In tropical countries, high ambient temperature (HTa) is a fundamental factor that cause a decline in the mammary gland function in dairy animals. Dairy animals fed under HTa decreased milk yield (MY) as well as milk compositions. The current study focuses on the effect of HTa to decrease milk synthesis from natural and in vitro HTa condition. The first experiment was performed to investigate the effect of HTa from the natural condition in early lactating Saanen goat, 2-4 weeks of postpartum (2 PP-4 PP). Five animals were investigated during the winter season as control ambient condition. Six animals as the …
Automatic Model Identification For Time Series Analysis Using Deep Learning, Paisit Khanarsa
Automatic Model Identification For Time Series Analysis Using Deep Learning, Paisit Khanarsa
Chulalongkorn University Theses and Dissertations (Chula ETD)
Most time series data can be characterized by a linear process via the autoregressive integrated moving average model requiring a three-component vector which are the autoregressive, differencing, and moving average orders before fitting coefficients. A model identification which determines those orders is analyzed via the partial autocorrelation function to identify the autoregressive order, the autocorrelation function to identify the moving average order and an extended sample autocorrelation function to identify both orders which is a challenging problem for statisticians. Accordingly, the auto-ARIMA model was proposed to automatically vary those orders and estimates their corresponding coefficients. This thesis proposes three architectures …

Chulalongkorn University Theses and Dissertations (Chula ETD)
L-Phenylalanine (L-Phe) is an important commercial amino acid. It is widely used in food and pharmaceutical industries. Currently, the requirement of L-Phe is increased according to the great demand for the low-calorie sweetener, aspartame. In Escherichia coli, the synthesis of L-Phe is controlled by the multi-hierarchical regulations. AroG isoform of 3-deoxy- D-arabino-heptulosonate-7-phosphate synthase (DAHP synthase) and chorismate mutase/prephenate dehydratase (PheA), two important enzymes, are feedback inhibited by LPhe. Co-expression of feedback-resistant pheA (pheA[superscript L359D]) with other pivotal genes in L-Phe biosynthesis pathway: aroB, aroL, phedh, tktA, aroG, pheA, yddG, and glpF in pRSFDuet-1 (pPTFBLYA[superscript L359D]) elevated L-Phe production of E. …

Fructooligosaccharide Synthesis By Immobilized Inulosucrase From Lactobacillus Reuteri 121 And Improvement Of Product Pattern Using Site-Directed Mutagenesis, Thanapon Charoenwongpaiboon
Chulalongkorn University Theses and Dissertations (Chula ETD)
Inulin-type fructooligosaccharides (IFOS) are well-known prebiotics which can be produced from sucrose using bacterial inulosucrase (E.C. 2.4.1.9). However, the use of inulosucrase for IFOS synthesis has some limitations because it also synthesizes some polysaccharides and exhibits lower stability than the fungal enzymes. To overcome these limitations, improvement of inulosucrase was made by using enzyme immobilization and enzyme engineering. First, inulosucrase from Lactobacillus reuteri 121 was immobilized on a novel, high capacity core-shell chitosan beads using glutaraldehyde as a cross-linker. The results showed that both immobilized and free inulosucrase had the same optimum pH (5.5), while the optimum temperature was shifted …

Ecosystem Service Assessment Of Green Roofs In Bangkok, Rattanapan Phoomirat
Chulalongkorn University Theses and Dissertations (Chula ETD)
Green roofs are constructed ecosystems that can provide four main types of ecosystem services (regulating, provisioning, cultural, and supporting services). However, most green roof studies have focused on the regulating services and each ecosystem service type was usually evaluated separately. Moreover, there is a lack of assessment tool that can provide comparable outputs of total and different ecosystem services on green roofs, resulting in the difficulty to improve ecosystem service provision by green roofs. Therefore, this research aimed to (i) assess four main categories of ecosystem services provided by seven green roofs in Bangkok using direct measurement techniques and a …
Argonaute 4 Promotes Genomic Methylation, Kanwalat Chalertpet
Argonaute 4 Promotes Genomic Methylation, Kanwalat Chalertpet
Chulalongkorn University Theses and Dissertations (Chula ETD)
RNA-directed DNA methylation (RdDM) is the major small RNA-mediated epigenetic pathway in plants. Similar to genome and epigenome editing with the CRISPR-Cas9 system, RdDM is composed of a regulatory protein and a “guide” RNA (gRNA), which recruits the complex to specific genomic locations containing DNA sequences homologous to the RNA sequence. In humans, we found that Argonaute-4 (AGO4) colocalization has a strong correlation to promoter methylation of AGO4-binding genes. Here, we engineered cell-penetrating tagged oligoarginines (R9) tagged AGO4 (R9-AGO4) or xentry tagged AGO4 (X-AGO4) loaded with single guided-RNA (sgRNA) as a tool to specifically add methylation at genome locations where …

Diabetes and diabetic nephropathy are two noticeable disorders in modern life. Chalcones are considered as a feasible candidate for these medication disorders due to the capability to activate AMPK relating to these two diseases. A series of chalcones with A-ring possessing 2'-hydroxy-, 3',4',5'-trimethoxyacetophenones or other types and B-ring decorated with mono-, di-, trisubstitution were accomplished by Claisen–Schmidt, etherification and hydrogenation reactions in almost moderate to high yield. 3-Methoxy group was found to be the best substituent for the activity in case of monosubstitution on B-ring. 3,4-Dimethoxy and 3,4-methylenedioxy groups on B-ring (63 and 65) enhanced the activity of disubstituted chalcones …

The investigation of chemical constituents of lichen Usnea baileyi (Stirt.) Zahlbr. led to the isolation of ten new bisxanthones (US1−3, 5-11) and a new depsidone (US4). The structures were unambiguously established by the spectroscopic evidence including HRESIMS, 1D and 2D NMR, as well as comparison to literature data. Moreover, the absolute configurations were elucidated through ECD analyses, DFT-NMR calculations and subsequent DP4 probability score. The biological activities of isolated bisxanthones were evaluated for antiparasitic, cytotoxic (US1-3), antibacterial, and enzymatic inhibitory (tyrosinase and α-glucosidase) (US5-11) activities. The results revealed null to mild bioactivities against Plasmodium falciparum (antiparasitic activity) as well as …

Aiming to discover novel bioactive secondary metabolites from genus Garcinia, we performed phytochemical and biological investigations of three Garcinia species collected from Indonesia. A series of chromatographic techniques and spectroscopic data analysis were used to isolate and characterize the Garcinia phytochemicals. The cytotoxicity of the isolated compounds was evaluated against five human cancer cell lines (HeLa S3, Hep G2, HT-29, MCF-7, and KB) using MTT assay. Four new xanthones, cylindroxanthones D–G, and two new biphenyls, cylindrobiphenyls A and B, together with 28 known compounds were successfully isolated from G. cylindrocarpa stems. From the stem bark of G. tetrandra, nine new …
