การผลิตและลักษณะสมบัติของรีคอมบิแนนท์แอนติบอดี Scfv-Fc ต่อ Notch1 และ Notch2 โดยใช้เซลล์ไลน์ Hek-293t, วรวรรณ บุญรัศมี

วิถีสัญญาณ Notch ประกอบด้วย รีเซบเตอร์ 4 ชนิด (Notch 1-4) และลิแกนด์ 5 ชนิด (Delta-1, 3, 4 และ Jagged 1, 2) การเกิดอันตรกิริยาระหว่างลิแกนด์และรีเซบเตอร์ จะส่งผลให้เกิดการเปลี่ยนแปลงบริเวณ NRR ทำให้เกิดการเผยตำแหน่งที่มีการตัดโดยเอนไซม์ซึ่งนำไปสู่การตัดโดยแกมมาซีครีเทส ดังนั้น NRR จึงเป็นตำแหน่งสำคัญเพื่อรักษาเสถียรภาพของ NRR ไว้เพื่อป้องกันการตัดของ Notch รีเซบเตอร์ วิถีสัญญาณ Notch ทำหน้าที่ควบคุมการเพิ่มจำนวน การเปลี่ยนสภาพ ตลอดจนถึงการตายของเซลล์หลายประเภท ความผิดปกติของสัญญาณ Notch ส่งผลให้เกิดการพัฒนาเป็นมะเร็งชนิดต่าง ๆ ได้ มีการใช้ยากดการทำงานของเอนไซม์แกมมาซีครีเทส (GSI) เพื่อการยับยั้งวิถีสัญญาณ Notch แต่มีรายงานว่า GSI ทำให้เกิดผลข้างเคียงที่ไม่พึงประสงค์ เนื่องจากมีซับสเตรทหลายชนิด รีคอมบิแนนท์แอนติบอดีในรูป scFv-Fc ซึ่งเป็นโปรตีนลูกผสมประกอบด้วย scFv และ Fc ของอิมมูโนโกลบูลินจีของมนุษย์ ได้ถูกนำมาประยุกต์ใช้ทางการรักษาบำบัดเนื่องจากมีคุณสมบัติที่ดีกว่ามอโนคลอนอลแอนติบอดีทั้งโมเลกุลหลายด้าน ในงานวิจัยนี้มีวัตถุประสงค์ในการผลิตรีคอมบิแนนท์แอนติบอดีชนิด scFv-Fc ต่อบริเวณ NRR ของ Notch1 และ Notch2 โดยใช้เซลล์ไลน์ HEK-293T และศึกษาลักษณะสมบัติของรีคอมบิแนนท์แอนติบอดี scFv-Fc โดยทดสอบการจับกับ Notch1 และ Notch2 และผลกระทบต่อวิถีสัญญาณ Notch ในเซลล์มะเร็งเม็ดเลือดขาว Jurkat จากผลการทดลองพบว่าสามารถผลิตรีคอมบิแนนท์แอนติบอดี scFv-Fc ต่อ Notch1 และ 2 ได้ในเซลล์ไลน์ HEK-293T เมื่อทำการทรานส์เฟคชันพลาสมิดที่มีชิ้นยีนของ scFv-Fc เข้าสู่เซลล์ โดยนำโปรตีนที่ได้ในอาหารเลี้ยงเซลล์ ไปแยกบริสุทธิ์ด้วยคอลัมน์โครมาโทกราฟีสัมพรรคภาพ พบว่าบน SDS-PAGE ปรากฏโปรตีนเพียงแถบเดียว เมื่อผ่านการแยกบริสุทธิ์ด้วยสองขั้นตอนโดยใช้คอลัมน์ต่อ His-tag และ คอลัมน์โปรตีน A นำแอนติบอดีที่ได้ไปทดสอบรีคอมบิแนนท์แอนติบอดีต่อบริเวณ NRR โดย anti-Notch1 …

