Acute myeloid leukemia (AML) is a heterogeneous group of fast growing cancers of myeloid progenitor cells, for which effective treatments are still lacking. Identification of signaling inhibitors that block their proliferation could reveal the proliferative mechanism of a given leukemia cell, and provide small molecule drugs for targeted therapy for AML. In this study, kinase inhibitors that block the majority of cancer signaling pathways are evaluated for their inhibition of two AML cell lines of the M5 subtypes, CTV-1 and THP-1. While THP-1 cells do not respond to any of these inhibitors, CTV-1 cells are potently inhibited by dasatinib, bosutinib, …

Tryptophan and tyrosine are aromatic amino acids that play significant roles in the folding processes of proteins at water-membrane interfaces because of their amphipathic structures. Employing appropriate heteroaromatic molecular structures are essential for obtaining accurate dynamics and predictive capabilities in molecular simulations of these amino acids. In this study, molecular dynamics simulations that applied the most recent version of the CHARMM36 force field were conducted on aqueous solutions of tryptophan and of tyrosine. Geometric analysis and dynamics quantified how aromatic rings deviated from planar structures and exhibited out-of-plane fluctuations. Radial distribution functions showed possible biological significance because extent of ring …

The RTS,S/AS01 malaria vaccine will undergo a pilot vaccination study in sub-Saharan Africa beginning in 2019. RTS,S/AS01 Phase III trials reported an efficacy of 28.3% (children 5–17 months) and 18.3% (infants 6–12 weeks), with substantial variability across study sites. We postulated that the relatively low efficacy of the RTS,S vaccine and variability across sites may be due to lack of T-cell epitopes in the vaccine antigen, and due to the HLA distribution of the vaccinated population, and/or due to ‘immune camouflage’, an immune escape mechanism. To examine these hypotheses, we used immunoinformatics tools to compare T helper epitopes contained in …

Ponatinib is a multi-target protein tyrosine kinase inhibitor, and its effects on hepatocellular carcinoma cells have not been previously explored. In the present study, we investigated its effects on hepatocellular carcinoma cell growth and the underlying mechanisms. Toward SK-Hep-1 and SNU-423 cells, ponatinib induces apoptosis by upregulation of cleaved caspase-3 and -7 and promotes cell cycle arrest in the G1 phase by inhibiting CDK4/6/CyclinD1 complex and phosphorylation of retinoblastoma protein. It inhibits the growth-stimulating mitogen-activated protein (MAP) kinase pathway, the phosphorylation of Src on both negative and positive regulation sites, and Jak2 and Stat3 phosphorylation. Surprisingly, it also activates the …

Cyber-attacks have increased greatly in recent years. Therefore, the identification of various network attacks has been an important research area. An Intrusion Detection System (IDS), can identify an ongoing invasion or an intrusion which has already occurred. Intrusion Detection is a classification problem. It identifies whether the network traffic behavior is normal or anomalous or identifies the attack types. Various approaches have been proposed to improve the accuracy of classifiers for identifying the intrusion types. Recently, deep learning has emerged as a successful approach in IDSs having a high accuracy rate with its distinctive learning mechanism. In this research, Long …

Reinforcement Learning Reward Design For Low-Level Controllers, Evan J. Olney
Theses
The past few decades have produced many successful applications of machine learning. In the area of automated controls, machine learning controllers have been developed to take advantage of the tendency for computers to stumble upon avenues to solutions people either overlooked, or simply do not have the computational capability to explore. Reinforcement learning is a machine learning technique for learning how to choose actions in an environment to increase its expected reward. The reward is the signal that indicates whether or not proper actions were chosen, therefore a behavioral goal is achieved from implicit rewards rather than explicit direction. The …

Abstract: The eastern subterranean termite, Reticulitermes flavipes, currently inhabits previously glaciated regions of the northeastern U.S., as well as the unglaciated southern Appalachian Mountains and surrounding areas. We hypothesized that Pleistocene climatic fluctuations have influenced the distribution of R. flavipes, and thus the evolutionary history of the species. We estimated contemporary and historical geographic distributions of R. flavipes by constructing Species Distribution Models (SDM). We also inferred the evolutionary and demographic history of the species using mitochondrial (cytochrome oxidase I and II) and nuclear (endo-beta-1,4-glucanase) DNA sequence data. To do this, genetic populations were delineated using Bayesian spatial-genetic clustering, competing …

