Several years ago in graduate school, I was a teaching assistant for a course titled, “Great Philosophers,” which at many universities really means, “Favorite Philosophers of the Professor.” But professors will usually make a case for why their favorites should be considered “great.” In this particular course, we read the work of Ruth Millikan, who wrote a lot of interesting things about language and biology and what it means for a word to ‘stand for’ for something in the world. As cells age, they accumulate DNA damage that can lead to genomic instability, mutations, and cancer. DNA repair proteins, such as Werner (WRN), have essential roles preventing and repairing DNA damage caused by stress from the environment, DNA replication errors, and free radicals. In humans, mutations in WRN lead to Werner syndrome (WS), a heritable disease characterized by patients’ early onset of aging, increased risk of cancer and other age-related pathologies. My lab uses Drosophila mutant in the fly homolog of WRN, WRNexo, to elucidate its role in DNA repair and aging. While human WRN has both an exonuclease and …

The Cognitive Reflection Test (CRT; Frederick, 2005) is the dominant measure of adult individual differences in analytic vs. intuitive cognitive style. The task was designed to measure a person's tendency to override and inhibit an intuitive response that is incorrect and engage in deliberate reflection that leads to a correct response. The CRT is a meaningful predictor of a diverse range of adult psychological and behavioral outcomes, including conceptual change in science and math learning (Shtulman & McCallum, 2014; Gómez-Chacón et al., 2014). Cancer remains a leading cause of death in the United States. Although chemotherapy and radiation therapy are being currently used to treat cancer, they have from detrimental side effects. Hence, several studies have focused on natural herbs and compounds as alternate therapies. Zingiber officinale, commonly known as ginger, is a root that has been used for centuries in food and in medicine due to its an anti-diabetic, anti-inflammatory anti-apoptotic, and anti- carcinogenic properties. Organisms have a finite amount of energy with which to invest in survival, growth, and reproduction. Reproductive strategies can range from those that require minimal energy to produce eggs (oviparity) that then develop unassisted, those that require some energy to produce eggs that then hatch internally (ovoviviparity), and those that require significant energy to fully develop internally before birth (viviparity). Females of species that invest large amounts of energy to give birth to live young (viviparous species) may thus have more to lose in choosing low-quality mates than females of species that lay eggs (oviparous species). Polymer foams are cellular solids composed of solid and gas phases, whose mechanical, thermal, and acoustic properties are determined by the composition, volume fraction, and connectivity of both phases. A new high-throughput additive manufacturing method, referred to as direct bubble writing, for creating polymer foams with locally programmed bubble size, volume fraction, and connectivity is reported. Direct bubble writing relies on rapid generation and patterning of liquid shell–gas core droplets produced using a core–shell nozzle. The printed polymer foams are able to retain their overall shape, since the outer shell of these bubble droplets consist of a low-viscosity monomer that …

Leaf shape in plants plays important roles in water use, canopy structure, and physiological tolerances to abiotic stresses; all important traits for the future development and sustainability of grapevine cultivation. Historically, researchers have used ampelography, the study of leaf shape in grapevines, to differentiate Vitis species and cultivars based on finite leaf attributes. However, ampelographic measurements have limitations and new methods for quantifying shape are now available. We paired an analysis of finite trait attributes with a 17-point landmark survey and generalized Procrustes analysis (GPA) to reconstruct grapevine leaves digitally from five interspecific hybrid mapping families. The DNA-binding protein PU.1 is a myeloid lineage– determining and pioneering transcription factor due to its ability to bind “closed” genomic sites and maintain “open” chromatin state for myeloid lineage–specific genes. The precise mechanism of PU.1 in cell type–specific programming is yet to be elucidated. The melanoma cell line B16BL6, although it is nonmyeloid lineage, expressed Toll-like receptors and activated the transcription factor NF-B upon stimulation by the bacterial cell wall component lipopolysaccharide. The Yukon–Alaska Highway corridor in southern Yukon is subject to geohazards ranging from landslides to floods and earthquakes on faults in the St. Elias Mountains and Shakwak Valley. Here we discuss the late Holocene seismic history of the Denali fault, located at the eastern front of the St. Elias Mountains and one of only a few known seismically active terrestrial faults in Canada. Holocene faulting is indicated by scarps and mounds on late Pleistocene drift and by tectonically deformed Pleistocene and Holocene sediments. Allele-specific expression (ASE) at single-cell resolution is a critical tool for understanding the stochastic and dynamic features of gene expression. However, low read coverage and high biological variability present challenges for analyzing ASE. We demonstrate that discarding multi-mapping reads leads to higher variability in estimates of allelic proportions, an increased frequency of sampling zeros, and can lead to spurious findings of dynamic and monoallelic gene expression. Here, we report a method for ASE analysis from single-cell RNA-Seq data that accurately classifies allelic expression states and improves estimation of allelic proportions by pooling information across cells. Purpose: Dilute intravenous (IV) admixtures of dexamethasone sodium phosphate (DSP) are becoming increasingly used in antiemetic regimens to prevent chemotherapy-induced nausea and vomiting (CINV). Based on its chemical structure and previous studies, DSP is known to be susceptible to hydrolysis and oxidation under certain conditions. There are limited data to directly support the selection of IV diluents, storage conditions, and beyond-use dates for the dilute IV solutions of DSP used in the antiemetic regimens. Reconstructing the phylogenetic relationships within Caryophyllaceae tribe Sileneae has been obscured by hybridization and incomplete lineage sorting. Silene is the largest genus in the Caryophyllaceae, and unraveling its evolutionary history has been particularly challenging. In order to infer the phylogenetic relationships among the five species in Silene section Psammophilae, we have performed a genome skimming approach to acquire the complete plastid genome (cpDNA), nuclear ribosomal cistron (nrDNA), and partial mitochondrial genome (mtDNA). We have included 26 populations, representing the range of each species' distribution. This dissertation explores the revalorization of Italian extra virgin olive oils after a food scare that revealed some oils to be inauthentic or adulterated. In the process of distinguishing products, producers and tasters looked toward terroir or the “taste of place” as proof of their declarations of authenticity and as a method for differentiating oils. However, in this attempt, they engaged with other pervasive tropes of difference including those that intersected with belonging, the local and global, and race. Seventeenth-Century Spanish Colonial Identity In New Mexico: A Study Of Identity Practices Through Material Culture, Caroline Marie Gabe

