Mantle cell lymphoma (MCL) is a comparatively rare non-Hodgkin’s lymphoma characterised by overexpression of cyclin D1.Many patients present with or progress to advanced stage disease within 3 years. MCL is considered an incurable disease withmedian survival between 3 and 4 years. We have investigated the role(s) of CCN1 (CYR61) and cell cycle regulators inprogressive MCL. We have used the human MCL cell lines REC1 < G519 < JVM2 as a model for disease aggression. Themagnitude of CCN1 expression in human MCL cells is REC1 > G519 > JVM2 cells by RQ-PCR, depicting a decrease in CCN1expression with disease progression. Investigation of CCN1 isoform expression by western blotting showed that whilst expres-sion of full-length CCN1 was barely altered in the cell lines, expression …

Protein prenylation is a post-translational modification that involves the addition of lipid groups to the end of a target protein and is necessary for protein activity. Prenylation has important roles in the cell: targeting and localizing proteins to subcellular compartments and promoting protein-protein interactions. Recently, we have found Protein Prenyltransferase Alpha Subunit-like (PPAL), which shares structural similarities to known prenylation enzymes. However, the biochemical function of PPAL is still unknown. PPAL is present in a single copy in other plants examined to date but is present in two copies in moss. Knockouts in our lab of either PpPAL1 or PpPAL2 …

Human Ancestry Indentification Under Resource Constraints -- What Can One Chromosome Tell Us About Human Biogeographical Ancestry?, Tanjin T. Toma, Jeremy M. Dawson, Donald A. Adjeroh
Clinical and Translational Science Institute
Background: While continental level ancestry is relatively simple using genomic information, distinguishing between individuals from closely associated sub-populations (e.g., from the same continent) is still a difficult challenge. Methods: We study the problem of predicting human biogeographical ancestry from genomic data under resource constraints. In particular, we focus on the case where the analysis is constrained to using single nucleotide polymorphisms (SNPs) from just one chromosome. We propose methods to construct such ancestry informative SNP panels using correlation-based and outlier-based methods. Results: We accessed the performance of the proposed SNP panels derived from just one chromosome, using data from the …

Climactic Niche Model For Overwintering Monarch Butterflies In A Topographically Complex Region Of California, Ashley Fisher, Kiana Saniee, Charis Van Der Heide, Jessica Griffiths, Daniel Meade, Francis X. Villablanca
Biological Sciences
We use climatic conditions that are associated with known monarch butterfly overwintering groves in California to build a Maxent model, and focus on the fine scale probability of overwintering grove occurrence in a topographically complex region of the state (Santa Barbara County). Grove locations are known from recent and historical surveys and a long-term citizen science database. The climatic niche model performs well, predicting that overwintering habitat is most likely to occur along the coast and at low elevations, as shown by empirical data. We then use climatic variables in conjunction with climate change scenarios to model the future location …

Mutualisms are essential for life, yet it is unclear how they arise. A two-stage process has been proposed for the evolution of mutualisms that involve exchanges of two costly resources. First, costly provisioning by one species may be selected for if that species gains a benefit from costless byproducts generated by a second species, and cooperators get disproportionate access to byproducts. Selection could then drive the second species to provide costly resources in return. Previously, a synthetic consortium evolved the first stage of this scenario: Salmonella enterica evolved costly production of methionine in exchange for costless carbon byproducts generated by …

Early Life History Response Of Reef Building Coral, Orbicella Faveolata, To Ocean Acidification And Warming, Kelly A. Pitts
HCNSO Student Theses and Dissertations
Ocean warming and acidification pose major threats to coral reef organisms. It is unknown how the early life history stages of Atlantic corals cope with the combined effects of these two global environmental stressors. Here, I investigate how these stressors influence the fertilization success, larval survivorship, and settlement of the threatened Atlantic coral, Orbicella faveolata. Gametes from O. faveolata were subjected to a factorial combination of present and future scenarios of oceanic temperatures (28.5° C and 30° C) and pH (8.2 and 8.0) predicted to occur by 2050. Results indicate that treatment type did not significantly affect fertilization success. …