Additives such as vinylene carbonate (VC) and fluoroethylene carbonate (FEC) are commonly added to lithium-ion battery electrolytes in order to form a solid electrolyte interphase (SEI) on the anode, suppressing continuous solvent reduction. In this work, we directly compare VC and FEC by analyzing the SEI with FTIR and XPS, and the evolved gases with on-line electrochemical mass spectrometry (OEMS) in different model systems. Since both additives evolve mainly CO2 during formation, the effect of CO2 as an additive is compared to the addition of VC and FEC. While Li2CO3 is as expected the main SEI compound found due to …

Milwaukee-Downer College Written Histories
The decision of the Milwaukee-Downer College Board of Trustees to sell their campus and associated buildings to the University of Wisconsin-Milwaukee was a historic event for both institutions and for Lawrence College. Sherwin Howard, assistant to the president at Lawrence University from 1971 to 1980, investigated the trustee decision-making process. He wrote a thesis on the subject and submitted the results of his investigation to the University of Wisconsin-Madison as partial fulfillment of the requirements for the degree of Doctor of Philosophy (1980). Much of Part I of this report has been derived from Howard’s thesis. This includes many of …

Prediction techniques are important in plant breeding as they provide a tool for selection that is more efficient and economical than traditional phenotypic and pedigree based selection. The conventional genomic prediction models include molecular marker information to predict the phenotype. With the development of new phenomics techniques we have the opportunity to collect image data on the plants, and extend the traditional genomic prediction models where we incorporate diverse set of information collected on the plants. In our research, we developed a hybrid matrix model that incorporates molecular marker and canopy coverage information as a weighted linear combination to predict …

Background: The Golgi apparatus undergoes disorganization in response to stress, but it is able to restore compact and perinuclear structure under recovery. This self-organization mechanism is significant for cellular homeostasis, but remains mostly elusive, as does the role of giantin, the largest Golgi matrix dimeric protein. Methods: In HeLa and different prostate cancer cells, we used the model of cellular stress induced by Brefeldin A (BFA). The conformational structure of giantin was assessed by proximity ligation assay and atomic force microscopy. The post-BFA distribution of Golgi resident enzymes was examined by 3D SIM high-resolution microscopy. Results: We detected that giantin …

Background: Clinical studies often track dose-response curves of subjects over time. One can easily model the dose-response curve at each time point with Hill equation, but such a model fails to capture the temporal evolution of the curves. On the other hand, one can use Gompertz equation to model the temporal behaviors at each dose without capturing the evolution of time curves across dosage
Results: In this article, we propose a parametric model for dose-time responses that follows Gompertz law in time and Hill equation across dose approximately. We derive a recursion relation for dose-response curves over time capturing the …

Cooking With Couples: A Grounded Theory Study On The Relational Aspects Found In The Cooking Interactions Of Couples, Nicole R. Gordon
Department of Family Therapy Dissertations and Applied Clinical Projects
Cooking is a universal activity which all humans can relate to on some level. Historically, cooking has continued to connect people across cultures and time, simultaneously providing nutritive sustenance as well as socio- and psychological benefits. Medical and mental health practitioners only in recent years have utilized cooking in a therapeutic process, and most of those cooking activities available tend to focus on cooking from an occupational and nutrition-education stance. A gap in the literature pertaining to cooking and its therapeutic applications exists around the relational nature underlying the cooking process, especially as it pertains to couples. While marriage and …

Additive manufacturing (AM) is becoming one of the most distinctively growing versatile technologies. In most AM techniques that requires the use of powder, the powder that is not fused into the 3D print can be replenished and used for another build. Unfused powder is generally collected on the powder-bed after an AM build is complete. In most cases, the ability to replenish unfused powder with virgin powder aids in limiting any significant change in powder’s properties that may cause compromising the quality of the 3D prints. If no powder is added to the recycled powder it is likely to shift …

Ultraviolet light absorber or stabilizer (UVLS) is one of the many additives that is incorporated into a polymer formulation which functions by preferentially absorbing the UV light to prevent its photodegradation. UVLS such as 2-hydroxyphenylbenzotriazole (2H-BZT) and its derivatives are commercially used UV-light absorbers in different polymers. The synthesis of 2H-benzotriazoles solely relies on the two-step azo-coupling of the diazonium salt and reductive cyclization where the former reaction suffers from poor yield.1 The presence of a phenolic hydrogen in the ortho position to the phenyl ring attached to the nitrogen atom on the benzotriazole imparts excellent photochemical stability to the …

