T cells can be controllably stimulated through antigen-specific or nonspecific protocols. Accompanying functional hallmarks of T cell activation can include cytoskeletal reorganization, cell size increase, and cytokine secretion. Photon-induced near-field electron microscopy (PINEM) is used to image and quantify evanescent electric fields at the surface of T cells as a function of various stimulation conditions. While PINEM signal strength scales with multiple of the biophysical changes associated with T cell functional activation, it mostly strongly correlates with antigen-engagement of the T cell receptors, even under conditions that do not lead to functional T cell activation. Change is not new, but the speed of change is new. Fueled by a combination of connecting and thinking technologies, the business environment is becoming increasingly dynamic and uncertain. Incumbent firms face a wider variety of threats that vary in frequency of occurrence and financial impact. Lead (Pb) is extremely toxic and a major cause of chronic diseases worldwide. Pb is associated with health disparities, particularly within low-income populations. In biological systems, Pb mimics calcium and, among other effects, interrupts cell signaling. Furthermore, Pb exposure results in epigenetic changes that affect multigenerational gene expression. Exposure to Pb has decreased through primary prevention, including removal of Pb solder from canned food, regulating lead-based paint, and especially eliminating Pb additives in gasoline. Introduction: Choristoma is tumor-like outgrowth of heterotopic and mature tissue located at anatomically unusual sites. Heterotopic salivary gland tissue (HSGT) has been most commonly described in head and neck region but can rarely involve gastrointestinal (GI) tract with a few cases reported in the literature.

Case Description/Methods: An 87-year-old female with past medical history of gastroesophageal reflux disease (GERD), hypertension, and chronic kidney disease was admitted for an acute DVT. The nucleus pulposus (NP) cells adapt to their physiologically hyperosmotic microenvironment through Tonicity-responsive enhancer binding protein (TonEBP/nuclear factor of activated T-cell5 [NFAT5])-mediated osmoregulation. Primary cilia in different organs serve diverse roles including osmosensing, but its contribution to NP cell osmoadaptive response is unknown. A high percentage of cultured primary NP cells possessed primary cilia that changed length in response to osmotic stimuli. Stable silencing of Intraflagellar Transport 88 (Ift88) or Kinesin Family Member 3 A (Kif3a) to inhibit the formation of primary cilia did not affect hyperosmotic upregulation of TonEBP. Oxytocin (OXT) is an important neuromodulator of social behaviors via activation of both oxytocin receptors (OXTR) and vasopressin (AVP) 1a receptors (AVPR1a). Marmosets are neotropical primates with a modified OXT ligand (Pro8-OXT), and this ligand shows significant coevolution with traits including social monogamy and litter size. Pro8-OXT produces more potent and efficacious responses at primate OXTR and stronger behavioral effects than the consensus mammalian OXT ligand (Leu8-OXT). Here, we tested whether OXT/AVP ligands show differential levels of crosstalk at primate AVPR1a. Changes In Southeastern Usa Summer Precipitation Event Types Using Instrumental (1940-2018) And Tree-Ring (1790-2018) Data, T.J. Mitchell, P.A. Knapp, T.W. Patterson

We examined short- and long-term changes in precipitation event types using instrumental (1940–2018) and tree-ring (1790–2018) data from North Carolina, USA. We documented the amount and frequency of summer (July–September) precipitation events using daily weather station data. Stationary front precipitation (SFP) represented 71% of total summer rainfall and SFP and convective uplift combined (i.e., quasi-stationary precipitation, QSP) represented 87%. SFP (r = 0.52, p < 0.01) and QSP (r = 0.61, p < 0.01) precipitation reconstructions from a montane longleaf pine latewood chronology both recorded significant declines during 1940–2018, matching the instrumental record. Conversely, no significant change in either SFP or QSP occured during the full reconstruction indicating the instrumental decline was unmatched throughout 1790–1939. Our method demonstrates that variations in latewood growth can be attributed to specific precipitation event types and that the relative contribution of each event type can be quantified over a multi-century period.

