Head and neck squamous cell carcinoma (HNSCC) is the sixth most common cancer worldwide and includes squamous cell carcinomas of the oropharynx and oral cavity. Patient prognosis has remained poor for decades and molecular targeted therapies are not in routine use. Here we showed that the overall expression of collagen subunit genes was higher in cancer-associated fibroblasts (CAFs) than normal fibroblasts. Focusing on collagen8A1 and collagen11A1, we showed that collagen is produced by both CAFs and tumour cells, indicating that HNSCCs are collagen-rich environments. We then focused on discoidin domain receptor 1 (DDR1), a collagen-activated receptor tyrosine kinase, and showed …

Objective: Lack of effective therapies remains a problem in the treatment of oral squamous cell carcinoma (OSCC), especially in patients with advanced tumors. OSCC development is driven by multiple aberrancies within the cell cycle pathway, including amplification of cyclin D1 and loss of p16. Hence, cell cycle inhibitors of the CDK4/6-cyclin D axis are appealing targets for OSCC treatment. Here, we determined the potency of palbociclib and identified genetic features that are associated with the response of palbociclib in OSCC. Methods: The effect of palbociclib was evaluated in a panel of well-characterized OSCC cell lines by cell proliferation assays and …

Research Publications (2016 to 2020)
Biodiesel or fatty acid methyl ester (FAME) is a green alternative fuel that degrades under long-term storage. Antioxidants are utilized in maintaining the oxidation stability of biodiesel by inhibiting the propagation of free radicals resulting in prolonged shelf life. Usage of natural antioxidants from renewable resources is preferred to sustain a green economy and potentially match synthetic antioxidants without being hazardous to the environment. In this study, the potential of antioxidants from crude extracts of Roselle seeds (Hibiscus sabdariffa) and cashew nut shells (Anacardium occidentale) which are agricultural residues that are commonly left unconsumed has been tested for biodiesel storage …

Research Publications (2016 to 2020)
The association of Epstein-Barr virus (EBV) with oral cancer has been widely reported in the past. However, previous studies mainly focused on the current infection of EBV without acknowledging the possibility of past infection in patients which may lead to oral cancer development. The present study aims to investigate the correlation between past EBV infections with Oral Squamous Cell Carcinoma (OSCC). Both Inununoglobulin M (IgM) and hnmunoglobulin G (IgG) antibodies against EBV were screened to detect the presence of EBV in sera of OSCC patients using Enzyme-Linked hnmunosorbent Assay (ELISA). The use of IgM antibody against EBV confirms current infection …

The Role Of Transient Receptor Potential (Trp) Channels In The Transduction Of Dental Pain, Mohammad Zakir Hossain, Marina Mohd Bakri, Farhana Yahya, Hiroshi Ando, Shumpei Unno, Junichi Kitagawa
Research Publications (2016 to 2020)
Dental pain is a common health problem that negatively impacts the activities of daily living. Dentine hypersensitivity and pulpitis-associated pain are among the most common types of dental pain. Patients with these conditions feel pain upon exposure of the affected tooth to various external stimuli. However, the molecular mechanisms underlying dental pain, especially the transduction of external stimuli to electrical signals in the nerve, remain unclear. Numerous ion channels and receptors localized in the dental primary afferent neurons (DPAs) and odontoblasts have been implicated in the transduction of dental pain, and functional expression of various polymodal transient receptor potential (TRP) …

Understanding Effects Of Engineering Outreach On Elementary Students, Thomas Cobb
Understanding Effects Of Engineering Outreach On Elementary Students, Thomas Cobb
Williams Honors College, Honors Research Projects
Currently, there is a desire to increase engineering understanding held by elementary students. As such, there needs to be a way to stimulate not only engineering, but other science, technology, engineering, and mathematics (STEM) topics in a young student’s lifestyle. One way to emphasize the importance of STEM while also showing how fun it can be is through different hands on activities led by individuals partaking in undergraduate studies or the industry. While having the students participate in the activities, it is important to understand what the students are learning and how the activity overall is viewed. With the help …

