The Deepwater Horizon (DWH) oil spill contaminated coastlines from Louisiana to Florida, burying oil up to 70 cm depth in sandy beaches, posing a potential threat to environmental and human health. The dry and nutrient-poor beach sand presents a taxing environment for microbial growth, raising the question how the biodegradation of the buried oil would proceed. Here we report the results of an in-situ experiment that (i) characterized the dominant microbial communities contained in sediment oil agglomerates (SOAs) of DWH oil buried in a North Florida sandy beach, (ii) elucidated the long-term succession of the microbial populations that developed in …

The Scaffolds To Guide Fast Angiogenesis, Tianhong Zhou
McKelvey School of Engineering Graduate Student Theses & Dissertations
Angiogenesis is one of the most challenging problems in bone structure repair. The different types of 3D fibrous morphology structure of the extracellular matrix for fasting angiogenesis have been studied. Due to the densely packed constructs and limited porosity of scaffold, the challenge still remains for scaffold fabrication. In this study, we fabricated PCL scaffold with growth factors and cells to guide fast angiogenesis by electro-spinning and electro-spray simultaneously. We developed a technique that electro-spinning encapsulates growth factors OPN(Recombinant Human Osteopontin) and SDF-1α(CXCL12) in poly(lactide-co-glycolide) (PLGA) microspheres into PCL 3D structures scaffold fibers. We also developed a new technique to …

Defects In Mating Behavior And Tail Morphology Are The Primary Cause Of Sterility In Caenorhabditis Elegans Males At High Temperature, Emily M. Nett, Nicholas B. Sepulveda, Lisa N. Petrella
Biological Sciences Faculty Research and Publications
Reproduction is a fundamental imperative of all forms of life. For all the advantages sexual reproduction confers, it has a deeply conserved flaw: it is temperature sensitive. As temperatures rise, fertility decreases. Across species, male fertility is particularly sensitive to elevated temperature. Previously, we have shown in the model nematode Caenorhabditis elegans that all males are fertile at 20°C, but almost all males have lost fertility at 27°C. Male fertility is dependent on the production of functional sperm, successful mating and transfer of sperm, and successful fertilization post-mating. To determine how male fertility is impacted by elevated temperature, we analyzed …
Characterization Of Immune Evasion Properties Of The Molluscum Contagiosum Virus Mc163 Protein, William De Martini
Characterization Of Immune Evasion Properties Of The Molluscum Contagiosum Virus Mc163 Protein, William De Martini
Seton Hall University Dissertations and Theses (ETDs)
Poxviruses utilize numerous strategies to prevent activation of the host pro-inflammatory transcription factor, NF-κB. For example, the Molluscum Contagiosum Virus (MCV) expresses several immune evasion molecules that either work directly at the level of NF-κB or IKK, the upstream kinase complex required for canonical NF-κB activation. Presumably these strategies allow MCV to evade host innate immune responses and cause persistent infections. In this dissertation I report the identification of a novel MCV inhibitor of NF-κB activation. Ectopic expression of the MC163 protein in HEK 293T cells resulted in a significant decrease in TNFα- induced NF-κB activation as measured by the …
Performance Of Nitrogen As A Carrier Gas In Capillary Gas Chromatography Using A Thin Film Column, Brittany A. Handzo
Performance Of Nitrogen As A Carrier Gas In Capillary Gas Chromatography Using A Thin Film Column, Brittany A. Handzo
Seton Hall University Dissertations and Theses (ETDs)
Gas chromatography (GC) is one of the most widely used analytical techniques for the separation and analysis of volatile compounds. Solids, liquids, and gases, organic and inorganic materials, and large molecular weight compounds can all be analyzed via this technique. Gas chromatographic separations are fast, accurate, and reliable. One of the reasons why these separations are so efficient is because of the carrier gas. The purpose of the carrier gas is to carry the injected sample through the column. It is known as the mobile phase and does not interact chemically with the sample. Common carrier gases include helium, hydrogen, …

A "Choose-Your-Own" Classroom-Based Activity That Promotes Scientific Inquiry About Rna Interference, Jeremy L. Hsu
Biology, Chemistry, and Environmental Sciences Faculty Articles and Research
RNA interference (RNAi), the process that results in the degradation of a target gene’s mRNA, is a fundamental part of eukaryotic gene regulation and is also an important molecular technique that allows for experimental manipulation of gene expression without altering DNA sequences. Despite the importance of RNAi, there have been relatively few lecture-based activities designed to teach about the consequences of this process and counter common misconceptions. I present here an inquiry-based activity that is centered around a “choose your own experiment” design where students generate hypotheses and critically evaluate their ideas by choosing several simulated experiments. The activity presents …