This dissertation explores how seventeenth-century Spanish colonial households expressed their group identity at a regional level in New Mexico. Through the material remains of daily practice and repetitive actions, identity markers tied to adornment, technological traditions, and culinary practices are compared between 14 assemblages to test four identity models. Seventeenth-century colonists were eating a combination of Old World domesticates and wild game on colonoware and majolica serving vessels, cooking using Indigenous pottery, grinding with Puebloan style tools, and conducting household scale production and prospecting. The 18O signature of atmospheric water vapour (δ18OV) is known to be transferred via leaf water to assimilates. It remains, however, unclear how the 18O‐signal transfer differs among plant species and growth forms. We performed a 9‐hr greenhouse fog experiment (relative humidity ≥ 98%) with 18O‐depleted water vapour (−106.7‰) on 140 plant species of eight different growth forms during daytime. The Tiny Desk concert series features live video-recorded performances of artists at the desk of NPR Music’s Bob Boilen, the series’ main host. This thesis interrogates NPR Music’s values and the ways artists both manifest and queer those ideals in performance. I argue, in light of the 2016 election, performers challenge NPR Music’s taste system through two modes of subversion. The first mode considers vocalities of outrage specifically in the performances of Saul Williams and the Drive-By Truckers. These performers shift their social positions in expressions of outrage through vocality—as the embodied materiality of the voice and its constructed meanings …

Internal parasites represent a significant obstacle to a profitable and sustainable US goat industry. In some states, internal parasites are a year-round problem, while in others, they are a more seasonal challenge. Traditionally, parasite control methods relied heavily on antiparasitic drugs called anthelmintics or dewormers. Continued use and misuse of these drugs have resulted in increasingly resistant worm populations, necessitating more holistic or integrated methods of parasite control. Background:

Men who have sex with men (MSM) continue to be the predominately impacted risk group in the United States HIV epidemic and are a priority group for risk reduction in national strategic goals for HIV prevention. Modeling studies have demonstrated that a comprehensive package of status-tailored HIV prevention and care interventions have the potential to substantially reduce new infections among MSM. However, uptake of basic prevention services, including HIV testing, sexually transmitted infection (STI) testing, condom distribution, condom-compatible lubricant distribution, and preexposure prophylaxis (PrEP), is suboptimal. The majority of the research studying punishment has focused on an aversive stimulus delivered immediately after an action. However, negative consequences often occur long after a decision has been made. The delayed punishment decision-making task was developed to address this gap in literature. Rats chose between a small reinforcer and a large reinforcer accompanied by a mild foot shock. The shock was preceded by a delay, which increased throughout the session. Rats discounted the negative value of delayed punishment, as indicated by increased choice of the punished reward as the delay preceding the shock lengthened. This article focuses on Laila Marrakchi’s film Rock the Casbah (2013), which reflects the exchange between global and local cultural and sociopolitical ideologies of a new Morocco. The film highlights the contradictions of globalization as it occurs through disjuncture. Arjun Appadurai’s theory of the world in motion and “a world of flows” provides a relevant framework for this analysis. The article uses Appadurai’s notion of “disjuncture” as a theoretical framework to discuss the dynamics and interrelationships involved in the protagonist’s movement between Western mediascapes as a filmstar and her Moroccan family’s local context. Salmon were among the first nonmodel species for which systematic population genetic studies of natural populations were conducted, often to support management and conservation. The genomics revolution has improved our understanding of the evolutionary ecology of salmon in two major ways: (a) Large increases in the numbers of genetic markers (from dozens to 104– 106) provide greater power for traditional analyses, such as the delineation of population structure, hybridization, and population assignment, and (b) qualitatively new insights that were not possible with traditional genetic methods can be achieved by leveraging detailed information …

The network organization of the human brain varies across individuals, changes with development and aging, and differs in disease. Discovering the major dimensions along which this variability is displayed remains a central goal of both neuroscience and clinical medicine. Such efforts can be usefully framed within the context of the brain's modular network organization, which can be assessed quantitatively using computational techniques and extended for the purposes of multi-scale analysis, dimensionality reduction, and biomarker generation.
