Nephritis is one of the major complications of systemic lupus erythematosus (SLE). While glucocorticoids (GC) are frequently used as the first line treatment for lupus nephritis, long-term GC usage is often complicated by severe adverse effects. To address this challenge, we have developed a polyethylene glycol (PEG)-based macromolecular prodrug (ZSJ-0228) of dexamethasone, which self-assembles into micelles in aqueous media. When compared to the dose equivalent daily dexamethasone 21-phosphate disodium (Dex) treatment, monthly intravenous administration of ZSJ-0228 for two months significantly improved the survival of lupus-prone NZB/W F1 mice and was much more effective in normalizing proteinuria, with clear histological evidence …

DNA damage is frequently induced in cells by both endogenous and exogenous agents. DNA damage, particular double strand breaks (DSBs) may lead to genomic instability, and the progression of cancer, aging, neurodegeneration, and other human diseases. The cell employs two major DSB repair pathways, including homologous recombination (HR) and Non-homologous end joining (NHEJ), but the detailed mechanisms of DSB repair remain to be further revealed.
In the first part of this study, we characterized a plasmid-based assay to investigate NHEJ repair in Xenopus egg extracts. Our data argued for a preference for the precise repair by the NHEJ machinery and …

Chronic pain afflicts millions of people worldwide. Particularly, the inflammatory conditions in the incurable disease rheumatoid arthritis (RA) generate persistent pain in its sufferers, for which a number of different analgesics have been prescribed, such as glucocorticoids (GCs) and opioids. However, administration of these pain- mitigating pharmaceutics is implicated in the development of adverse systemic effects due to their non-specific tissue distribution and quick excretion, eliciting the need for high dosing frequencies. Lutein + zeaxanthin are carotenoids most recognized in eye health, but less is known about their status and transfer during pregnancy. While quantified in maternal and umbilical cord blood, they have never been analyzed in placenta tissue. Therefore, the purpose of this dissertation is to firstly quantify lutein + zeaxanthin concentrations in human placenta and to later correlate with levels in maternal dietary intake, maternal serum, and umbilical cord blood. The purpose of this investigation was to evaluate the effects of Class II correctors and functional appliances on the dentoalveolar support of the mandibular incisors during orthodontic treatment. Twenty six patients who were treated with a ForsusTM, Herbst, or MARA participated this study. Lateral cephalometric radiographs, limited field-of-view CBCTs, and periodontal measurements were collected both the day of appliance activation and day of appliance deactivation. Purpose: The purpose of this investigation was to evaluate the immediate and long-term effects of palatal expansion on the transverse dimension of the nasomaxillary complex.
Materials and Methods: Twenty-eight patients’ CBCTs were obtained at four time points: pre-expansion (T1), post-expansion (T2), pre-treatment (T3), and middle or end of orthodontic treatment (T4). N-methyl-D-aspartate receptors (NMDARs) are ligand-gated ion channels consisting of two GluN1 subunits and two other subunits from among GluN2A-2D and GluN3A-3B subunits. NMDARs play critical roles in synaptic plasticity, learning and memory, and higher brain function such as cognition and perception. Dysfunction of NMDARs (hyper-function and hypo-function of NMDARs) are related to various diseases, including stroke, schizophrenia, Alzheimer’s disease, and others. However, to date, NMDARs antagonists have mostly failed in clinical trials due to adverse effects.

Breast cancer (BC) remains one of the most frequently diagnosed cancer worldwide. Bone is one of the most common sites and often the first clinical indication of metastatic progression of BC. Current treatments including radiation, surgery, and chemotherapy are rarely curative with limited effect in overall survival. The challenge is true especially for patients with triple-negative BC that are not responsive to receptor-targeted therapy, or patients who had exposure to chemotherapy before and might already gain the resistance to some of the drugs.

The aim of this study was to investigate the differences in nutrient limitation within the beach and marsh environments at Waties Island, SC. Conducting the experiment on Waties Island allowed most physical factors to be held as constant, meaning the marsh and beach environments would serve as the only variables. Experiments were performed in the winter, spring, summer and fall of 2018 to determine temporal changes in limitation to phytoplankton and cyanobacteria growth. Maturase K (MatK) is the only group II intron encoded protein in the chloroplast of land plants. Maturases are prokaryotic enzymes that catalyze formation of the lariat structure needed for intron removal from precursor RNAs. The chloroplast maturase MatK, is a descendant of prokaryotic maturases, however, unlike its prokaryotic relatives, MatK is thought to catalyze excision of, not only its own intron, but also the introns of other group II introns in the plastome. In the southern United States, institutional forest owners engaged in forest certification programs often retain unharvested or less-intensively harvested vegetation when clearcut harvesting intensively managed pine (Pinus spp.) forests (“IMPFs”), a practice called ‘green tree retention’. I investigated resultant patterns of land cover and retained structural elements in recently-harvested IMPF management units (“MUs”) and related them to avian biodiversity to provide information to support harvest decisions. White Crappie (Pomoxis annularis) and Black Crappie (P. nigromaculatus) are popular North American gamefish; however, frequent fluctuations in year class strength present a management challenge for recreational fisheries. Intensive aquaculture production has the potential to address this challenge through controlled hatchery reproduction for supplemental stocking, but further study is needed to refine and optimize techniques. Therefore, this study evaluated the effects of hormone injection timing on latency period and spawning success, examined effective cryopreservation techniques for black-stripe Black Crappie sperm (a preferred hatchery phenotype), and compared simulated spring duration on out-of-season spawning success. Background: Epigenetic modifications including DNA methylation and post-translational modifications of histones are known to regulate gene expression. Antagonistic activities of histone acetyltransferases (HATs) and histone deacetylases (HDACs) mediate transcriptional reprogramming during insect development as shown in Drosophila melanogaster and other insects. Juvenile hormones (JH) play vital roles in the regulation of growth, development, metamorphosis, reproduction and other physiological processes. However, our current understanding of epigenetic regulation of JH action is still limited. Hence, we studied the role of CREB binding protein (CBP, contains HAT domain) and Trichostatin A (TSA, HDAC inhibitor) on JH action.

