Tumour necrosis factor (TNF) is a cytokine belonging to a family of trimeric proteins; it has been shown to be a key mediator in autoimmune diseases such as rheumatoid arthritis and Crohn's disease. While TNF is the target of several successful biologic drugs, attempts to design small molecule therapies directed to this cytokine have not led to approved products. Here we report the discovery of potent small molecule inhibitors of TNF that stabilise an asymmetrical form of the soluble TNF trimer, compromising signalling and inhibiting the functions of TNF in vitro and in vivo. This discovery paves the way for …

New Paradigm New Practices And New Professionalism Needed To Meet Modern World Library Challenges, Muhammad Ramzan Dr, Ashfaq Hussain, Zaheer Ahmad
Library Philosophy and Practice (e-journal)
Since their existence libraries have been acquiring, organizing, disseminating and preserving recorded human knowledge only. However, inventions enforced through information and communication technologies have brought drastic changes in the medium of information storage, dissemination and use. There have been a number of studies in the developed world, which investigated and reported how they created, managed, adopted and reacted to the challenges and opportunities provided by the new information and communication technologies. Now the question is what type of systems, skills and services we need to establish and convert existing libraries into hybrid 21st Centry Libraries. This paper traces the …
Protocol Efficiently Measuring The Swelling Rate Of Hydrogels, Katherine Zhang, Wuxiang Feng, Congrui Jin
Protocol Efficiently Measuring The Swelling Rate Of Hydrogels, Katherine Zhang, Wuxiang Feng, Congrui Jin
Department of Civil and Environmental Engineering: Faculty Publications
Hydrogels are polymeric materials which can swell in water and retain a significant fraction of water within their structure without dissolving in water. Swelling rate is one of the most important properties of hydrogels. To measure the swelling rate, the profile of swelling capacity versus time of a hydrogel sample is obtained by performing free-absorbency capacity measurements at consecutive time intervals. Traditionally, either the tea-bag method, the sieve method, or the filtration method is used for the free-absorbency capacity measurements depending on the amount of the available sample and the desired precision. However, each method has its own systematic drawbacks. …

A Different Master Of War: The Influence Of The Folk Music Revival On The Antiwar Movement During The Vietnam Era, Isabelle Gillibrand
A Different Master Of War: The Influence Of The Folk Music Revival On The Antiwar Movement During The Vietnam Era, Isabelle Gillibrand
Pell Scholars and Senior Theses
Although the folk revival diminished as the United States entered the height of the Vietnam War, it lived on through antiwar activists. Music historians note how folk music was one of the early sparks of antiwar sentiment during the Vietnam era. This research builds on the ideas of previous scholars by analyzing the evolution of folk music from the 1930s to 1960s, how the folk revival's young audience connected to the music, and the influence the folk revival later had during the rise of the Vietnam antiwar movement. The antiwar sentiment expressed in the folk revival carried into antiwar activism, …

Likely Nh Dem Voters Happy With Debate And Continued Early Nh Primary; Pessimistic About Defeating Trump 12/19/2019, Unh Survey Center
All UNH Survey Center Polls
Ahead of the sixth Democratic Primary debate, likely Democratic Primary voters in New Hampshire are modestly satisfied with the criteria used by the DNC to determine who can participate in debates and most think the seven candidates who have qualified is too many or just about the right amount. A majority of likely Democratic voters are not concerned about hosting the first-in-the-nation primary despite a relative lack of racial/ethnic diversity in the state and more than half support state law requiring New Hampshire to retain the first primary. Likely Democratic voters are pessimistic about defeating Donald Trump next November, as …

Application Development For Goods Management System Towards Waste Minimization Options, Wan Ahmad Nadzim
Application Development For Goods Management System Towards Waste Minimization Options, Wan Ahmad Nadzim
Student Works (2020-2029)
Waste is a global concern. Increasing waste generation levels are causing the increase of waste management costs. In fact, the greatest costs of waste are measured by its impacts to the environment, society and economy. Prevention is better than cure—since all wastes come from goods, thus the best way to manage wastes is to manage goods. An application model of goods management system has been developed using GIS (Geographic Information System) and app prototyping. The primary purpose of goods management is to maximize goods and minimize waste by promoting reduction and reuse. GIS was used to perform spatial analyses on …