วิทยานิพนธ์นี้เป็นการนำเสนองานวิจัย การพัฒนาวิธีการใหม่เพื่อควบคุมและเฝ้าระวังโรคติดเชื้อ โดยประกอบด้วยเนื้อหาสำคัญสองส่วน ได้แก่ (i) การพัฒนาแบบจำลองเซลล์ลำไส้ของมนุษย์ที่เพาะเลี้ยงบนกระดาษที่มีการดัดแปรพื้นผิว เพื่อใช้ประโยชน์ในการศึกษากลไกการบุกรุกของเชื้อจุลชีพก่อโรคในระบบทางเดินอาหาร และ (ii) การพัฒนาต้นแบบไบโอเซนเซอร์แบบไร้สายเพื่อการตรวจติดตามการเจริญของเชื้อจุลชีพก่อโรค โดยในส่วนแรก เซลล์ลำไส้ของมนุษย์ HT-29 จำนวน 200,000 เซลล์ ถูกเพาะเลี้ยงในบริเวณชอบน้ำของกระดาษกรอง Whatman No.1 โดยกระดาษถูกดัดแปรพื้นผิวด้วยสารแมทริกซ์นอกเซลล์ (Extracellular matrix: ECM) ชนิดต่าง ๆ ได้แก่ Matrigel, Collagen-1 และ Laminin พื้นที่เพาะเลี้ยงเซลล์มีขนาด 12.56 ตารางมิลลิเมตร ภายใต้สภาวะที่เหมาะสม เซลล์ HT-29 ที่ถูกเพาะเลี้ยงบนกระดาษ มีชีวิตรอดได้ถึง 28 วัน และแสดงลักษณะของเซลล์ที่เจริญเต็มที่ โดยมีการแสดงออกของวิลไล (villi) ที่ผิวเซลล์ด้านบน (apical) และมีการแสดงออกของโปรตีน ZO-1 ซึ่งแสดงถึงเซลล์มีการสร้างไทต์จังก์ชันเกิดขึ้น จากการใช้สารเชื่อมพันธะสำหรับเชื่อมพันธะ ECM แต่ละชนิด พบว่าเมื่อใช้สารเจนิพินในการเชื่อมพันธะ เซลล์มีชีวิตรอดทั้งหมด (100 -112.8%) ในขณะที่มีเพียง 1.1 - 67.3 % ของเซลล์รอดชีวิต หากใช้สารกลูตารัลดีไฮด์ในการเชื่อมพันธะ แต่การใช้สารเชื่อมพันธะทั้งสองชนิด ทำให้เซลล์ไม่สามารถแบ่งตัวมีรูปร่างแบบสามมิติได้ เมื่อสังเกตด้วย SEM เซลล์มีลักษณะแบน การดัดแปรพื้นผิวกระดาษด้วยสาร ECM ชนิด Matrigel ส่งผลให้เซลล์มีชีวิตรอดมากที่สุดและสาร ECM ชนิด Laminin ทำให้เซลล์เกิดการแปรสภาพ (cell differentiation) ได้ดีที่สุด งานวิจัยส่วนที่ 2 เป็นการพัฒนาไบโอเซนเซอร์ไร้สายสำหรับติดตามการเจริญของเชื้อจุลชีพก่อโรค ได้แก่ เชื้อ Escherichia coli, Staphylococcus aureus, Pseudomonas aeruginosa, Streptococcus gordonii และ Candida albicans พบว่าเชื้อดังกล่าวสามารถสร้างไบโอฟิล์มบนผิวของขั้วไฟฟ้าคาร์บอนอสัณฐาน (glassy carbon electrode) และสามารถให้ค่าศักย์ไฟฟ้าที่เป็นลบได้ …

ผลของกาวติดฟันเทียมต่อประสิทธิภาพการบดเคี้ยวและแรงบดเคี้ยวสูงสุดในผู้ป่วยที่ใส่ฟันเทียมทั้งปาก, หยาดฝน พลศรี

