Railspace: A Geocritical Study Of The Railroad Through American Literature And Culture, Michael A. Smith

This dissertation uses geocriticism to argue that the American railroad is best understood as a set of discursively constructed railspaces formed through a variety of viewpoints, a polysensorial awareness of space, and stratified social relationships and power struggles. This study takes up four railspaces, the constituent texts of which demonstrate how intertextual discourse shapes and is shaped by the railroad. The observation car, charted through California Zephyr advertisements and Muriel Rukeyser’s “Campaign,” is an apparatus that produces perpetual spectacle. These organizations have partnered with companies to create monetary value for the team and the companies, which has led sponsorship from traditional advertising in the arena or venue of the professional sports organization to their team’s Twitter account. Breast cancers of the claudin-low subtype make up a substantial portion of triple-negative breast cancers and have stem cell-like and mesenchymal features. Although it has been recognized for some time that this breast cancer subtype is highly aggressive and difficult to treat, the molecular drivers and cellular origin of claudin-low breast cancer have been poorly defined. The lack of suitable in vivo models has prohibited the study of tumor initiation and progression of this subtype. A critical barrier to achieving a functional cure for infection by human immunodeficiency virus type one (HIV-1) rests in the presence of latent proviral DNA integrated in the nuclei of host CD4+ T cells and mononuclear phagocytes. Accordingly, HIV-1-infected patients must adhere to lifelong regimens of antiretroviral therapy (ART) to prevent viral rebound, CD4+ T cell decline, and progression to acquired immunodeficiency syndrome (AIDS). Classroom management, although considered one of the essentials of the optimal learning experience, is often a challenge for teachers. Certain classroom management strategies, such as group contingency interventions and token economies, are evidence-based strategies to aid teachers in gaining successful management of the classroom. The utilization of Class Dojo, a popular classroom management website, as a mechanism to implement an interdependent group contingency (IGC) intervention with randomized components may increase both the effectiveness of the intervention and the social validity of the intervention when compared to a manual implementation.

Mystical texts often present themselves as possessing a “donative” intent, in which the writer aims to share the significance of their own experience. This dissertation will show how a consideration of donative intent and its textual results can enrich the study of medieval and early-modern mystical writings, approaching them from the broad concept of triangular mimetic desire in conjunction with other theoretical insights in order to examine the methods through which writers seek to portray their mediator-object relationship with God and inspire reading subjects’ mimetic desire.

High fluoride concentrations in drinking water affect millions of people around the world. The World Health Organization recommended a fluoride concentration in drinking water of ≤ 1.5 mg/L; fluoride above this concentration can cause long-term dental and skeletal fluorosis. Fluoride imparts no taste to water; therefore, people do not realize that they are ingesting high fluoridated water. Researchers sought to develop a material to remove fluoride from drinking water through sorption. Aluminum was used to form a double ionic layer on the bentonite clay surface. The inverse relationship of plasma bilirubin levels with liver fat accumulation has prompted the possibility of bilirubin as a therapeutic for non-alcoholic fatty liver disease. Here, we used diet-induced obese mice with non-alcoholic fatty liver disease treated with pegylated bilirubin (bilirubin nanoparticles) or vehicle control to determine the impact on hepatic lipid accumulation. The bilirubin nanoparticles significantly reduced hepatic fat, triglyceride accumulation, de novo lipogenesis, and serum levels of liver dysfunction marker aspartate transaminase and ApoB100 containing very-low-density lipoprotein. The bilirubin nanoparticles improved liver function and activated the hepatic β-oxidation pathway by increasing PPARα and acyl-coenzyme A oxidase 1. …

Cerebral palsy (CP) is a permanent, non-progressive neuromuscular disorder diagnosed early in childhood. Frequently the lesion that causes the motor impairments in individuals with CP concurrently disrupts the visual networks, placing them at a high risk of cerebral visual dysfunctions. Cerebral visual impairment (CVI) often remains unrecognized or misdiagnosed in people with CP. Despite the crucial role of visual function in the development of movement and cognition, the neurophysiological basis of the cerebral visual dysfunctions is almost entirely unknown. Antiretroviral therapy (ART) has improved the quality and duration of life for people living with human immunodeficiency virus (HIV) infection. However, limitations in drug efficacy, the emergence of viral mutations and the paucity of cell-tissue targeting remain. We posit that to maximize ART potency and therapeutic outcomes newer drug formulations that reach HIV cellular reservoirs need to be created. In a step towards achieving this goal we discovered the aggregation-induced emission (AIE) property of the non-nucleoside reverse transcriptase inhibitor Rilpivirine (RPV) and used it as a platform for drug cell and subcellular tracking. Genome integrity is necessary to prevent mutations and disease. During eukaryotic DNA replication, DNA polymerases ε (Polε) and δ (Polδ) synthesize the leading and lagging strand, respectively. Polε and Polδ also have exonuclease activity that acts in series with post-replicative mismatch repair (MMR) to remove replication errors. Defects in proofreading and MMR lead to an increase in mutations and cause cancer in humans. This dissertation focuses on several unresolved issues involving the relationship between Polε and Polδ in replication error avoidance. The deregulation of cell surface glycoproteins, such as mucins, is a hallmark of many tumors of epithelial origin. The transmembrane mucin MUC4, is differentially overexpressed in several malignancies, including pancreatic, breast, ovarian, lung, cervical, and head and neck cancers. Of all the aforementioned cancers, the role of MUC4 has been the most thoroughly in pancreatic cancer (PC), which is the 4th leading cause of cancer-related deaths in the United States. While MUC4 is undetectable in the normal or inflamed pancreas (pancreatitis), its expression progressively increases during PC progression and its higher expression correlates with poor survival. Diabetes mellitus (DM) increases the risk of heart failure 2-4-fold. Intensive glycemic control does not abate this risk in DM patients, suggesting that DM-induced heart failure is beyond hyperglycemia. DM silently induces cardiac muscle disorder termed diabetic cardiomyopathy (DMCM), which progresses to heart failure. The objective of this dissertation is to understand the mechanistic underpinnings of DMCM to develop new solutions to diagnose and treat heart failure in DM.

The heart pumps blood due to the contraction and relaxation of terminally differentiated cardiomyocytes with limited regenerative capacity. The DM environment of hyperglycemia, hyperlipidemia, and inflammation causes cardiomyocyte death and leads …