A high-speed 3D imaging method is developed by integrating ultrafast laser pulse shaping, temporal focusing microscopy and defocused imaging. This system does not require mechanical movement of either the stage or laser beam. Axial scanning is achieved by manipulation of group velocity dispersions on the femtosecond laser spectrum via pulse shaping method by applying modulation functions on an acoustic optical modulator which diffracts the laser spectrum. The scanning depth becomes only dependent on electronic signals which can be tuned to kHz speeds. The volumetric high-speed scanning capability was demonstrated on fluorescent microspheres suspended in a volume of 100 x100 x …
Identification Of The Cell Type Expressing The Glycine Transporter 1 (Glyt1) In The Thalamus, Patricia A. Lozano
Identification Of The Cell Type Expressing The Glycine Transporter 1 (Glyt1) In The Thalamus, Patricia A. Lozano
Open Access Theses & Dissertations
Glycine acts as an inhibitory neurotransmitter and a co-agonist of NMDAR at the glutamatergic synapses. Termination of glycinergic neurotransmission is achieved by glycine transporter 1 (GlyT1) and glycine transporter (GlyT2). Both transporters are expressed in the spinal cord, brainstem and cerebellum, but GlyT1 is also expressed in the retina and the forebrain. Previous studies have described the role and cyto- architecture of GlyT1 in the caudal areas of the brain and retina, but not in the structures of the forebrain. Based on our preliminary data, GlyT1 is highly expressed in the thalamus, an area involved in sensory and motor signal …
Ultrafine Black Carbon: A Cumulative Assessment On The Physiology Of Lung - Derived Cells, M Esther Salinas
Ultrafine Black Carbon: A Cumulative Assessment On The Physiology Of Lung - Derived Cells, M Esther Salinas
Open Access Theses & Dissertations
Combustion-derived nanomaterials are noxious ultrafine (<100 nm) aerosolized by-products generated by human activity. They pose threats to pulmonary health due to their small size, allowing them to penetrate deep into the alveolar regions causing detrimental responses downstream. Indeed, an examination between nanocarbon particle exposure and poor pulmonary health via cellular behavior was needed. I hypothesized that low-dose and long-term administrations of carbonaceous nanoparticles contribute to respiratory conditions by irritating lung-derived cells. Responses to ultrafine black carbon (UBC), a key component of airborne pollutants, by human lung A549 and murine lung LA4 epithelial cells, human peripheral-blood monocytes THP1, and murine macrophages RAW264.7 were investigated. These lines were chosen because epithelial cells and mononuclear cells (monocytes and macrophages) are the first to come in contact with inhaled particulate matter (PM) and contribute to stress and inflammation of pulmonary tissues. The cells were first plated on day zero and fed fresh UBC suspended in culture media on days one, four, and seven. The exposure regimen included three different low-level concentrations of UBC. On day ten, all cells were prepared for subsequent assays. Effects on cellular viability revealed that UBC was modestly cytotoxic while ATPase and esterase activity (termed enzyme vitality for the purpose of this study) were significantly diminished in a dose-dependent manner. Additionally, beta-galactosidase proportionally increased with UBC concentration compared to untreated cells, indicating that cellular senescence was promoted across all cell types. Multiple markers of oxidative stress were subsequently measured, showing non-uniform trends and displaying moderate accumulation of oxidative damage, contingent upon cell line and UBC treatment. However, a significant induction of inner mitochondrial membrane depolarization of human UBC-treated cells was observed; an aberrant cell-cycle progression was found across all cells. The implemented regimen of UBC exposure elicited an imbalance of free (reduced) glutathione, altered mitochondrial potential, and abnormal cell-cycle activity. This method demonstrated that modifications across both species induced changes to cellular fitness overall. The data supported that compounding nanosized black carbon exposure could negatively affect overall pulmonary cell health by distinctively modifying intracellular activity. Additionally, we further surmised that sustained exposure to nanocarbon could impair innate immunity. Therefore, reactive oxygen species (ROS), reactive nitrogen species (RNS), and cytokine levels upon continuous administrations to 3.0 μg/mL UBC after brief challenges to microbial proxies were assessed; on day 9, the cells were succumbed to three different microbial insults (LPS, Poly I:C, and Zymosan). On day 10, measurements demonstrated a moderate accumulation of oxidative stress, conditional to each cell type. Thus, UBC plus microbe approach triggered pro-oxidant levels and cytokine production across different cell types and diminished phagocytic response by murine macrophages. Based on the results, ultrafine-PM exposure may impair inflammatory function in response to pathogenic infections. In summation, our findings indicated that additive nanosized UBC in combination with microbe-like challenges may compromise cellular behavior, suggesting potential ways by which inhalation of ultrafine-PM may contribute to or exacerbate poor pulmonary health.

