Authorship is not merely an act of putting pen to paper or fingers to keyboard; it is a social identity performance that includes the use of multiple media. Authors must be hyper- visible to cut through the dearth of information, entertainment options, and personae vying for attention in our supersaturated media environment. As they enter the literary world, writers consciously create characters and narratives around themselves, and through the consistent and believable enactment of these features, authors are born. Periodontitis is an inflammatory disease of the oral cavity induced by anaerobic bacteria, that remains to be the primary cause of tooth loss in adults worldwide. Finding an anti-microbial therapeutic to selectively target periodontal pathogens has proven to be difficult, and current treatment modalities only provide a transient benefit. Amixicile is a non-toxic, readily bioavailable novel antimicrobial that targets strict anaerobes through inhibition of the activity of Pyruvate Ferredoxin Oxidoreductase (PFOR), a major enzyme mediating oxidative decarboxylation of pyruvate, a critical step in metabolism. Head and neck cancer accounts for approximately 3 percent of all cancers in the United States, and over 90% of them are head and neck squamous cell carcinoma (HNSCC). Chemotherapeutic drugs that treat HNSCC can activate BCL-2 family dependent apoptosis. Pro-apoptotic NOXA induced by adenovirus (Ad-NOXA) or fenretinide inactivates anti-apoptotic MCL-1, while ABT-263 can inactivate other anti-apoptotic BCL-2 family members such as BCL-2 and BCL-XL. We used p53 inactive HN8 and HN12, p53 wild-type UMSCC1, and HPV-positive UMSCC47 human HNSCC cell lines and five mouse HNSCC cell lines. Studies on repeated exposure to opioids have been carried out for decades yet the mechanisms for certain phenomena such as tolerance are still not fully understood. Furthermore, different medications, such as frequently prescribed benzodiazepines, or different disease states, such as HIV, have their own effects and interactions with chronic opioid exposure that are not fully understood. The overall objective of this dissertation was to investigate the complexities of chronic opioid exposure and how different disease states and medications may modulate the effects of chronic opioids. The ultimate goal of this project has been to develop a computational model for quantifying the interactions between of a body of fluid and a fibrous surface. To achieve this goal, one has to develop a model to create virtual structures that resemble the morphology of a fibrous surface (Objective-1) as well as a model that can simulate the flow of a fluid over these virtual surfaces (Objective-2). In urban population genetic studies, the "urban fragmentation model" predicts that urbanization acts as a barrier that isolates native populations, and can lead to reduced gene flow and increased genetic drift between populations. The “urban facilitation model” predicts urban areas act as corridors to increase dispersal among urban areas, and can lead to higher genetic diversity within and lower differentiation between urban areas.

In a review of the current literature, we found that there is no consistent signature of reduced within-population genetic diversity or increased between-population genetic differentiation. Analyses that investigate the urban barriers to gene flow also found no …

The aim of this study was to determine the trends in protocol, success rates, and consistency in follow up of revascularization procedures in a controlled environment. Patients of the Virginia Commonwealth University School of Dentistry were identified who were offered revascularization therapy as a treatment option on immature permanent teeth from January 1, 2010 to May 31, 2017. A total of 77 patients and 78 teeth were evaluated for revascularization therapy. For patients accepting treatment, records were reviewed for outcome assessment and consistency of follow up. Unmanned aerial vehicle (UAV) technology was used to determine tidal extent in Kimages Creek, a restored tidal wetland located in Charles City County, Virginia. A Sensefly eBee Real-Time Kinematic UAV equipped with the Sensor Optimized for Drone Applications (SODA) camera (20-megapixel RGB sensor) was flown during a single high and low tide event in Summer 2017. Collectively, over 1,300 images were captured and processed using Pix4D. Horizontal and vertical accuracy of models created using ground control points (GCP) ranged from 0.176 m to 0.363 m. Black individuals have been disproportionately targeted by law enforcement, most notably in shooting deaths at the hands of police, and has contributed to a general distrust of law enforcement. A rise in citizen journalism has helped document excessive use of force by police on video; however, little is known how individuals respond to viewing this type of media. A history of race-based stress is likely to contribute to an expectation of racism and may impact how individuals respond to videos of the treatment of unarmed Black men at the hands of police, with some evidence suggesting ethnic identity may moderate …

