A comprehensive understanding of spaceflight factors involved in immune dysfunction and the evaluation of biomarkers to assess in-flight astronaut health are essential goals for NASA. An elevated neutrophil-to-lymphocyte ratio (NLR) is a potential biomarker candidate, as leukocyte differentials are altered during spaceflight. In the reduced gravity environment of space, rodents and astronauts displayed elevated NLR and granulocyte-to-lymphocyte ratios (GLR), respectively. To simulate microgravity using two well-established ground-based models, we cultured human whole blood-leukocytes in high-aspect rotating wall vessels (HARV-RWV) and used hindlimb unloaded (HU) mice. Background: Congenital muscular dystrophies (CMD) are a clinically and genetically heterogeneous group of neuromuscular disorders characterized by muscle weakness. The two most prevalent forms of CMD, collagen VI-related myopathies (COL6RM) and laminin α2 deficient CMD type 1A (MDC1A), are both caused by deficiency or dysfunction of extracellular matrix proteins. Previously, we showed that an intramuscular transplantation of human adipose-derived stem cells (ADSC) into the muscle of the Col6a1-/- mice results in efficient stem cell engraftment, migration, long-term survival, and continuous production of the collagen VI protein, suggesting the feasibility of the systemic cellular therapy for COL6RM. Diarrheal disease remains a major cause of childhood mortality and morbidity causing poor health and economic outcomes. In low-resource settings, young children are exposed to numerous risk factors for enteric pathogen transmission within their dwellings, though the relative importance of different transmission pathways varies by pathogen species. The objective of this analysis was to model associations between five household-level risk factors-water, sanitation, flooring, caregiver education, and crowding-and infection status for endemic enteric pathogens in children in five surveillance studies. The anti-diabetic property of aqueous leaf extract of Moringa oleifera was performed in streptozocin-induced diabetic rats using serum chemistry, histology and immunochemical parameters as indices of diabetes. The blood glucose level of the diabetic untreated group continues to increase while that of the treated group after 21 days decreased. While the animals in the diabetic untreated group experienced increase in the levels of markers of organ damage when compared to the control group (P values < 0.0001). ALT increased from 61.83±1.5 to 96.1±22.4, AST was 225.1±26.6 from 172.6±13.9, ALP 13.5±0.006 to 13.6±0.002, UREA 1.0±0.08 to 3.0±0.4, their reduction was observed in the extract-treated groups. ALT reduced from 96.1±22.4 to 73.70±9.7; AST from 225.1±26.6 to 184.4±18.2; ALP from 13.6±0.002 to 13.6±0.01; UREA from 3.0±0.4 to 2.0±0.4. Treatment with the extract significantly reduced markers of oxidative stress in the kidney [hydrogen peroxide (898.8±6.26 to 688.0±13.7), malondialdehyde (640±0.1 to 600±0.2) and protein carbonyl (548.4±1.5 to 458.1±1.6)]; heart [hydrogen peroxide (389.4±1.8 to 358.2±1.5), malondialdehyde (264.0±0.5 to 122.0±0.3), protein carbonyl (196.8±0.5 to 162.7±3.5)]; and liver [hydrogen peroxide (119.36±3.2 to 103.94±10.7), malondialdehyde (236.0±0.4 to 73.0±0.2), protein carbonyl (269.3±1.0 to 174.2±1.1) respectively. The levels of antioxidants were reduced in the diabetic untreated group but there was increase in the Moringa treated group. Glucose transporter 4 (GLUT 4) was down regulated in the diabetic untreated group while it was well expressed in the treated groups. The histology of pancreas and liver showed varied levels of infiltration of inflammatory cells, congestion and necrotic lesions, but these were mild in the treated groups. The result shows that the extract does have an anti-diabetic effect with the decrease in the levels of blood glucose and markers of oxidative stress as well as increase in the amount of antioxidants in the treated group when compared to the diabetic untreated group. More importantly, the extract caused upregulation of GLUT 4, which is relevant in reversing insulin resistance in the same manner as pioglitazone, the standard antidiabetic agent used in this study.