Rigorous Analysis Of An Edge-Based Network Disease Model, Sabrina Mai
Honors Undergraduate Theses
Edge-based network disease models, in comparison to classic compartmental epidemiological models, better capture social factors affecting disease spread such as contact duration and social heterogeneity. We reason that there should exist infinitely many equilibria rather than only an endemic equilibrium and a disease-free equilibrium for the edge-based network disease model commonly used in the literature, as there do not exist any changes in demographic in the model. We modify the commonly used network model by relaxing some assumed conditions and factor in a dependency on initial conditions. We find that this modification still accounts for realistic dynamics of disease spread …

A simple fused silica capillary interferometric (FSCI) sensor has been proposed and investigated for the detection and analysis of multiple chemical compounds content in aqueous solutions. The sensor was fabricated by splicing a commercially available fused silica capillary (FSC) with two single mode fibers to create a 0.7 cm long air cavity. The fiber surface was functionalized with two different polymers: poly (allylamine hydrochloride) (PAH) and sol-gel silica in sequence using a layer-by-layer deposition method. The operating principle of the sensor relies on light interference in the fused silica capillary cavity due to adhesion of the different chemical compounds on …

A black phosphorus (BP) functionalized optical fiber sensor based on a microfiber coil resonator (MCR) for Pb2+ ion detection in an aquatic environment is presented and experimentally demonstrated. The MCR-BP sensor is manufactured by winding a tapered microfiber on a hollow rod composed of a low-refractive-index polycarbonate (PC) resin with the BP deposited on the internal wall of the rod. Based on the propagation properties of the MCR, the chemical interaction between the Pb2+ ions and the BP alters the refractive index of the ambient environment and thus results in a detectable shift in the transmission spectrum. The resonance wavelength …

Establishing baseline activity budgets for zoo-housed captive animals can be important in tracking behavior changes that may indicate medical or other concerns, as well as assessing overall welfare and the need for implementation of interventions such as increased amounts of enrichment. This study sought to calculate activity budgets for the current spider monkeys residing at the Central Florida Zoo, a father-daughter pair of Ateles geoffroyi. Having a baseline activity budget is also valuable because major changes in behavior may accompany the upcoming introduction of new individuals to this particular group of spider monkeys. BORIS behavior coding software was used to …

The association between dietary diversity (DD) and psychological resilience among older people is an underdeveloped area of research. This cross-sectional study explored the associations of DD with psychological resilience among 8571 community-based elderly individuals. The intake frequencies of food groups were collected, and dietary diversity was assessed based on the mean DD score. Psychological resilience was assessed using a simplified resilience score (SRS). Data were analyzed using multiple linear regression and logistic regression models. Poor DD was significantly associated with psychological resilience, with a β (95% CI) of −0.94 (−1.07, −0.81) for the SRS (p < 0.01) and an odds ratio (95% CI) of 1.83 (1.66, 2.01) for low SRS status. The interaction effects of age with DD were observed for the SRS (p < 0.001) and low SRS status (p < 0.001). Based on separate analyses by age group, the association of a low SRS with poor DD was more prominent in the younger elderly than the oldest old, with OR (95% CI) 2.32 (1.96, 2.74) and 1.61 (1.43, 1.82), respectively. Compared with younger participants with good DD, the risk of a low SRS was greater for younger participants with poor DD, the oldest old with good DD, and the oldest old with poor DD, with OR (95% CI) 2.39 (2.02, 2.81), 1.28 (1.09, 1.51), and 2.03 (1.72, 2.39), respectively. The greatest contribution to DD was from a high consumption of vegetables, fruits, and nuts. Our study suggested that poor DD was associated with a low psychological resilience among the Chinese elderly, especially the younger elderly. These findings suggest that augmentation of DD might promote psychological resilience.

Urbanization is accelerating worldwide and creates novel habitat conditions including increases in environmental temperature and changes in presence and abundance of predators, prey and parasites. For species that use urban habitats, these changes can have strong impacts on phenotypes. Anolis lizards commonly exploit urban habitats and, as ectotherms, are likely to experience pressures from these novel environments. Previous research shows that anoles may adapt to some aspects of urban habitats, but we lack an understanding of the breadth of traits that may shift in response to urbanization and how widespread these changes may be. To assess effects of urban habitat …
