Abdominal aortic aneurysm (AAA) is a life-threatening immunological disease responsible for 1 to 2% of all deaths in 65 year old or older individuals. Although mononuclear cell infiltrates have been demonstrated in AAA lesions and autoimmunity may be responsible for the initiation and account for the propagation of the disease, the information available about the pathogenesis of AAA is limited. To examine whether AAA lesions from patients with AAA contain clonally expanded α-chain TCR transcripts, we amplified by the non-palindromic adaptor-PCR (NPA-PCR)/Vα-specific PCR and/or the Vα-specific PCR these α-chain TCR transcripts. In this dissertation, I investigate the characteristics and quality of liminality among the Tibetan exile community in Dharamsala, India, and the United States. I argue that the quality of their liminality defines this exile community’s ability to maneuver and voice their influence to geo-political community of states that surround them, all while within their liminal condition. The Tibetan exile people live as stateless foreigners in India but have a better standard of living and better opportunities to acquire transnational resources than their surrounding host community. In the U.S., Tibetan diaspora people live as asylum-seekers and naturalized Tibetan-Americans but have established …

Musical instrument makers in the state of West Virginia in the United States pursue “singing,” lively instruments that capture ideals of musical tone and “re-enchant” their work and lives through relationships with craft materials and the forest landscape. Suitable tonewoods that grow in the region, such as red spruce (Picea rubens), intersect with makers’ desires to craft instruments in the style of famed makers such as the C.F. Material culture is an understudied aspect of social life in Appalachian Studies, the multi- disciplinary investigation of social life in the Appalachian region. Historically, material culture in the region has been largely studied for its semiotic properties, decoded as a tangible symbol of “a region apart,” lagging behind the rest of America in terms of moral, mental, economic, and social development. Critical material studies from archaeology and other disciplines paint a different picture, however, and construct a region as American as any other. Though some researchers have argued that the Big Barrens grasslands of Kentucky were the product of anthropogenic land clearing practices by Native Americans, heretofore, this hypothesis had not been tested archaeologically. More work was needed to refine chronologies of fire activity in the region, determine the extent to which humans played a role in the process, and integrate these findings with the paleoenvironmental and archaeological record. With these goals in mind, I conducted archaeological and geoarchaeological investigations at Crumps Sink in the Sinkhole Plain of Kentucky. The proliferation of precarious work represents a sea change in the opportunity structure of the new economies of the American South. As the role of the State has shifted from guarantor of rights and services to anxious custodian of economic liberalization, Americans have been enjoined to shoulder ever-larger shares of the responsibilities and risks associated with wage labor. As a result, the working poor have been left to weather the vicissitudes of the unfettered market with the increasingly paltry social membership guaranteed through waged employment. “Makers” around the world cohere in a digital and physical network of technology hobbyists. “Makers" are open-source hardware enthusiasts who use machines like 3D printers and laser cutters - manufacturing tools that have only recently become accessible to laypeople - to make things. “Makers" share a vision for a world where everyone would be able to make almost anything, supplanting top-down economic systems and channels of production. This ethnographic research examines a subset of the “maker” community: “makers” in “FabLabs” in Japan. Usipa Engraulicypris sardella is the most abundant small pelagic species in Lake Malawi. It plays an important part in the lake communities’ economy and food security. However, much remains unknown on their stock status and bioeconomic importance. This study is carried out to estimate the maximum economic yield and maximum sustainable yield for Usipa fishery in the South-east arm of Lake Malawi. Structured quantitative questionnaire was used to collect information from 139 informants on the price of usipa landings and cost of fishing effort. Hemendranath Mazumdar, popularly referred to as Hemen Mazumdar, was born 1894 in Gachihata village of Mymensingh district, which is currently part of Bangladesh. Coming from a relatively wealthy landowning family, at the age of sixteen, Hemen dropped out of school and ran away to Calcutta to pursue his passion for painting. He enrolled at the Government College of Art in 1911, but left in 1912 for another institution, Jubilee Art Academy. By 1915, he left Jubilee Art Academy to start earning his living through portrait painting. Abanindranath Tagore’s coterie had banished any artist following the western academic approach. Heuristics are often viewed as inferior to “rational” strategies that exhaustively search and process information. Introducing the theoretical perspective of ecological rationality, we challenge this view and argue that under conditions of uncertainty common to managerial decision making, managers can actually make better decisions using fast-and-frugal heuristics. Within the context of personnel selection, we show that a heuristic called Δ-inference can more accurately predict which of two job applicants would perform better in the future than logistic regression, a prototypical rational strategy. Using data from 236 applicants at an airline company, we demonstrate in Study 1 that despite searching less …