This narrative study examined (1) how women raising school age children with disabilities in one of Zimbabwe’s urban areas, articulated and conceptualized (a) their children’s disabilities and (b) their experiences as mothers of children with disabilities and (2) how their social status, marital status, age, ethnicity, and/or race intersected with their experiences of raising children with disabilities. A localized understanding of how mothers view their children’s disabilities was important because it influences results of any interventions their children might receive. In this article, the authors describe a multi-site case study of five 4-year postsecondary institutions in Missouri that used evidence-based strategies to support first-generation students in their efforts to persist in and complete college. In the case study, data from qualitative interviews of university administrators in academic and student support services, as well as focus groups of underrepresented students, were analyzed and coded using grounded theory. The resulting themes were compared to prior research on first-generation students, resulting in three broad areas of evidence-based practice with potential to increase college success for first-generation students. The land snails and slugs have the highest level of endemism among all major animal groups on the California Channel Islands, with nearly 75% of the native terrestrial species confined to one or more of the 8 islands. In spite of this endemism, and in spite of the rarity of some species, the snails and slugs are one of the most poorly known groups. We present the first comprehensive overview of the land mollusk fauna of the Channel Islands, along with the results of recent intensive inventory studies. Vesicular stomatitis virus (VSV) is a negative sense, single-stranded RNA (ssRNA) virus that is currently studied for its uses as a vaccine viral vector and its potential in oncolytic therapy. The M protein of VSV is the active participant in the virus’s abrogation of the host innate immune response. This activity is thought to be due to the M protein’s ability to downregulate overall host transcription by blocking nuclear transport. In this project, we explore using a musical space to represent the properties of amino acids. We consider previous mappings and explore the limitations of these mappings. In this exploration, we will propose a new method of mapping into musical spaces that extends the properties that can be represented. For this work, we will use amino acid sequences as our example mapping. The amino acid properties we will use include mass, charge, structure, and hydrophobicity. Finally, we will show how the different musical properties can be compared for similarity.

Purpose: The purpose of this project is to identify if individuals with first episode of schizophrenia would utilize computerized cognitive trainings after the study. Using this information, we can identify better ways to support individuals with first episode of schizophrenia in utilizing cost-effective interventions to improve cognition such as attention, memory, and problem solving among ages 18-35 individuals.

The chicken bursa of Fabricius is a primary lymphoid tissue important for B-cell development. Our long-term goal is to understand the role of bursal microenvironment in an early B-cell differentiation event initiating repertoire development through immunoglobulin gene-conversion in the chick embryo. We hypothesize that early bursal B-cell differentiation is guided by signals through cytokine receptors. Our theory is based on previous evidence for expression of the receptor tyrosine kinase superfamily members and interleukin receptors in unseparated populations of bursal B-cells and bursal tissue. Men are most susceptible to prostate cancer in the United States. The general treatment options include surgery, hormone therapy, chemotherapy, and radiation therapy. But in recent days, the nanoformulation have shown promising applications in overcoming the drawbacks of the currently available treatment options. To complement, we tried to enhance the capability of the nanoparticle formulation by loading them with a novel drug combination for the treatment of prostate cancer. Magnetic Resonance Imaging (MRI) is increasingly being used as a diagnostic tool for cancer. We propose a novel molecular probe, Gadolinium-DTPA disulfide-bonded Doxorubicin (Gd-DTPA-SS-Doxo) encapsulated in iron oxide which could provide a dual modality for detecting malignant growth while simultaneously targeting treatment options. We first synthesized Gd-DTPA-SS-Doxo followed by encapsulation within the poly (acrylic acid) (PAA) coating of iron oxide nanoparticle (IONP), producing a nanoprobe (IO-Gd-DTPA-SS-Doxo) with quenched longitudinal spin lattice magnetic relaxation (T1). Introduction: Ambulation can be used to monitor the healing of lower extremity fractures. However, the ambulatory behavior of tibia fracture patients remains unknown due to an inability to continuously quantify ambulation outside of the clinic. The goal of this study was to design and validate an algorithm to assess ambulation in tibia fracture patients using the ambulatory tibial load analysis system during recovery, outside of the clinic.
