Targeted Insertion Of The Mping Transposable Element, Ashley E. Strother, Stephanie S. Diaz, Mary E. Baker, C. Nathan Hancock
Journal of the South Carolina Academy of Science
Class II DNA Transposable Elements (TEs) are moved from one location to another in the genome by the action of transposase proteins that bind to repeat sequences at the ends of the elements. Although the location TE insertion is mostly random, the addition of DNA binding domains to the transposase proteins has allowed for targeted insertion of some elements. In this study, the Gal4 binding domain was added to the transposase proteins, ORF1 and TPase, which mobilize the mPing element from rice. The Gal4:TPase construct was capable of increasing the number of mPing insertions into the Gal2 and Gal4 promoter …

Arkansas’S Rosenwald Schools 1917-1924, Chelsea Deserea Mcnutt
Student Theses and Dissertations
This work explores Arkansas Rosenwald Schools in rural African American communities from 1917-1924. Jim Crow laws governed the lives of black southerners throughout the region. Such laws unjustly marginalized them, their communities, and their schools. In 1868, Arkansas passed Act 52, which mandated segregated and unequal schools for black and white Arkansans. Black Arkansans, both children and adults, lacked educational opportunities, especially those residing in rural areas. African Americans, of all socioeconomic backgrounds, understood the importance of education. Education allowed black Arkansans to reshape their lives and futures. Much of this effort was aided by resources from the Rosenwald Fund. …

The Island Night Lizard (Xantusia riversiana) was removed from the federal list of threatened species in May 2014. This strongly differentiated species is endemic to 3 of the southern California Channel Islands—San Clemente, San Nicolas, and Santa Barbara. Suitable habitat for Island Night Lizards is extensive on San Clemente Island, and the species is abundant there. Habitat is limited and fragmented, however, on San Nicolas Island and small Santa Barbara Island. Bringing together extensive field surveys and mark-recapture sampling, we synthesize available data for Island Night Lizards on San Nicolas Island and calculate population and density estimates for …

Capture-recapture methods are commonly used to estimate abundance and density of wild animal populations. Although a variety of sophisticated analytical techniques are available to evaluate capture-recapture data, vertebrate monitoring programs often lack the resources (e.g., time, personnel, and/or analytical expertise) to apply these methods. As an alternative, simple population indices, such as counts of unique individuals, may provide sufficient information to detect meaningful changes in population size. In this study we investigated whether a population index, easily generated from mark-recapture data under all conditions, might be used to provide valid ecological information for managers interested in long-term population trends of …

Dual Gain And Loss Of Cullin 3 Function Mediates Familial Hyperkalemic Hypertension, Ryan J. Cornelius, Chang Zhang, Kayla J. Erspamer, Larry N. Agbor, Curt D. Sigmund, Jeffrey Singer, Chao-Ling Yang, David H. Ellison
Biology Faculty Publications and Presentations
Familial hyperkalemic hypertension is caused by mutations in with-no-lysine kinases (WNKs) or in proteins that mediate their degradation, kelch-like 3 (KLHL3) and cullin 3 (CUL3). Although the mechanisms by which WNK and KLHL3 mutations cause the disease are now clear, the effects of the disease-causing CUL3Δ403-459 mutation remain controversial. Possible mechanisms, including hyperneddylation, altered ubiquitin ligase activity, decreased association with the COP9 signalosome (CSN), and increased association with and degradation of KLHL3 have all been postulated. Here, we systematically evaluated the effects of Cul3Δ403-459 using cultured kidney cells. We first identified that the catalytically active CSN subunit jun activation domain-binding …

Serotonin (5-HT) is an important regulator of numerous aspects of flatworm biology, ranging from neuromuscularfunctionto sexualmaturationandegglaying. Intheparasiticblood flukeSchistosomamansoni,5-HTtargets several G-protein coupled receptors (GPCRs), one of which has been demonstrated to couple to cAMP and regulate parasite movement. This receptor, Sm.5HTRL, has been successfully co-expressed in mammalian cells alongside a luminescent cAMP-biosensor, enabling pharmacological profiling for candidate anti-schistosomal drugs. Here, we have utilized this assay to perform structure-activity investigations of 143 compounds containing previously identified alkaloid natural product pharmacophores (tryptamines, aporphines and protoberberines) shown to regulate Sm.5HTRL. These experiments mapped regions of the tryptamine pharmacophore amenable and intolerant to substitution, highlighting …
