Deletion Of Mgr2p Affects The Gating Behavior Of The Tim23 Complex, Oygul Mirzalieva, Shinhye Jeon, Kevin Damri, Ruth Hartke, Layla Drwesh, Keren Demishtein-Zohary, Abdussalam Azem, Cory D. Dunn, Pablo M. Peixoto

Publications and Research

The TIM23 complex is a hub for translocation of preproteins into or across the mitochondrial inner membrane. This dual sorting mechanism is currently being investigated, and in yeast appears to be regulated by a recently discovered subunit, the Mgr2 protein. Deletion of Mgr2p has been found to delay protein translocation into the matrix and accumulation in the inner membrane. This result and other findings suggested that Mgr2p controls the lateral release of inner membrane proteins harboring a stop-transfer signal that follows an N-terminal amino acid signal. However, the mechanism of lateral release is unknown. Here, we used patch clamp electrophysiology …

© 2018, Springer Nature B.V. Abstract: The insufficient resolution of conventional methods has long limited the structural elucidation of cellulose and its derivatives, especially for those with relatively low crystallinities or in native cell walls. Recent 2D/3D solid-state NMR studies of 13 C uniformly labeled plant biomaterials have initiated a re-investigation of our existing knowledge in cellulose structure and its interactions with matrix polymers but for unlabeled materials, this spectroscopic method becomes impractical due to limitations in sensitivity. Here, we investigate the molecular structure of unlabeled cotton cellulose by combining natural abundance 13 C– 13 C 2D correlation solid-state NMR …

Reaction Of Titanium Subhydride With Multiple Passivating Gasses, Erik C. Strobert Jr Jan 2019

Reaction Of Titanium Subhydride With Multiple Passivating Gasses, Erik C. Strobert Jr

Chemical and Biological Engineering ETDs

Sandia National Laboratories has used substoichiometric titanium hydride as a pyrotechnic fuel for decades. Production of titanium subhydride requires partial dehydriding of titanium hydride powder and subsequent chemical passivation to render the resultant bare titanium surface nonreactive and the powder non-pyrophoric. Novel passivation techniques, using a gas other than air, have been suggested in the past but have never been investigated. This work examines the reaction kinetics of titanium subhydride with three passivating agents, nitrogen, oxygen, and propane. These passivation experiments took place under isothermal conditions (420 ˚C) with varied dehydriding and passivation time periods. It was found that the …

The Effect Of Nonassociative Learning On Oxidative Stress In Caenorhabditis Elegans: A Potential Application For Alzheimer’S Disease Research, Gillian Patton Jan 2019

The Effect Of Nonassociative Learning On Oxidative Stress In Caenorhabditis Elegans: A Potential Application For Alzheimer’S Disease Research, Gillian Patton

Journal of the South Carolina Academy of Science

Alzheimer’s disease results in learning and memory deficits due to toxic changes in the brain caused by amyloid-β plaques and oxidative stress. Oxidative stress is caused by the overproduction of free radicals, which are uncharged molecules containing an unpaired valence electron. Nonassociative learning is a type of cognitive processing that only uses one stimulus instead of two related stimuli. Cognitive processing promotes neural growth and learning, while Alzheimer’s inhibits it. The purpose of this study was to test how nonassociative learning impacts oxidative stress and thus, if it might be considered as a potential treatment option for Alzheimer’s disease. It …

The Effect Of Flavonoid Galangin On The Cell Viability And Toxicity Of Mcf-7 Human Breast Cancer Cells Exposed To Polycyclic Aromatic Hydrocarbons (Pahs), Abhimanyu Sailesh

Journal of the South Carolina Academy of Science

Polycyclic aromatic hydrocarbons (PAHs) are chemical compounds ubiquitous in the environment; many of which are known human and animal carcinogen. The purpose of this study was to investigate MCF-7 epithelial breast cancer cells’ viability and toxicity following a short-term in vitro exposure to a cocktail of PAHs and benzo[a]pyrene (B[a]P) separately in the presence and absence of varying concentrations of flavonoid galangin. It was predicted that presence of galangin will completely or partially block the damaging effects of PAHs. It was hypothesized that exposing the cells to 7 µM cocktail of PAHs or 10 µM B[a]P in the presence of …

The Effect Of Hordeum Vulgare, Rice Hull, And Oak Leaf Litter On The Prevalence Of Microcystis, Sreya Varanasi

