Crystallographic And Computational Characterization Of Methyl Tetrel Bonding In S-Adenosylmethionine-Dependent Methyltransferases, Raymond C. Trievel, Steve Scheiner

Tetrel bonds represent a category of non-bonding interaction wherein an electronegative atom donates a lone pair of electrons into the sigma antibonding orbital of an atom in the carbon group of the periodic table. Prior computational studies have implicated tetrel bonding in the stabilization of a preliminary state that precedes the transition state in SN2 reactions, including methyl transfer. Notably, the angles between the tetrel bond donor and acceptor atoms coincide with the prerequisite geometry for the SN2 reaction. Background: The National Sleep Research Resource (NSRR) is a large-scale, openly shared, data repository of de-identified, highly curated clinical sleep data from multiple NIH-funded epidemiological studies. Although many data repositories allow users to browse their content, few support fine-grained, cross-cohort query and exploration at study-subject level. We introduce a cross-cohort query and exploration system, called X-search, to enable researchers to query patient cohort counts across a growing number of completed, NIH-funded studies in NSRR and explore the feasibility or likelihood of reusing the data for research studies.

Objective: The objective of this study was to evaluate the effects of intraoperative epidural anesthesia combined with balanced general anesthesia on intraoperative hemodynamics and fluid requirement, and on postoperative patient outcome.

Design: The study design was a retrospective data analysis of patients undergoing open hepatectomy at a single tertiary care center from May, 2013 to June, 2016. Patients undergoing hepatectomies were separated into two groups: patients not receiving epidural local anesthetic intraoperatively (either no epidural or epidural catheter not used intraoperatively) were designated the control group and patients receiving epidural local anesthetic intraoperatively (bolus and/or continuously). The status signalling hypothesis aims to explain within-species variation in ornamentation by suggesting that some ornaments signal dominance status. Here, we use multilevel meta-analytic models to challenge the textbook example of this hypothesis, the black bib of male house sparrows (Passer domesticus). We conducted a systematic review, and obtained primary data from published and unpublished studies to test whether dominance rank is positively associated with bib size across studies. Contrary to previous studies, the overall effect size (i.e. meta-analytic mean) was small and uncertain. Eleven in-depth life history interviews with respondents who identified as former self-injurers and a thematic analysis of the existing qualitative literature on self-injury constitute the data for this research. Self-injury, a growing public health concern, has typically been framed by researchers as an individual level, psychological, phenomenon with largely negative connotations. Edgework, a theoretical orientation which has been used to explain voluntary risk-taking such as skydiving and mountain climbing, has been applied to the activity of self-injury. The interviews and qualitative research on self-injury were coded for the presence of edgework as a vocabulary of motive. Framed as edgework, this …

The thermal performance of typical insulating materials is a limiting factor in the cost effectiveness of transporting temperature sensitive, biological materials, also known as cold chain bio-logistics. Biologicals that need to be maintained at near freezing or cryogenic temperatures rely on active refrigeration systems or passive solutions, such as dewars, gel packs, dry ice, urethane foam and/or extruded polystyrene. Lowering of the estuarine Environmental Quality Standard for zinc in the UK to 121 nM reflects rising concern regarding zinc in ecosystems and is driving the need to better understand its fate and behavior and to develop and parametrize speciation models to predict the metal species present. For the first time, an extensive data set has been gathered for the speciation of zinc within an estuarine system with supporting physicochemical characterization, in particular dissolved organic carbon. WHAM/Model VII and Visual MINTEQ speciation models were used to simulate zinc speciation, using a combination of measured complexation variables and available defaults. Data …

