Plasmodium Secretion Induces Hepatocyte Lysosome Exocytosis And Promotes Parasite Entry., Kamalakannan Vijayan, Igor Cestari, Fred D Mast, Elizabeth K K Glennon, Suzanne M Mcdermott, Heather S Kain, Alyssa M Brokaw, John D Aitchison, Kenneth Stuart, Alexis Kaushansky

Articles, Abstracts, and Reports

The invasion of a suitable host hepatocyte by Plasmodium sporozoites is an essential step in malaria infection. We demonstrate that in infected hepatocytes, lysosomes are redistributed away from the nucleus, and surface exposure of lysosome-associated membrane protein 1 (LAMP1) is increased. Lysosome exocytosis in infected cells occurs independently of sporozoite traversal. Instead, a sporozoite-secreted factor is sufficient for the process. Knockdown of SNARE proteins involved in lysosome-plasma membrane fusion reduces lysosome exocytosis and Plasmodium infection. In contrast, promoting fusion between the lysosome and plasma membrane dramatically increases infection. Our work demonstrates parallels between Plasmodium sporozoite entry of hepatocytes and infection …

Duty Of Care And Risk Management For Institutions Of Higher Education, Jorge De La Torre Nov 2019

Duty Of Care And Risk Management For Institutions Of Higher Education, Jorge De La Torre

CUP Ed.D. Dissertations

Institutions of Higher Education (IHE) authority has evolved from nearly absolute power in the hands of colleges and universities to an approach more focused on individual students and their rights. Yet, while college authority over campus life and student conduct has diminished, the duty of care required in the form of additional student affairs services has increased. The history of the complicated relationship between students and IHE starts with the concept “in loco parentis,” which is Latin for “in place of parents.” A lack of proper and continued risk assessment and a lack of effective and manageable policies and procedures …

Bitcoin Is Speech: Notes Toward Developing The Conceptual Contours Of Its Protection Under The First Amendment, Justin S. Wales, Richard J. Ovelmen Nov 2019

Bitcoin Is Speech: Notes Toward Developing The Conceptual Contours Of Its Protection Under The First Amendment, Justin S. Wales, Richard J. Ovelmen

University of Miami Law Review

Bitcoin permits users to engage in direct expressive activity with one another without the need for centralized intermediaries. It does so by utilizing an open and community-managed global database called a blockchain. While much of the literature about Bitcoin has focused on its use as a form of digital payment, this Article suggests an expanded understanding by demonstrating its use as a protocol network, not unlike the internet, that can be used to extend the possible range of human expression. After developing an appreciation of the technology, this Article recommends a framework for applying the First Amendment to Bitcoin and …

Glycerol-3-Phosphate Mediates Rhizobia-Induced Systemic Signaling In Soybean, M. B. Shine, Qing-Ming Gao, R. V. Chowda-Reddy, Asheesh K. Singh, Pradeep Kachroo, Aardra Kachroo Nov 2019

Glycerol-3-Phosphate Mediates Rhizobia-Induced Systemic Signaling In Soybean, M. B. Shine, Qing-Ming Gao, R. V. Chowda-Reddy, Asheesh K. Singh, Pradeep Kachroo, Aardra Kachroo

Plant Pathology Faculty Publications

Glycerol-3-phosphate (G3P) is a well-known mobile regulator of systemic acquired resistance (SAR), which provides broad spectrum systemic immunity in response to localized foliar pathogenic infections. We show that G3P-derived foliar immunity is also activated in response to genetically-regulated incompatible interactions with nitrogen-fixing bacteria. Using gene knock-down we show that G3P is essential for strain-specific exclusion of non-desirable root-nodulating bacteria and the associated foliar pathogen immunity in soybean. Grafting studies show that while recognition of rhizobium incompatibility is root driven, bacterial exclusion requires G3P biosynthesis in the shoot. Biochemical analyses support shoot-to-root transport of G3P during incompatible rhizobia interaction. We describe …

Collaborative Features Of Graphic Narratives: Research For "Lovely, Satisfied, Indifferent Eyes", Brigid K. Fitzpatrick Nov 2019

Collaborative Features Of Graphic Narratives: Research For "Lovely, Satisfied, Indifferent Eyes", Brigid K. Fitzpatrick

University Honors Theses

Graphic narratives, such as comics and graphic novels, innately include formal features that invite reader participation. Reading a graphic narrative means contributing to the work. I created a graphic novelette, Lovely, Satisfied, Indifferent Eyes, that prioritizes the conceptual collaboration between author and reader. I did this by enhancing the formal features of the gutter, visual voice, braiding, and temporal representation (time). Creating a graphic narrative that relies on reader participation allows the author and the work to benefit from the vast networks of diverse readers, made possible by the internet and the increased communication in our world as a …

Chemical Tools And Treatments For Neurological Disorders And Infectious Diseases, Andrea Lemus

USF Tampa Graduate Theses and Dissertations

Neurological disorders have multiple etiologies such as genetics and age. Stress related disorders are becoming more common and diagnosed more often. Mental health disorders are no longer being treated as purely psychological. Mechanisms into the underlying factors of these disorders continue to bring this area to new frontiers. Chapter 2 of this dissertation focuses on the efforts made to prove the association of the protein FKBP51 to the stress response. We synthesized and used a molecular tool to discover and prove these interactions.

