Restructuring Of The Axon Initial Segment In Mouse Models Of Neurodevelopmental Disorders, Rachel Ali Rodriguez Dec 2019

Restructuring Of The Axon Initial Segment In Mouse Models Of Neurodevelopmental Disorders, Rachel Ali Rodriguez

UNLV Theses, Dissertations, Professional Papers, and Capstones

Neurodevelopmental disorders (NDDs) affect more than 36% of children in countries with low- and middle- incomes (Boivin, 2015; McCoy, 2016). Interestingly, these heterogeneous disorders share a high incidence of epileptic seizures, suggesting a shared pathology. Seizures result when neuronal firing activity becomes disturbed and neurons fire excessively or in unregulated patterns. A key site in the control of neuronal firing patterns is the axon initial segment (AIS), where the local density of proteins and the morphology of the AIS in part determine the firing of neurons. We hypothesized that a disruption in the morphology and/or composition of the AIS can …

Cyclotron Resonance In Graphene Heterostructurescyclotron Resonance In Graphene Heterostructures, Billy Jordan Russell Dec 2019

Cyclotron Resonance In Graphene Heterostructurescyclotron Resonance In Graphene Heterostructures, Billy Jordan Russell

Arts & Sciences Graduate Student Theses and Dissertations

We present observations of cyclotron resonance in graphene Van der Waals heterostructure devices. Such devices provide dramatic improvements in sample quality and allow for ad- vanced electronic control, opening windows on previously inaccessible physics. The design and construction of a dedicated system for the measurement of electronic transport and infrared magnetospectroscopy in microscopic samples of atomically thin materials at cryogenic temperatures is presented. In high-mobility encapsulated monolayer graphene, electron- electron interaction effects are unambiguously observed to impact the interband cyclotron resonance as the Landau level filling factor is varied in a quantizing magnetic field. Additionally, a splitting of transitions involving …

Longitudinal Acoustic Traps: Design, Fabrication, And Evaluation For Biological Applications, Michael Moore Binkley Dec 2019

Longitudinal Acoustic Traps: Design, Fabrication, And Evaluation For Biological Applications, Michael Moore Binkley

McKelvey School of Engineering Graduate Student Theses & Dissertations

Acoustofluidics combine ultrasonic actuation with small-volume microfluidic channels to enable precise, contactless object manipulation for a range of applications from serial chemical processing to blood component separation and single-cell analysis. Micron- to millimeter-scale vibrational waves generate reproducible pressure fields within the microfluidic channels and chambers. By exploiting the material property mismatch between a particle (polymeric and silica beads, cells, etc.) and a suspending fluid, the acoustic radiation force is used to move particles toward regions of low (nodes) or high pressure (antinodes). An understanding of these field-particle interactions is applied to design and implement complicated channel architectures for preferential segregation …

Rhodococcus Opacus Pd630 Genetic Tool Development To Enable The Conversion Of Biomass, Drew Michael Delorenzo Dec 2019

Rhodococcus Opacus Pd630 Genetic Tool Development To Enable The Conversion Of Biomass, Drew Michael Delorenzo

McKelvey School of Engineering Graduate Student Theses & Dissertations

The discovery of fossil fuels facilitated a new era in human history and allowed many firsts, such as the mass production of goods, the ability to travel and communicate long distances, the formation of population dense cities, and unprecedented improvements in quality of life. Alternative sources of energy and chemicals are needed, however, as hydrocarbon reserves continue to deplete and the effects of burning fossils on the planet become better understood. Lignocellulosic biomass is the most abundant raw material in the world and a viable alternative to petroleum-derived products. The pre-treatment of lignocellulose (e.g., thermocatalytic depolymerization, enzymatic hydrolysis, pyrolysis, etc.) …

A Microfluidic Platform To Investigate The Mechanism By Which Gdnf Overexpression In Schwann Cells Causes Neuronal Axon Entrapment, Ze Zhong Wang

