This paper seeks to address a knowledge gap concerning how flood events impact beaver dams over time. To address this gap, I took four time-snapshots of beaver dams, mapping them across the Utah portion of the Logan-Little Bear watershed, from 2009 to 2016 to bookend a large flood event in 2011. I assessed dam status (intact, breached, or blown out) for each dam mapped to assess the impact of the large spring runoff on the dam status. Modeling Lake Temperature Response To Climate Change In The Alaskan Arctic, Thomas Balkcom

This thesis study focuses on simulating lake temperature and ice duration for four lakes at the Arctic Long-Term Ecological Research site, near the Toolik Field Station in Alaska. Model projections were driven by the representative global climate model outputs under different carbon emission scenarios. Results show that my simple lake model can reproduce historical lake temperature and ice duration observations, indicating the reliability of the model for future projections. Model projections show that JuneSeptember lake temperatures would increase by 4.3-5.8 °C from the historical period with most progressive carbon emission scenarios, but by 0.7-2.2 °C in the conservative scenarios. Becoming Ted Speakers In School: Student Development Through Participation In Ted-Ed Clubs, Melina Furman, Inés Taylor, Ariel Merpert, Tobías Yatche

This case study explores adolescents’ experiences of participating in TED-Ed Clubs in Buenos Aires, Argentina. TED-Ed Clubs is a school-based, extracurricular program in which 12- to 18-year-old youth develop and deliver a “TED-style” talk to their peers, teachers and parents. In particular, we explore changes in participants’ self-development, communication skills and overall school experience. We conducted in-depth interviews with teachers, principals and focus groups of students from 8 representative schools. We find that students struggled considerably to establish what they had to say that was worth sharing with others, and—after a long process of self-reflection—ended up developing talks which were …

Background

Immature mammalian oocytes are held arrested at prophase I of meiosis by an inhibitory phosphorylation of cyclin-dependent kinase 1 (CDK1). Release from this meiotic arrest and germinal vesicle breakdown is dependent on dephosphorylation of CDK1 by the protein, cell cycle division 25B (CDC25B). Evidence suggests that phosphorylated CDC25B is bound to YWHA (14-3-3) proteins in the cytoplasm of immature oocytes and is thus maintained in an inactive form. The importance of YWHA in meiosis demands additional studies.

Results

The prenatal environment plays a major role in influencing the health of adult offspring. Maternal food restriction (MFR) during pregnancy is a common stressor that correlates to long-term consequences, including increased risk of cardiovascular and metabolic diseases in adult offspring. Numerous studies report persistent changes following MFR, with an emphasis on the heart and kidney. These consequences are thought to occur via programming, in which a stressor, during critical developmental windows, permanently alters the structure and function of selective fetal tissues. Novel, effective, and accessible therapeutic interventions for treating Posttraumatic Stress Disorder (PTSD) symptoms are in demand given the significant physical and psychosocial impairment associated with the disorder. Although PTSD is largely treated using various forms of cognitive behavioral therapy, treatment-resistance, or non-response, rates continue to remain high. Research has shown that talk-therapies can often trigger the limbic system, keeping it in a continual state of fight or flight. Salivary gland adenoid cystic carcinoma (ACC) is an aggressive tumor with a tendency to infiltrate surrounding nerves and metastasize to distant sites. The standard treatment often fails to control local tumor recurrence and distant metastases and no approved targeted therapeutic options exist for these tumors. The goal of our studies was to reveal the molecular mechanisms driving ACC tumor development and novel drug targets to improve patient morbidity and mortality.

NK-cell resistance to transduction is a major technical hurdle for developing NK-cell immunotherapy. By using Baboon envelope pseudotyped lentiviral vectors (BaEV-LVs) encoding eGFP, we obtained a transduction rate of 23.0 ± 6.6% (mean ± SD) in freshly-isolated human NK-cells (FI-NK) and 83.4 ± 10.1% (mean ± SD) in NK-cells obtained from the NK-cell Activation and Expansion System (NKAES), with a sustained transgene expression for at least 21 days. BaEV-LVs outperformed Vesicular Stomatitis Virus type-G (VSV-G)-, RD114- and Measles Virus (MV)- pseudotyped LVs (p < 0.0001). mRNA expression of both BaEV receptors, ASCT1 and ASCT2, was detected in FI-NK and NKAES, with higher expression in NKAES. Transduction with BaEV-LVs encoding for CAR-CD22 resulted in robust CAR-expression on 38.3 ± 23.8% (mean ± SD) of NKAES cells, leading to specific killing of NK-resistant pre-B-ALL-RS4;11 cell line. Using a larger vector encoding a dual CD19/CD22-CAR, we were able to transduce and re-expand dual-CAR-expressing NKAES, even with lower viral titer. These dual-CAR-NK efficiently killed both CD19KO- and CD22KO-RS4;11 cells. Our results suggest that BaEV-LVs may efficiently enable NK-cell biological studies and translation of NK-cell-based immunotherapy to the clinic.

The cultural and legal landscape in the United States has shifted towards increased recognition of LGBQ-parent families. This shift raises questions about the everyday experiences of LGBQ parents and whether the cultural and legal changes also manifest in diminished experiences of discrimination. Drawing on data from 74 interviews with LGBQ parents, we analyze their accounts of whether they are read as a parent by others in their daily interactions. Our findings reveal the ways in which heterosexuality is a key component of how membership to the category of ‘parent’ is produced in social interactions. Environmental phenomena such as temperature, photoperiod, tidal cycle, and lunar rhythm act as external cues that stimulate the reproductive activity of marine organisms. In this study, I report the annual and lunar reproductive cycles, and changes in nutritive phagocytes (NPs) in relation to gonadal maturation of Atlantic sea urchin (Arbacia punctulata, a primeval species). The current research explored the styles of criminal thinking among juvenile delinquents in Pakistani cultural context. Initially, 40 juvenile delinquents were interviewed individually to generate an item pool of 34 distinct thought statements. After excluding repetitive items a list of 19 items was piloted on 30 juvenile delinquents as a self-report measure of 5-point rating scale (Juvenile Criminal Thinking Styles’ Inventory). Finally, a sample of 211 juvenile delinquents were given the final list of 19 items, Measure of Criminal Social Identity (Boduszek, Adamson, Shevlin, & Hyland, 2012; Shagufta, 2015), and a demographic form. In this paper, an experimentally derived ensemble made by a set of six face detectors is presented that maximizes the number of true positives while simultaneously reducing the number of false positives produced by the ensemble. False positives are removed using different filtering steps based primarily on the characteristics of the depth map related to the subwindows of the whole image that contain candidate faces. Oral history is the only past record in many Aboriginal groups in Canada. In 1997, in Delgamuukw, the Supreme Court of Canada recognized that the strict approach to evidence law with respect to oral history had to be relaxed for Aboriginal peoples to be able to pursue claims to Aboriginal rights or Aboriginal title. This was a necessary element of the attempt to achieve reconciliation between Aboriginal and non-Aboriginal peoples. Active Learning Classrooms (ALCs) are learning spaces specially designed to optimize the practice of active learning and amplify its positive effects in learners from young children through university-level learners. As interest in and adoption of ALCs has increased rapidly over the last decade, the need for grounded research in their effects on learners and schools has grown proportionately.