Obesity is a metabolic disorder characterized by excess energy storage in adipose tissue and it increases the risk for Type 2 diabetes. Impaired or altered adipocyte function during obesity can contribute to metabolic dysfunction. DBC1 (deleted in breast cancer 1) and EHMTs 1 and 2 (euchromatic histone lysine methyltransferases 1 and 2) have been shown to have a role in fat cell development and function. DBC1 is typically localized in the nucleus and can regulate a variety of epigenetic modifiers, transcription factors, and nuclear receptors. Urban centers are not where one expects to find new species. Yet, we report here a newly recognized ant, Strumigenys ananeotes sp. nov., in an urban yard in Salt Lake City, Salt Lake County, Utah, USA. The genus Strumigenys is a major radiation of litter ants that are found primarily in the wet tropics and subtropics. They are extremely rare in western North America, known only from sites hundreds of kilometers south and west of Salt Lake City. The integrity and organization of animal tissues depend upon specialized protein complexes that mediate adhesion between cells with each other (cadherin-based adherens junctions), and with the extracellular matrix (integrin-based focal adhesions). Reconstructing how and when these cell junctions evolved is central to understanding early tissue evolution in animals. We examined focal adhesion protein homologs in tissues of the freshwater sponge, Ephydatia muelleri (phylum Porifera; class Demospongiae). Our principal findings are that (1) sponge focal adhesion homologs (integrin, talin, focal adhesion kinase, etc.) co-precipitate as a complex, separate from adherens junction proteins; (2) that actin-based structures resembling focal adhesions form at …

This data article provides the methods and procedures followed to collect and analyse soil, vegetation and bird data on three different treatment islands in Guntersville Reservoir, Alabama. The evolution of microbial magnetoreception (or magnetotaxis) is of great interest in the fields of microbiology, evolutionary biology, biophysics, geomicrobiology, and geochemistry. Current genomic data from magnetotactic bacteria (MTB), the only prokaryotes known to be capable of sensing the Earth’s geomagnetic field, suggests an ancient origin of magnetotaxis in the domain Bacteria. INTRODUCTION: Despite evidence that people with serious mental illness benefit from receiving primary care within mental health care settings, there is little research on this type of integration. The objective of this study was to characterize how providers and patients experienced implementation of primary care into specialty mental health services.

METHODS: During site visits, study team members interviewed staff and conducted focus groups with patients at 10 United States community mental health centres then beginning to integrate primary into their practices. This small-scale ethnographic study looks at the how queer women living in Florida imagine navigating family building decisions under the current climate of policies such as a lack of federal non-discrimination protections and the largely unregulated use of assisted reproductive technologies. Despite the federal legalization of same-sex marriage in the United States in 2015, state and county legislations continue to vary greatly on the extent of support they will provide for LGBTQ families. In Emergent Masculinities, Ndubueze L. Mbah argues that the Bight of Biafra region’s Atlanticization—or the interaction between regional processes and Atlantic forces such as the slave trade, colonialism, and Christianization—between 1750 and 1920 transformed gender into the primary mode of social differentiation in the region. He incorporates over 250 oral narratives of men and women across a range of social roles and professions with material culture practices, performance traditions, slave ship data, colonial records, and more to reveal how Africans channeled the socioeconomic forces of the Atlantic world through their local ideologies and practices. Muscle function is regulated by Ca2+, which mediates excitation-contraction coupling, energy metabolism, adaptation to exercise, and sarcolemmal repair. Several of these actions rely on Ca2+ delivery to the mitochondrial matrix via the mitochondrial Ca2+ uniporter, the pore of which is formed by mitochondrial calcium uniporter (MCU). MCU's gatekeeping and cooperative activation are controlled by MICU1. Loss-of-protein mutation in MICU1 causes a neuromuscular disease. To determine the mechanisms underlying the muscle impairments, we used MICU1 patient cells and skeletal muscle-specific MICU1 knockout mice. We focus on estimating the average treatment effect in clinical trials

that involve stratified randomization, which is commonly used. It is

important to understand the large sample properties of estimators that

adjust for stratum variables (those used in the randomization

procedure) and additional baseline variables, since this can lead to

A history of allergies doubles the risk of vulvodynia-a chronic pain condition of unknown etiology often accompanied by increases in numbers of vulvar mast cells. We previously established the biological plausibility of this relationship in mouse models where repeated exposures to the allergens oxazolone or dinitrofluorobenzene on the labiar skin or inside the vaginal canal of ND4 Swiss Webster outbred mice led to persistent tactile sensitivity and local increases in mast cells. Recent advances in metabolic engineering have enabled the production of chemicals via bio-conversion using microbes. However, downstream separation accounts for 60–80% of the total production cost in many cases. Previous work on microbial production of extracellular chemicals has been mainly restricted to microbiology, biochemistry, metabolomics, or techno-economic analysis for specific product examples such as succinic acid, xanthan gum, lycopene, etc. In these studies, microbial production and separation technologies were selected apriori without considering any competing alternatives. However, technology selection in downstream separation and purification processes can have a major impact on the overall costs, product recovery, and purity. To this …

Many parent and family education programs lack a clearly articulated program theory that is solidly founded in the social science literature and used to guide rigorous evaluation. This article describes the program theory for Parents Forever, a divorce education program developed by the Minnesota Extension to serve divorcing parents. The Parents Forever program theory is theoretically based and empirically-informed. The program theory is based on three significant frameworks, which serve as the theoretical foundation for the program: human ecosystems, life course development, and family resilience.