Can a large system be fully characterized using its subsystems via inductive reasoning? Is it possible to completely reduce the behavior of a complex system to the behavior of its simplest “atoms”? In this paper we answer these questions in the negative for a specific class of systems and measurements. After a general introduction of the topic, we present the main idea with a simple two-particle example, where strong correlations arise between two apparently empty boxes. Platelet α-granule cargo release is fundamental to both hemostasis and thrombosis. Granule matrix hydration is a key regulated step in this process, yet its mechanism is poorly understood. In endothelial cells, there is evidence for 2 modes of cargo release: a jack-in-the-box mechanism of hydration-dependent protein phase transitions and an actin-driven granule constriction/extrusion mechanism. The third alternative considered is a prefusion, channel-mediated granule swelling, analogous to the membrane “ballooning” seen in procoagulant platelets. Today’s state of the art additive manufacturing (AM) systems have the ability to fabricate multi-material devices with novel capabilities that were previously constrained by traditional manufacturing. AM machines fuse or deposit material in an additive fashion only where necessary, thus unlocking advantages of mass customization, no part-specific tooling, near arbitrary geometric complexity, and reduced lead times and cost. This study aimed to examine address term usage as a communicative component of blended/step-families by examining the address terms that step-children use to address their step-parent and whether address term usage can allude to the quality of relationship reported by step-children within the step-child step-parent relationship. Rowan University undergraduate students (n=67) were recruited to complete questionnaires on address term usage and quality of relationship using the Quality of Relationships Inventory (QRI). Interleukin-27 (IL-27) plays an important role in regulation of anti-inflammatory responses and autoimmunity; however, the molecular mechanisms of IL-27 in modulation of immune tolerance and autoimmunity have not been fully elucidated. Dendritic cells (DCs) play a central role in regulating immune responses mediated by innate and adaptive immune systems, but regulatory mechanisms of DCs in CD4+ T cell-mediated immune responses have not yet been elucidated. Here we show that IL-27 treated mature DCs induced by LPS inhibit immune tolerance mediated by LPS-stimulated DCs. Allogeneic hematopoietic cell transplantation (allo-HCT) is a potentially curative treatment of chronic myeloid leukemia (CML). Optimal conditioning intensity for allo-HCT for CML in the era of tyrosine kinase inhibitors (TKIs) is unknown. Using the Center for International Blood and Marrow Transplant Research database, we sought to determine whether reduced-intensity/nonmyeloablative conditioning (RIC) allo-HCT and myeloablative conditioning (MAC) result in similar outcomes in CML patients. We evaluated 1395 CML allo-HCT recipients between the ages of 18 and 60 years. BACKGROUND: In muscular dystrophy and old age, skeletal muscle repair is compromised leading to fibrosis and fatty tissue accumulation. Therefore, therapies that protect skeletal muscle or enhance repair would be valuable medical treatments. Hypoxia-inducible factors (HIFs) regulate gene transcription under conditions of low oxygen, and HIF target genes EPO and VEGF have been associated with muscle protection and repair. We tested the importance of HIF activation following skeletal muscle injury, in both a murine model and human volunteers, using prolyl hydroxylase inhibitors that stabilize and activate HIF.

Applying science to the current art of producing engineering and research knowledge has proven difficult, in large part because of its seeming complexity. We posit that the microscopic processes underlying research are not so complex, but instead are iterative and interacting cycles of divergent (generation of ideas) and convergent (testing and selecting of ideas) thinking processes. This reductionist framework coherently organizes a wide range of previously disparate microscopic mechanisms which inhibit these processes. We give examples of such inhibitory mechanisms and discuss how deeper scientific understanding of these mechanisms might lead to dis-inhibitory interventions for individuals, networks and institutional levels.

An essential event during early pregnancy is the invasion of trophoblasts into the maternal decidua, which is necessary for proper implantation and establishment of maternal-fetal interface and ultimately allows for proper nutrient exchange and immunological tolerance of the growing fetus. For this invasion to occur, cells originating from the trophectoderm undergo an epithelial to mesenchymal transition to become invasive extravillous trophoblasts and begin invading the uterine decidual tissue. Through the secretion of matrix metalloproteinases and through interactions with many cytokines and cell-adhesion molecules, this well-orchestrated process of trophoblast invasion results in extensive remodeling of the maternal spiral vasculature by the …

Pancreatic adenocarcinoma (PDAC) remains one of the most lethal cancer types, with 92% of patients succumbing to the disease within five years. One of the hallmarks of this intractable disease is its nearly universal resistance to chemotherapeutic intervention. The deoxycytidine analog gemcitabine persists as the primary chemotherapy for advanced PDAC patients despite little improvement to its toxicity and tenuous efficacy over the past two decades. Purpose: Positron emission tomography (PET) scanning is a widely used method of proton therapy verification. In this study, a proton radiotherapy accuracy verification process was developed by comparing predicted and measured PET data to verify the correctness of PET prediction and was tested at the Shanghai Proton and Heavy Ion Center.

Method: Irradiation was performed on a polymethyl methacrylate (PMMA) phantom. There were two dose groups, to which 2 and 4 Gy doses were delivered, and each dose group had different designed dose depths ranging from 5 to 20 cm. The predicted PET results were obtained using a PET prediction …