Background: Parents are the primary providers of nurturing care for young children’s healthy early development. However, the literature on parenting in early childhood, especially in low- and middle-income countries, has primarily focused on mothers. In this study, we investigate how parents make meaning of fathers’ parenting roles with regards to their young children’s early health and development in rural Pakistan.
Methods: Data were collected between January and March 2017 through in-depth interviews with fathers (N = 33) and their partners (N = 32); as well as separate focus group discussions with fathers (N = 7) and mothers (N = 7). …

2:1 cocrystals of amino acids and Li+ salts crystallize from hot water to afford water stable cationic networks based upon tetrahedral lithium cations: bilayered square grids, a lithium zeolitic metal-organic material (LiZMOM) and several lithium diamondoid metal-organic materials (LiDMOMs). The compositions may be used as a pharmaceutical for the treatment of suicidality and other disorders that require lithium to penetrate the blood brain barrier and exert therapeutic effects in the CNS. Advantageously, the novel cocrystal forms described herein may be used to lower the oral dose required to achieve therapeutic concentrations of lithium in the brain, thus reducing the peripheral …

Analysis Of Environment-Marker Associations In American Chestnut, Markus Müller, C. Dana Nelson, Oliver Gailing
Michigan Tech Publications, Part 1
American chestnut (Castanea dentata Borkh.) was a dominant tree species in its native range in eastern North America until the accidentally introduced fungus Cryphonectria parasitica (Murr.) Barr, that causes chestnut blight, led to a collapse of the species. Different approaches (e.g., genetic engineering or conventional breeding) are being used to fight against chestnut blight and to reintroduce the species with resistant planting stock. Because of large climatic differences within the distribution area of American chestnut, successful reintroduction of the species requires knowledge and consideration of local adaptation to the prevailing environmental conditions. Previous studies revealed clear patterns of genetic diversity …

Biostatistics, Epidemiology & Environmental Health Sciences: Faculty Publications
Background
Acute infections are known cardiovascular disease (CVD) triggers, but little is known regarding how CVD risk varies following inpatient versus outpatient infections. We hypothesized that in‐ and outpatient infections are associated with CVD risk and that the association is stronger for inpatient infections.
Methods and Results
Coronary heart disease (CHD) and ischemic stroke cases were identified and adjudicated in the ARIC (Atherosclerosis Risk in Communities Study). Hospital discharge diagnosis codes and Medicare claims data were used to identify infections diagnosed in in‐ and outpatient settings. A case‐crossover design and conditional logistic regression were used to compare in‐ and outpatient …

Iris sanguinea is a perennial flowering plant that is typically cultivated through seeds or bulbs. However, due to limitations in conventional propagation, an alternate regeneration system using seeds was developed. The protocol included optimization of sterilization, stratification and scarification methods as iris seeds exhibit physiological dormancy. In addition to chlorine-based disinfection, alkaline or heat treatment was used to break seed dormancy and reduce contamination. When seeds were soaked in water at 80 °C overnight, and sterilized with 75% EtOH for 30 s and 4% NaOCl solution for 20 minutes, contamination was reduced to 10% and a 73.3% germination was achieved. …

Wide And Deep Neural Networks In Remote Sensing: A Review, Okan Ersoy
Wide And Deep Neural Networks In Remote Sensing: A Review, Okan Ersoy
Department of Electrical and Computer Engineering Technical Reports
Wide and deep neural networks in multispectral and hyperspectral image classification are discussed. Wide versus deep networks have always been a topic of intense interest. Deep networks mean large number of layers in the depth direction. Wide networks can be defined as networks growing in the vertical direction. Then, wide and deep networks are networks which have growth in both vertical and horizontal directions. In this report, several directions in order to achieve such networks are described. We first review a methodology called Parallel, Self-Organizing, Hierarchical Neural Networks (PSHNN’s) which have stages growing in the vertical direction, and each stage …

A Comprehensive Reentry Policy For Student Registered Nurse Anesthetists With Substance Use Disorder, Thomas M. Nigro
College of Science and Health Theses and Dissertations
Substance use disorder (SUD) is a common problem in anesthesia. While there are policies in place for practicing anesthetists, there are no known studies to date discussing reentry policies specific to the student registered nurse anesthesia (SRNA) population. The purpose of this research study was to describe key stakeholders’ knowledge and perspectives surrounding policies for reentry into academic programs for SRNAs in Illinois with SUD.
The theoretical framework used to drive this research was based off George L. Engel’s Biopsychosocial Theory. The theory examines the biological component of a person’s disease process, the psychological component, and the social component.