Journal of the South Carolina Academy of Science

Cyanobacterial harmful algal blooms are becoming more prevalent issues throughout the world, threatening ecosystems, posing risks to human health, and resulting in millions of dollars in economic damage. Conventionally, treatment approaches have been chemical or physical, but these are harmful to aquatic life or inefficient, respectively, which presents the biological approach as a favorable alternative. To assess the effects of low-effort and cost-effective treatments, Hordeum vulgare, rice hull, and oak leaf litter were individually applied to Microcystis cultures and compared to two negative controls (Microcystis alone and cotton on Microcystis) and a positive control (CuSO4). …

Epha4b Expression In The Craniofacial Development Of African Cichlid Fishes, Chelse Vanatter Jan 2019

Epha4b Expression In The Craniofacial Development Of African Cichlid Fishes, Chelse Vanatter

Journal of the South Carolina Academy of Science

Epha4b gene expression may contribute to variances in facial formation including functional differences such as nose shape and clinical conditions such as cleft palate. During craniofacial development, neural crest cells migrate to the pharyngeal arches then differentiate to form bone, muscle, and cartilage cells. The Eph/ephrin signaling pathway guides the streams of migrating neural crest cells into the pharyngeal arches; epha4b, a gene encoding an Eph receptor, contributes to this signaling pathway. To determine how epha4b expression differs between species at different developmental stages, in situ hybridization, a process that stains areas of gene expression, was performed. African cichlid fish …

The Effects Of Matthew’S Storm Surge On Coastal Loblolly Pine And Wax Myrtle Understory Regeneration, Brynn D. Wilkinson

Journal of the South Carolina Academy of Science

Hurricanes have detrimental effects on the environment and the economy. Not only are these effects felt immediately after the hurricane, but they last long after the winds have died down. They can affect future plant growth and mortality. In October of 2016 South Carolina was hit by Hurricane Matthew, a category 5 hurricane. Hobcaw Barony, our study site, was in the direct path of the storm. This created an opportunity to study the immediate effects on regeneration after a hurricane. Regenerations rates were measured along and around a surge line to determine regeneration and mortality rates. This data also allowed …

Offshore Oil Leasing: Trump Administration’S Environmentally Dangerous Energy Policy, Carol J. Miller, Bonnie B. Persons Jan 2019

Offshore Oil Leasing: Trump Administration’S Environmentally Dangerous Energy Policy, Carol J. Miller, Bonnie B. Persons

William & Mary Environmental Law and Policy Review

The Trump administration’s Executive orders on Promoting Energy Independence and Economic Growth (“Energy Independence Order”) and Implementing an America-First Offshore Energy Strategy (“Offshore Energy Order”) set the stage to open over 90% of the continental shelf to offshore oil drilling from 2019–2024. The Offshore Energy Order ignores the statutory requirements of the Outer Continental Shelf Lands Act (“OCSLA”) and the National Environmental Policy Act (“NEPA”) to balance energy exploration with safeguards for marine life and the environment. We analyze the lack of express authority in OCSLA for the President to rescind its protective designations, in comparison to other laws that …

Multivariate Information Fusion With Fast Kernel Learning To Kernel Ridge Regression In Predicting Lncrna-Protein Interactions, Cong Shen, Yijie Ding, Jijun Tang, Fei Guo

Faculty Publications

Long non-coding RNAs (lncRNAs) constitute a large class of transcribed RNA molecules. They have a characteristic length of more than 200 nucleotides which do not encode proteins. They play an important role in regulating gene expression by interacting with the homologous RNA-binding proteins. Due to the laborious and time-consuming nature of wet experimental methods, more researchers should pay great attention to computational approaches for the prediction of lncRNA-protein interaction (LPI). An in-depth literature review in the state-of-the-art in silico investigations, leads to the conclusion that there is still room for improving the accuracy and velocity. This paper propose a novel …

Complementary and alternative treatment approaches are becoming more common among children with chronic conditions. The prevalence of CAM use among US adults was estimated to be around 42% in 2015 and around 44% to 50% among adults with neurologic disorders. Studies demonstrate that children with certain chronic illnesses such as asthma, cancer, genetic disorders, attention-deficit/hyperactivity disorder (ADHD), and other neurodevelopmental disorders are treated with complementary and alternative treatments at higher rates. Dietary therapies are gaining increasing popularity in the mainstream population. Although the majority of "fad" diets do not have enough supporting evidence, some dietary therapies have been utilized for …

