Development Of Cell-Penetrating Peptides Targeting Nuclear And Extra-Nuclear Signaling Pathways In Cancer Cells, Panthita Kaewjanthong Jan 2019

Development Of Cell-Penetrating Peptides Targeting Nuclear And Extra-Nuclear Signaling Pathways In Cancer Cells, Panthita Kaewjanthong

Chulalongkorn University Theses and Dissertations (Chula ETD)

Growth factors and hormones often mediate their biological effects through series of protein-protein interaction between cytoplasmic signaling molecules or transcription factors. Interfering of these protein-protein interactions has been shown to reduce or inhibit biological and growth responses to various growth factors and hormones. We previously demonstrated that progesterone receptor (PR) contains a polyproline domain (PPD) which directly interact to Src homology 3 (SH3) domain-containing molecules and expression of PR-PPD inhibits EGFR-mediated NSCLC cell proliferation. In this study, we investigated that the introduction of PR-PPD by cell-penetrating peptide could inhibit EGF-induced NSCLC cell proliferation. PR-PPD was attached to a cancer-specific CPP, …

Roles Of Lin28 Proteins In Cholangiocarcinoma, Nattapong Puthdee Jan 2019

Roles Of Lin28 Proteins In Cholangiocarcinoma, Nattapong Puthdee

Chulalongkorn University Theses and Dissertations (Chula ETD)

LIN28 is an RNA-binding protein, a known as regulator of microRNAs biogenesis and play a role in multiple cellular functions such as stem cell maintenance, cell metabolism and implicated in oncogenesis. In cancer study, LIN28 protein is frequently expressed and associated with tumor aggressiveness and poor disease outcome. Previous studies have shown that paralog-LIN28B was upregulated in during CCA development. However, the role of LIN28B in cholangiocarcinogenesis remains largely unknown. The present study, we found LIN28B is overexpressed in CCA patients' tissues. Overexpression of LIN28B in cholangiocytes-MMNK-1 cells showed that LIN28B enhanced cell proliferation, clonogenic potential, and promoted epithelial-to mesenchymal …

Cyp3a And Drug Transporters Activity Changes In Thai Elderly With Or Without Chronic Kidney Disease Using A Microdose Cocktail, Punyabhorn Rattanacheeworn

Chulalongkorn University Theses and Dissertations (Chula ETD)

Thai general population has become more “aged society” (>60 years old) and chronic kidney disease (CKD) is one of major public health problems in Thailand. Ageing and CKD are known to affect pharmacokinetics which generally determines drug exposure in plasma and site of action. Due to the related mechanisms are still unclear, cytochrome P450 (CYP)3A and drug transporters activity changes were investigated in Thai elderly with or without CKD compared to healthy adults using a microdose cocktail. This study was conducted in 3 groups of subjects: Group 1, healthy young subjects; Group 2, healthy elderly subjects; Group 3; elderly …

Development Of Niosome Containing Ace Inhibitor/Cyclodextrin Complexes As An Ophthalmic Delivery For Lowering Intraocular Pressure, Hay Marn Hnin Jan 2019

Development Of Niosome Containing Ace Inhibitor/Cyclodextrin Complexes As An Ophthalmic Delivery For Lowering Intraocular Pressure, Hay Marn Hnin

Chulalongkorn University Theses and Dissertations (Chula ETD)

The purpose of this study was to develop a stable niosomal eye drop containing angiotensin converting enzyme (ACE) inhibitor for lowering intraocular pressure. Three ACE inhibitors i.e., captopril, quinapril and fosinopril (FOS) were used in this study. Their inclusion complexes formation with parent CDs i.e., alphaCD, betaCD and gammaCD were determined. All drugs represented 1:1 stoichiometry inclusion complex with each CD. Of these, FOS/gammaCD complex showed the highest stability constant. Kinetic degradation study confirmed that FOS showed the lowest degradation in the presence of gammaCD. The obtained results of solution- and solid-state characterizations including molecular docking supported the true inclusion …

Development And Optimization Of Self-Microemulsifying Astaxanthin Delivery System Using The Design Of Experiment (Doe) Approach, Mo Mo Ko Zin

Chulalongkorn University Theses and Dissertations (Chula ETD)

The purpose of this research was to develop and optimize a self-microemulsifying delivery system (SMEDS) to improve dissolution rate of poor soluble compound AST by mixture experimental design. The solubility of AST was analyzed with various excipients so that the appropriate oil, surfactant, and cosurfactant were figured out. Through the findings about the microemulsion existence area, the pseudoternary phase diagrams were constructed for selecting the optimum combination of excipients in a formulation of SMEDS. The optimized LCT-SMEDS obtained from the design space was composed of 19.59% castor oil (oil; X1), 62.34% Cremophor® RH 40 (surfactant; X2), and 18.03% Tween® 80 …

Chemical Modifications Of Α-Mangostin For Cytotoxic Study On Lung Cancer Cell Lines, Nan Yadanar Lin Pyae Jan 2019

Chemical Modifications Of Α-Mangostin For Cytotoxic Study On Lung Cancer Cell Lines, Nan Yadanar Lin Pyae

Chulalongkorn University Theses and Dissertations (Chula ETD)

α-Mangostin is a natural oxygenated and prenylated xanthone, which is mainly isolated from the pericarps of Garcinia mangostana. It possesses numerous pharmacological properties and promising therapeutic effects. However, the limitations of α-mangostin such as highly hydrophobicity and poor bioavailability hider its therapeutic applications. To overcome these drawbacks of α-mangostin, chemical modifications have been performed to discover the improved analogs. Herein, two semi-synthetic approaches to obtain new α-mangostin derivatives were investigated by the modifications of the phenolic hydroxy groups at C-3 and C-6 positions involving Smiles rearrangement to install amine functional groups and the addition reaction of isocyanate to form the …

Genetic And Pathogenic Characterizations Of Newly Emerged Duck Tembusu Virus Isolated In Thailand, Patchareeporn Ninvilai

Chulalongkorn University Theses and Dissertations (Chula ETD)

Duck Tembusu virus (DTMUV), a mosquito-borne Flavivirus, caused an emerging disease in ducks in Asia since 2010, resulting in significant economic loss in duck producing industry. After the initial outbreak in 2013, DTUMV cases have been continuously detected in several duck farms in Thailand. Thus, the disease surveillance and the genetic characterization of the Thai DTMUV are necessary. Therefore, the first objective of this dissertation was to investigate the genetic characteristic of DTMUVs circulating in Thailand. Our retrospective study demonstrated the presence of DTMUV in the Thai ducks since 2007. Phylogenetic analysis of the polyprotein gene sequence revealed that the …

Comparative Study Of The Effectiveness Between Used And Non-Used Noise Warning Application On Promoting The Wearing Of Hearing Protection Among Steel Industry Workers In Samut Prakan Province Thailand, Petcharat Kerdonfag

Chulalongkorn University Theses and Dissertations (Chula ETD)

The exposure to loud noises from occupational performance is a major problem for workers in steel mills. The operators have to be exposed to loud noises from various types of steel production machinery at all times. These noises may affect to the operator's hearing perception in spite the factories themselves have set up hearing conservation program whereas one element is the procurement of loud noise protection equipment to workers working in loud noise area. But it is still found that the operators hardly prefer wearing hearing protection devices during their working hours to work without wearing them. The main objective …
