The Intersection Of Aging And Pet Guardianship: Influences Of Health And Social Support, Ranell L. Mueller, Elizabeth G. Hunter
People and Animals: The International Journal of Research and Practice
Studies of the human-animal bond show many positive health effects for pet guardians including a sense of companionship, reduced depression and loneliness, and higher activity levels, yet few studies have examined factors such as how the pet guardians’ health, age, and social networks influence their relationship with and ability to care for their pet. These health factors may affect the ability of older adults to care for their pets, therefore inhibiting them from reaping positive benefits associated with pet guardianship. This qualitative study involved 21 in-depth interviews with older adults, aged 60+, who were pet guardians. Four themes emerged from …

Effects Of Short-Term Human-Horse Interactions On Human Heart Rate Variability: A Multiple Single Case Study, Saan Ecker, Amy Lykins
People and Animals: The International Journal of Research and Practice
Influences from human-horse interactions form the basis of the emerging field of equine-assisted psychotherapy (EAP). However, the psychophysiological effects of horses on humans in the EAP context have been underinvestigated. This multiple single case design study examined the effects of short-term human-horse interactions on human heart rate variability (HRV). Nine adults with limited prior experience with horses participated in time-limited in vivo exposures to five different free-roaming horses in a yard. Results were mixed with HRV improving from a preexposure baseline in 40% and deteriorating in 23% of the 43 ten-minute horse-human interactions. In the remaining horse-human interactions, HRV was …

“We’Re Doing Things That Are Meaningful”: Student Perspectives Of Project-Based Learning Across The Disciplines, Emily E. Virtue, Brandi N. Hinnant-Crawford
Interdisciplinary Journal of Problem-Based Learning
Supporters of project-based learning (PBL) argue that outcomes from the model include better performance in academic (Bell, 2010; Thomas, 2000) and non-academic outcomes (Saunders-Steward, Gyles, & Shore, 2010). The New Tech Network (NTN) is a school improvement network that provides training and development for high school faculty who commit to using project and/or problem-based learning as the primary instruction methodology (New Tech Network, 2017). This study uses qualitative data to investigate student perspectives of PBL across multiple disciplines at the high school level in NTN schools. Results suggest that students find value in the “hard work” they engaged in whilst …

The mechanism of fluid migration in stratium - reservoirs is revealed. The relevance of the study of the mechanism of migration of fluids is determined by the high (up to 90%) water cut of recoverable oil. It has been established that the migration of water and oil is mainly influenced by the type, nature and parameters of their interaction with host rocks, which have different values of wettability and sorption capacity. The amount of wettability is influenced by the pH values of water and oil. It is shown that pores and capillaries often have a mixed type of wettability, depending …

Background
Mitochondrial calcium overload and oxidative stress during ischemia reperfusion (I/R) injury remains a major obstacle during percutaneous coronary intervention after acute myocardial infarction. It often leads to an increased susceptibility for mitochondria permeability transition pore (mPTP) opening leading to cell death. Mitochondrial calcium overload and ROS have been identified as key triggers to open the mPTP for over 30 years, yet the exact mechanism has remained elusive. Additionally, glycogen synthase kinase 3β; (GSK-3β;) is proposed as one of the key molecules that regulate mitochondrial dysfunction and injury during I/R. Indeed inhibition of GSK-3β has been shown to be required …

TRPM2 is a non-selective cation channel located in the plasma membrane of the cell. Upon activation, the channel opens, allowing calcium to enter into the cytosol of the cell, leading ultimately to the phosphorylation and activation of the enzyme Pyk2 (proline-rich tyrosine kinase 2). Once phosphorylated, Pyk2 translocates from the cytosol to the mitochondria, where it regulates the formation of the pore component of the mitochondrial calcium uniporter (MCU) complex. Consequently, this interaction is a key factor in mitochondrial calcium uptake and therefore mitochondrial bioenergetics.

MicroRNAs form a class of short, non-coding RNA molecules which are essential for proper development in tissue-based plants and animals. To help explain their role in gene regulation, a number of mathematical and computational studies have demonstrated the potential canalizing effects of microRNAs. However, such studies have typically focused on the effects of microRNAs on only one or a few target genes. Consequently, it remains unclear how these small-scale effects add up to the experimentally observed developmental outcomes resulting from microRNA perturbation at the whole-genome level. To answer this question, we built a general computational model of cell differentiation to …

Oral anticoagulants are a critical component of stroke prevention, but carry a risk of brain hemorrhage. These hemorrhagic complications tend to occur in elderly individuals, especially those with predisposing conditions such as cerebral amyloid angiopathy (CAA). Clinical evidence suggests that non-vitamin K antagonist oral anticoagulants are safer than traditional oral anticoagulants. We analyzed whether the anticoagulant dabigatran produces cerebral microhemorrhage (the pathological substrate of MRI-demonstrable cerebral microbleeds) or intracerebral hemorrhage in aged mice with and without hemorrhage-predisposing angiopathy. We studied aged (22 months old) Tg2576 (a model of CAA) and wild-type (WT) littermate mice. Mice received either dabigatran etexilate (DE) …