This thesis examines the work of nonprofit organizations collaborating with communities to build civic capacity in North Highland Park, a neighborhood in Richmond, Virginia. Place-focused planning strategies during the twentieth century diminished civic capacity in the community and fostered isolation. Today, collaborative community work in Highland Park is incorporating the assets, resources and knowledge held in the community into strategies to improve quality of life using collaborative learning. A case study approach closely analyzes community engagement and revitalization processes in North Highland Park between 2011 and 2017. This thesis is an examination of autobiographical solo performance. It explores the use of personal trauma and illness in the dramatic form. In addition to investigating how other solo performing artists utilize their medical conditions in their work, this thesis gives some historical context to the author’s own process and development. The thesis culminates in the author’s solo performance script and a desire for its audience to find solace and compassion through the experience of witnessing it being performed.

Periodontal disease affects 47% of Americans over 30 and is a growing global concern. Current treatments for periodontal disease focus on the mechanical elimination of periodontal biofilms. Very few treatments are available that target the rampant, unregulated host immune response that is ultimately responsible for tissue degradation. BET proteins have been shown to play critical roles in inflammatory gene regulation and are therefore potentially ideal therapeutic targets for treating periodontal disease. Self-cleaning, anti-corrosion, anti-icing, dropwise-condensation, and drag-reduction are some applications in which superhydrophobic surfaces are implemented. To date, all the studies associated with superhydrophobic surfaces have been dedicated to understanding the liquid interaction with surfaces that are macroscopically smooth. The current study investigates the solid-liquid interaction of such surfaces which are fully decorated with macroscopic ridges (ribbed surfaces). In particular, the drop motion and impact on our newly designed non-wettable ribbed surface have been investigated in this work. Ubiquitin (Ub) is a small, 76 amino acid, and post-translational modification (PTM) protein in eukaryotes. Modification of a substrate protein via the covalent attachment of the C-terminal glycine of Ub to the ε-amino group of lysine residues in a substrate is termed ubiquitination. Unlike, other PTM proteins, Ub can form polyUb chains at one or more of its seven lysine residues. (K6, K11, K27, K29, K33, K48, and K68). The metabolic pathways required for adenosine triphosphate (ATP) production within the cell are well understood, however recent publications suggest that metabolic pathways are closely linked to immune cell activation and inflammatory diseases. There has been little examination of the metabolic pathways that modulate mast cell activation and the feedback regulator lactic acid. The vaginal microbiome plays an important role in reproductive health and pregnancy. It has coevolved with humans and has direct effects on reproductive success, rendering selective pressure more pronounced at this site. Herein, we probe coevolution of the vaginal microbiome using a systems-level approach. In Chapter 2, we examine the evolutionary trajectory of two vaginal Veillonellaceae phylotypes evolved from an ancestral gastrointestinal lineage to inhabit the vaginal niche. We present evidence of their divergence and subniche specification and describe their differential associations with vaginal infection and pregnancy. Today 49 million individuals in the US are affected by food insecurity (Whittle et. al., 2015). Low-income populations tend to depend solely on convenient stores for calorically-dense, nutrient poor sustenance, and suffer from health problems that drastically shorten or debilitate lives (Dhurandhar et al., 2016). In Richmond, Virginia there is a dramatic divide between wealthy and low-income communities in terms of the accessibility as well as the types and quality of foods available. Music, it has been shown, has the potential to both create social cohesion and encourage conflict resolution (Koelsch, Offermanns, & Franzke, 2010). In a city with deep roots in slavery, music may be more powerful than language or the toppling of monuments to help create a cultural identity that every demographic can fit. Not only can music improve mood in an individual, but musical contagion affects large groups at once. Creating a space to equalize access to and share the experience of music—both making and appreciation—would give the city an opportunity for reconciliation and community bonding.