วัตถุประสงค์เพื่อศึกษาผลของกาวติดฟันเทียมต่อประสิทธิภาพการบดเคี้ยว (masticatory performance) และแรงบดเคี้ยวสูงสุด (maximum occlusal force) ของผู้ป่วยฟันเทียมทั้งปาก ผู้ป่วยสันเหงือกว่างที่ใส่ฟันเทียมทั้งปากบนและล่างจำนวน 65 คน อายุเฉลี่ย 68.9±7.2 ปี เป็นเพศชาย 39 คน เพศหญิง 26 คน มีคุณภาพฟันเทียมอยู่ในระดับที่ยอมรับได้ (กลุ่ม A) และยอมรับไม่ได้ (กลุ่ม UA) จำนวน 44 และ 21 คน ตามลำดับ มีลักษณะของเนื้อเยื่อรองรับฟันเทียมตามระบบของ American College of Prosthodontists กลุ่ม I, II, III และ IV จำนวน 19 30 11 และ 5 คนตามลำดับ ผู้ป่วยเข้ารับการทดสอบประสิทธิภาพการบดเคี้ยวด้วยการเคี้ยวถั่วลิสงจำนวน 20 รอบวงเคี้ยว ถั่วลิสงที่ผ่านการเคี้ยวแล้วจะถูกร่อนผ่านเครื่องสั่น ด้วยตะแกรงทดสอบมาตรฐานจำนวน 12 ชั้น เพื่อวิเคราะห์หาขนาดอนุภาคกลางของถั่วลิสง (median particle size) และเข้ารับการทดสอบแรงบดเคี้ยวสูงสุดด้วยการกัดแผ่นฟิล์มที่อ่อนไหวต่อความดัน (pressure sensitive film) โดยวางแผ่นฟิล์มระหว่างด้านบดเคี้ยวของฟันเทียมบนและล่างในตำแหน่งสบสนิทที่สุด (maximum intercuspal position) วิเคราะห์และแปรผลด้วยโปรแกรมสถิติสำเร็จรูป เพื่อเปรียบเทียบประสิทธิภาพการบดเคี้ยวและแรงบดเคี้ยวสูงสุดระหว่างก่อน (T1) และหลังการใช้กาวติดฟันเทียมอย่างต่อเนื่องเป็นระยะเวลา 1 เดือน (T2) สถิติที่ใช้ในการทดสอบคือ paired t-test ที่ระดับความเชื่อมั่นร้อยละ 95 ผลการศึกษาพบว่าเมื่อเปรียบเทียบก่อนและหลังการใช้กาวติดฟันเทียม ประสิทธิภาพการบดเคี้ยวเพิ่มขึ้นอย่างมีนัยสำคัญในกลุ่ม A (P = 0.035) แต่ไม่มีความแตกต่างอย่างมีนัยสำคัญทางสถิตในกลุ่ม UA (P = 0.313) แรงบดเคี้ยวสูงสุดลดลงอย่างมีนัยสำคัญในกลุ่ม A (P = 0.020) แต่ไม่มีความแตกต่างอย่างมีนัยสำคัญทางสถิติในกลุ่ม UA (P …

ประสิทธิภาพของซิลเวอร์ไดเอมีนฟลูออไรด์ในการคืนแร่ธาตุในรอยผุชั้นเนื้อฟัน: การวิจัยเชิงทดลองในห้องปฏิบัติการ, กันตพร คุณพนิชกิจ

การศึกษานี้มีวัตถุประสงค์เพื่อเปรียบเทียบประสิทธิภาพของซิลเวอร์ไดเอมีนฟลูออไรด์ที่มีจำหน่ายในท้องตลาด ในการคืนแร่ธาตุในรอยผุชั้นเนื้อฟันของฟันน้ำนม การศึกษานี้เป็นการศึกษาในห้องปฏิบัติการโดยใช้ชิ้นฟันตัวอย่างจากฟันกรามน้ำนมที่มีรอยผุตามธรรมชาติชั้นเนื้อฟันจำนวน 36 ชิ้น วัดความหนาแน่นแร่ธาตุของรอยผุเริ่มต้นด้วยเครื่องถ่ายภาพรังสีคอมพิวเตอร์ระดับไมโครเมตรแล้วนำมาคำนวณความลึกรอยผุและความหนาแน่นแร่ธาตุเฉลี่ย แบ่งชิ้นฟันเป็น 3 กลุ่ม กลุ่มที่ 1 ทา Saforide® (Toyo Seiyaku Kesei, Japan) กลุ่มที่ 2 ทา Advantage ArrestTM (Elevate Oral Care, USA) และกลุ่มที่ 3 ทาน้ำปราศจากไอออน (กลุ่มควบคุม) นำไปผ่านกระบวนการสลับกรด-ด่างโดยใช้เชื้อแบคทีเรียเพื่อจำลองสภาวะในช่องปากเป็นเวลา 5 วัน จากนั้นนำมาวัดความหนาแน่นแร่ธาตุหลังการทดลองด้วยเครื่องถ่ายภาพรังสีคอมพิวเตอร์ระดับไมโครเมตร คำนวณความลึกรอยผุ ความหนาแน่นแร่ธาตุเฉลี่ยและร้อยละการเปลี่ยนแปลงความหนาแน่นแร่ธาตุหลังการทดลอง ผลการทดลองพบว่าชิ้นฟันตัวอย่างกลุ่มที่ทา Saforide® และ Advantage ArrestTM มีความลึกรอยผุหลังการทดลองลดลงอย่างมีนัยสำคัญทางสถิติ (p= 0.033 และ 0.021 ตามลำดับ) และมีความหนาแน่นแร่ธาตุเฉลี่ยหลังการทดลองเพิ่มขึ้นอย่างมีนัยสำคัญทางสถิติ (p= 0.002 และ 0.002 ตามลำดับ) พบว่าร้อยละการเปลี่ยนแปลงความหนาแน่นแร่ธาตุระหว่างกลุ่มทดลองทั้งสองกลุ่มไม่แตกต่างกันแต่แตกต่างจากกลุ่มควบคุมอย่างมีนัยสำคัญทางสถิติ (p< 0.001) สรุปได้ว่า Saforide® และ Advantage ArrestTM มีประสิทธิภาพในการคืนแร่ธาตุในรอยผุชั้นเนื้อฟันของฟันน้ำนมใกล้เคียงกัน