Deep fractured rock ecosystems across most of North America have not been studied extensively. However, the US Great Basin, in particular the Nevada National Security Site (NNSS, formerly the Nevada Test Site), has hosted a number of influential subsurface investigations over the years. This investigation focuses on resident microbiota recovered from a hydrogeologically confined aquifer in fractured Paleozoic carbonate rocks at 863 – 923 m meters below land surface. Analysis of the microorganisms living in this oligotrophic environment provides a perspective into microbial metabolic strategies required to endure prolonged hydrogeological isolation deep underground. Here we present a microbiological and physicochemical …

A surveillance system in mammals constantly monitors cell activity to protect against aberrant proliferation in response to damage, injury and oncogenic stress. Here we isolate and culture connective tissue fibroblasts from highly regenerative mammals (Acomys and Oryctolagus) to determine how these cells interpret signals that normally induce cellular senescence in non-regenerating mammals (Mus and Rattus). While H2O2 exposure substantially decreases cell proliferation and increases p53, p21, p16, and p19 in cells from mice and rats, cells from spiny mice and rabbits are highly resistant to H2O2. Quantifying oxygen consumption …

Age At Menarche, Measures Of The Midlife Gut Microbiome, And Cardiovascular Risk Factors, Yan Wang
Theses and Dissertations
Introduction Early menarche has been associated with greater risks of cardiovascular disease (CVD) and CVD-related mortality but the mechanisms remain unclear. We hypothesized that early menarche and CVD may share as a common cause, changes in the natural history of the gut microbiome. Methods We obtained data on 2,400 adult female twins from the TwinsUK registry. Participants reported their age at menarche retrospectively and underwent anthropometric and clinical laboratory measures in 1992-2014. They provided fecal samples in 2007-2012. Microbial DNA sequences represent the V4 region of the 16S rRNA gene. OTU-picking was done using open-reference strategy in QIIME and the …

Humpback whale populations migrate extensively between winter breeding grounds and summer feeding grounds, however known links to remote Antarctic feeding grounds remain limited in many cases. New satellite tracks detail humpback whale migration pathways from Western Australia into the Southern Ocean. These highlight a focal feeding area during austral spring and early summer at the southern Kerguelen plateau, in a western boundary current where a sharp northward turn and retroflection of ocean fronts occurs along the eastern plateau edge. The topographic steering of oceanographic features here likely supports a predictable, productive and persistent forage ground. The spatial distribution of whaling …

Although in recent years there has been an increased awareness of the widespread nature of biofluorescence in the marine environment, the diversity of the molecules responsible for this luminescent phenotype has been mostly limited to green fluorescent proteins (GFPs), GFP-like proteins, and fluorescent fatty acid-binding proteins (FABPs). In the present study, we describe a previously undescribed group of brominated tryptophan-kynurenine small molecule metabolites responsible for the green biofluorescence in two species of sharks and provide their structural, antimicrobial, and spectral characterization. Multi-scale fluorescence microscopy studies guided the discovery of metabolites that were differentially produced in fluorescent versus non-fluorescent skin, as …

A critical step in the development of chronic inflammatory diseases is the accumulation of proinflammatory macrophages in the extracellular matrix (ECM) of peripheral tissues. The adhesion receptor integrin αDβ2 promotes the development of atherosclerosis and diabetes by supporting macrophage retention in inflamed tissue. We recently found that the end product of docosahexaenoic acid (DHA) oxidation, 2-(ω-carboxyethyl)- pyrrole (CEP), serves as a ligand forαDβ2.CEPadduct withECM is generated during inflammation-mediated lipid peroxidation. The goal of this project was to identify a specific inhibitor for αDβ2-CEP interaction that can prevent macrophage accumulation. Using a specially designed peptide library, Biacore-detected protein-protein interaction, and adhesion …

During Drosophila melanogaster gastrulation, the invagination of the prospective mesoderm is driven by the pulsed constriction of apical surfaces. Here, we address the mechanisms by which the irreversibility of pulsed events is achieved while also permitting uniform epithelial behaviors to emerge. We use MSD-based analyses to identify contractile steps and find that when a trafficking pathway initiated by Sbf is disrupted, contractile steps become reversible. Sbf localizes to tubular, apical surfaces and associates with Rab35, where it promotes Rab GTP exchange. Interestingly, when Sbf/Rab35 function is compromised, the apical plasma membrane becomes deeply convoluted, and nonuniform cell behaviors begin to …

We have used a transposon insertion sequencing (TIS) approach to establish the fitness landscape of the African Salmonella enterica serovar Typhimurium ST313 strain D23580, to complement our previous comparative genomic and functional transcriptomic studies. We used a genome-wide transposon library with insertions every 10 nucleotides to identify genes required for survival and growth in vitro and during infection of murine macrophages. The analysis revealed genomic regions important for fitness under two in vitro growth conditions. Overall, 724 coding genes were required for optimal growth in LB medium, and 851 coding genes were required for growth in SPI-2-inducing minimal medium. These …
