The Potential Of Organ On Chip Technology For Replacing Animal Testing, Malcolm Wilkinson Jan 2019

The Potential Of Organ On Chip Technology For Replacing Animal Testing, Malcolm Wilkinson

Human Clinical Trials Collection

The term organ on a chip is used to describe the latest stage of development of in vitro cell culture technology. Figure 26.1 shows its steady development since the 1960s. Each step forward has improved our ability to model human-clinical response to new drugs or therapies and has enabled safety risk assessment of existing cosmetics, personal care products, or other chemicals in the environment. Scientific evidence that the predictive power of in vitro tests is superior to the use of animals will trigger a major shift in the way that medical research, in many areas, is carried out. In this …

Development Of A Functional Testing Platform For The Sensory Segment Of The Neuromuscular Reflex Arc, Alisha Colon Jan 2019

Development Of A Functional Testing Platform For The Sensory Segment Of The Neuromuscular Reflex Arc, Alisha Colon

Electronic Theses and Dissertations

Investigations of human biology and disease have been hindered by the use of animal models. The information obtained from such studies often results in clinically irrelevant results and drug trial failures. Additionally, several governing bodies have been formulating legislation to move away from animal models and toward more ethical and efficient testing platforms for drug discovery and cosmetic research. As an answer to these issues, "body-on-a-chip" systems have been a rapidly developing field which easily recapitulates in vivo functionality, providing a more relevant, repeatable, and ethical testing platform to better predict biology. These systems can be used as human-based testing …

Effects Of Resistant Starch Type 4 On Gut Microbiota Derived Medium And Long Chain Fatty Acids And Bile Acids, Elizabeth Nagel Jan 2019

Effects Of Resistant Starch Type 4 On Gut Microbiota Derived Medium And Long Chain Fatty Acids And Bile Acids, Elizabeth Nagel

Electronic Theses and Dissertations

The human gastrointestinal tract is populated by a diverse and mutualistic microbial flora that has profound effect on human health. Identification and characterization of microbial-metabolites produced in response to human dietary intakes may provide insights into host-microbiota interactions to better understand the role of the microbiota in human health and diseases. However, much of this interaction is poorly understood as many metabolites remain unidentified or understudied at the present time. Starch is ubiquitously present in human diet and is a major energy source to all living cells. Resistant starch (RS) is the starch portion that is not absorbed in the …

Investigation Of Intergroup Bias In Two Neuromaturationally Distinct Age Cohorts: An Erp Study, Reuven M. Hanna Jan 2019

Investigation Of Intergroup Bias In Two Neuromaturationally Distinct Age Cohorts: An Erp Study, Reuven M. Hanna

Electronic Theses and Dissertations

Currently, sociological investigation of adolescent behavior focuses on the intersection of biography, history, and structure to explain adolescent risk-taking, reward-seeking, impulsivity, novelty-seeking and peer-salience. However, the preponderance of the evidence points away from social ecology and to a significant neuromaturational restructuring event between the 12th and 25th years of life as the root of adolescent behavioral tendencies. As a result, sociological social psychology can benefit from engaging in basic research using neuroscience methods. The present study expands the dual systems model of brain development to account for maturational changes in the social brain network as a way to explain social …

Physiological Approach To Characterize Drowsiness In Simulated Flight Operations During Window Of Circadian Low, Bijay Guragain Jan 2019

Physiological Approach To Characterize Drowsiness In Simulated Flight Operations During Window Of Circadian Low, Bijay Guragain

Theses and Dissertations

Drowsiness is a psycho-physiological transition from awake towards falling sleep and its detection is crucial in aviation industries. It is a common cause for pilot’s error due to unpredictable work hours, longer flight periods, circadian disruption, and insufficient sleep. The pilots’ are prone towards higher level of drowsiness during window of circadian low (2:00 am- 6:00 am). Airplanes require complex operations and lack of alertness increases accidents. Aviation accidents are much disastrous and early drowsiness detection helps to reduce such accidents. This thesis studied physiological signals during drowsiness from 18 commercially-rated pilots in flight simulator. The major aim of the …