การศึกษานี้มีวัตถุประสงค์เพื่อศึกษาผลของการปรับเปลี่ยนชีวภาพเนื้อฟันด้วยสารสกัดจากเมล็ดองุ่นในการลดปริมาณแอคทีฟเจลาติเนส เมื่อใช้ร่วมกับสารยึดติดยูนิเวอร์แซล เอทช์แอนด์รินส์โหมด โดยใช้ผงเนื้อฟันที่เตรียมจากฟันกรามใหญ่ซี่ที่สามของมนุษย์ในการวัดปริมาณแอคทีฟเอ็มเอ็มพี ชนิดที่ 2 และใช้ชิ้นตัวอย่างการยึดติดเรซินและเนื้อฟันเพื่อศึกษาตำแหน่งที่มีแอคทีฟเจลาติเนสในชั้นไฮบริด แบ่งกลุ่มการทดลองเป็น 6 กลุ่ม คือ เนื้อฟันที่ไม่ปรับสภาพผิวใด ๆ กลุ่มเนื้อฟันที่กัดด้วยกรดฟอสฟอริก กลุ่มเนื้อฟันที่ใช้สารยึดติดยูนิเวอร์แซล เอทช์แอนด์รินส์โหมด ซิงเกิลบอนด์ ยูนิเวอร์แซล หรือ จี พรีมิโอ บอนด์ และกลุ่มที่มีการปรับสภาพเนื้อฟันด้วยสารสกัดจากเมล็ดองุ่นนาน 1 นาที หลังกัดด้วยกรดฟอสฟอริก ร่วมกับการใช้สารยึดติดดังกล่าว วิเคราะห์ข้อมูลทางสถิติด้วยการวิเคราะห์ความแปรปรวนทางเดียวและการทดสอบบองเฟอโรนี ที่ระดับนัยสำคัญทางสถิติ 0.05 พบว่า กระบวนการยึดติดเนื้อฟันด้วยสารยึดติดยูนิเวอร์แซล เอทช์แอนด์รินส์โหมด มีผลลดปริมาณแอคทีฟเอ็มเอ็มพี ชนิดที่ 2 และการปรับเปลี่ยนชีวภาพเนื้อฟันด้วยสารสกัดจากเมล็ดองุ่นยิ่งทำให้ลดปริมาณแอคทีฟเอ็มเอ็มพี ชนิดที่ 2 โดยเฉพาะอย่างยิ่ง เมื่อใช้สารยึดติด จี พรีมิโอ บอนด์ สอดคล้องกับผลจากการศึกษาตำแหน่งที่พบแอคทีฟเจลาติเนสในชั้นไฮบริดลดลงเมื่อใช้สารสกัดจากเมล็ดองุ่นก่อนการทาสารยึดติด จี พรีมิโอ บอนด์ อาจสรุปได้ว่าสารสกัดจากเมล็ดองุ่นช่วยส่งผลดีในการลดปริมาณแอคทีฟเจลาติเนส โดยเฉพาะอย่างยิ่งเมื่อใช้ร่วมกับสารยึดติดที่มีความไม่ชอบน้ำสูง แต่อย่างไรก็ตาม ยังต้องศึกษาเพิ่มเติมเกี่ยวกับกลไกการยับยั้งการทำงานของเอนไซม์ย่อยโปรตีน ผลต่อค่ากำลังการยึดติดและประสิทธิภาพของสารสกัดจากเมล็ดองุ่นในการยืดอายุของการยึดติดเนื้อฟัน