Cardiac failure induced by myocardial infarction is believed to be one of the primary causes of morbidity and mortality all over the world. Affecting more than 26 million people worldwide, its prevalence has been observed to increase steadily. Myocardial infarction, which intitially begins as the occlusion of of a coronary artery, results in the death of millions of cardiomyocytes by obstructing blood flow to the serviced regions of the myocardium. The dead myocardium is replaced by a dense, collagenous scar which reduces the contractility of the heart, gradually leading to heart attacks. Most of the existing treatment therapies are palliative in …

Study Of Blinking Statistics In Silver Coated Cobalt Ferrite Single Nanoprticle Using Two-Photon Fluorescence Microscopy, Rajen Kumar Goutam
Open Access Theses & Dissertations
Two-photon fluorescence microscopy is a powerful tool to study the molecular and cellular interactions. This technology is a non-invasive approach with the advantage of three-dimensional imaging up to submicron resolution. Two-photon excitation process is the result of simultaneous absorption of two photons that has special features of reduced photodamaged and elongated penetration depth on samples. In this work, blinking statistics of silver-coated Cobalt ferrite (CoFe2O4) single nanocrystal is studied using two-photon fluorescence microscopy. By defining the intensity threshold, the observed fluorescence is divided into two distinct stages: OFF and ON states. Both of these states followed the inverse power law. …
Improving Dopamine Monitoring With Ncam And The Effects Of Intranasal Oxytocin On Dopamine Signaling In The Rat Brain, Darren Earl Ginder
Improving Dopamine Monitoring With Ncam And The Effects Of Intranasal Oxytocin On Dopamine Signaling In The Rat Brain, Darren Earl Ginder
EWU Masters Thesis Collection
Dopamine (DA) is a neurohormone highly involved in learning and memory. Oxytocin (OXT), another neurohormone, has also been implicated in learning and memory. Fast-scan cyclic voltammetry (FSCV) is a method used for the real-time examination of DA neurotransmission in the brain. Implanting FSCV electrodes is an invasive technique that likely results in an inflammatory response (i.e. gliosis) that can restrict FSCV recording of DA signals. Neural cell adhesion molecule (NCAM) may reduce gliosis and potentially improve the ability of FSCV electrodes to monitor DA signaling. Chapter 1 Methods: FSCV electrodes were coated with NCAM and implanted in the rat brain. …

Neuromuscular Electrical Stimulation: A Novel Treatment Intervention For Improving Metabolic Health In An Overweight/Obese Population, Michelle J. Galvan
Open Access Theses & Dissertations
Background: Most U.S. adults (80%) do not meet minimum exercise recommendations by ACSM (CDC, 2015). Using an in vitro primary cell culture model, we and others have shown that muscle contraction induced by electrical stimulation results in increased glucose transporter 4 (GLUT4) protein, glucose uptake and mitochondrial content. Neuromuscular electrical stimulation (NMES) is a novel alternate strategy to induce muscle contraction, using electrical impulses. However, effectiveness of NMES induced muscle contraction to improve insulin sensitivity and energy expenditure is not clear. The purpose of this study was to investigate the effects of four weeks of NMES on insulin sensitivity in …