Structure Determination By Single-Particle Cryo-Electron Microscopy: Only The Sky (And Intrinsic Disorder) Is The Limit, Emeka Nwanochie, Vladimir N. Uversky Jan 2019

Structure Determination By Single-Particle Cryo-Electron Microscopy: Only The Sky (And Intrinsic Disorder) Is The Limit, Emeka Nwanochie, Vladimir N. Uversky

Molecular Medicine Faculty Publications

Traditionally, X-ray crystallography and NMR spectroscopy represent major workhorses of structural biologists, with the lion share of protein structures reported in protein data bank (PDB) being generated by these powerful techniques. Despite their wide utilization in protein structure determination, these two techniques have logical limitations, with X-ray crystallography being unsuitable for the analysis of highly dynamic structures and with NMR spectroscopy being restricted to the analysis of relatively small proteins. In recent years, we have witnessed an explosive development of the techniques based on Cryo-electron microscopy (Cryo-EM) for structural characterization of biological molecules. In fact, single-particle Cryo-EM is a special …

The Effect Of Maternal Dietary Habits During Pregnancy On Neonate Leptin Methylation Patterns And Gestational Age, Sean Fitzpatrick Jan 2019

The Effect Of Maternal Dietary Habits During Pregnancy On Neonate Leptin Methylation Patterns And Gestational Age, Sean Fitzpatrick

Legacy Theses & Dissertations (2009 - 2024)

The health of a newborn baby is inextricably linked to the health status of its mother and in turn the mother’s diet during pregnancy. Leptin (LEP) is an adipokine hormone involved in metabolism regulation and has been linked fetal development through the hypothalamic-pituitary-adrenal axis (HPA). Prior work suggests that gestational epigenetic alterations the LEP gene may be sensitive to adverse exposures during pregnancy, which in turn could explain variation in neonate outcomes. However, no prior work has examined this possibility explicitly. The objective of this study was to investigate the association between dietary patterns of mothers during pregnancy and their …

Force Perception At The Shoulder After A Unilateral Suprascapular Nerve Block, David Phillips, Peter Kosek, Andrew Karduna Jan 2019

Force Perception At The Shoulder After A Unilateral Suprascapular Nerve Block, David Phillips, Peter Kosek, Andrew Karduna

Department of Exercise Science and Physical Education Scholarship and Creative Works

There are two key sources of information that can be used to match forces—the centrally generated sense of effort and afferent signals from mechanical receptors located in peripheral tissues. There is currently no consensus on which source of information is more important for matching forces. The corollary discharge hypothesis argues that subjects match forces using the centrally generated sense of effort. The purpose of this study was to investigate force matching at the shoulder before and after a suprascapular nerve block. The nerve block creates a sensory and muscle force mismatch between sides when matching loads. The torque matching accuracy …

Refusing Care As A Legal Pathway To Medical Assistance In Dying, Jocelyn Downie, Matthew J. Bowes Jan 2019

Refusing Care As A Legal Pathway To Medical Assistance In Dying, Jocelyn Downie, Matthew J. Bowes

Articles, Book Chapters, & Popular Press

Can a competent individual refuse care in order to make their natural death reasonably foreseeable in order to qualify for medical assistance in dying (MAiD)? Consider a competent patient with left-side paralysis following a right-brain stroke who is not expected to die for many years; normally his cause of death would not be predictable. However, he refuses regular turning, so his physician can predict that pressure ulcers will develop, leading to infection for which he will refuse treatment and consequently die. Is he now eligible for MAiD? Consider a competent patient with spinal stenosis (a non-fatal condition) who refuses food …

A Hybrid Cognitive Architecture With Primal Affect And Physiology, Christopher L. Dancy Jan 2019