Lcp1 Mutant Zebrafish: A Look At Neutrophils, Cancer, And Gene Compensation, Taylor Mitchell
College of Science and Health Theses and Dissertations
Lymphocyte cytosolic protein 1 (lcp1 or L-plastin) is a small actin-bundling protein that is typically only expressed in motile leukocytes, such as neutrophils and macrophages. However, it is also overexpressed in cancer cells, which may be related to tumor metastasis. Using CRISPR/Cas9 gene editing, our lab has created zebrafish that are genetic knockouts for lcp1 in order to better understand the relationship between L-plastin and cell motility.
Previous studies on L-plastin knockout mice have shown that the mutants have a decreased immune response, and therefore I predicted that our zebrafish mutants might have impaired development or distribution of immune cells. …

Latent Classes Of Exposure To Potentially Traumatic And Stressful Life Events In A Sample Of Young Predominantly Ethnic Minority Women, Alyssa Donovan
College of Science and Health Theses and Dissertations
Women are disproportionately affected by specific types of potentially traumatic and stressful life events that are strongly linked to PTSD and depressive symptoms (Tolin & Foa, 2006; U.S. Census Bureau, 2016). Yet, while many studies have investigated patterns of potentially traumatic events (PTEs) and their associations with clinical outcomes, few have assessed PTE typologies with exclusively female samples (e.g., Cavanaugh et al., 2013). This study examined profiles of childhood and adulthood potentially traumatic and stressful life events in a predominantly ethnic minority, community sample of 191 young women. Using latent class analysis, we found four distinct profiles of exposure to …

Systemic lupus erythematosus (SLE) is an autoimmune disorder with heterogeneous clinical presentation and complex etiology involving the interplay between genetic, epigenetic, environmental and hormonal factors. Many common SNPs identified by genome wide-association studies (GWAS) explain only a small part of the disease heritability suggesting the contribution from rare genetic variants, undetectable in GWAS, and complex epistatic interactions. Using targeted re-sequencing of coding and conserved regulatory regions within and around 215 candidate genes selected on the basis of their known role in autoimmunity and genes associated with canine immune-mediated diseases, we identified a rare regulatory variant rs200395694:G > T located in intron …

Dormancy In The Amphistegina Gibbosa Holobiont: Ecological And Evolutionary Implications For The Foraminifera, Benjamin J. Ross
USF Tampa Graduate Theses and Dissertations
Dormancy, a state of severely decreased or suspended metabolism, is a widespread survival strategy in nature. In the Foraminifera, one of the most studied groups of marine organisms, its presence had been suggested by circumstantial evidence, but rarely studied directly until recently. Despite the lack of research, stressor-induced dormancy can significantly alter the way in which foraminiferal ecology is understood, especially in marginal environments. In this dissertation, I reviewed the evidence for dormancy in the foraminiferal literature, concluding that evidence for dormancy is widespread across the Phylum. I then explored the role of dormancy in the survival of the diatom-bearing …

Biofilm-sediment aggregate (BSA) contains a high water content, either within internal pores and channels or bound by extracellular polymeric substances (EPS) forming a highly hydrated biofilm matrix. Desiccation of BSAs alters the biofilm morphology and thus the physical characteristics of porous media, such as the binding matrix within BSA and internal pore geometry. Observing BSAs in their naturally hydrated form is essential but hampered due to the lack of techniques for imaging and discerning hydrated materials. Generally, imagery techniques (scanning electron microscopy (SEM), transmission electron microscopy (TEM), and focused ion beam nanotomography (FIB-nt)) involve the desiccation of BSAs (freeze-drying or …
