Curcumin diethyl disuccinate (CDD) is an ester prodrug of the active compound, curcumin. CDD has been improved chemical stability and intestinal permeability more than curcumin. CDD demonstrated biological activity higher than curcumin. The aim of this study is to investigate a suitability of using liver S9 fraction (LS9) in drug metabolism profiling as function of animal sources of enzyme regarding possible translation of in vitro outcome to clinical study. Molecular pharming represents the innovative development of plants as alternative production platforms for recombinant proteins. Human vascular endothelial growth factor (VEGF) is one of the growth factors that has displayed eminent roles in wound healing. This study aimed to develop optimized expression conditions and rapid production of recombinant VEGF in Nicotiana benthamiana. Herein, four plant expression constructs, particularly pBYR2e-SP-VEGF-His-SEKDEL (S-V-H-K), pBYR2e-SP-VEGF-His (S-V-H), pBYR2e-SP-His-VEGF-SEKDEL (S-H-V-K) and pBYR2e-SP-His-VEGF (S-H-V), were utilized and inserted into geminiviral vector pBYR2e-K2Md for transient expression in N. benthamiana. Cordycepin, an adenosine analogue, has displayed numerous pharmacological and therapeutic implications including enhanced apoptosis and inhibition of proliferation of cancer cells, antioxidant property and improving sexual activity. However, oral administration of cordycepin has limited practical use due to its poor bioavailability in the intestine. Here, we developed a novel hybrid nanoparticle system for effective delivery of cordycepin in the gastrointestinal tract by using the bile salt-based liposome/niosome hybrid system (designated as biloniosome). The purpose of this study was to develop pickering nanoemulsions (NEs) containing amphotericin B (AmB) for fungal keratitis therapy. The phase-solubility profiles of AmB in various aqueous cyclodextrin (CD) solutions were determined. The AmB solubilization in oils and surfactants was also examined. Two CDs i.e., gCD and 2-hydroxypropyl-g-CD (HPgCD), medium-chain triglycerides (MCT oil) and phosphatidylcholine (lecithin) were chosen because of their high solubility increment of AmB. Solution-state by 1H-NMR studies demonstrated the formation of MCT oil/αCD, AmB/gCD and AmB/HPgCD complexes. This study aimed to express recombinant protein human Dentin Matrix Protein 1 (hDMP1) in Escherichia coli and Nicotiana benthamiana and investigated the osteogenic differentiation effects in human Periodontal Ligament Stem Cells (hPDLSCs). The gene encoding hDMP1 was ligated into the pET22b and pBY2R2e expression vectors. Recombinant hDMP1 protein has been productively expressed in E. coli and N. benthamiana. E. coli-produced hDMP1 resulted in 60 kDa and 100, and the optimal condition to produce highly expression was demonstrated at 0.5 mM IPTG and incubation at 37oC for 6 hr. Trend of regenerative therapy for diabetes in human and veterinary practice has conceptually been proven according to Edmonton protocol and animal models. Establishing an alternative insulin-producing cell (IPC) resource is a challenge task for further clinical application. In this study, IPC generation from two practical canine mesenchymal stem cells (cMSCs), canine bone marrow-derived MSCs (cBM-MSCs) and canine adipose-derived MSCs (cAD-MSCs), was of interest. The results illustrated that cBM-MSCs and cAD-MSCs contained distinct pancreatic differentiation potential and required the tailor-made induction protocols. Lung cancer is a major cause of cancer-related death. Tumor growth is a fundamental process of cancer progression. Vital characteristics that potentiate tumor growth are cancer stem cell (CSC) and enhance cell proliferation. หมึกกล้วย (Indian squid) สายพันธุ์ Loligo duvauceli เป็นสัตว์ทะเลทางเศรษฐกิจที่สำคัญของประเทศไทย พบว่าครีบหมึกจัดว่าเป็นของเสียในกระบวนการผลิตจากหมึกกล้วยคิดเป็น 12% และมีราคาถูก โดยครีบหมึกกล้วยแห้งประกอบด้วยความชื้น 4.62% โปรตีน 77.08% ไขมัน 15.60% และ เถ้า 3.03% ในงานวิจัยนี้ได้สกัดคอลลาเจนจากครีบหมึกกล้วยบดโดยใช้สารละลายโซเดียมไฮดรอกไซด์ความเข้มข้น 0.5 โมลต่อลิตรเพื่อกำจัดไขมันและโปรตีนอื่นๆ จากนั้นนำไปสกัดต่อด้วยกรดแอซิติกความเข้มข้น 0.5 โมลต่อลิตรที่อุณหภูมิ 4 องศาเซลเซียส แยกส่วนของแข็งและสารละลายออกจากกันด้วยการกรอง และนำสารละลายกรดที่ได้ไปทำแห้งด้วยวิธีแช่เยือกแข็ง โดยของแข็งที่ได้จะเรียกว่า คอลลาเจนที่ละลายในกรด (ASC) ส่วน คอลลาเจนที่ละลายในเอนไซม์เปปซิน (APSC) สกัดได้โดยใช้สารละลายเปปซินปริมาณ 1% โดยน้ำหนักต่อปริมาตรในกรดแอซิติก 0.5 โมลต่อลิตร ในขั้นตอนการสกัดพบว่าผลได้ของคอลลาเจน ASC เท่ากับ 0.69 ± 0.25% และสามารถสกัดคอลลาเจน APSC ได้ 3.02 ± 00.16% เมื่อสกัดที่ 48 ชั่วโมงและ 1.42 ± 0.41% เมื่อสกัดที่ 24 ชั่วโมง ตรวจสอบน้ำหนักโมเลกุลของคอลลาเจนด้วยเทคนิค SDS-Page เทียบกับคอลลาเจนชนิดที่ 1 จากหนังหมูพบว่าคอลลาเจนที่สกัดได้จากครีบหมึกเป็นคอลลาเจนชนิด SQ ([α1]2[α2]1) ซึ่งมีลักษณะคล้ายกับคอลลาเจนชนิดที่ 1 โดย ASC ประกอบด้วยสาย α1 (202.39 kDa), α2 (159.90 kDa) และ β (281.50 kDa) และ APSC ประกอบด้วยสาย α1 (184.19 kDa), α2 (145.52 kDa) และ β (281.50 kDa) แสดงถึงส่วนเทโลเปปไทด์ถูกเอนไซม์เปปซินย่อยส่วนเทโลเปปไทด์ ฟูเรียร์ทรานส์ฟอร์มอินฟราเรดสเปกตรัมของ ASC และ …