Objective: To investigate oro-dental characteristics and amelogenesis imperfecta (AI)-causing genomic variants associated with AI. Methods: Three Thai families affected with autosomal dominant hypocalcified amelogenesis imperfecta (ADHCAI) were recruited. Whole exome sequencing was used to identify mutations, confirmed by sanger sequencing. Clinical and radiographic features were recorded. The extracted teeth obtained from 3 patients were subjected for investigations of their properties and ultrastructure by micro-computerized tomography, surface profilometer, nano-base indentation system, EDX analysis, and SEM. The aim of this research was to investigate the synthesis and characterization of activated carbon from bacterial cellulose (BC-AC) and its applications. The research was divided into three parts. The study includes effects of contact time of the solution on the surface, flow velocity, particles size and particle concentration in the solution, on the thickness of rust particle film on the surface. Gamma-Hydroxybutyrate (GHB) is an endogenous short-chain fatty acid formed from Gamma-aminobutyric acid (GABA). Clinically, GHB is marketed in the United States as Xyrem to treat narcolepsy with cataplexy and in Europe for the treatment of alcohol withdrawal and narcolepsy. However, the illicit use and abuse of GHB occurs due to its sedative/hypnotic and euphoric effects. Monocarboxylate transporters (MCTs and SMCTs) are integral membrane proteins that control the bidirectional transport of endogenous substrates including lactate, acetate and pyruvate. The spread of drug-resistant bacterial infections has increased the need for novel antimicrobial agents. One potential but unexplored target is the de novo purine biosynthetic pathway. PurK, found only in bacteria, yeast, and fungi, catalyzes the sixth step in purine biosynthesis and has no human homolog. Herein we disclose the discovery of the first PurK inhibitor with submicromolar potency.

PurK is a member of the ATP-grasp superfamily of enzymes and recently, nanomolar inhibitors of biotin carboxylase, a related enzyme, were published. We hypothesized that those inhibitors, which target the ATP-binding site, could also inhibit PurK. To explore this hypothesis, four …

Introduction: In this study, we are testing the overarching hypothesis that preconditioning the myometrial UPR allows for the maintenance of non-apoptotic CASP3 activity and thus sustains uterine quiescence. We have previously demonstrated that the pregnant uterus facilitates uterine quiescence through UPR mediated activation of non-apoptotic CASP3, yet the mechanism in which CASP3 utilizes to avoid its apoptotic cell fate is unresolved. A Narrative Approach To Helping Families And Their Children Who Identify As Transgender Or Gender Nonconforming, Samantha Klassen-Bolding

Counselors can help families of transgender or gender nonconforming youth adjust after a child or adolescent expresses differences in gender identity. We propose a community based narrative group therapy which may increase family cohesion and acceptance within the changing family dynamic. The program is psychoeducational and experiential, lasting eight sessions, and begins with psychoeducation about transgender individuals and heteronormative societies. It culminates in re-authoring and witnessing the family’s story in a manner which honors the youth’s gender identity. We propose this program as a strategy for helping practitioners connect diverse LGBTQ+ families with other families experiencing similar transitions.

We propose a SAGE model for social psychological research. Encapsulated in our acronym is a proposal to have a synthetic approach to social psychological research, in which qualitative methods are augmentative to quantitative ones, qualitative methods can be generative of new experimental hypotheses, and qualitative methods can capture experiences that evade experimental reductionism. We remind social psychological researchers that psychology was founded in multiple methods of investigation at multiple levels of analysis. Motivational Interviewing (MI) is a known effective intervention for alcohol use disorder (AUD). MI's mechanisms of action remain inconsistently substantiated, and research in this area has been reliant on identifying relationships through strength of association rather than experimental manipulation of active ingredients. In two previous studies, a pilot and a larger replication study, we disaggregated MI into its hypothesized active ingredients by creating three conditions: MI, Spirit Only MI (SOMI, in which evocation of change talk was proscribed), and a non-therapy condition (NTC). The disease results in raised and pitted lesions seen on the surface of the potato and is caused by the soil pathogen, Streptomyces scabies. The S. scabies genome has two cyclic-di-GMP phosphodiesterases, RmdA and RmdB, both regulators of morphology and development. These proteins break down the ubiquitous second messenger molecule cyclic di-GMP, a molecule in charge of cell cycle progression and virulence. Larvae and nymphs of many species of ticks select small mammals, reptiles, and birds as hosts. Since 2011, we have examined more than 1,000 small mammals of 10 species and collected a small tissue sample as well as all visible ticks; rates of infestation range from 18% in harvest mice to 53% in meadow voles. Identification of immature ticks is challenging and requires molecular or genetic methods beyond classification as being from the genus Ixodes or another genus. Many tick species are invading new areas because of anthropogenic changes in the landscape, shifting climatic variables and increasing populations of suitable host species and tick habitat. However, the relative influences of habitat and hosts in tick dispersal and tick population establishment remain in question. Maximal repetitions or runs in strings have a wide array of applications and thus have been extensively studied. In this paper, we extend this notion to 2-dimensions, precisely defining a maximal 2D repetition. We provide initial bounds on the number of maximal 2D repetitions that can occur in a matrix. The main contribution of this paper is the presentation of the first algorithm for locating all maximal 2D repetitions in a matrix.