McKelvey School of Engineering Graduate Student Theses & Dissertations

Twenty million Americans suffer from peripheral nerve injury (PNI) caused by trauma and medical disorders, with approximately $150 billion spent in annual health-care dollars in the United States. Even with proper surgical reconstruction, less than 50% of the patients achieve satisfactory functional recovery. The gold standard surgical repair for long gaps (>3cm) is the autologous nerve graft, despite its disadvantages such as donor site morbidity, risk of infection, and increased cost. Alternative methods, such as acellular nerve grafts (ANA), are ineffective for large lesion gaps because of the lack of cells and regeneration factors. Recent efforts have been focused …

Three-Dimensional Image Reconstruction In Transcranial Photoacoustic Computed Tomography, Joemini Poudel Dec 2019

Three-Dimensional Image Reconstruction In Transcranial Photoacoustic Computed Tomography, Joemini Poudel

McKelvey School of Engineering Graduate Student Theses & Dissertations

Photoacoustic computed tomography (PACT) is an emerging imaging modality that exploitsoptical contrast and ultrasonic detection principles to form images of the photoacousticallyinduced initial pressure distribution within tissue. The PACT reconstruction problemcorresponds to an inverse source problem in which the initial pressure distribution is recoveredfrom measurements of the radiated pressure wavefield. A major challenge in transcranialPACT brain imaging is compensation for aberrations in the measured data due to the presenceof the skull. Ultrasonic waves undergo absorption, scattering and longitudinal-to-shear wavemode conversion as they propagate through the skull. To properly account for these effects, awave-equation-based inversion method should be employed that can …

The Role Of Multi-Charged Responses: Construction And Application Of A Tandem Differential Mobility Analyzer (Tdma), Christopher Ray Oxford Dec 2019

The Role Of Multi-Charged Responses: Construction And Application Of A Tandem Differential Mobility Analyzer (Tdma), Christopher Ray Oxford

McKelvey School of Engineering Graduate Student Theses & Dissertations

Atmospheric aerosols impact health outcomes, visibility, and the energy balance of the earth. The atmosphere contains a variety of compounds, and the volatility (phase change enthalpy and vapor pressure) of each compound determines its partitioning between the gas phase and the particle phase. The hygroscopicity (an aerosol’s affinity for water) of an atmospheric aerosol particle is determined by the many compounds present in the particle, and thus, the volatility impacts hygroscopicity. Changes in hygroscopicity alter the fraction of the aerosol deposited in the lungs and the fraction of the aerosol activated into cloud droplets. Thus, understanding the volatility and hygroscopicity …

Dynamics And Control In Spiking Neural Networks, Fuqiang Huang Dec 2019

Dynamics And Control In Spiking Neural Networks, Fuqiang Huang

McKelvey School of Engineering Graduate Student Theses & Dissertations

In the brain, neurons (brain cells) produce electrical impulses, or spikes, that are thought to be the substrate of information processing and computation. Through enigmatic processes, these spikes are ultimately decoded into perceptions and actions. The nature of this encoding and decoding is one of the most pervasive questions in theoretical neuroscience. In other words, what are the specific functions enacted by neural circuits, through their biophysics and dynamics? This thesis examines the dynamics of neural networks from the perspective of control theory and engineering. The pivotal concept is that of the normative synthesis of neural circuits, wherein neural dynamics …

Structural And Tissue-Level Adaptations Of Lumbar Intervertebral Discs In Humans With Inducible Low Back Pain Symptoms, Christian Isaiah Weber Dec 2019

Structural And Tissue-Level Adaptations Of Lumbar Intervertebral Discs In Humans With Inducible Low Back Pain Symptoms, Christian Isaiah Weber

McKelvey School of Engineering Graduate Student Theses & Dissertations

Low back pain (LBP) is a devastating musculoskeletal ailment that impacts majority of the global population. Up to 95% of cases are considered nonspecific where the origin is unclear. Although there is a diagnosis with these cases, there lacks specificity that would help individuals with treatment and pain management. Imaging techniques such as magnetic resonance imaging (MRI) are primarily utilized in the diagnosis of LBP as it illuminates internal structures of the spine. One important structure clearly identified with MRI is the intervertebral disc (IVD) that is susceptible to injury and disease. Further, diagnostic imaging is performed lying down where …

Anion Exchange And Bipolar Membranes For Electrochemical Energy Conversion And Storage, Zhongyang Wang Dec 2019