Background: GMC1 (2-(1H-benzimidazol-2-ylsulfanyl)-N-[(Z)-(4-methoxyphenyl) methylideneamino] acetamide) effectively inhibits androgen receptor function by binding directly to FKBP52. This is a novel mechanism for the treatment of castration resistant prostate cancer (CRPC). Methods: an LC-MS/MS method was developed and validated to quantify GMC1 in plasma and urine from pharmacokinetics studies in rats. Background: Splicing factor 1 (SF1) is a conserved alternative splicing factor expressed in many different mammalian cell types. The genetically modified Sf1+/− (or Sf1β-geo/+ ) mice express reduced levels of SF1 protein in mouse tissues, including in cells of the intestines. Mutational inactivation of human adenomatous polyposis coli (APC) gene deregulates the Wnt signaling pathway and is a frequent genetic event in colon cancers. Genetic mutations and aberrant epigenetic alterations are the triggers for carcinogenesis. The emergence of the drugs targeting epigenetic aberrations has provided a better outlook for cancer treatment. Histone deacetylases (HDACs) are epigenetic modifiers playing critical roles in numerous key biological functions. Single-cell RNA-sequencing (scRNAseq) technologies are rapidly evolving. Although very informative, in standard scRNAseq experiments, the spatial organization of the cells in the tissue of origin is lost. Conversely, spatial RNA-seq technologies designed to maintain cell localization have limited throughput and gene coverage. Mapping scRNAseq to genes with spatial information increases coverage while providing spatial location. However, methods to perform such mapping have not yet been benchmarked. PURPOSE: This study assessed how individuals compensate for energy expended during a 12-wk aerobic exercise intervention, elucidating potential mechanisms and the role exercise dose plays in the compensatory response.

PARTICIPANTS AND DESIGN: Three-arm, randomized controlled trial among sedentary adults age 18 to 40 yr, body mass index of 25 to 35. Groups included six exercise sessions per week, two sessions per week, and sedentary control.

Development of numerical models to predict stormwater-mediated transport of pathogenic spores in the environment depends on an understanding of adhesion forces that dictate detachment after rain events. Zeta potential values were measured in the laboratory for Bacillus globigii and Bacillus thuringiensis kurstaki, two common surrogates used to represent Bacillus anthracis, in synthetic baseline ultrapure water and laboratory prepared stormwater. Zeta potential curves were also determined for materials representative of urban infrastructure (concrete and asphalt). In response to extreme violence by Myanmar, over one million Rohingya refugees have flooded into Bangladesh, resulting in massive scale deforestation, land erosion, farmland degradation, and destruction of animal habitat, along with the Rohingya being denied basic environmental protections. KDM5B (lysine[K]-specific demethylase 5B) is frequently upregulated in various human cancers including prostate cancer. KDM5B controls H3K4me3/2 levels and regulates gene transcription and cell differentiation, yet the contributions of KDM5B to prostate cancer tumorigenesis remain unknown. In this study, we investigated the functional role of KDM5B in epigenetic dysregulation and prostate cancer progression in cultured cells and in mouse models of prostate epithelium–specific mutant Pten/Kdm5b. Kdm5b deficiency resulted in a significant delay in the onset of prostate cancer in Pten-null mice, whereas Kdm5b loss alone caused no morphologic abnormalities in mouse prostates. Human immunodeficiency virus (HIV) is associated with co-morbid affective and stress-sensitive neuropsychiatric disorders that may be related to dysfunction of the hypothalamic-pituitary-adrenal (HPA) stress axis. The HPA axis is perturbed in up to 46% of HIV patients, but the mechanisms are not known. The neurotoxic HIV-1 regulatory protein, trans-activator of transcription (Tat), may contribute. We hypothesized that HPA dysregulation may contribute to Tat-mediated interactions with oxycodone, a clinically-used opioid often prescribed to HIV patients. Ecotourism opportunities in the marine environment often rely heavily on provisioning to ensure the viewing of cryptic species by the public. However, intentional feeding of wildlife can impact numerous aspects of an animals’ behavior and ecology. Southern stingrays (Hypanus americana) provisioned at Stingray City Sandbar (SCS) in Grand Cayman have altered diel activity patterns and decreased measures of health. This study looked at seasonal changes in stable isotope (SI) and fatty acid (FA) profiles of provisioned stingrays at SCS. Plasma δ15N was higher in male stingrays (11.86 ± 1.71‰) compared to females (10.70 ± 1.71‰). Lower values for …

Coral reefs are home to over 2 million species and provide habitat for roughly 25% of all marine animals, but they are being severely threatened by pollution and climate change. Proteins that regulate the coagulation cascade, including thrombin, are elevated in the brains of Alzheimer's disease (AD) patients. While studies using amyloid-based AD transgenic mouse models have implicated thrombin as a protein of interest, the role of thrombin in tau-based animal models has not been explored. The current study aims to determine how inhibiting thrombin could alter oxidative stress, inflammation, and AD-related proteins in a tau-based mouse model, the Tg4510. In this chapter, we argue that to make sure that human-like robots exhibit human-like behavior, we need to use fuzzy techniques -- and we also provide details of this usage. The chapter is intended both for researchers and practitioners who are very familiar with fuzzy techniques and also for researchers and practitioners who do not know these techniques -- but who are interested in designing human-like robots.