This research investigated the effects of microplastic accumulation on the floc characteristics and mechanism of membrane fouling in a sequencing batch membrane bioreactor (SB-MBR). At first, the jar test experiment was performed for 120 min. The results indicate that spiking microplastics of 100-1,000 MPs/L resulted in an increase in floc size by 27.9±9.2% to 70.9±5.1%, while the control set (without microplastic) showed a greater increase in the floc size at 113.3±5.6% of the initial floc size. There are many enzymes required for efficient and proper pyrimidine biosynthesis. The two that are most important and were discussed in this thesis are aspartate transcarbamoylase (ATCase) and dihydroorotase (DHOase). Both play an important role in not only pyrimidine biosynthesis production, but also mechanistic regulation of de novo synthesis. Anthrax is an infection caused by Bacillus anthracis. Cystic Fibrosis (CF) is the most common, lethal autosomal recessive disorder, and is caused by mutations in the cystic fibrosis transmembrane conductance regulator protein (CFTR), an anion channel that is found in most epithelial cells lining the airway and gut. The most common mutation of CFTR is deletion of phenylalanine at position 508 (CFTR-F508del), which produces a misfolded protein. Through the ubiquitin proteasome system (UPS), this misfolded protein is ubiquitinated and signaled for degradation via the cytosolic proteasome. Previous studies demonstrating experimental restoration of F508DEL-CFTR trafficking to the plasma membrane showed partial function of the chloride channel, raising therapeutic speculations. …