Anion Exchange And Bipolar Membranes For Electrochemical Energy Conversion And Storage, Zhongyang Wang

McKelvey School of Engineering Graduate Student Theses & Dissertations

Anion exchange and bipolar membrane fuel cells generate electrical energy directly from chemical fuels and have attracted considerable interests as alternate power sources for large market applications, such as transportation (hydrogen fuel cells) and unmanned vehicles (sodium borohydride fuel cells). Anion exchange membrane (AEM), generally composed of a polymer with covalently tethered ionic groups, is the central component of the fuel cell serving as the electrolyte, conducting hydroxide ions from cathode to anode, where fast ionic conduction is directly related to power output. However, AEMs currently used in fuel cells (H2 fuel cells and sodium borohydride fuel cells) exhibit ion …

Activation And Regulation Of The Alkbh3-Ascc Alkylation Repair Pathway, Josh Brickner Dec 2019

Activation And Regulation Of The Alkbh3-Ascc Alkylation Repair Pathway, Josh Brickner

Arts & Sciences Graduate Student Theses and Dissertations

DNA repair is essential to prevent the cytotoxic or mutagenic effects of various types of DNA lesions. These lesions are sensed by distinct pathways to recruit repair factors specific to type of damage. In particular, the ALKBH family of proteins recognizes and repairs specific alkylated lesions, including 1-methyladenine (m1A) and 3-methylcytosine (m3C). A major outstanding question in the field is how the AlkB homologue ALKBH3 and its associated protein partners are recruited to sites of alkylation damage and how this repair activity is regulated. Understanding the upstream signaling events that mediate recognition and repair of DNA alkylation damage is particularly …

Wheelchair Propulsion For Everyday Manual Wheelchair Users: Repetition Training And Machine Learning-Based Monitoring, Pin-Wei Chen Dec 2019

Wheelchair Propulsion For Everyday Manual Wheelchair Users: Repetition Training And Machine Learning-Based Monitoring, Pin-Wei Chen

Arts & Sciences Graduate Student Theses and Dissertations

Upper limb pain and injuries are prevalent among manual wheelchair users and can restrict their participation and daily activities. Due to the high repetition and force in wheelchair propulsion, chronic wheelchair propulsion has been linked to the risk of upper limb pain and injury. Prevention of upper limb pain and injury is a high priority in wheelchair-related research. Decades of research in wheelchair propulsion biomechanics have led to clinical practice guidelines (CPG). Unfortunately, a decade after the publication of the CPG, CPG-recommended propulsion is still uncommon. Hence, for the first aim, a randomized controlled trial pilot study with two groups …

Transcriptional Regulation Of Adipose Tissue Development By Pexrap And Med19, John Dean Dec 2019

Transcriptional Regulation Of Adipose Tissue Development By Pexrap And Med19, John Dean

Arts & Sciences Graduate Student Theses and Dissertations

Targeting adipose tissue function to decrease adiposity and improve insulin sensitivity could treat obesity and diabetes. How the nuclear receptor PPARγ regulates the development of two functionally distinct types of adipose tissue, brown and white fat (BAT and WAT), as well as the browning of white fat, remains unclear. Our previous studies suggest that PexRAP, a peroxisomal lipid synthetic enzyme, regulates PPARγ signaling and white adipogenesis. Here, we show that PexRAP is an inhibitor of brown adipocyte gene expression. PexRAP inactivation promoted adipocyte browning, increased energy expenditure, and decreased adiposity. Identification of PexRAP-interacting proteins suggests that PexRAP function extends beyond its …

Young Children’S Knowledge About The Role Of Print In Reading, Molly Farry-Thorn Dec 2019

Young Children’S Knowledge About The Role Of Print In Reading, Molly Farry-Thorn

Arts & Sciences Graduate Student Theses and Dissertations

Children begin to learn about the print in books and the role it plays in reading well before the onset of formal literacy instruction. Young children’s knowledge about precisely what readers are reading when they read books and who is able to read books has been studied primarily through interviews, but conclusions from this research are limited by methodological concerns. Three experiments examined whether pre-readers understand what part of a book is read and whether they distinguish between the skill of reading and the activity of reading. Although pre-readers were typically able to locate the print in a book, they …

