions by the ministers of rival parishes to the Statistical Account of Scotland, giving particular attention to the roles in the "battle over Ramsay country" of two local landowners, Alexander Fraser Tytler (Lord Woodhouselee) and Robert Brown of Newhall; providing maps and illustrations to clarify …

'Some Pastoral Improvement' In The Gentle Shepherd: Mediation, Remediation, And Minority, Steve Newman Dec 2020

'Some Pastoral Improvement' In The Gentle Shepherd: Mediation, Remediation, And Minority, Steve Newman

Studies in Scottish Literature

This essay shows how in The Gentle Shepherd Allan Ramsay engages in the complex work of "pastoral improvement" on an individual and national scale and foresees--to a point--how his work will be received in the decades and even centuries to come. After situating his work within the uprising of the Galloway Levellers, pastoral, and the early work of agricultural improvement, I consider how the concept of improvement shapes the reception of his work in the Linley-Tickell production of the 1780s--including a surprising appearance from the Shakespearean forger, William Henry Ireland--and the key role The Gentle Shepherd plays in "The Young …

Morphological Assessment And Mitochondrial Dna Analysis Of Roadside-Skippers (Genus Amblyscirtes, Family Hesperiidae) Occurring In Arkansas, Sachith Srikanth

Theses and Dissertations

Amblyscirtes linda is a candidate for federal listing under the Endangered Species Act (1973). The present study, funded by the U.S. Fish and Wildlife Service and the Arkansas Game and Fish Commission, aims to gather data that will enable a decision regarding federal listing to be made. It is understood that the conservation status of Amblyscirtes linda cannot be accurately assessed without examining other congeneric species with which it cooccurs and can be easily confused. This confusion is due to morphological characteristics being very similar in the genus. We therefore studied morphological and molecular means of identification, as well as …

Biological Pretreatment Of Biomass For Enhanced Biogas Production, Skylar F. Van Horne Dec 2020

Biological Pretreatment Of Biomass For Enhanced Biogas Production, Skylar F. Van Horne

Undergraduate Honors Theses

The biogas output efficiency of controlled anaerobic digestion (AD) can be enhanced by several different pretreatment methods to produce renewable energy. According to the literature, there are pros and cons to each pretreatment method. Biological pretreatment provides environmentally safe and renewable means for enhancing biodegradability and net energy output. Current research has yet to reliably demonstrate the ability of bacterial strains, such as Caldicellulosiruptor bescii (C. bescii), to digest common waste substrates in efficient ways. Novel methods in key intermediate molecular quantification and alkalinity tracking are used here to explore C. bescii’s ability to digest lignocellulosic material in …

Influence And Innovation: Beethoven's Impact On The Sonatas For Piano And Cello By Mendelssohn And Chopin, Patrick T. Bellah Dec 2020

Influence And Innovation: Beethoven's Impact On The Sonatas For Piano And Cello By Mendelssohn And Chopin, Patrick T. Bellah

Dissertations, 2020-current

The bulk of the scholarship in this paper centers around Beethoven’s five sonatas written for piano and cello and how he established a new normal within the genre. This is evidenced by what are arguably the two most noteworthy sonatas for the same instrumental medium, written by Mendelssohn and Chopin, following Beethoven’s death. I posit that the five sonatas written by Beethoven establish a series of models upon which the latter two works by his successors are based.

Chapters two and three of this document are separated into subsections that detail the plausibility of Beethoven’s influence through circumstantial evidence, musical …

Long-Acting Nanoformulated Antivirals For The Treatment And Prevention Of Hiv-1 And Hbv Infections, Dhruvkumar M. Soni Dr.

Theses & Dissertations

While antiretroviral therapy (ART) has revolutionized treatment and prevention of human immunodeficiency virus type one (HIV-1) infection, regimen adherence, viral mutations, drug toxicities, stigma and pill fatigue are limitations. These have led to the development of long acting (LA) ART. These include, but are not limited to, implantable devices, new chemical entities, prodrug modifications and nanoformulations. To these ends, this thesis focues on the transformation of nucleoside reverse transcriptase inhibitors (NRTIs) into LA parenterals, While elusive, data from our laboratories demonstrated that modifications to the PROdrug and nucleoTide technology (ProTide) enables improvements in drug apparent half-life and tissue and cell …

Functional roles of neutrophil elastase (NE) have not been examined in distinct steps of the metastatic cascade. NE, delivered to primary tumors as a purified enzyme or within intact neutrophils or neutrophil granule content, enhanced human tumor cell intravasation and subsequent dissemination via NE-mediated formation of dilated intratumoral vasculature. These effects depended on picomole range of NE activity, sensitive to its natural inhibitor, α1PI. In

Endoplasmic reticulum (ER) stress is known to induce pro-inflammatory response and ultimately leads to cell death. Mesencephalic astrocyte-derived neurotrophic factor (MANF) is an ER-localized protein whose expression and secretion is induced by ER stress and a crucial survival factor. However, the underlying mechanism of how MANF exerts its cytoprotective activity remains unclear due to the lack of knowledge of its receptor. Here we show that Neuroplastin (NPTN) is such a receptor for MANF. Biochemical analysis shows the physiological interaction between MANF and NPTN on the cell surface. Binding of MANF to NPTN mitigates the inflammatory response and apoptosis via suppression …

A better understanding of the negative impact of general anesthetics on gastrointestinal motility requires thorough knowledge of their molecular targets. In this respect the muscarinic cationic current (mICAT carried mainly via TRPC4 channels) that initiates cholinergic excitation-contraction coupling in the gut is of special interest. Here we aimed to characterize the effects of one of the most commonly used “dissociative anesthetics”, ketamine, on mICAT. Patch-clamp and tensiometry techniques were used to investigate the mechanisms of the inhibitory effects of ketamine on mICAT in single mouse ileal myocytes, as well as …