This Article examines specialty courts, including drug, alcohol, and mental health courts, which proponents claim created a revolution in criminal justice. Defendants whose underlying crime is the result of a substance use disorder or a mental health disorder can choose to be diverted into a specialty court, where they receive treatment instead of punishment. Many of these individuals, however, do not just suffer from a substance use disorder or a mental health disorder; instead, many have a “co-occurring disorder.” Approximately 8.9 million American adults have co-occurring mental health and substance use disorders, and almost half of individuals who meet diagnostic …
Ringing Changes: Systems Thinking About Legal Licensing, Joan W. Howarth, Judith Welch Wegner
Ringing Changes: Systems Thinking About Legal Licensing, Joan W. Howarth, Judith Welch Wegner
Scholarly Works
Part I examines core assumptions associated with licensing systems as well as associated ambiguities. In particular, it acknowledges multiple understandings about what “competence” is and differing assumptions about how to evaluate or measure it. Part I thus sets forth important predicates for our argument that only a multi-faceted licensing system can do what is needed in assuring minimal competence, and that not all forms of competence are best measured by traditional licensing examinations.
Part II raises the possibility of creating a post-first-year examination designed to assess critical thinking in the context of the first-year curriculum. It also considers ways in …

Mediation: An Unlikely Villain, Thomas O. Main
Scholarly Works
Professor Main argues that the modem ADR movement (and mediation in particular), rather than some (other) ideology, beget the pleading and summary judgment standards that exemplify contemporary practice and procedure in the fourth era in the history of American civil procedure. The other key reforms of the fourth era-the vanishing trial, the embrace of ADR, judicial case management and the pursuit of settlement by any means necessary-are more obviously tied to the modem ADR movement. Blame for all of the key fourth era reforms is thus traceable to the modern ADR movement. This, in turn, matters because it is generally …

Judicial decision-making is not a neutral and logical enterprise that involves applying clear rules to agreed-upon facts. Legal educators can and should help students learn more about how judges actually go about making their decisions. The study of re-imagined judicial decisions, such as the alternative judgments from various Feminist Judgments Projects, can enrich the study of law in multiple ways. First, seeing a written decision that differs from the original can help students think “outside the box” constructed by the original opinion by showing them a concrete example of another perspective written in judicial language. Second, the rewritten judgments show …

Broken Bodies And Broken Dreams: How Social Safety Net Programs Subsidize Professional Boxing And The Need To Improve Legal And Health Protections For Prizefighters, Robert I. Correales
Scholarly Works
This article explores the lack of basic health and insurance protections for professional boxing participants and proposes changes in policy that will reflect the dangerous nature of professional boxing and the modem cost of medical services. Recognizing that a legislative remedy may never arrive, this article also examines previously unexplored or underutilized legal doctrines such as tort and workers' compensation law that may provide an alternative to inadequate insurance protection, and suggests a more aggressive approach along those lines to compel fuller protection for prizefighters.

Thus Sayeth The Matthean Lord: Exploring The Intersection Of The Quest For The Historical Jesus And The Proclaimation Of The Biblical Jesus, Edward T. Wright
The Asbury Journal
The themes of Biblical Studies and mission meet in a significant way at the intersection of the quest for the historical Jesus (an aspect of biblical studies) and the proclamation of the biblical Jesus (an aspect of the mission of the Church). This intersection undoubtedly occurs in the context we currently find ourselves, the evangelical seminary setting. This paper explores a passage, Matthew 4:-11, that has both significant theological content but also some rather difficult historical issues. When confronted with this type of problem, is it possible to navigate the passage in such a way that the historical Jesus and …

Filamentous Bacteriophage Associated With Shaping Community Structure And Fitness Of Invasive Vibrio Parahaemolyticus St36, Jillian Means
Master's Theses and Capstones
Vibrio parahaemolyticus a ubiquitous coastal inhabitant, is the leading cause of bacterial seafood-borne illnesses in the United States. An increasing number of reported cases and rapid expansion into new areas has led to the classification of V. parahaemolyticus as an emergent pathogen. Most strains of V. parahaemolyticus are not virulent; however, the spread of virulent lineages from their native ranges to new locations has contributed drastically to the increase in vibriosis attributed to V. parahaemolyticus in recent years. In the United States (US), sequence type (ST) 36, a virulent strain endemic to the Pacific Northwest (PNW), spread from its native …