The study of neurodegenerative diseases is of continued importance as the world reaches record numbers of an …

Justice Scalia Got It Right, But For The Wrong Reasons: Scalia’S Recognition Of The Supreme Court’S “Southern Exception” In U.S. Constitutional Jurisprudence And The Connection Of “Southern Exceptionalism” To “American Exceptionalism", James D. Wilets

University of Miami Law Review

The late Justice Scalia has repeatedly and sardonically noted that the Supreme Court has discounted the views of Southern states in determining whether there is a consensus among the states with regards to a Constitutional norm. This Article has termed that Supreme Court position as “Southern Exception” and can be viewed as an effort by some Justices to address the unique social, economic, religious and cultural traditions in the South engendered by its unique" and “exceptional” history. This Article will also explore how this "Southern Exception" affected American jurisprudence to the point of rendering it "exceptional" from much of the …

A Modest Proposal: The Federal Government Should Use Firing Squads To Execute Federal Death Row Inmates, Stephanie Moran Nov 2019

A Modest Proposal: The Federal Government Should Use Firing Squads To Execute Federal Death Row Inmates, Stephanie Moran

University of Miami Law Review

The Eighth Amendment prohibits cruel and unusual punishment in the criminal justice system. As the federal government looks to reinstate the death penalty, this Note argues that it should include firing squad as an option for carrying out executions. While firing squads may shock the senses, this Note argues that they are in fact the only way to comport with the requirements of the Eighth Amendment.

Design As Entrepreneurship: Towards A Design-Specific Entrepreneurship Framework, Con Kennedy Nov 2019

Design As Entrepreneurship: Towards A Design-Specific Entrepreneurship Framework, Con Kennedy

Conference papers

Current thinking on entrepreneurship states there is no specific link between the entrepreneur and their enterprise. Design is an individual creative act; therefore, the practitioner is not separate from their idiosyncratic process. The Designer is the product offering. Designers form Design Enterprises, meaning that with this aspect of inseparability, the Design Entrepreneur is a different kind of entrepreneur than currently discussed. The literature on the topic discusses design as a creative output, not as entrepreneurship output. Like any enterprise, Design Enterprises must be profitable. There are many entrepreneurship processes, none of which seem to address the needs of the Design …

Bone quality in diabetic patients is compromised, leading to weaker bones and increased fracture risk. However, the mechanism by which this occurs in diabetic bone remains to be fully elucidated. We hypothesized that elevated glucose and glucose variation would affect the function of osteocytes, essential regulators of bone homeostasis and quality. To first test this hypothesis, we used the IDG-SW3 osteocyte-like cell line to examine the effects of glucose levels on osteocyte function and viability in vitro. We confirmed our in vitro findings using the in vivo streptozotocin-induced (STZ) diabetic rat model and ex-vivo cultured osteocytes …

An Aggregation Of Survey Questionnaires In A Fuzzy Environment And Its Application, Dae Young Choi Nov 2019

An Aggregation Of Survey Questionnaires In A Fuzzy Environment And Its Application, Dae Young Choi

Malaysian Journal of Computing (MJoC)

In the response process of survey questionnaires, respondents generally express their opinions (assessment) in a fuzzy environment, i.e., assessment with a degree of membership, and in many cases, an aggregation of survey questionnaires is generally computed mathematically. In this paper, we propose a new aggregation method using the FEV (Fuzzy Expected Value) in summarizing survey questionnaires. As an application of proposed aggregation method, to quantify accurately the current state of opinions and the reasons for national IT investment increasing the rate of economic growth, we decided to circulate a survey questionnaire on national IT investment to interested parties throughout the …

Network-Based Apt Profiler, Benjamin Bornholm

Theses

Constant innovation in attack methods presents a significant problem for the security community which struggles to remain current in attack prevention, detection and response. The practice of threat hunting provides a proactive approach to identify and mitigate attacks in real-time before the attackers complete their objective. In this research, I present a matrix of adversary techniques inspired by MITRE’s ATT&CK matrix. This study allows threat hunters to classify the actions of advanced persistent threats (APTs) according to network-based behaviors.

Detection Of Salient Video Segments And Associated Comments Via Reinforcement Learning, Aditya Shah, Jared M. Zimmerman

Defensive Publications Series

The application of computer vision and techniques that rely on machine intelligence has improved automated determination of notable segments within video content. However, further refinement in these techniques can improve the quality and accuracy of the marked segments. Also, it can be difficult for automated techniques to surface the most notable text comments associated with video content. This disclosure describes techniques that combines initial independent automated determination of notable video segments and text comments to refine and improve each aspect based on the other. The techniques leverage audience engagement with live and pre-recorded video content as captured via user comments …
