Xylanases decrease the xylan content in pretreated biomass releasing it from hemicellulose, thus improving the accessibility of cellulose for cellulases. In this work, an endo-β-1,4-xylanase from Aspergillus fumigatus var. niveus (AFUMN-GH10) was successfully expressed. The structural analysis and biochemical characterization showed this AFUMN-GH10 does not contain a carbohydrate-binding module. The enzyme retained its activity in a pH range from 4.5 to 7.0, with an optimal temperature at 60°C. AFUMN-GH10 showed the highest activity in beechwood xylan. The mode of action of AFUMNGH10 was investigated by hydrolysis of APTS-labeled xylohexaose, which resulted in xylotriose and xylobiose as the main products. AFUMN-GH10 …

The performances of six time-domain reflectometry (TDR) and frequency-domain reflectometry (FDR) type soil moisture sensors were investigated for measuring volumetric soil-water content (θv) in two different soil types. Soil-specific calibration equations were developed for each sensor using calibrated neutron probe-measured θv. Sensors were also investigated for their performance response in measuring θv to changes in soil temperature. The performance of all sensors was significantly different (P<0.05) than the neutron probe-measured θv, with the same sensor also exhibiting variation between soils. The present work studied the optimization of aeration rate, agitation rate and oxygen transfer and the use of various batch fermentation strategies for xylanase production from a recombinant Aspergillus nidulans strain in a 3 L stirred tank reactor. Maximum xylanase production of 1250 U/mL with productivity of 313 U/mL/day was obtained under an aeration rate of 2 vvm and an agitation rate of 400 rpm using batch fermentation. The optimum volumetric oxygen transfer coefficient (kLa) for efficient xylanase production was found to be 38.6 h1. Land application of manure introduces gastrointestinal microbes into the environment, including bacteria carrying antibiotic resistance genes (ARGs). Measuring soil ARGs is important for active stewardship efforts to minimize gene flow from agricultural production systems; however, the variety of sampling protocols and target genes makes it difficult to compare ARG results between studies. We used polymerase chain reaction (PCR) methods to characterize and/or quantify 27 ARG targets in soils from 20 replicate, long-term no-till plots, before and after swine manure application and simulated rainfall and runoff. Hydrogen peroxide (H2O2) is an important molecule within the human body, but many of its roles in physiology and pathophysiology are not well understood. To better understand the importance of H2O2 in biological systems, it is essential that researchers are able to quantify this reactive species in various settings, including in vitro, ex vivo and in vivo systems. This review covers a broad range of H2O2 sensors that have been used in biological systems, highlighting advancements that have taken place since 2015.

Grain yield, crop evapotranspiration (ETc), basal evapotranspiration (ETb), ETc-yield production functions (ETYPF), and crop water use efficiency (CWUE) response of three drought-tolerant (DT) and one non-drought-tolerant (NDT) maize (Zea mays L.) hybrids to two plant population densities (PPDs) [84,000 plants ha-1 (high PPD) and 59,300 plants ha-1 (low PPD)] and three irrigation levels were researched at two semi-arid locations: North Platte (WCREC) and Scottsbluff (MAL), Nebraska, in 2010, 2011, and 2012. Sexual selection is widely hypothesized to facilitate the evolution of reproductive isolation through divergence in sexual traits and sexual trait preferences among populations. However, direct evidence of divergent sexual selection causing intraspecific trait divergence remains limited. Using the wolf spider Schizocosa crassipes, we characterized patterns of female mate choice within and among geographic locations and related those patterns to geographic variation in male display traits to test whether divergent sexual selection caused by mate choice explains intraspecific trait variation. The competition-colonization trade-off, by which species can partition spatial niches, is a potentially important mechanism allowing the maintenance of species diversity in plant communities. We examined whether there was evidence for this trade-off among tree species in a subtropical forest and how it correlated with eight functional traits. We developed and estimated a metric for colonization ability that incorporates both fecundity and seed dispersal based on seed trap data and the sizes and distributions of adult trees. Spiders in the genus Tetragnatha feed on emerging aquatic insects, including mosquitoes and midges, but there is little known about the foraging behavior of these spiders. We hypothesized that female spiders actively developing egg sacs would increase food consumption to provide more energy to produce and provision their eggs. We tested this hypothesis by measuring foraging rates of Tetragnatha spiders kept in jars and provisioned with different levels of midges. 1. Metabolism is the fundamental process that powers life. Understanding what drives metabolism is therefore critical to our understanding of the ecology and behavior of organisms in nature.

2. Metabolic rate generally scales with body size according to a power law. However, considerable unexplained variation in metabolic rate remains after accounting for body mass with scaling functions.