Over the years, various governmental, employment, and academic organizations have identified a list of skills to successfully master the challenges of the 21st century. So far, an adequate assessment of these skills across countries has remained challenging. Limitations inherent in the use of self-reports (e.g., lack of self-insight, socially desirable responding, response style bias, reference group bias, etc.) have spurred on the search for methods that could complement or even substitute self-report inventories. In this dissertation, we develop the finite integration method by using Chebyshev polynomial expansion (FIM-CPE) for solving one- and two-dimensional nonlinear differential equations. The developed FIM-CPE can be used on any domains. Then, we utilize our FIM-CPE to deal with the spatial variable and the forward difference quotient to handle the derivative involving temporal variable. Thus, the numerical algorithms based on this idea are devised to overcome three nonlinear problems including one-dimensional Burgers' equation with shock wave, time-fractional Benjamin-Bona-Mahony-Burgers' equation and two-dimensional nonlinear Poisson equation over irregular domains. Beta-cardiotoxin (ß-CTX), a novel protein isolated from the King cobra (Ophiophagus hannah) venom has previously been proposed as a beta-blocker candidate. However, cellular mechanisms of ß-CTX on cardiomyocyte is unknown. This study aimed to evaluate the impact of ß-CTX on isolated rat cardiomyocyte function and the cardiac myofibrillar activity, and to explore involvement of ß-adrenergic receptor (ß-AR) signaling pathway. ß-CTX was isolated and purified using two-step chromatographic method and confirmed by N-terminal sequencer. The function of ß-CTX on cardiomyocyte was compared to propranolol in 3 conditions, basal state, with isoproterenol (ISO), and with forskolin (FSK). ß-CTX exhibited more potency than …

The most widely used area-level model in small area estimation is the Fay-Herriot model, proposed by Fay and Herriot. It was used first to estimate average per capita income for small places (population less than 1,000) of the USA. In the context of the Fay-Herriot model, the traditional method in obtaining estimation of the population mean is the empirical best linear unbiased prediction (EBLUP) estimator. Most time series data can be characterized by a linear process via the autoregressive integrated moving average model requiring a three-component vector which are the autoregressive, differencing, and moving average orders before fitting coefficients. A model identification which determines those orders is analyzed via the partial autocorrelation function to identify the autoregressive order, the autocorrelation function to identify the moving average order and an extended sample autocorrelation function to identify both orders which is a challenging problem for statisticians. Initially, the World Bank used the ELL method to produce poverty maps for used in designing, targeting, prioritizing interventions and allocating the budgets for underdeveloped countries. Tilapia population with harvesting in the tilapia farm can be modeled by an ordinary differential equation (ODE). In real life, tilapia population can be affected by many factors. To make the model for tilapia population more realistic, we develop the ODE into the stochastic differential equation (SDE) together with jumps that represent the epidemic occuring for tilapia. Furthermore, we study the effect of some important parameters in the model to the number of tilapia in the tilapia farm via simulation. Both SDEs for tilapia population with and without jumps are simulated by using Euler-Maruyama and jump-adapted Euler methods.

In this study, we construct the discrete-time risk models based on integer-valued time series models by incorporating the concepts of surrender and investment. The surrender considered in this study is the situation that the policyholder decides to exit the policy before maturity date. In our study, we provide the probabilistic properties of the model. Moreover, we derive approximation of ruin probabilities of the constructed risk model. Finally, we discuss the trends of the ruin probability and the value at risk of the model by numerical simulations.

The basic idea of an iterative jump method is moving a feasible point along the direction that improves the objective value maintaining the feasibility. It is applied for solving a linear programming (LP) model without artificial variables by applying the iterative jump on the LP relaxation having only acute constraints with respect to the objective direction and reinsert all non-acute constraints to find the optimal solution which is named SAJS. In this study, we introduce the first order integer-valued autoregressive models for count data with the two-parameter generalized Poisson-Lindley distribution. The thinning operators considered in this study are the binomial and negative binomial thinning operators. Some important probabilistic and statistical properties such as moments, stationarity and autocorrelation functions are derived. Moreover, parameter estimations and numerical simulations to examine the constructed model are performed. Finally, applications of the models for real data sets are also discussed.

L-Phenylalanine (L-Phe) is an important commercial amino acid. It is widely used in food and pharmaceutical industries. Currently, the requirement of L-Phe is increased according to the great demand for the low-calorie sweetener, aspartame. In Escherichia coli, the synthesis of L-Phe is controlled by the multi-hierarchical regulations. AroG isoform of 3-deoxy- D-arabino-heptulosonate-7-phosphate synthase (DAHP synthase) and chorismate mutase/prephenate dehydratase (PheA), two important enzymes, are feedback inhibited by LPhe.