Pharmaceuticals And Personal Care Products In Pacific Northwest Coastal Ecosystems: Spatial Variation, Organism Effects, And Consumer Perspectives, Amy Lynne Ehrhart

Dissertations and Theses

Anthropogenic pollution poses a threat to marine organisms and ecosystems worldwide. Common chemical pollutants that enter the marine environment include legacy contaminants, which are well known and heavily regulated or banned pollutants, and emerging contaminants, which are more recently recognized as pollutants and often lack regulatory limits for their use and discharge. Wastewater treatment plant (WWTP) effluent is a major source of various contaminants of concern, particularly pharmaceutical and personal care products (PPCPs) that are not fully removed during treatment. PPCPs exist at low concentrations in the environment and may have unknown and subtle effects on marine life. Data gaps …

Mechanisms By Which Mnte-2-Pyp Suppresses Prostate Cancer Cell Growth, Yuxiang Zhu Dec 2020

Mechanisms By Which Mnte-2-Pyp Suppresses Prostate Cancer Cell Growth, Yuxiang Zhu

Theses & Dissertations

Prostate cancer patients are often treated with radiotherapy. MnTE-2-PyP, is a superoxide dismutase (SOD) mimic and a known radioprotector of normal tissues. Our recent work demonstrates that MnTE-2-PyP also inhibits prostate cancer progression with radiotherapy; however, the mechanisms remain unclear. In this thesis, we identified that MnTE-2-PyP-induced intracellular H2O2 levels are critical in inhibiting growth of prostate cancer cells. We found that MnTE-2-PyP induced protein oxidations in PC3 cells and one major group of oxidized protein targets were involved in energy metabolism. The oxidative phosphorylation rates were significantly enhanced in both PC3 and LNCaP cells with MnTE-2-PyP treatment, but mitochondrial …

Comparison Of Cardiac Muscle Between Male And Female Rats With Exercise And Aging: Analyzing Vitals, Gdnf Expression, Ngf Expression, Rna Expression, Sympathetic Innervation, Parasympathetic Innervation, And Sensory Innervation, Katelyn Kurrie

Honors Theses

The direction of my research-based thesis answers the question of how exercise affects the cardiovascular system with aging in male and female rats. My study would highlight the important differences of the cardiovascular system of both sexes which could lead to sex-specific insight on the cardiovascular disease epidemic which prevails as the most common cause of death in the United States. The studies completed will test the hypothesis: neurotrophic factor will be higher in females than males due to the higher presence of estrogen in females. The trophic factor is predicted to increase in both males and females with regular …

A Rapid Response Vaccine Development Strategy For Newcastle Disease In Poultry, Sr R. Maremagae, Cc C. Bezuidenhout, Am M. Murphy, Ma A. Jarvis, P Naicker, P. J. Lebea

School of Biomedical Sciences

Poultry is a major source of protein in sub-Saharan Africa and many other lower-income regions. Newcastle disease virus (NCDV) comprises a significant threat toward poultry production. While NCDV vaccines are routinely used in developed countries, those used in sub-Saharan Africa are mostly imported and are not specific to locally circulating strains. Indeed, the lack of rapid, field-based NCDV detection and the absence of cost-effective production methods for pure, strain-specific vaccines hampers efficient poultry production throughout these regions. This remains a major problem for both subsistence and commercial farming. The aim for this study was firstly, to develop a field-based isothermal PCR …

Intrahistiocytic Storage Of Clofazimine Crystals In A Cat, Nathan D. Helgert, Debra L. Miller, Jacqueline C. Whittemore, Mee-Ja M. Sula

Faculty & Staff Research and Creative Activity

A 13-year-old castrated male Maine coon cat with a 5-year history of atypical mycobacteriosis was euthanized and submitted for necropsy. The cat had been kept in clinical remission since diagnosis using a combination of the antimycobacterial drug clofazimine and additional multimodal antimicrobial therapy. Grossly, tissues were diffusely discolored red-brown to yellow. Histologically, the myocardial interstitum was expanded by numerous, often multinucleated cells, which were distended by uniformly shaped acicular cytoplasmic spaces. These cells were immunopositive for CD18 and immunonegative for desmin, suggesting a histiocytic rather than muscular origin. Macrophages in other tissues contained similar acicular spaces. Ultrastructurally, the spaces were …

Expanding The Boundaries Of Food Policy: The Turn To Equity In New York City, Nevin Cohen, Rositsa Ilieva

Publications and Research

Policymakers acknowledge that the food system is multidimensional and that social determinants affect diet-related health outcomes, yet cities have emphasized programs and policies narrowly connected to food access and nutritional health. Over the past fifteen years, the boundaries of food governance have expanded to include a wider range of issues and domains not previously considered within the purview of food policy, like labor, housing, and education policies. This paper illustrates the processes by which this shift occurs by presenting the case of New York City, which has broadened its food governance to a larger set of issues, requiring cross-sectoral initiatives …

Age and sex differences in brain metabolite concentrations in early life are not well under- stood. We examined the associations of age and sex with brain metabolite levels in healthy neonates, and investigated the associations between neonatal brain metabolite concentrations and developmental outcomes. Forty-one infants (36–42 gestational weeks at birth; 39% female) of predominantly Hispanic/Latina mothers (mean 18 years of age) underwent MRI scanning approximately two weeks after birth. Multiplanar chemical shift imaging was used to obtain voxel-wise maps of N-acetylaspartate (NAA), creatine, and choline concentrations across the brain. The Bayley Scales of Infant and Toddler Development, a measure of …