A major challenge for astronauts in long-duration space travel is combatting the hazardous spaceflight environment caused by microgravity and increased levels of ionizing radiation. Microgravity damages cellular DNA by increasing the production of harmful reactive oxygen species, while ionizing radiation damages DNA by creating double-stranded DNA (dsDNA) breaks. Cellular damage due to microgravity and radiation has been investigated using ground-based models, but most models consider microgravity and ionizing radiation alone, or asynchronously. Synchronous modeling better mimics spaceflight conditions and can be used to understand the combined effects of microgravity and ionizing radiation. However, commercially available devices to model microgravity and …

Biostatistics Collaborative Core: Faculty Publications and Other Works
Children’s metabolic energy expenditure is central to evolutionary and epidemiological frameworks for understanding variation in human phenotype and health. Nonetheless, the impact of a physically active lifestyle and heavy burden of infectious disease on child metabolism remains unclear. Using energetic, activity, and biomarker measures, we show that Shuar forager-horticulturalist children of Amazonian Ecuador are ~25% more physically active and, in association with immune activity, have ~20% greater resting energy expenditure than children from industrial populations. Despite these differences, Shuar children’s total daily energy expenditure, measured using doubly labeled water, is indistinguishable from industrialized counterparts. Trade-offs in energy allocation between competing …

Background: Although male and female rats differ in their patterns of alcohol use, little is known regarding the neural circuit activity that underlies these differences in behavior. The current study used a machine learning approach to characterize sex differences in local field potential (LFP) oscillations that may relate to sex differences in alcohol-drinking behavior. Methods: LFP oscillations were recorded from the nucleus accumbens shell and the rodent medial prefrontal cortex of adult male and female Sprague-Dawley rats. Recordings occurred before rats were exposed to alcohol (n = 10/sex × 2 recordings/rat) and during sessions of limited access to alcohol (n …

In the early 90s military operations and United Nations Special Commission “UNSCOM” teams have been destroyed the past Iraqi chemical program. Both operations led an extensive number of scattered remnants of contaminated areas. The quantities of hazardous materials, incomplete destructed materials, and toxic chemicals were sealed in two bunkers. Deficiency of appropriate destruction technology led to spreading the contamination around the storage site. This paper aims to introduce the environmental detection of the contamination in the storage site area using geospatial analysis technique. The environmental contamination level of nutrients and major ions such as sulphate (SO4), potassium(K), sodium(Na), magnesium(Mg), calcium(Ca), …

The Continuous Classical Boundary Optimal Control Of Couple Nonlinear Hyperbolic Boundary Value Problem With Equality And Inequality Constraints, Jamil A. Ali Al-Hawasy
Baghdad Science Journal
The paper is concerned with the state and proof of the existence theorem of a unique solution (state vector) of couple nonlinear hyperbolic equations (CNLHEQS) via the Galerkin method (GM) with the Aubin theorem. When the continuous classical boundary control vector (CCBCV) is known, the theorem of existence a CCBOCV with equality and inequality state vector constraints (EIESVC) is stated and proved, the existence theorem of a unique solution of the adjoint couple equations (ADCEQS) associated with the state equations is studied. The Frcéhet derivative derivation of the "Hamiltonian" is obtained. Finally the necessary theorem (necessary conditions "NCs") and the …

Bladder cancer (BCa) is the 4th most common cancer in men, and more often presents as advanced disease in women. Management of BCa is complicated and costly due to high rates of recurrence and progression, multifocality, and heterogeneity of tumors. The hyaluronic acid-HYAL-1 system is a molecular driver of BCa. Tumor cell-derived hyaluronidase, HYAL-1, degrades hyaluronic acid (HA) into angiogenic fragments (AGF: 10-12 disaccharides). AGF support tumor growth and progression. In preclinical models of BCa, we evaluated whether o-sulfated AGF (sHA-F) inhibits HYAL-1 HAase activity and has antitumor activity. At IC50 for HAase activity inhibition (5-20 µg/ml [0.4-1.7 µM]), sHA-F …