Impaired Nuclear Export Of Polyglutamine-Expanded Androgen Receptor In Spinal And Bulbar Muscular Atrophy., Frederick J. Arnold, Anna Pluciennik, Diane E. Merry Jan 2019

Impaired Nuclear Export Of Polyglutamine-Expanded Androgen Receptor In Spinal And Bulbar Muscular Atrophy., Frederick J. Arnold, Anna Pluciennik, Diane E. Merry

Department of Biochemistry and Molecular Biology Faculty Papers

Spinal and bulbar muscular atrophy (SBMA) is a neuromuscular disease caused by polyglutamine (polyQ) expansion in the androgen receptor (AR). Prior studies have highlighted the importance of AR nuclear localization in SBMA pathogenesis; therefore, in this study, we sought to determine the role of AR nuclear export in the pathological manifestations of SBMA. We demonstrate here that the nuclear export of polyQ-expanded AR is impaired, even prior to the formation of intranuclear inclusions of aggregated AR. Additionally, we find that promoting AR export with an exogenous nuclear export signal substantially reduces its aggregation and blocks hormone-induced toxicity. Moreover, we show …

Spatial Heterogeneity Of Temporal Shifts In Extreme Precipitation Across India, Manas Khan, Francisco Munoz-Arriola, Rehana Shaik, Penny Greer

Department of Agricultural and Biological Systems Engineering: Faculty Publications

When analyzing trends and patterns of extreme precipitation, one can easily generalize the shifts caused by external climate forcings and map a single temporal shift of annual precipitation. However, the complexity of a changing environment evidences spatially distributed shifts particularly those of extreme precipitation which are essential in planning and designing enterprises and, ultimately, in managing infrastructure across scales. The goal of this study is to characterize the spatial heterogeneity of shifts in the increasing pace of extreme events over India. The study has a two-part hypothesis: (1) the number of grid cells with significant trends in annual precipitation (P), …

Determining The Efficacy Of Insulin For Use In Factor Delivery Device For Tendon Healing And Regeneration, Daisy Ramos Jan 2019

Determining The Efficacy Of Insulin For Use In Factor Delivery Device For Tendon Healing And Regeneration, Daisy Ramos

Doctoral Dissertations

Tendon injuries account for roughly half of the 33 million musculoskeletal injuries that occur every year. Of all patients seeking nonsurgical treatment, 29% eventually require surgery. The use of autografts, allografts, and xenografts have limitations associated with donor site morbidity, availability, and immunogenicity. Tissue engineering has emerged as a feasible approach to find treatments for injuries requiring tissue replacement through biodegradable scaffolds, stem cells, and biomolecules.

Tissue engineering scaffolds serve to provide a temporary, biomimetic template for cells, as well as act as delivery vehicles for biomolecules, such as growth factors. Growth factors have been widely popular in tissue engineering …

What Do Ip25 And Related Biomarkers Really Reveal About Sea Ice Change?, Simon T. Belt

School of Geography, Earth and Environmental Sciences

© 2018 Elsevier Ltd Significant changes to Arctic and Antarctic sea ice in recent decades has prompted the development and application of novel approaches to the reconstruction of past sea ice conditions over much longer timeframes. One such approach is based on the variable distribution of certain source-specific highly branched isoprenoid (HBI) lipid biomarkers in well-dated marine sediment records. Thus, IP25 and IPSO25 have emerged as useful proxy measures of seasonal sea ice in the Arctic and Antarctic, respectively. An overview of the salient features of IP25, IPSO25 and related biomarkers is presented, together with aspects that are currently less …

Oxygen plays a key role in human mesenchymal stem cell growth. Without adequate oxygen (hypoxic condition), cells are not able to survive, proliferate, and migrate. The objective of the present study is to investigate oxygen transfer through the cell-seeded scaffolds stored in static or dynamic bioreactors using a mathematical model. The effects of porosity, cell type, scaffold architecture and cell distribution as potential effective parameters on oxygen transfer kinetics were examined. The results suggest the substantial effect of porosity and cell type on the oxygen concentration within the scaffold compared to scaffold architecture (homogeneous vs. gradient). The obtained data …