Xenograft models are invaluable tools in establishing the current paradigms of hematopoiesis and leukemogenesis. The zebrafish has emerged as a robust alternative xenograft model but, like mice, lack specific cytokines that mimic the microenvironment found in human patients. To address this critical gap, we generated the first humanized zebrafish that express human hematopoietic-specific cytokines (GM-CSF, SCF, and SDF1α). Termed GSS fish, these zebrafish promote survival, self-renewal and multilineage differentiation of human hematopoietic stem and progenitor cells and result in enhanced proliferation and hematopoietic niche-specific homing of primary human leukemia cells. Using error-corrected RNA sequencing, we determined that patient-derived leukemias transplanted …

BACKGROUND: The cytokine IL-7 is critical for T cell development and function. We performed a Phase Ib study in patients with type 1 diabetes (T1D) to evaluate how blockade of IL-7 would affect immune cells and relevant clinical responses.
METHODS: Thirty-seven subjects with T1D received s.c. RN168, a monoclonal antibody that blocks the IL -7 receptor α (IL7Rα) in a dose-escalating study.
RESULTS: Between 90% and 100% IL-7R occupancy and near-complete inhibition of pSTAT5 was observed at doses of RN168 1 mg/kg every other week (Q2wk) and greater. There was a significant decline in CD4+ and CD8+ effector and central …

Seafloor backscatter mosaics are now routinely produced from multibeam echosounder data and used in a wide range of marine applications. However, large differences (>5 dB) can often be observed between the mosaics produced by different software packages processing the same dataset. Without transparency of the processing pipeline and the lack of consistency between software packages raises concerns about the validity of the final results. To recognize the source(s) of inconsistency between software, it is necessary to understand at which stage(s) of the data processing chain the differences become substantial. To this end, willing commercial and academic software developers were …
Accuracy Matters For The Benefits Of Sleep After Retrieval Practice, Steven Dessenberger
Accuracy Matters For The Benefits Of Sleep After Retrieval Practice, Steven Dessenberger
Arts & Sciences Graduate Student Theses and Dissertations
Previous research suggests that while sleep and retrieval practice can each improve memory on their own, their benefits cannot be combined to produce an additive effect unless feedback is given during the initial test. These previous findings would seem to support a retrieval-as-consolidation of the testing effect, which states that the benefits of retrieval are the result of memory consolidation, a process that normally occurs during the sleep cycle. The present study sought to determine whether the retrieval-as-consolidation account held true when initial test accuracy was considered as a factor. Using foreign language word pairs, we examined the combined effects …

Supply Shock Versus Demand Shock: The Local Effects Of New Housing In Low-Income Areas, Brian J. Asquith, Evan Mast, Davin Reed
Supply Shock Versus Demand Shock: The Local Effects Of New Housing In Low-Income Areas, Brian J. Asquith, Evan Mast, Davin Reed
Upjohn Institute Working Papers
We study the local effects of new market-rate housing in low-income areas using microdata on large apartment buildings, rents, and migration. New buildings decrease nearby rents by 5 to 7 percent relative to locations slightly farther away or developed later, and they increase in-migration from low-income areas. Results are driven by a large supply effect—we show that new buildings absorb many high-income households—that overwhelms any offsetting endogenous amenity effect. The latter may be small because most new buildings go into already-changing areas. Contrary to common concerns, new buildings slow local rent increases rather than initiate or accelerate them.

Genome-Wide Analysis And Characterization Of F-Box Gene Family In Gossypium Hirsutum L, S Zhang, Z Tian, H Li, Y Guo, Et Al
School of Biological and Marine Sciences
Background F-box proteins are substrate-recognition components of the Skp1-Rbx1-Cul1-F-box protein (SCF) ubiquitin ligases. By selectively targeting the key regulatory proteins or enzymes for ubiquitination and 26S proteasome mediated degradation, F-box proteins play diverse roles in plant growth/development and in the responses of plants to both environmental and endogenous signals. Studies of F-box proteins from the model plant Arabidopsis and from many additional plant species have demonstrated that they belong to a super gene family, and function across almost all aspects of the plant life cycle. However, systematic exploration of F-box family genes in the important fiber crop cotton (Gossypium hirsutum) …