The gut microbiome of domestic horses (Equus ferus caballus) can be broken down into two primary divisions of interest: (1) prokaryotic composition, consisting of bacteria and archaea and (2) fungal composition. Within the gut, there are also eukaryotic parasite communities that may interact with the microbiome. While each has been the subject of various scientific explorations, in this study we analyze the potential interconnectedness of these categories. Recently, there has been a surge in awareness of the gastrointestinal microbiome (GM) and its role in health and disease. Of particular note is an association between the GM and Parkinson’s disease (PD) and the realisation that the GM can act via a complex bidirectional communication between the gut and the brain. Compelling evidence suggests that a shift in GM composition may play an important role in the pathogenesis of PD by facilitating the characteristic ascending neurodegenerative spread of α-synuclein aggregates from the enteric nervous system to the brain. Atrial fibrillation (AF) is the most common type of sustained cardiac arrhythmia encountered in clinical practice, and its burden is expected to increase in most developed countries over the next few decades. Because AF can be silent, it is often not diagnosed until an AF-related complication occurs, such as stroke. AF is also associated with increased risk of heart failure, lower quality of life, and death. We aimed to identify practitioners’ perspectives on current research priorities in grief and bereavement care. Grief and bereavement care providers were invited to participate in a three-phase Delphi study to create expert consensus on the top priorities for grief and bereavement research. A total of 140 participants completed Phase 1, 84 completed Phase 2, and 70 completed Phase 3. These top 10 research priorities form the basis of a practice-based research agenda for grief and bereavement care to enable researchers to respond to key issues in grief and bereavement care that will ultimately improve the lives of bereaved people.

After similar extent of injury there is considerable variability in scarring between individuals, in part due to genetic factors. This study aimed to identify genetic variants associated with scar height and pliability after burn injury. An exome-wide array association study and gene pathway analysis were performed on a prospective cohort of 665 patients treated for burn injury. Outcomes were scar height (SH) and scar pliability (SP) sub-scores of the modified Vancouver Scar Scale (mVSS). DNA was genotyped using the Infinium® HumanCoreExome-24 BeadChip. Context: There are growing reports of dopamine agonist (DA)-induced impulse control disorders (ICDs) in hyperprolactinemic patients. However, the magnitude of this risk and predictive factors remain uncertain.

Objective: To determine ICD prevalence and risk factors in DA-treated hyperprolactinemic patients compared to community controls.

Design, Setting and Participants: Multicenter cross-sectional analysis of 113 patients and 99 healthy controls.

Main Outcome Measures: Participants completed a neuropsychological questionnaire consisting of the Depression Anxiety Stress Scale (DASS21), Questionnaire for Impulsive-Compulsive Disorders in Parkinson’s disease (QUIP-S), Hypersexual Behavior Inventory (HBI), Hypersexual Behavior Consequences Scale and Social Desirability Response Set Scale. Background: Despite WHO Framework Convention of Tobacco Control (FCTC) adoption, effective implementation of national smoking bans remains pending in several countries. This study quantified the association of second hand smoke (SHS) exposure and 10-year cardiovascular disease (CVD) among never smokers in such settings.

Methods: In 2001–2002, a sample of 1514 males and 1528 females (range: 18–89 years old) were randomly selected in Greece. The association between lipoprotein (a) (Lp(a)) and 10-year first fatal/nonfatal cardiovascular disease (CVD) risk in apparently healthy men and women was evaluated. The ATTICA prospective study was conducted during 2001-2012 and included n = 1514 men and n = 1528 women (age >18 years) from the greater Athens area, Greece. Follow-up CVD assessment (2011-2012) was achieved in n = 2020 participants (n = 317 cases); baseline Lp(a) was measured in n = 1890 participants. The GP has a key role in identifying patients presenting with anxiety symptoms and ensuring appropriate acute and long-term management. There are two key messages for GPs to follow: once you have made a diagnosis of an anxiety disorder, tell the patient you have a treatment for it. Second, do not let your anxiety lead you to prescribe inappropriately or overinvestigate for all possible differential diagnoses.