3. We measured resting metabolic rates (oxygen consumption) of 227 field-caught wolf spiders. Then, we tested for effects of body mass, species, and body condition on metabolic rate.

Document recommendation systems for locating relevant literature have mostly relied on methods developed a decade ago. This is largely due to the lack of a large offline gold-standard benchmark of relevant documents that cover a variety of research fields such that newly developed literature search techniques can be compared, improved and translated into practice. Groundwater is vital worldwide for water supply, agriculture and industry. Nearly 60% of all water use in Nebraska is from groundwater. Over 90% of groundwater is used for irrigation in Nebraska, which has the largest area of irrigated land in the United States. Many Nebraskans depend on groundwater for drinking water, both from private wells and municipal wells. The sustainability of groundwater resources is dependent on groundwater recharge. The recharge processes, as well as climatic patterns, influence the stable isotope signatures.

English: As a part of an ongoing project to inventory the helminth parasites of rodents in Mexico, 30 individuals of Aspiculuris americana were collected inhabiting the intestine from three specimens of the rock mouse Peromyscus difficilis, collected from Cerro Xihuingo, Municipality of Tepeapulco, Hidalgo State, Mexico. Magnetic Co-doped Fe3O4@FeOOH nanocomposites were prepared in one step using the hydrothermal synthesis process for catalyzing peroxymonosulfate (PMS) to degrade refractory methylene blue (MB) at a wide pH range (3.0–10.0). The catalysts' physiochemical properties were characterized by different equipment; Fe3+/Fe2+ and Co3+/Co2+ were confirmed to coexist in the nanocomposite by X-ray photoelectron spectroscopy. The nanocomposite effectively catalyzed PMS's decoloration (99.2%) and mineralization (64.7%) of MB. A new promising protocol has been developed for the synthesis of scarce oxocine derivatives 3a–e and 6 through addition of amine-based nucleophiles such as hydroxylamine hydrochloride, primary amine and hydrazide to chromonylidene benzothiazol-2-ylacetonitrile 2 in refluxing dioxane under metal free reaction conditions in moderate to good yields. Other nitrogen nucleophiles such as piperidine, hydrazine and thiosemicarbazide failed to afford the corresponding oxocinols, and instead pyridine derivatives 7, 8 and 10 were obtained exclusively. In this study, the hydroxyapatite powder is investigated for both of methylene blue and thymol blue in aqueous solution. The physical and chemical properties of the adsorbent were evaluated systematically using the different techniques including Microsoft Excel programming, linear regression model and also the coefficient of determination. Batch adsorption experiments were conducted to determine the effect of contact time, solution pH, initial dye concentrations, and also the adsorbent dosage on adsorption. Limited information about nutrition exists on American White Pelicans (Pelecanus erythrorhynchos) from hatching to fledging. To detail immunity, metabolism and nutrition of juvenile American White Pelicans, during 22-23 July 2011, 103 samples of regurgitate matter were collected at five Chase Lake, North Dakota, USA, and three Bitter Lake, South Dakota, USA, sub-colonies. We report the results of a survey on wild pigs (Sus scrofa) damage to livestock producers in 13 US states (Alabama, Arkansas, California, Florida, Georgia, Louisiana, Mississippi, Missouri, North Carolina, Oklahoma, South Carolina, Tennessee, Texas). After decades of biodiversity loss and economic burden caused by the brown treesnake invasion on the Pacific island of Guam, relief hovers on the horizon. Previous work by USDA Wildlife Services (WS) and its National Wildlife Research Center (NWRC) demonstrated that brown treesnake numbers in forested habitats can be dramatically suppressed by aerial delivery of dead newborn mouse (DNM) baits treated with 80 mg of acetaminophen. Social learning has important ecological and evolutionary consequences but the role of certain factors, such as social rank, neophobia (i.e., avoidance of novel stimuli), persistence, and task-reward association, remain less understood. We examined the role of these factors in social learning by captive coyotes (Canis latrans) via three studies. Study 1 involved individual animals and eliminated object neophobia by familiarizing the subjects to the testing apparatus prior to testing. Influenza A viruses (IAVs) are a public-health, veterinary, and agricultural concern. Although wild birds are considered the primary reservoir hosts for most IAVs, wild-bird IAV strains are known to spill over into poultry, domestic or wild mammals, and humans. Occasionally, spillover events may result in adaptation or reassortment with other strains. Wild pigs (Sus scrofa) damage agricultural and natural resources throughout their nearly global distribution. Subsequently, population control activities (e.g., trapping, shooting, or toxic baiting) frequently involve the deployment of bait to attract wild pigs. A better understanding of how wild pigs respond to bait sites can help maximize efficiency of baiting programs and identify any potential pitfalls.