T Cell Immunity In Pancreatic Cancer Is Undermined By Dendritic Cell Dysfunction, Samarth Hegde Dec 2019

T Cell Immunity In Pancreatic Cancer Is Undermined By Dendritic Cell Dysfunction, Samarth Hegde

Arts & Sciences Graduate Student Theses and Dissertations

Pancreatic cancer carries a dismal prognosis, and desperately needs viable therapeutic interventions beyond chemo-radiation. T cell-dependent immunotherapies have shown great promise in several tumor types, but have not been effective for the vast majority of pancreatic cancer patients. This is, in part, due to our limited understanding of how antigenicity of pancreatic lesions is recognized, and how adaptive immunity is overcome in this disease. We sought to study tumor-immune interactions and identify mechanisms for this immune-failure using several spontaneous and unperturbed mouse models of pancreatic adenocarcinoma. We found that early pancreatic lesions fail to elicit tumor-limiting CD4+ TH1 and CD8+ …

Crip Time In Fin-De-Siècle Spain: Disability, Degeneration, And Eugenics, Erika Rodriguez Dec 2019

Crip Time In Fin-De-Siècle Spain: Disability, Degeneration, And Eugenics, Erika Rodriguez

Arts & Sciences Graduate Student Theses and Dissertations

A period of intense nation-building, the late nineteenth century was marked by the search for medical and legal solutions to the increasing number of bodies that did not align with culturally constructed expectations of productivity and reproduction in Spanish modernity. Authors of this time used representations of disability to engage in urgent political questions about population control and the rights of individuals in the face of increasing medical intervention. In carrying out this analysis, I raise the question of how representations of disability created a space to reconfigure the social values that determined what lives matter. Focusing on canonical realist …

Antibiotic resistance is an increasing threat in today’s society. In order to overcome resistant bacteria, it is necessary to discover new drugs with novel mechanisms of action. This work focuses on the sideromycin pathway, encompassing the biosynthetic production, mechanism of entry and hydrolysis-mediated drug release. Sideromycins are an interesting approach to combat the rise of antibiotic resistance since they provide a different avenue that overcomes problems that arise when entering the cell. The dissertation is separated into distinct sections dealing with the various areas of interest in the sideromycin pathway, particularly for the sideromycin, salmycin, produced by Streptomyces violaceus. The …

Generation And Utilization Of Diverse Memory B Cells After Flavivirus Challenges, Rachel Wong Dec 2019

Generation And Utilization Of Diverse Memory B Cells After Flavivirus Challenges, Rachel Wong

Arts & Sciences Graduate Student Theses and Dissertations

Over the course of an effective immune response to an infection, two distinct B cell populations are generated to provide protection against reinfection, long-lived plasma cells (LLPCs) and memory B cells (MBCs). LLPCs and MBCs originate from germinal center (GC) B cells that have undergone B cell receptor (BCR) affinity maturation through iterative rounds of somatic hypermutation, proliferation, and selection. Thus, LLPCs and MBCs can bind to their antigen with higher affinity than their naïve B cell precursors. LLPCs constitutively secrete antibodies and can provide sterilizing immunity that pre-exists subsequent infections. MBCs, on the other hand, are quiescent and provide …

We discovered that •CF3 …

Genomic Contributors To Individual Differences In Reward-Related Neural Activity, Lindsay Jane Michalski Dec 2019

Genomic Contributors To Individual Differences In Reward-Related Neural Activity, Lindsay Jane Michalski

Arts & Sciences Graduate Student Theses and Dissertations

Aberrant reward-related behavior, including impulsive and risk-taking behaviors, is a common feature of externalizing psychopathology (e.g., attention deficit hyperactivity disorder, antisocial personality disorder, and substance-use disorders). Through imaging studies, these behaviors have been linked to dysregulated reactivity within a diffuse reward-related corticostriatal neural network, including the striatum, frontal regions (namely orbital, ventromedial, and dorsolateral cortices), the insula, and the hippocampus. Because variability in risk-taking behavior and related psychopathology is moderately-to-largely heritable (i.e., with estimates ranging from 40 – 80%), a genetically-informed approach is well-positioned to provide valuable insight into the etiology of reward-related neural and behavioral phenotypes that characterize externalizing …
