Organisms adapt to their environments by adjusting their biochemistry and physiology; such adaptation is limited by resource availability and physiological constraints. The freshwater crustacean Daphnia magna inhabits a wide range of environments and must survive and reproduce within a range of temperatures. One limit to low-temperature adaptation is thought to be the availability of unsaturated fatty acids necessary to maintain proper fluidity of cellular membranes. D. magna maintained at 10 ºC on a diet poor in unsaturated fatty acids have been observed to produce clutches that fail to develop. However, this has not been observed on a diet rich in …

Animal and human wastes introduce pathogens into rivers and streams, creating human health and economic burdens. While direct monitoring for pathogens is possible, it is impractical due to the sporadic distribution of pathogens, cost to identify, and health risks to laboratory workers. To overcome these issues, fecal indicator organisms are used to estimate the presence of pathogens. Although fecal indicators generally protect public health, they fall short in their utility because of difficulties in public health risk characterization, inconsistent correlations with pathogens, weak source identification, and their potential to persist in environments with no point sources of fecal pollution. Amyloid fibrils from milk proteins have recently been the subject of extensive investigations in biophysics. The purpose of this thesis is to investigate the dynamics of fibrillation and the mechanical properties of β-LG and β-CN fibers using AFM-Quantitative Nanomechanical Mapping (QNM) and AFM-force spectroscopy. β-LG required one day of heating at 80 °C , pH 2 to fibrillate in solution; β-CN required three days. Using the AFM-QNM mode an average elastic modulus of 4.3 GPa was determined for the β-LG fibers and 3.1 GPa for β-casein fibers. The term co-infection describes an event when multiple pathogens infect a single host. For vector-borne diseases, a vector may become co-infected by feeding either on a single host that has multiple pathogens or feeding multiple hosts, each of which may be infected. Co-infection in non-vector hosts has been shown to influence immune response and eventual outcome of disease, and is likely to have a similar impact on vector species. Radio frequency (RF) ion traps based on the quadrupole device developed by Paul and Steinwedel utilize a dynamic electric field to spatially confine the trajectory of charged particles and may be employed as mass spectrometers by selectively ejecting trapped molecules based on the mass to charge ratio. Rhodococcushas been recognized as a potential antibiotic producer. Recently, a strain of Rhodococcussp. MTM3W5.2 was isolated from a soil sample collected in Morristown, Tennessee and was found to produce an inhibitory compound which is active against other related species. The purpose of this research is to extract, purify and analyze the active metabolite. The compound was extracted from RM broth cultures and purified by preliminary fractionation of crude extract through a Sephadex LH-20 column. Many construction projects need to pump turbid water from borrow pits or other excavations into stilling basins or sediment filter bags prior to discharge. This study evaluated the effectiveness of these devices with polyacrylamide (PAM) injection to reduce effluent turbidity. Results from laboratory jar tests using two coastal plain sediments of North Carolina suggested that a cationic PAM was the most effective in reducing turbidity, followed by a nonionic PAM. Anionic PAM was effective in whole-soil jar tests but not when turbid supernatant was tested. A stilling basin was not effective in reducing the turbidity of the pumped water without …

Campylobacter jejuni is a prevalent infectious enteritis mainly foodborne from chickens. Despite of reducing C. jejuni food contamination dramatically decreases campylobacteriosis, few effective approaches are available for the bacterial reduction in chickens. The aim of this study was to use microbial metabolic product deoxycholic acid (DCA) to reduce C. jejuni chicken colonization. Broiler chicks were fed 0 or 1.5 g/kg DCA, lithocholic acid (LCA), or urodeoxycholic acid (UDCA) in diets or orally gavaged with cholic acid (CA, 1.5g/kg body weight). Background: Predicting disease causative genes (or simply, disease genes) has played critical roles in understandingthe genetic basis of human diseases and further providing disease treatment guidelines. Premature cervical remodeling is a critical precursor of spontaneous preterm birth, and the remodeling process is characterized by an increase in tissue hydration. Nevertheless, current clinical measurements of cervical remodeling are subjective and detect only late events, such as cervical effacement and dilation. Here, we present a photoacoustic endoscope that can quantify tissue hydration by measuring near-infrared cervical spectra. We quantify the water contents of tissue-mimicking hydrogel phantoms as an analog of cervical connective tissue. Applying this method to pregnant women in vivo, we observed an increase in the water content of the cervix throughout pregnancy. The application of this …

This paper explores the nature of emotions by considering what appear to be two differing, perhaps even conflicting, approaches to affectivity—an evolutionary functional account, on the one hand, and a phenomenological view, on the other. The paper argues for the centrality of the notion of function in both approaches, articulates key differences between them, and attempts to understand how such differences can be overcome.

The Modifier Model of autism spectrum disorder (ASD) suggests that phenotypic variability within ASD is rooted in modifier processes, such as the behavioral inhibition system (BIS) and behavioral activation system (BAS). Among a sample of 53 adolescents with ASD, this study examined associations between (a) self-reported BIS/BAS and frontal and parietal alpha electroencephalogram asymmetry and whether these indices related to (b) ASD severity (via the Autism Quotient), and/or (c) co-occurring anxiety and attention-deficit hyperactivity disorder (via Youth Self Report and Child Behavior Checklist). At Amgen, the Automated Vision Inspection (AVI) systems capture the movement of unwanted particles in Amgen's drug product containers. For quality inspection, the AVI system must detect these undesired particles using a high speed spin-stop agitation process. Ectotherms are excellent indicators of potential climatic ramifications on biodiversity because of their heavy reliance on the environment for their thermoregulation. Studies have historically looked at thermal tolerance values to establish predictive models for population and species extinctions.

Two sets of triplicate pilot algal raceway ponds (1000-L, 0.30-m deep, paddle wheel mixed) were operated for 14 months at a California wastewater treatment plant to treat wastewater and generate algal-bacterial biomass as biofuel feedstock. Two experiments were run to determine the effect on biomass productivity of (1) hydraulic residence time (HRT: 2, 3, 4, or 4.5 days) and (2) feeding schedule (18 small pulses during 8 AM-4 PM [diurnal] versus 20 large pulses during 4 AM-12 AM [diel]). While transcriptomic analyses have been applied to cultured mammary epithelial cells (MEC) and to whole gland samples, few have studied purified MEC isolated directly from the gland in vivo. This thesis considers the use of Young Adult Chicana Literature in the classroom to help young Chicanas work through their process of finding their identities. It begins by making the case that Chicana identities are complex because of their intersectional borderland positioning between Mexican and U.S. American cultures, which makes the identity formation process more difficult for them than others. By relating these complex issues facing young Chicanas to literature that is more relevant to them and their struggles, it is argued that teachers can help ease some of the tensions that exist within their students and help them work …