A Hybrid Cognitive Architecture With Primal Affect And Physiology, Christopher L. Dancy

Faculty Journal Articles

Though computational cognitive architectures have been used to study several processes associated with human behavior, the study of integration of affect and emotion in these processes has been relatively sparse. Theory from affective science and affective neuroscience can be used to systematically integrate affect into cognitive architectures, particularly in areas where cognitive system behavior is known to be associated with physiological structure and behavior. I introduce a unified theory and model of human behavior that integrates physiology and primal affect with cognitive processes in a cognitive architecture. This new architecture gives a more tractable, mechanistic way to simulate affect-cognition interactions …

Detection Of Human Plasma Glucose Using A Self-Powered Glucose Biosensor, Gymama Slaughter, Tanmay Kulkarni Jan 2019

Detection Of Human Plasma Glucose Using A Self-Powered Glucose Biosensor, Gymama Slaughter, Tanmay Kulkarni

Bioelectrics Publications

This work presents the characterization of a self-powered glucose biosensor using individual sequential assays of human plasma glucose obtained from diabetic patients. The self-powered glucose biosensor is exploited to optimize the assay parameters for sensing plasma glucose levels. In particular, the biofuel cell component of the system at pH 7.4, 37 °C generates a power density directly proportional to plasma glucose and exhibited a maximum power density of 0.462 mW·cm−2 at a cell voltage of 0.213 V in 5 mM plasma glucose. Plasma glucose is further sensed by monitoring the charge/discharge frequency (Hz) of the integrated capacitor functioning as …

Inhibition Of The Akt1-Mtorc1 Axis Alters Venous Remodeling To Improve Arteriovenous Fistula Patency, Arash Fereydooni Jan 2019

Inhibition Of The Akt1-Mtorc1 Axis Alters Venous Remodeling To Improve Arteriovenous Fistula Patency, Arash Fereydooni

Yale Medicine Thesis Digital Library

Arteriovenous fistulae (AVF) are the most common access created for hemodialysis, but up to 60% do not sustain dialysis within a year, suggesting a need to improve AVF maturation and patency. In a mouse AVF model, Akt1 regulates fistula wall thickness and diameter. We hypothesized that inhibition of the Akt1-mTORC1 axis alters venous remodeling to improve AVF patency. Daily intraperitoneal injections of rapamycin reduced AVF wall thickness with no change in diameter. Rapamycin decreased smooth muscle cell (SMC) and macrophage proliferation; rapamycin also reduced both M1 and M2 type macrophages. AVF in mice treated with rapamycin had reduced Akt1 and …

The bond between pet and owner has been shown to decrease stress-related physical, mental, and emotional disorders by lowering blood pressure and heart rate. The purpose of this study was to examine whether a nonpet canine would have the same effect in reducing physical symptoms of stress as a known dog. Human-animal bond theory, Human pressor test. Data were analyzed using repeated measures, analyses of variance, and t tests. Results indicated that interaction with a strange dog lowered heart rate and blood pressure significantly faster in the experimental group than in the control group. The positive social change implications of …

A Possible Link Between R-Wave Amplitude Alternans And T-Wave Alternans In Ecgs, Sahar Alaei Jan 2019

A Possible Link Between R-Wave Amplitude Alternans And T-Wave Alternans In Ecgs, Sahar Alaei

Theses and Dissertations--Biomedical Engineering

Sudden Cardiac Death (SCD) is the largest cause of natural deaths in the USA, accounting for over 300,000 deaths annually. The major reason for SCD is Ventricular Arrhythmia (VA). Therefore, there is need for exploration of approaches to predict increased risk for VA. Alternans of the T wave in the ECG (TWA) is widely investigated as a potential predictor of VA, however, clinical trials show that TWA has high negative predictive value but poor positive predictive value. A possible reason that TWA has a large number of false positives is that a pattern of alternans known as concordant alternans, may …
